Multiple symmetric lipomatosis: a defect in adrenergic-stimulated lipolysis.

The cellularity of normal and lipomatous adipose tissue and its response to different lipolytic agents have been studied in a group of 10 patients with multiple symmetric lipomatosis (MSL). In MSL patients, fat cells from lipomatous tissue are smaller than normal, uninvolved adipocytes. Fat cells from lipomata show minimal variations in size following conspicuous increase of lipomatous masses. These findings suggest that the growth of lipomata can be attributed to the neoformation of adipocytes rather than to an enlargement in the single fat cells. The incidence of reduced glucose tolerance an

Lipolysis gone rogue: the HSL connection in feeding cancer.

Lipolysis, a tightly regulated metabolic process, is hijacked by cancer cells to meet their energy and biosynthetic demands under stress. Central to this process is hormone-sensitive lipase (HSL), a key enzyme that orchestrates lipid mobilization by hydrolyzing diacylglycerols into free fatty acids (FFAs). This review explores the pivotal and multifaceted role of HSL in cancer metabolism, focusing on its dual function acting as both a tumor promoter and suppressor depending on the cancer type and microenvironment. Lipolysis, the breakdown of triglycerides into free fatty acids (FFAs), is essen

Rapid orbital lipolysis associated with critical illness and colectomy.

Orbital lipolysis typically develops in the setting of a chronic catabolic state. The acute development and rapid progression of orbital lipolysis are much less commonly described. In this report, we present a rare case of a 64-year-old male who progressed from normal orbital fat content to marked orbital lipolysis in less than one month following episodes of undifferentiated shock, colonic perforation, and total colectomy. We outline the clinical course, describe the characteristic imaging findings, and provide a review of the cellular mechanisms underlying lipolysis. Our case suggests that m

Impaired lipolysis in propionic acidemia: A new metabolic myopathy?

The objective of this study was to investigate the fat and carbohydrate metabolism in a patient with propionic acidemia (PA) during exercise by means of indirect calorimetry and stable isotope technique. A 34-year-old patient with PA performed a 30-minute submaximal cycle ergometer test. Data were compared to results from six gender- and age-matched healthy controls. Main findings are that the patient with PA had impaired lipolysis, blunted fatty acid oxidation, compensatory increase in carbohydrate utilization, and low work capacity. Our findings indicate that PA should be added to the list o

Effects of laser-assisted lipolysis on nipple-areola complex.

BACKGROUND: Gynecomastia, as a most common benign condition, represents itself as the enlargement of the male breast and also nipple-areola complex as the severity of the condition increases. With this study, we aimed to clarify the effects of 980-nm diode laser on nipple-areola complex (NAC). OBJECTIVES: Although numerous open techniques have been described to correct gynecomastia, nowadays trends have shifted to minimally invasive techniques such as laser-assisted lipolysis (LAL). METHODS: A total of 25 patients with bilateral gynecomastia treated with LAL by using a 980-nm diode laser. R

Apelin inhibits adipogenesis and lipolysis through distinct molecular pathways.

Apelin is an adipokine secreted by adipocytes. Co-expression of apelin and apelin receptor (APJ) in adipocytes implies the autocrine regulations of apelin on adipocyte functions through yet unknown molecular mechanisms. In the present study, we provide evidence that apelin, through its interaction with APJ receptor, inhibits adipogenesis of pre-adipocytes and lipolysis in mature adipocytes. The detailed molecular pathways underlying apelin signaling is proposed based on our experimental observations. Specifically, we show that apelin suppresses adipogenesis through MAPK kinase/ERK dependent pa

Lipolysis of triglyceride-rich lipoproteins, vascular inflammation, and atherosclerosis.

Epidemiological and interventional studies have implicated elevated triglyceride-rich lipoprotein (TGRL) levels as a risk factor for cardiovascular disease and vascular inflammation, though the results have not been entirely consistent. This appears particularly relevant in model systems where the lipolysis occurs in the setting of established inflammation (e.g., in pre-existing atherosclerotic plaques), rather than in the tissue capillary beds where lipolysis normally occurs. Two main mechanisms seem to link TGRL lipolysis to vascular inflammation. First, lipolysis of TGRL leaves behind parti

Hypothalamic histamine neurons activate lipolysis in rat adipose tissue.

The contribution of hypothalamic histamine neurons to the central regulation of peripheral lipid metabolism was investigated in rats using in vivo microdialysis system. A bolus infusion of L-histamine at doses of 10--10(3) nmol/rat into the third cerebral ventricle (i3vt) dose-dependently increased glycerol concentration in the perfusate from the epididymal adipose tissue. I3vt infusion of 10(2) nmol/rat thioperamide, an autoinhibitory H(3) receptor antagonist that activates histamine neurons to increase synthesis and release of neuronal histamine, convincingly mimicked histamine action in the

Homocysteine suppresses lipolysis in adipocytes by activating the AMPK pathway.

Hyperhomocysteinemia (HHcy) is an independent risk factor for coronary artery disease. Emerging evidence suggests that HHcy is also associated with adipocyte tissue dysfunction. One of the principal functions of adipose tissue is to provide energy substrate via lipolysis. In the present study, we investigated the effects of homocysteine (Hcy) on lipolysis in adipocytes. We found that Hcy inhibited release of glycerol and fatty acids, two typical indicators of the lipolytic response, in primary adipocytes and fully differentiated 3T3-L1 adipocytes in a dose-dependent manner under both basal and

Effects of glucagon on lipolysis and ketogenesis in normal and diabetic men.

The effect of glucagon (50 ng/kg/min) on arterial glycerol concentration and net splanchnic production of total ketones and glucose was studied after an overnight fast in four normal and five insulin-dependent diabetic men. Brachial artery and hepatic vein catheters were inserted and splanchnic blood flow determined using indocyanine green. The glucagon infusion resulted in a mean circulating plasma level of 4,420 pg/ml. In the normal subjects, the glucagon infusion resulted in stimulation of insulin secretion indicated by rising levels of immunoreactive insulin and C-peptide immunoreactivity.

Lipolysis in Health and Disease: Pathways, Regulation, and Metabolic Consequences.

PURPOSE OF REVIEW: Lipolysis regulates lipid distribution, energy availability, and metabolic homeostasis in organisms that store triacylglycerols. In multicellular organisms, adipose tissue evolved as a specialized organ that centralizes lipid storage and release, buffers nutrient fluctuations, and protects non-adipose tissues from lipid overload. Dysregulated adipocyte lipolysis occurs across diverse metabolic conditions, including obesity, diabetes, lipodystrophy, and inflammatory states. This review synthesizes evidence that defective suppression of basal lipolysis and impaired responsiven

Inhibition of adipocyte lipolysis by vaspin impairs thermoregulation in vivo.

Altered activity of brown adipose tissue (BAT) contributes to obesity, insulin resistance, and cardiovascular disease. BAT secretes endocrine factors ("batokines") that regulate thermogenesis. We identify the serpin vaspin as a batokine that modulates adrenergic control of lipolysis and thermogenesis. Adipocyte-specific vaspin overexpression in mice reduces BAT activation and impairs thermoregulation during cold exposure or fasting. Mechanistically, vaspin binds low-density lipoprotein receptors (LRP1, LDLR, vLDLR), inhibiting adrenergic signaling and lipolysis in brown and white adipocytes by

TLR4 and prostaglandin pathways at the crossroads of endotoxemia-induced lipolysis.

BACKGROUND: During endotoxemia, immune activation rapidly increases energy needs. To meet these demands, white adipose tissue (WAT) mobilizes fatty acids through lipolysis. While fatty acids serve as energy-dense substrates, they also act as precursors for lipid mediators, including prostaglandins (PGs), that drive inflammation. The dual role of WAT is crucial during endotoxemia, wherein both energy needs and inflammatory signals are amplified. However, the mechanisms by which WAT regulates lipolysis during endotoxemia are not well understood. Dairy cows serve as an excellent model for studyin

The efficacy and safety of cold-induced lipolysis in the treatment of pseudogynecomastia.

BACKGROUND: The treatment options for pseudogynecomastia have been limited. Cold-induced lipolysis provides a noninvasive, localized subcutaneous adipocyte destruction by inducing adipocyte apoptosis. OBJECTIVE: This study has been designed to evaluate the efficacy of cold-induced lipolysis as a treatment modality for pseudogynecomastia. METHODS: In this 28-week prospective trial, a total of 12 male pseudogynecomastia patients (Korean) were treated twice with cold-induced lipolysis. Efficacy was determined by chest circumference, ultrasonographic measurement of fat thickness, Simon's Gynecom

Biphasic effects of orexin-A on autonomic nerve activity and lipolysis.

Previously, we showed that orexin-A, a 33-aa peptide, influences renal sympathetic nerve activity. Because the autonomic nervous system plays an important role in the regulation of lipid metabolism, we investigated the in vivo effects of orexin-A on the sympathetic nerve activity innervating white adipose tissue (WAT-SNA) and lipolysis. We found that intracerebroventricular (icv) administration of orexin-A at doses of 1 microg/rat and 10 ng/rat elevated and suppressed WAT-SNA, respectively. The effect of the high dose of orexin-A (1 microg/rat) was eliminated by pretreatment with diphenhydrami

Adrenergic regulation of lipolysis in a patient with lipoatrophy of the upper body.

A patient with lipoatrophy of the upper body had laboratory evidence of increased activity of the adrenergic nervous system. Because the adrenergic nervous system is important in the regulation of lipolysis, studies were performed to determine whether lipolysis of the upper body was ongoing in this patient and whether pharmacologic reduction of activity of the adrenergic nervous system would affect lipolysis of adipose tissue. Release of free fatty acids from systemic and forearm adipose tissue was measured before and after the administration of guanfacine hydrochloride. Forearm adipose tissue
