Comment on the ratio of chromosome-type dicentric interchanges to centric rings for track-clustered compared with random breaks.

Evidence has been published suggesting that the ratio of chromosome-type dicentric interchanges to centric rings (D/R or F) is significantly lower for neutrons than for X or gamma rays, and it is proposed that a low D/R could be used as a "fingerprint" for high-LET radiations. One explanation offered for this observation is that the closely spaced, clustered breaks confined to linear tracks will favor intrachanges, as opposed to interchanges, leading to a lower D/R, while the more scattered, random breaks of low-LET radiations will favor the reverse situation and elevate the D/R. We have teste

Estimation of LET distributions ofα-particles from measured scintillator-based trajectory images.

Objective. Linear energy transfer (LET) measurements ofα-particles are increasingly important for micro-dosimetric evaluation in targetedα-radionuclide therapy. However, direct LET measurements remain challenging due to the extremely short range ofα-particles in matter. In this study, we propose a method to estimate LET distributions using quenching effects observed inα-particle trajectory images.Approach. α-particle trajectories were imaged using an ultra-high-resolution imaging system consisting of a Gd3Al2Ga3O12scintillator, an optical magnification unit, and an electron-multiplying charge-

Carbon ion dosimetry on a fluorescent nuclear track detector using widefield microscopy.

Fluorescent nuclear track detectors (FNTDs) are solid-state dosimeters used in a wide range of dosimetric and biomedical applications in research worldwide. FNTDs are a core but currently underutilized dosimetry tool in the field of radiation biology which are inherently capable of visualizing the tracks of ions used in hadron therapy. The ions that traverse the FNTD deposit their energy according to their linear energy transfer and transform colour centres to form trackspots around their trajectory. These trackspots have fluorescent properties which can be visualized by fluorescence microscop

The influence of hypoxia on LET and RBE relationships with implications for ultra-high dose rates and FLASH modelling.

Objective. To investigate relationships between linear energy transfer (LET), fluence rates, changes in radiosensitivity and the oxygen enhancement ratio (OER) in different ion beams and extend these concepts to ultra-high dose rate (UHDR) or FLASH effects.Approach.LET values providing maximum relative biological effect (RBE), designated as LETU, are found for neon, carbon and helium beams. Proton experiments show reduced RBEs with depth in scattered (divergent) beams, but not with scanned beams, suggesting that instantaneous fluence rates (related to track separation distances) can modify RBE

Radiation-induced neoplastic transformation in vitro: evidence for a protective effect at low doses of low LET radiation.

The study of radiation-induced transformation in vitro has long been an experimental approach to examine mechanisms underlying radiation carcinogenesis. Even though the major concern of exposure to radiation is the risk of cancer induction at low radiation doses, most laboratory mechanistic studies have focused on high dose effects. This, coupled with the fact that epidemiologic data are rarely powerful enough to accurately discriminate this risk at doses <5 cGy, has led in recent years to an increased effort to study low dose effects using the endpoint of neoplastic transformation in vitro. High throughput screen of small molecules as potential countermeasures to galactic cosmic radiation induced cellular dysfunction.

Space travel increases galactic cosmic ray exposure to flight crews and this is significantly elevated once travel moves beyond low Earth orbit. This includes combinations of high energy protons and heavy ions such as 56Fe or 16O. There are distinct differences in the biological response to low-energy transfer (x-rays) or high-energy transfer (High-LET). However, given the relatively low fluence rate of exposure during flight operations, it might be possible to manage these deleterious effects using small molecules currently available. Virtually all reports to date examining small molecule man

Characterizing diamond detectors for various dose and dose rate measurements in scanned carbon and oxygen beams.

BACKGROUND: The emerging FLASH radiotherapy technique employs "Ultra-High Dose Rate" (UHDR) irradiations and offers the potential to spare normal tissue while maintaining iso-effective tumor treatment. Given the physical and biological advantages inherent to high "Linear Energy Transfer" (LET) particles, the combination of UHDR and high LET has the capability to enhance the normal tissues sparing, as indicated by initial in vivo trials. However, to ensure a safe implementation of this combined modality, it is essential to establish robust dosimetric protocols utilizing dose-, dose rate-, and L

Motion energy versus position tracking: spatial, temporal, and chromatic parameters.

The fundamental question in motion perception is whether motion is an interpretation imposed on an object or feature perceived at separate positions at sequential instants, or whether it is the response of direction-sensitive detectors that can extract the motion-energy in the stimulus, i.e. the orientation of spatio-temporal energy. To answer this question we constructed stimuli whose position changed in one direction while the motion energy contained in the same spatial frequency moved in the same or the opposite direction (by superimposing moving sinusoidal gratings on stationary gratings o

Radium deposition in human brain tissue: A Geant4-DNA Monte Carlo toolkit study.

NASA has encouraged studies on 226Ra deposition in the human brain to investigate the effects of exposure to alpha particles with high linear energy transfer, which could mimic some of the exposure astronauts face during space travel. However, this approach was criticized, noting that radium is a bone-seeker and accumulates in the skull, which means that the radiation dose from alpha particles emitted by 226Ra would be heavily concentrated in areas close to cranial bones rather than uniformly distributed throughout the brain. In the high background radiation areas of Ramsar, Iran, extremely hi

Radiation quality and intra-chromosomal aberrations: Size matters.

The shift from plant to mammalian cell models in radiation cytogenetics hastened the development of methods suitable for the analysis of chromosome-type aberrations. These included methods to detect interchanges that take place between different chromosomes (dicentrics and translocations), and intrachanges occurring within a given chromosome (rings, interstitial deletions and inversions). In this review we consider the relationship between chromosome-type interchanges and intrachanges in response to changes in ionization density (linear energy transfer; LET). In that context, we discuss advant

Harnessing Particle Ions for FLASH Radiotherapy.

FLASH radiotherapy continues to be an emerging clinical treatment that utilises ultra-high dose rate radiation (>40 Gy/s) to elicit normal tissue sparing compared to conventional (CONV) dose rate delivery. This is termed the FLASH effect, which has also been demonstrated to lead to effective tumour control. FLASH sparing has been demonstrated in many studies using electrons and now increasingly with particle ions, particularly protons. Due to unique properties such as deeper penetrance and a narrow range of energy deposition providing more precision targeting, this makes hadron radiotherapy us

A Monte Carlo simulation framework for investigating the effect of inter-track coupling on H 2 O 2 ${\rm H}_{2}{\rm O}_{2}$ productions at ultra-high dose rates.

BACKGROUND: Lower production of H 2 O 2 ${\rm H}_{2}{\rm O}_{2}$ in water is a hallmark of ultra-high dose rate (UHDR) compared to the conventional dose rate (CDR). However, the current computational models based on the predicted yield of H 2 O 2 ${\rm H}_{2}{\rm O}_{2}$ are the opposite of the experimental data. PURPOSE: To present a multi-scale formalism to reconcile the theoretical modeling and the experimental observations of H 2 O 2 ${\rm H}_{2}{\rm O}_{2}$ production and provide a mechanism for the suppression of H 2 O 2 ${\rm H}_{2}{\rm O}_{2}$ at FLASH-UHDR. Inverse dose protraction effects of low-LET radiation: Evidence and significance.

Biological effects of ionizing radiation vary not merely with total dose but also with temporal dose distribution. Sparing dose protraction effects, in which dose protraction reduces effects of radiation have widely been accepted and generally assumed in radiation protection, particularly for stochastic effects (e.g., solid cancer). In contrast, inverse dose protraction effects (IDPEs) in which dose protraction enhances radiation effects have not been well recognized, nor comprehensively reviewed. Here, we review the current knowledge on IDPEs of low linear energy transfer (LET) radiation. To

Inverse dose protraction effects of high-LET radiation: Evidence and significance.

Biological effects of ionizing radiation vary with radiation quality, which is often expressed as the amount of energy deposited per unit length, i.e., linear energy transfer (LET). For acute irradiation, high-LET radiation generally produces greater biological effects than low-LET radiation, but little knowledge exists as to how dose protraction modifies effects. In this regard, inverse dose protraction effects (IDPEs) are phenomena in which dose protraction enhances effects, contrasting with sparing dose protraction effects in which dose protraction reduces effects. Here, we review the curre

[Molecular Mechanisms of Antitumor Effect in Targeted Alpha-radionuclide Therapy].

Targeted α-radionuclide therapy (TAT) is a systemic therapy for cancer with cancer-targeting compounds conjugated with α-emitters. Since the linear energy transfer (LET) of α-particles are high, relative biological effectiveness is higher than low LET radiation, such as X-rays and β-particles, and the damage to the normal tissues surrounding the tumor is minimal because path length of α-particle is short (a few cells). TAT has demonstrated remarkable therapeutic effects in patients and has attracted considerable attention from physicians and researchers worldwide. In particular, a study publis

Lighting up FLASH and beyond: TLD response across four orders of magnitude in proton dose rate.

BACKGROUND: Thermoluminescent dosimeters (TLDs) are widely used for proton therapy dosimetry and remote audits. Yet their response under pulsed delivery and the ultrahigh dose rates relevant to proton FLASH remains incompletely characterized. In proton beams, apparent dose-rate effects can be masked by changes in linear energy transfer (LET), so we designed measurements to separate true dose-rate dependence from LET-driven changes in TLD sensitivity. PURPOSE: To characterize TLD-100 (LiF:Mg,Ti) detector response to synchrotron-based proton beams across in-pulse dose rates spanning approximate

Dosimetric and microdosimetric analyses for blood exposed to reactor-derived thermal neutrons.

Thermal neutrons are found in reactor, radiotherapy, aircraft, and space environments. The purpose of this study was to characterise the dosimetry and microdosimetry of thermal neutron exposures, using three simulation codes, as a precursor to quantitative radiobiological studies using blood samples. An irradiation line was designed employing a pyrolytic graphite crystal or-alternatively-a super mirror to expose blood samples to thermal neutrons from the National Research Universal reactor to determine radiobiological parameters. The crystal was used when assessing the relative biological effe
