A simulation study of confounding in generalized linear models for air pollution epidemiology.

Confounding between the model covariates and causal variables (which may or may not be included as model covariates) is a well-known problem in regression models used in air pollution epidemiology. This problem is usually acknowledged but hardly ever investigated, especially in the context of generalized linear models. Using synthetic data sets, the present study shows how model overfit, underfit, and misfit in the presence of correlated causal variables in a Poisson regression model affect the estimated coefficients of the covariates and their confidence levels. The study also shows how this

Predicting clinical events using Bayesian multivariate linear mixed models with application to scleroderma.

BACKGROUND: Scleroderma is a serious chronic autoimmune disease in which a patient's disease state manifests in several irregularly spaced longitudinal measures of lung, heart, skin, and other organ systems. Threshold crossings of pulmonary and cardiac measures indicate potentially life-threatening key clinical events including interstitial lung disease (ILD), cardiomyopathy, and pulmonary hypertension (PH). The statistical challenge is to accurately and precisely predict these events by using all of the clinical history for the patient at hand and for a reference population of patients. METH

Identification of Affine Linear Parameter Varying Models for Adaptive Interventions in Fibromyalgia Treatment.

There is good evidence that naltrexone, an opioid antagonist, has a strong neuroprotective role and may be a potential drug for the treatment of fibromyalgia. In previous work, some of the authors used experimental clinical data to identify input-output linear time invariant models that were used to extract useful information about the effect of this drug on fibromyalgia symptoms. Additional factors such as anxiety, stress, mood, and headache, were considered as additive disturbances. However, it seems reasonable to think that these factors do not affect the drug actuation, but only the way in

Cumulative input function method for linear compartmental models and spectral analysis in PET.

Compartmental modeling and spectral analysis are often used for tracer kinetic modeling in positron emission tomography (PET). The concentrations in kinetic equations are usually considered to be instantaneous, whereas PET data are inherently integrated over time, which leads to uncertainties in the results. A new formalism for kinetic analysis that uses cumulative tracer concentrations and avoids approximating the image-derived input function and PET measurements with midframe instantanous values was developed. We assessed the improvements of the new formalism over the midframe approximation

Searching for genotype-phenotype structure: using hierarchical log-linear models in Crohn disease.

There has been considerable recent success in the detection of gene-disease associations. We consider here the development of tools that facilitate the more detailed characterization of the effect of a genetic variant on disease. We replace the simplistic classification of individuals according to a single binary disease indicator with classification according to a number of subphenotypes. This more accurately reflects the underlying biological complexity of the disease process, but it poses additional analytical difficulties. Notably, the subphenotypes that make up a particular disease are ty

Characterization of the growth curve of free-range turkeys using linear and non-linear models in locally adapted versus commercial hybrid genotypes.

Organic and free-range farming are becoming increasingly common practices in the turkey meat industry, although limited research has been conducted in uncontrolled environments. Therefore, the present work aims to define the growth pattern of an indigenous landrace and a commercial strain grown outdoors in southern Spain. To achieve this aim, the weights of Andalusian turkeys (1,826 and 1,797 records for males and females, respectively) and commercial toms (584 records) were recorded to test 19 linear and nonlinear mathematical functions. The suitability of the models was defined using the coe

Model-free prognostication of non-linear time series.

OBJECTIVE: The COVID-19 pandemic has highlighted the importance of studying the course of infectious progression. Similar needs exist for time series of other origins. While models are commonly devised and fitted to the observed data, we recently demonstrated the feasibility to directly evaluate the noisy non-linear time series that characterize the occurrence. However, for practical utility, analytics alone has limited value. The requirement of forecasting - at least in the short term - needs to be met. METHODS: We initially utilized normalized new infections per day (7-day moving average fo

High-dimensional partial linear model with trend filtering.

Understanding the links between diet, metabolic changes, and health outcomes is a key focus in nutritional science and broader biological research. Analyzing relationships, such as those between ultra-processed food (UPF) intake and metabolites, offers insights into potential biomarkers for diet-related diseases and public health applications. However, these analyses are challenging due to high-dimensional data structures and complex, often nonlinear associations between covariates and health outcomes. Traditional linear models and conventional nonparametric methods often lack the flexibility

Linear No-Threshold model and standards for protection against radiation.

In response to the three petitions by Carol S. Marcus, Mark L. Miller, and Mohan Doss, dated February 9, February 13, and February 24, 2015, respectively, the Nuclear Regulatory Commission (NRC or the Commission) has announced that it is considering assessing its choice of dose-response model, the Linear No-Threshold (LNT) model, for exposure to ionizing radiation. This comment is designed to assist the Commission in evaluating the merits of a review of the default dose-response model it uses as the basis for the Standards for Protection against Radiation regulations. It extends the petitioner

A noninvasive estimation of hypernasality using a linear predictive model.

The pronunciation of a speaker with a defective soft palate is marked by hypernasality and an operation may be necessary to repair the defective soft palate to reduce this hypernasality. An assessment of hypernasality is necessary to quantify the effect of the surgery. The current clinical methods for assessing hypernasality are uncomfortable or require expensive equipment. In this paper, a new quantitative method is proposed to estimate hypernasality. This method requires only a microphone and a personal computer equipped with a sound card. Application of the linear-quadratic model to myelotoxicity associated with radioimmunotherapy.

The purposes of this study were: (1) to use the linear-quadratic model to determine time-dependent biologically effective doses (BEDs) that were delivered to the bone marrow by multiple infusions of radiolabeled antibodies, and (2) to determine whether granulocyte and platelet counts correlate better with BED than administered radioactivity, which does not take stem cell repopulation, i.e., time, into consideration. Twenty patients with B-cell malignancies that had progressed despite intensive chemotherapy and who had a significant number of malignant cells in their bone marrow were treated wi

Toward predictive prosthetic socket modeling: Preliminary linear regression models of transradial limb-socket geometry.

Socket fit is essential for successful upper-limb prosthetic devices, yet current design workflows remain labor-intensive and dependent on clinician experience. Several digital and algorithmic approaches have been proposed to partially automate socket design, many of which rely on characterizing conventional design practice. In upper-limb prosthetics, prior research has quantified transradial socket rectification practices, but the direct geometric relationship between the residual limb and final socket remains unclear despite its importance to digital socket design workflows. This study exami

From linear models to deep learning: statistical advances in genomic selection for animal breeding.

Genomic selection (GS) has revolutionized animal breeding by accelerating genetic gain through genome-wide marker data. As genotyping technologies advance and data dimensionality grows, the statistical foundations of GS are shifting from classical linear frameworks, which assume additive genetic effects, toward advanced computational models that capture complex nonlinear relationships in genomic data. The commercialization of genotyping arrays for livestock and poultry, coupled with steadily declining sequencing costs, has led to an exponential increase in the availability of high-density geno

Penalized estimation of linear transformation models for interval-censored data with time-dependent covariates.

We investigate efficient estimation strategies for partially linear transformation models with time-dependent covariates under interval censoring. The unknown monotone function is approximated using a monotone B-spline basis to enable flexible semiparametric modeling, and we develop a computationally efficient nested hybrid EM algorithm that integrates Newton's method with isotonic regression. To support large-sample inference, we propose a straightforward variance-covariance estimation procedure for the regression parameters and introduce a score test to assess the adequacy of the proportiona

Generalized Functional Linear Regression Models With Functional and Scalar Covariates Prone to Measurement Error.

Most methods for adjusting for biases due to measurement errors in covariates in generalized linear regression models focus on scalar covariates. Less work exists to correct for biases due to measurement error in a mixture of functional and scalar covariates. To address this limitation, we develop joint functional simulation extrapolation (FSIMEX) and mixed effects model-based (MEM) approaches in a mixture of functional and scalar covariates prone to classical measurement errors in generalized functional linear regression. The proposed methods were compared with the Oracle estimator, which ser

Determinants of old age disability in Botswana: an empirical investigation using generalized linear models.

BACKGROUND: As Botswana experiences a demographic transition marked by an expanding population of older adults, understanding the determinants of disability among older adults becomes critical for shaping inclusive health and social policies. Disability in later life often stems not only from biological ageing but also from intersecting personal, household, and community-level conditions. Despite increasing attention to ageing in sub-Saharan Africa, few studies have comprehensively assessed the multilevel factors influencing functional limitations in older populations. METHODS: Drawing on nat

A linear noise approximation for stochastic epidemic models fit to partially observed incidence counts.

Stochastic epidemic models (SEMs) fit to incidence data are critical to elucidating outbreak dynamics, shaping response strategies, and preparing for future epidemics. SEMs typically represent counts of individuals in discrete infection states using Markov jump processes (MJPs), but are computationally challenging as imperfect surveillance, lack of subject-level information, and temporal coarseness of the data obscure the true epidemic. Analytic integration over the latent epidemic process is impossible, and integration via Markov chain Monte Carlo (MCMC) is cumbersome due to the dimensionalit

Modeling dengue vector population with earth observation data and a generalized linear model.

Mosquitoes propagate many human diseases, some widespread and with no vaccines. The Ae. aegypti mosquito vector transmits Zika, Chikungunya, and Dengue viruses. Effective public health interventions to control the spread of these diseases and protect the population require models that explain the core environmental drivers of the vector population. Field campaigns are expensive, and data from meteorological sites that feed models with the required environmental data often lack detail. As a consequence, we explore temporal modeling of the population of Ae. aegypti mosquito vector species and en

Multiple linear regression models for individualized radiation exposure planning in dark-field chest radiography.

BACKGROUND: Clinical X-ray dark-field radiography has shown to be promising for visualizing different lung pathologies. To keep the radiation dose as low as reasonably achievable (ALARA principle), individualized exposure planning is necessary. However, the current scanning-based implementation of dark-field radiography complicates the use of automatic exposure control. Previously, a BMI-based linear regression model was proposed as a substitute. Here, we aim to improve this proposed model by investigating multiple linear regression for patient-individual exposure planning of dark-field chest

Enhancing Machine-Learned Electrostatic Models with Linear Response Theory for Multiscale Simulations in Enzymes.

To accelerate computationally expensive quantum mechanical (QM) calculations, integrating machine learning potentials (MLPs) with molecular mechanics (MM) has become a promising approach, which can be employed to perform multiscale ML/MM-MD simulations and estimate reaction free energy for enzymatic catalysis. However, the ML/MM frameworks face the challenge of intuitive chemical interpretability in QM/MM electrostatic coupling, which limits their transferability in unlearned MM environments. Therefore, their practical application in enzyme engineering is hindered by the bottleneck of MLP retr
