Acute monocytic leukemia in an irradiated Beagle.

A purebred female Beagle dog that had received 2,000 R of protracted wholebody gamma-irradiation from 60Co when 14 months old had hematologic changes consistent with a myeloproliferative disorder 3 years after the termination of radiation exposure. Peripheral blood and bone marrow findings during the 7-month period before death showed progressive anemia with increased numbers of platelets; immature granulocytes, monocytes and promonocytes. A period of partial remission occurred during which time the peripheral blood was aleukemic, although there was marked thrombocytosis and abnormal erythropo

Orbital meningioma after cranial radiation for acute lymphocytic leukemia.

PURPOSE: To describe the ophthalmic findings of cranial radiation-induced orbital meningioma after acute lymphocytic leukemia therapy. METHODS: This is a prospective clinical report. RESULTS: We describe two patients recently evaluated with orbital meningiomas many years after cranial radiation for acute lymphocytic leukemia. CONCLUSIONS: There will probably be more of these cases in the next several years. Unlike primary meningiomas, these neoplasms have a tendency to be more diffuse, multiple, and may undergo malignant degeneration.

Empty sella/pituitary atrophy and endocrine impairments as a consequence of radiation and chemotherapy in long-term survivors of childhood leukemia.

Radiation-induced empty sella (ES) or pituitary atrophy/small pituitary and endocrine impairments, including pituitary and gonadal dysfunction, can manifest decades after radiation and chemotherapy in childhood-onset leukemia patients who received prophylactic cranial irradiation or total body irradiation in preparation for bone marrow transplant. Six childhood-onset leukemia patients (age at diagnosis of leukemia; 2.7-10.2 years) participated in this study. Magnetic resonance imaging (MRI) of the pituitary gland and endocrinological studies were performed 10.5-32.1 years after cranial irradia

Reactions to ionizing radiation.

This review deals with essential physical and biologic aspects of ionizing radiation as related to dermatology. The discussion of major physical factors and basic biophysics includes the production of x rays, dose definitions and recent changes in terminology, various factors affecting dose and tissue damage and current radiotherapy methods. The biologic effects and potential risks of ionizing radiation are reviewed in detail, particularly the major nonstochastic and stochastic somatic effects, such as radiation-induced skin cancer, thyroid carcinoma, breast cancer, and other radiogenic neopla

Leukemia: lessons from the Japanese experience.

Probably more has been learned about radiation-induced leukemia from intensive study of Japanese atomic bomb survivors than about radiation damage to any other organ system. This has been the result of an intensive binational effort at the Atomic Bomb Casualty Commission and the Radiation Effects Research Foundation in Hiroshima and Nagasaki to monitor the occurrence of leukemia in a large group of atomic bomb survivors over a period of more than 50 years (the Life Span Study). The focus in the leukemia studies has been on disease latency, time of peak incidence rates, clinical course, shape o

Myeloid leukaemia frequency after protracted exposure to ionizing radiation: experimental confirmation of the flat dose-response found in ankylosing spondylitis after a single treatment course with X-rays.

The dose-response for leukaemia induction by exposure to ionizing radiation protracted over several weeks was largely independent of dose not only in X-rayed patients with ankylosing spondylitis but also in experimentally gamma-rayed CBA/H mice. In the experiment the induced leukaemia frequency of acute myeloid leukaemia was independent of a several thousand-fold variation in physical dose rate. Any difference in leukaemia induction between brief and protracted exposures must therefore depend on specifically biological consequences of protracted exposures. Experimental analysis is required to

Lessons from Chernobyl.

The Chernobyl disaster on April 26th, 1986, led to the emission of radioactive substances such as iodine-131 and radioactive cesium. As the Soviet Union did not control food distribution and intake, residents were exposed to high levels of internal radiation, leading to the internal radiation exposure of the thyroid gland by iodine-131. As a result, the number of people who had thyroid cancer increased drastically among those who had been under 15 years old at the time of the accident. The age predilection is about to move to 25 or older. However, there has been no scientific evidence of impac

Mouse models of radiation-induced cancers.

Radiation-induced (RI) secondary cancers were not a major clinical concern even as little as 15 years ago. However, advances in cancer diagnostics, therapy, and supportive care have saved numerous lives and many former cancer patients are now living for 5, 10, 20, and more years beyond their initial diagnosis. The majority of these patients have received radiotherapy as a part of their treatment regimen and are now beginning to develop secondary cancers arising from normal tissue exposure to damaging effects of ionizing radiation. Because historically patients rarely survived past the extended

Is the Linear No-Threshold Dose-Response Paradigm Still Necessary for the Assessment of Health Effects of Low Dose Radiation?

Inevitable human exposure to ionizing radiation from man-made sources has been increased with the proceeding of human civilization and consequently public concerns focus on the possible risk to human health. Moreover, Fukushima nuclear power plant accidents after the 2011 East-Japan earthquake and tsunami has brought the great fear and anxiety for the exposure of radiation at low levels, even much lower levels similar to natural background. Health effects of low dose radiation less than 100 mSv have been debated whether they are beneficial or detrimental because sample sizes were not large eno

The proportion of childhood leukaemia incidence in Great Britain that may be caused by natural background ionizing radiation.

The aetiology of childhood leukaemia remains generally unknown, although exposure to moderate and high levels of ionizing radiation, such as those experienced during the atomic bombings of Japan or from radiotherapy, is an established cause. Risk models based primarily on studies of the Japanese atomic bomb survivors imply that low-level exposure to ionizing radiation, including ubiquitous natural background radiation, also raises the risk of childhood leukaemia. Using two sets of recently published leukaemia risk models and estimates of natural background radiation red-bone-marrow doses recei

Deficiency of Bruton's tyrosine kinase in B cell precursor leukemia cells.

Bruton's tyrosine kinase (BTK) deficiency results in a differentiation block at the pre-B cell stage. Likewise, acute lymphoblastic leukemia cells are typically arrested at early stages of B cell development. We therefore investigated BTK function in B cell precursor leukemia cells carrying a BCR-ABL1, E2A-PBX1, MLL-AF4, TEL-AML1, or TEL-PDGFRB gene rearrangement. Although somatic mutations of the BTK gene are rare in B cell precursor leukemia cells, we identified kinase-deficient splice variants of BTK throughout all leukemia subtypes. Unlike infant leukemia cells carrying an MLL-AF4 gene rea

Reanalysis of cancer mortality using reconstructed organ-absorbed dose: J-EPISODE 1991‒2010.

The Japanese Epidemiological Study on Low-Dose Radiation Effects (J-EPISODE) has been conducted since 1990 by the Radiation Effects Association to analyse health effects for nuclear workers. It uses the recorded doses, i.e. dosimeter readings, evaluated inHp(10) for estimation of radiation risk; however, the International Commission on Radiological Protection does not recommend the use of effective doses for epidemiological evaluation and instead recommends the use of organ-absorbed doses for assessing cancer risk. Recently, the J-EPISODE has developed a conversion factor that can convert dosi

The epidemiology of leukemias (past, present, future).

There are three periods of the history of the epidemiology of leukemias. The first period is characterized by 3 features: the description of the occupational leukemias (radiations, benzene) with more recently the war leukemias (Hiroshima), the leukemia induced by chemotherapy; the description of the leukemias of animals induced by viruses (chicken, mice and more over cats); and the concept of the geographical hematology. The second period may be called "Burkitt period" so important are the consequences of the discoveries made in Uganda. The third period is Japanese and American with the leukem

Non-targeted and delayed effects of exposure to ionizing radiation: II. Radiation-induced genomic instability and bystander effects in vivo, clastogenic factors and transgenerational effects.

The goal of this review is to summarize the evidence for non-targeted and delayed effects of exposure to ionizing radiation in vivo. Currently, human health risks associated with radiation exposures are based primarily on the assumption that the detrimental effects of radiation occur in irradiated cells. Over the years a number of non-targeted effects of radiation exposure in vivo have been described that challenge this concept. These include radiation-induced genomic instability, bystander effects, clastogenic factors produced in plasma from irradiated individuals that can cause chromosomal d

Risk of Leukemia Associated with Protracted Low-Dose Radiation Exposure: Updated Results from the National Registry for Radiation Workers Study.

While the link between risk of leukemia and acute radiation exposure is well established for large doses received acutely, uncertainty remains around the translation of these risk estimates to occupational exposure scenarios where the doses are low and accumulated over time, possibly over many years. We present leukemia incidence and mortality radiation risk estimates derived from the National Registry for Radiation Workers, which is a large cohort of occupationally exposed workers from the United Kingdom (UK). The cohort comprised 173,081 workers from the UK who were monitored for occupationa

Aleukaemic leukaemia cutis as an initial manifestation of myeloid/NK cell precursor acute leukaemia.

Aleukaemic leukaemia cutis is a rare condition characterized by infiltration of leukaemic cells into the skin before they appear in the peripheral blood. We report a case of an aleukaemic leukaemia cutis, which had a history of exposure to atomic bomb radiation. A 57-year-old Japanese woman initially presented with a 20-week history of multiple red papules and plaques mainly over the trunk. Histological examination revealed the infiltration of atypical monocytic cells in the dermis, but no leukaemic cells were detected in the peripheral blood. Twenty-three weeks after the appearance of the eru

[Concepts of dosimetry].

The effects of exposure to ionizing radiation are determined by the absorbed dose D. The equivalent dose H takes into consideration the variation in probability of a stochastic effect (cancer, leukemia, genetic mutation) based on the quality of the exposure. The effective dose E takes into account the sensitivity of tissues (T) to stochastic effects from radiation exposure. Optimization of radiation exposure to patients in diagnostic radiology in based on diagnostic reference levels (DRL): entrance surface dose or dose-area product for conventional radiology; CT dose index and DLP for CT.

A case-control analysis of leukemia in accident emergency workers of Chernobyl.

We estimated the radiation-induced risk of leukemia in 162,684 Chernobyl accident emergency workers (EWs) using the data of the Russian National Medical and Dosimetric Registry (RNMDR). A system was established for the collection and verification of data in 55 cases of leukemia from 1986 to 1995. The principal analysis included 41 leukemia cases that occurred more than 2 years after the first exposure to radiation. The case-control methodology was used to evaluate the risk of leukemia associated with various factors. Radiation dose, effective exposure dose rate, date of entry into the Chernoby
