Properties and functions of TRPM1 channels in the dendritic tips of retinal ON-bipolar cells.

An increase in light intensity induces a depolarization in retinal ON-bipolar cells via a reduced glutamate release from presynaptic photoreceptor cells. The underlying transduction cascade in the dendritic tips of ON-bipolar cells involves mGluR6 glutamate receptors signaling to TRPM1 proteins that are an indispensable part of the transduction channel. Several other proteins are recognized to participate in the transduction machinery. Deficiency in many of these leads to congenital stationary night blindness, because rod bipolar cells, a subgroup of ON-bipolar cells, constitute the main route

Nonlinear distributed sensing of light patterns leads to perceptual distortions in plants.

Many organisms lack centralized sensory-processing systems, navigating complex environments through local integration of spatially distributed stimuli (e.g., chemotaxis of cells, bacteria, or growing neurons). Here we propose a general physical and geometric framework to describe how such distributed sensing translates into integrated directional responses. We study plants, multicellular decentralized systems that grow toward light (phototropism), which can come from multiple directions and at different intensities. We develop a model in which light is sensed locally on the shoot circumference

Light Hygiene for Circadian Health: A Molecular Perspective.

In discussing the interplay between light exposure/blue light exposure (LE/BLE) and circadian health, we emphasize the role of light hygiene and its effects on maintaining sleep, mood, and metabolic health, among other physiological processes. We define compromised circadian light hygiene as low dynamic range and/or irregular 24-hour patterns of LE. Poor light hygiene interferes with circadian entrainment and weakens circadian robustness alignment, thereby increasing health hazards. We provide an overview of the complex molecular pathways underlying light perception and downstream signaling. G

Rapid blue light regulation of a Trichoderma harzianum photolyase gene.

Photolyases and blue light receptors belong to a superfamily of flavoproteins that make use of blue and UVA light either to catalyze DNA repair or to control development. We have isolated a DNA photolyase gene (phr1) from Trichoderma harzianum, a common soil fungus that is of interest as a biocontrol agent against soil-borne plant pathogens and as a model for the study of light-dependent development. The sequence of phr1 is similar to other Class I Type I eukaryotic photolyase genes. Low fluences of blue light rapidly induced phr1 expression both in vegetative mycelia, which lack photoprotecti

Monogenic Retinal Diseases Associated With Genes Encoding Phototransduction Proteins: A Review.

Phototransduction, the process by which captured photons elicit electrical changes in retinal rod and cone cells, represents the first neuronal step in vision and involves interactions between several highly specialised proteins. Pathogenic variants in genes encoding many of these proteins can give rise to significant vision impairment, accounting for a substantial portion of inherited retinal disease. Such genes include RHO, OPN1LW, OPN1MW, GNAT1, GNAT2, GNB3, PDE6A, PDE6B, PDE6G, PDE6C, PDE6H, CNGA1, CNGB1, CNGA3, CNGB3, GRK1, SAG, ARR3, RGS9, RGS9BP, GUCY2D, GUCA1A and SLC24A1. Many of thes

Induction of the unfolded protein response by constitutive G-protein signaling in rod photoreceptor cells.

Phototransduction is a G-protein signal transduction cascade that converts photon absorption to a change in current at the plasma membrane. Certain genetic mutations affecting the proteins in the phototransduction cascade cause blinding disorders in humans. Some of these mutations serve as a genetic source of "equivalent light" that activates the cascade, whereas other mutations lead to amplification of the light response. How constitutive phototransduction causes photoreceptor cell death is poorly understood. We showed that persistent G-protein signaling, which occurs in rod arrestin and rhod

Impact of Natural Dietary Agents on Multiple Myeloma Prevention and Treatment: Molecular Insights and Potential for Clinical Translation.

Chemoprevention is based on the use of non-toxic, pharmacologically active agents to prevent tumor progression. In this regard, natural dietary agents have been described by the most recent literature as promising tools for controlling onset and progression of malignancies. Extensive research has been so far performed to shed light on the effects of natural products on tumor growth and survival, disclosing the most relevant signal transduction pathways targeted by such compounds. Overall, anti-inflammatory, anti-oxidant and cytotoxic effects of dietary agents on tumor cells are supported eithe

Whole mount labeling of cilia in the main olfactory system of mice.

The mouse olfactory system comprises 6-10 million olfactory sensory neurons in the epithelium lining the nasal cavity. Olfactory neurons extend a single dendrite to the surface of the epithelium, ending in a structure called dendritic knob. Cilia emanate from this knob into the mucus covering the epithelial surface. The proteins of the olfactory signal transduction cascade are mainly localized in the ciliary membrane, being in direct contact with volatile substances in the environment. For a detailed understanding of olfactory signal transduction, one important aspect is the exact morphologica

Three distinct roles of aquaporin-4 in brain function revealed by knockout mice.

Aquaporin-4 (AQP4) is expressed in astrocytes throughout the central nervous system, particularly at the blood-brain and brain-cerebrospinal fluid barriers. Phenotype analysis of transgenic mice lacking AQP4 has provided compelling evidence for involvement of AQP4 in cerebral water balance, astrocyte migration, and neural signal transduction. AQP4-null mice have reduced brain swelling and improved neurological outcome in models of (cellular) cytotoxic cerebral edema including water intoxication, focal cerebral ischemia, and bacterial meningitis. However, brain swelling and clinical outcome are

G protein signaling in UV protection: methods for understanding the signals in young etiolated seedlings.

A seed is competent to respond to light soon after imbibition. A new developmental program begins in or on the ground where the young seedling may be exposed to heat, cold, drought, flooding (anoxia), salts, varying levels of visible light, and the topic of this paper, ultraviolet radiation. Herein what is described is a method for growing and maintaining seedlings, then methods of UV irradiation in order to measure discrete effects of UV wavelengths in signal transduction, very early in seedling development. The physiological response to an abiotic signal is partly dependent on the developmen

Enhanced Electrochemical Detection of Circularly Polarized Light Enabled by Spin-Selective Oxygen Evolution Reaction.

The interaction between circularly polarized light (CPL) and chiral matter generates spin-polarized charge carriers; however, efficiently converting this microscopic spin imbalance into a macroscopic electrical signal remains a central challenge in CPL detection. Herein, we present an electrochemical CPL detection platform based on a metalated chiral covalent organic framework (CCOF), where CPL-induced spin polarization is coupled with the spin-selective oxygen evolution reaction (OER) to enable nonlinear transduction of spin polarization into an amplified electrical response. The nonlinear sp

Host-based antipoxvirus therapeutic strategies: turning the tables.

The potential threat of the smallpox virus as a bioterror weapon has long been recognized, and the need for developing suitable countermeasures has become especially acute following the events of September 2001. Traditional antiviral agents interfere with viral proteins or functions. In a new study, Yang et al. focus instead on host cellular pathways used by the virus. A drug that interferes with the cellular ErbB-1 signal transduction pathway, activated by smallpox growth factor, sheds new light on how the virus replicates in the cell. Drugs that target the ErbB-signaling pathways represent a

Light-induced phosphorylation of crystallins in the retinal pigment epithelium.

Protein phosphorylations have essential regulatory roles in visual signaling. Previously, we found that phosphorylation of several proteins in the retina and retinal pigment epithelium (RPE) is involved in anti-apoptotic signaling under oxidative stress conditions, including light exposure. In this study, we used a phosphoprotein enrichment strategy to evaluate the light-induced phosphoproteome of primary bovine RPE cells. Phosphoprotein-enriched extracts from bovine RPE cells exposed to light or dark conditions for 1h were separated by 2D SDS-PAGE. Serine and tyrosine phosphorylations were vi

Photic regulation of melatonin in humans: ocular and neural signal transduction.

Light is a potent stimulus for regulating the pineal gland's production of melatonin and the broader circadian system in humans. It initially was thought that only very bright photic stimuli (> or = 2500 lux) could suppress nocturnal melatonin secretion and induce other circadian responses. It is now known that markedly lower illuminances (< or = 200 lux) can acutely suppress melatonin or entrain and phase shift melatonin rhythms when exposure conditions are optimized. The elements for physical/biological stimulus processing that regulate photic influences on melatonin secretion include the ph

Light-Inducible Activation of TrkA for Probing Chronic Pain in Mice.

Chronic pain is a prevalent problem that plagues modern society, and better understanding its mechanisms is critical for developing effective therapeutics. Nerve growth factor (NGF) and its primary receptor, Tropomyosin receptor kinase A (TrkA), are known to be potent mediators of chronic pain, but there is a lack of established methods for precisely perturbing the NGF/TrkA signaling pathway in the study of pain and nociception. Optobiological tools that leverage light-induced protein-protein interactions allow for precise spatial and temporal control of receptor signaling. Previously, our lab

Acetylcholine modulates ganglion cell activity in the trout pineal organ.

In the teleost pineal organ light activates functional photoreceptors, which transmit electrical activity to the brain via axons of intermediate ganglion cells. To investigate whether acetylcholine plays a role in the transduction of pineal photoreceptor signals, extracellular recordings were performed from ganglion cells of intact superfused pineal organs of the rainbow trout. Bath-applied acetylcholine increased the spike discharge rate of 96% of achromatic ganglion cells in a dose-dependent manner. The light response curve of ganglion cells, which was obtained by plotting spike rate vs ligh

Transcriptome analysis of the medicinal mushroom Sanghuangporus vaninii in response to white light stress.

Light is a vital environmental factor that promotes the growth and development of edible fungi mycelium. Under white light, the mycelium color of Sanghuangporus vaninii shifts during its growth stages. To investigate the impact of visible light on mycelial morphogenesis, a comparative transcriptomic analysis was conducted. This analysis revealed the molecular processes that underpin mycelial growth and development in S. vaninii when cultured in both darkness and light conditions. From the analysis, 13,643 genes were aligned using Illumina raw reads. Of these, 596 genes exhibited significant ex

Monochromatic green light-mediated melatonin promotes cecal barrier function in broilers.

Light plays a crucial role as an environmental factor, greatly affecting chick behavior, production performance and health. Melatonin, an essential marker of photoelectric signaling, plays a role in regulating various physiological functions in broilers, such as gut health. However, whether monochromatic light can mediate melatonin secretion to regulate intestinal function and its regulatory mechanism are not known. In the present study, we showed that monochromatic green light significantly enhanced the mucosal barrier function of the broiler cecum, including the up-regulation of goblet cells
