Candidatus Liberibacter asiaticus (CLas), a pathogenic bacterium causing citrus Huanglongbing, forms a mutualistic relationship with the Asian citrus psyllid, Diaphorina citri. However, the underlying molecular mechanisms remain unclear. We find that CLas enhances the development of testes, ovaries, and mating behavior in D. citri. Transcriptome analysis of ovaries reveals that CLas infection upregulates ATP synthesis genes, increasing ATP levels in D. citri. Silencing the ATPSyn-β gene expression reduces ATP content in the ovary and triglyceride and glycogen content in the fat body, thus decr

Diaphorina citri Kuwayama is the primary natural vector of citrus Huanglongbing (HLB). Prolonged use of chemical pesticides leads to insect resistance, reduces control efficacy, and causes ecological pollution. RNA interference (RNAi) enables sequence-specific gene silencing, representing a promising precision strategy for managing D. citri. This review summarizes advances in RNAi-based control of D. citri, focusing on three groups of target gene screening strategies based on essential functional genes, behavioral regulation genes, and D. citri-Candidatus Liberibacter asiaticus (CLas) interact

Huanglongbing (HLB), caused by the phloem-restricted bacterium Candidatus Liberibacter asiaticus (C. Las), is vectored by the Asian citrus psyllid (ACP) and threatens global citrus production. We demonstrate that VU041 (C19H20F3N3O), a small-molecule inhibitor of inward rectifier potassium (Kir) channels, provides a dual-mechanism approach to management of Huanglongbing (HLB) disease by combining acute toxicity with antifeedant activity. Topical and tarsal contact bioassays revealed that VU041 induced rapid mortality to ACP (24 h LD50 = 25 ng/insect) and reduced salivary gland secretion to sup

Selenium treatment has been shown to enhance plant resilience against both biotic and abiotic stresses; however, whether it can improve citrus resistance to bacterial diseases remains unexplored. In this study, we systematically identified sulfate transporter (Sultr) genes in the Citrus sinensis genome that are involved in selenate uptake. Analysis of gene structure, conserved motifs, and protein domains revealed a high degree of conservation among CsSultrs. Collinearity analysis between C. sinensis and Arabidopsis thaliana Sultr genes suggested possible evolutionary duplication and divergence

Huanglongbing (HLB) disease, associated with the fastidious bacterium Candidatus Liberibacter asiaticus (CLas), has a significant impact on citrus production worldwide. Conventional biochemical and genetic evaluation studies to identify potential disease resistance strategies have been mainly hindered due to the inability to culture CLas in a defined medium and the general recalcitrance of Citrus cultivars (grapefruits and oranges) to Agrobacterium-mediated plant transformation. We previously demonstrated the utility of plant hairy roots to co-cultivate CLas. In this study, we developed a hair

Citrus production under enclosed structures can exclude the Asian citrus psyllid (ACP, Diaphorina citri) and eliminate the negative effects of citrus greening or huanglongbing (HLB) disease caused by Candidatus Liberibacter asiaticus to the grapefruit (Citrus paradisi) fresh fruit industry. Physically impeding the insect vector from accessing trees is a logical method to have disease-free groves. Our objectives were to assess the ability of enclosed screenhouses to exclude the ACP, stop HLB inoculation and dissemination, and improve fruit yield of in-ground and container-grown 6-year-old "Ray

UNLABELLED: Infection with pathogens can increase the fecundity and other fitness-related traits of insect vectors for their own advantage. Our previous research has reported the pivotal role of DcKr-h1 in the fecundity improvement of Diaphorina citri induced by the bacterium, "Candidatus Liberibacter asiaticus" (CLas), and the fungus, Cordyceps fumosorosea (Cf). However, the posttranscriptional regulation of this process remains poorly understood. Given the significance of miRNAs in gene regulation, we delved into their roles in shaping phenotypes and their underlying molecular mechanisms. Ou

The Asian citrus psyllid, Diaphorina citri Kuwayama (Hemiptera: Liviidae), is the primary vector of Candidatus Liberibacter asiaticus, the causal agent of huanglongbing, the most devastating disease of citrus worldwide. Understanding its population structure and developing lineage-specific markers are important for surveillance, quarantine and pest management. Here, 133 complete mitochondrial genomes were analyzed, comprising 77 previously published sequences, 13 newly assembled genomes and 43 metagenome-assembled mitogenomes. Phylogenetic analysis revealed two highly differentiated mitochondr

Diaphorina citri Kuwayama (Hemiptera: Liviidae) vector bacteria (Candidatus Liberibacter asiaticus) that cause huanglongbing, an economically devastating disease of citrus. Management of D. citri infestations is primarily through insecticides, but alternative control methods remain under consideration including the co-opting and disruption of D. citri mating-duet vibrational communication signals. This study applies generative adversarial network and convolutional neural network methods to distinguish among female and male duetting signals when the sex of the signaler is not visually verifiabl

"Candidatus Liberibacter asiaticus" (CLas) is a phloem-restricted Gram-negative bacterium that is the causal agent of citrus huanglongbing (HLB). In this study, we identified a CLas-encoded Sec-dependent secretory protein CLIBASIA_04405 that could contribute to the pathogenicity of this bacterium. The gene expression level of CLIBASIA_04405 was significantly higher in citrus than in psyllids. Transient overexpression of the mature CLIBASIA_04405 protein (m4405) in Nicotiana benthamiana leaves could suppress hypersensitive response (HR)-based cell death and H2O2 accumulation triggered by the mo

Huanglongbing (HLB), caused by Candidatus Liberibacter asiaticus (CLas), an uncultured α-proteobacterium, is the most destructive disease of citrus trees worldwide. In previous studies, trunk injections of penicillin reduced CLas titers and HLB symptoms in citrus. However, antibiotic effects on the whole plant microbial community, which include effects on taxa that interact with CLas, have not yet been addressed. In this study, we investigated the effects of penicillin injection (0, 1000, and 6000 mg L-1) on rhizospheric and endophytic bacterial communities of grapefruit trees in field and gre

Huanglongbing (HLB) is the most devastating disease of citrus. The global citrus industry is in urgent need of effective chemical treatments for HLB control because of its rapid spreading worldwide. Due to the fastidious nature of the pathogens, and the poor permissibility of citrus leaf surfaces, effective screening of chemicals for the HLB control can be challenging. In this study, we developed a graft-based chemotherapy method to rapidly screen potential HLB-controlling chemical compounds. In addition, we improved transmission efficiency by using the best HLB-affected scion-rootstock combin

BACKGROUND: The Asian Citrus Psyllid (ACP), Diaphorina citri, can transmit the bacterium Candidatus Liberibacter while feeding on citrus flush shoots. This bacterium causes Huanglongbing (HLB), a major disease of citrus cultivation worldwide necessitating the development of new tools for ACP surveillance and control. The olfactory system of ACP is sensitive to variety of odorants released by citrus plants and offers an opportunity to develop new attractants and repellents.
RESULTS: In this study, we performed single-unit electrophysiology to identify odorants that are strong activators, inhib

Huanglongbing (HLB), formerly known as citrus greening disease, is one of the most devastating bacterial diseases in citrus worldwide. HLB is caused by 'Candidatus Liberibacter asiaticus' bacterium and transmitted by Diaphorina citri. Both 'Ca. L. asiaticus' and its vector manipulate the host metabolism to fulfill their nutritional needs and/or to neutralize the host defense responses. Herein, we discuss the history of HLB and the complexity of its pathosystem as well as the geographical distribution of its pathogens and vectors. Recently, our recognition of physiological events associated wit

The Asian Citrus Psyllid (ACP), Diaphorina citri, severely threatens citrus production worldwide by transmitting the greening (= Huanglongbing)-causing bacterium Candidatus Liberibacter asiaticus. There is growing evidence that the push-pull strategy is suitable to partially mitigate HLB by repelling ACP with transgenic plants engineered to produce repellents and attracting the vector to plants with a minimal disease transmission rate. Species that pull ACP away from commercial citrus plants have been identified, and transgenic plants that repel ACP have been developed. The concept of a repell

BACKGROUND: Piercing-sucking insects secrete diverse repertoires of effectors into their hosts to weaken host defenses and promote infestation. The Asian citrus psyllid, Diaphorina citri Kuwayama, is the most destructive insect pest in citrus orchards because of its role as a vector for the huanglongbing pathogen, Candidatus Liberibacter asiaticus (CLas). However, specific effector proteins and their functions in D. citri remain unclear.
RESULTS: We demonstrate that DcE1, a salivary protein gene from D. citri, is predominantly expressed in the salivary gland tissues and is delivered into host

Huanglongbing (HLB), caused by Candidatus Liberibacter asiaticus (CLas), is a devastating citrus disease that severely threatens global citrus production, yet the metabolic mechanisms underlying citrus tolerance to HLB remain elusive. Citrus genotypes exhibit differential tolerance to HLB, with lemon (Citrus limon) showing greater resilience compared to susceptible sweet orange (Citrus sinensis). In this study, we integrated untargeted liquid chromatography-high resolution mass spectrometry metabolomics with a bootstrap-enhanced LASSO approach to characterize the metabolic responses of HLB-tol

Huanglongbing (HLB), a deadly citrus disease, is primarily associated with Candidatus Liberibacter asiaticus (CLas) and spread by the hemipteran insect Diaphorina citri. Control strategies to combat HLB are urgently needed. In this work, we developed and compared workflows for the extraction of the D. citri peptidome, a dynamic set of polypeptides produced by proteolysis and other cellular processes. High-resolution mass spectrometry revealed bias among methods reflecting the physiochemical properties of the peptides: while TCA/acetone-based methods resulted in enrichment of C-terminally amida
