APMIS : acta pathologica, microbiologica, et immunologica Scandinavica2026eng
Candida albicans has been recognized as a WHO-prioritized critical fungal pathogen that requires urgent attention, as it can potentially cause life-threatening infections. Antifungal agents may become ineffective in the future due to the rapid emergence of antifungal resistance, potentially rendering them unable to treat biofilm-associated infections. The present investigation employs various in vitro techniques, such as antifungal susceptibility, in vitro pharmacodynamics, advanced electron microscopy, zeta potential assessment, and rhodamine 6G efflux assay, to evaluate the anti-Candida and

The polymyxin and lipopeptide classes of antibiotics are membrane-targeting drugs of last resort used to treat infections caused by multi-drug-resistant pathogens. Despite similar structures, these two antibiotic classes have distinct modes of action and clinical uses. The polymyxins target lipopolysaccharide in the membranes of most Gram-negative species and are often used to treat infections caused by carbapenem-resistant species such as Escherichia coli, Acinetobacter baumannii and Pseudomonas aeruginosa. By contrast, the lipopeptide daptomycin requires membrane phosphatidylglycerol for act

Mycobacterium avium subspecies paratuberculosis (MAP), the causative agent of Johne disease in cattle and other ruminants, is proposed to be at least one of the causes of Crohn disease in humans. MAP and Mycobacterium avium subspecies avium, a closely related opportunistic environmental bacterium, share 95% of their genes and exhibit homologies of more than 99% between these genes. The identification of molecules specific for MAP is essential for understanding its pathogenicity and for development of useful diagnostic tools. The application of gas chromatography, mass spectrometry, and nuclear

Lipopeptide natural products are essential agents against multidrug-resistant bacteria, but their clinical utility is often constrained by toxicity and resistance. Here, we compare the mechanisms of action of two superficially similar lipopeptide antibiotics: colistin, a last-line treatment for Gram-negative infections, and turnercyclamycins, a new class active against certain colistin-resistant strains. Both antibiotics require lipopolysaccharide (LPS) biosynthesis, even when LPS transport to the outer membrane (OM) is impaired. Colistin rapidly disrupts both the OM and the cytoplasmic membra

Toxoplasmosis, caused by Toxoplasma gondii (T. gondii), is a serious zoonotic parasitic disease. We previously found that Lycosin-I exhibited anti-T. gondii activity, but its serum stability was not good enough. In this study, we aimed to improve the stability and activity of Lycosin-I through fatty acid chain modification, so as to find a better anti-T. gondii drug candidate. The α/ε-amino residues of different lysine residues of Lycosin-I were covalently coupled with lauric acid to obtain eight lipopeptides, namely L-C12, L-C12-1, L-C12-2, L-C12-3, L-C12-4, L-C12-5, L-C12-6, and L-C12-7. Several Bacillus strains have been well studied for their ability to control soil-borne plant diseases. This property is linked to the production of several families of lipopeptides. Depending of their structure, these compounds show antifungal and/or plant systemic resistance inducing activities. In this work, the biodiversity of lipopeptides produced by different antifungal Bacillus strains isolated from seeds, rhizospheric, and non-rhizospheric soils in Algeria was analyzed. Sixteen active strains were characterized by PCR for their content in genes involved in lipopeptide biosynthesis and

The self-assembly in aqueous solution and conformation of lipopeptides C16-WKK, C16-KWK, C16-YKK and C16-KYK is compared and examined. Remarkable differences are observed among the systems despite the small sequence changes comparing C16-XKK with the C16-KXK homologue (X = W or Y), depending on pH. These are rationalized using a molecular theory for amphiphile self-assembly (MOLT) to predict the morphology along with atomistic molecular dynamics simulations to probe local conformation and packing, along with new experimental data from small-angle X-ray scattering (SAXS) and FTIR spectroscopy.

China is the world's leading producer and consumer of eggplant, yet effective strategies are urgently needed to manage postharvest soft rot caused by Fusarium equiseti, a newly identified pathogen. This study explores the biocontrol potential and mechanism of Bacillus amyloliquefaciens SDTA 2-1 against F. equiseti-induced soft rot in eggplants. Strain SDTA 2-1 exhibited significant inhibitory activity against F. equiseti, reducing the decay rate from 66.67 % to 26.67 %. Its biofilm-forming ability promoted colonization on eggplant fruit, enhancing disease control efficacy. Furthermore, SDTA 2-

Foodborne pathogenic microorganisms and the escalating crisis of antibiotic resistance present significant threats to global public health and food safety. This necessitates the urgent development of sustainable, green, and safe antibiotic alternatives. Lipopeptides, a class of emerging natural antimicrobial agents, offer considerable promise due to their low toxicity, broad-spectrum antibacterial activity, and environmental compatibility. Bacillus smithii, a facultative anaerobic thermophilic bacterium, exhibits probiotic properties, including antibacterial and anti-inflammatory effects. Howe

Bacterial resistance to antibiotics is a crisis. Acinetobacter baumannii is among the CDC urgent threat pathogens in part for this reason. Lipopeptides known as turnercyclamycins are produced by symbiotic bacteria that normally live in marine mollusks, where they may be involved in shaping their symbiotic niche. Turnercyclamycins killed Gram-negative pathogens including drug-resistant Acinetobacter, but how do the mechanisms of resistance compare to other lipopeptide drugs? Here, we define resistance from a truncation of MlaA, a protein involved in regulating bacterial membrane phospholipids.

A new family of synthetic, membrane-active, ultrashort lipopeptides composed of only four amino acids linked to fatty acids was tested for the ability to induce systemic resistance and defense responses in plants. We found that two peptides wherein the third residue is a d-enantiomer (italic), C16-KKKK and C16-KLLK, can induce medium alkalinization of tobacco suspension-cultured cells and expression of defense-related genes in cucumber and Arabidopsis seedlings. Moreover, these compounds can prime systemic induction of antimicrobial compounds in cucumber leaves similarly to the plant-beneficia

Candida glabrata is an important opportunistic human pathogen well known to develop resistance to antifungal drugs. Due to their numerous desirable qualities, antimicrobial lipopeptides have gained significant attention as promising candidates for antifungal drugs. In the present study, two bioactive lipopeptides (AF4 and AF5 m/z 1071.5 and 1085.5, respectively), coproduced and purified from Bacillus subtilis RLID12.1, consist of seven amino acid residues with lipid moieties. In our previous studies, the reversed phased-HPLC purified lipopeptides demonstrated broad-spectrum of antifungal activ

Endophytes are mutualistic symbionts within healthy plant tissues. In this study we isolated Bacillus spp. from seeds of several varieties of maize. Bacillus amyloliquifaciens or Bacillus subtilis were found to be present in all maize varieties examined in this study. To determine whether bacteria may produce antifungal compounds, generally lipopeptides in Bacillus spp., bacterial cultures were screened for production of lipopeptides. Lipopeptides were extracted by acid precipitation from liquid cultures of Bacillus spp. Lipopeptide extracts from Bacillus spp. isolated from Indian popcorn and

BACKGROUND: Toll-like receptor (TLR)-2/TLR1 heterodimers recognize bacterial lipopeptides and initiate the production of inflammatory mediators. Adaptors and co-receptors that mediate this process, as well as the mechanisms by which these adaptors and co-receptors function, are still being discovered.
METHODOLOGY/PRINCIPAL FINDINGS: Using shRNA, blocking antibodies, and fluorescent microscopy, we show that U937 macrophage responses to the TLR2/1 ligand, Pam(3)CSK(4), are dependent upon an integrin, α(3)β(1). The mechanism for integrin α(3)β(1) involvement in TLR2/1 signaling is through its ro

The Gram-negative bacterium Dickeya solani causes potato soft rot and blackleg, and effective control measures remain limited. We evaluated the agricultural potential of Aspergillus sulphureoviridis, a recently described and chemically unexplored fungus. Bioactivity-guided molecular networking of extracts that inhibited D. solani led to the isolation of six metabolites: cladobotrin IV (1), two new analogs cladobotrin VII and cladobotrin VIII (2 and 3), and the orsellinic acid-derived metabolites F9775A (4), F9775B (5), and violaceol I (6). Among these, the polyketide F9775B (5) inhibited both

Honey represents a unique ecological niche that harbors microorganisms with unexplored antimicrobial potential. This study aimed to isolate, characterize, and genomically evaluate Bacillus subtilis strains from multifloral ginger honey to assess their capacity for producing antimicrobial compounds and peptides. Two strains, Miodo8 and Miodo10, were characterized using phenotypic assays and whole-genome sequencing. Crude extracts obtained from optimized yeast peptone dextrose (YPD) fermentations demonstrated significant antimicrobial activity primarily against Gram-positive pathogens, including

Pemuchiamides A and B (1 and 2) were isolated from a marine Hormoscilla sp. cyanobacterium collected from Pemuchi Beach on Hateruma Island, Japan. Although 1 and 2 existed as a complex mixture of rotamers in chloroform-d, detailed analyses of their 2D NMR and tandem mass spectra revealed their planar structures, respectively. The absolute configurations of 1 and 2 were established via the degradation and derivatization reactions. Pemuchiamide A (1) exhibited potent growth-inhibitory activity against Trypanosoma brucei rhodesiense, the causative organism of African sleeping sickness, while 2 sh

BACKGROUND AND AIMS: Certain micro-organisms can improve plant protection against pathogens. The protective effect may be direct, e.g. due to antibiotic compounds, or indirect, by priming of plant defence as induced systemic resistance (ISR). The plant growth-promoting rhizobacterium Bacillus amyloliquefaciens UCMB5113 shows potential for disease management of oilseed rape. To investigate the mode of action of this protection, especially in relation to jasmonic acid-dependent ISR, Bacillus UCMB5113 was tested with Arabidopsis thaliana mutants and several important fungal pathogens of Brassica

Biofilm-associated infections pose significant challenges due to the increased antibiotic resistance of microorganisms within these structures. This study involved isolating and characterizing the biosurfactant produced by Bacillus subtilis RSL-2 using molasses to develop a sustainable antibiofilm agent. The biosurfactant was identified as a surfactin-like lipopeptide (molecular weight: 1037 g/mol) based on FTIR, 1H NMR, and HRMS analyses. It demonstrated a low critical micelle concentration (CMC) of 70 mg/L and markedly decreased surface and interfacial tensions to 24.46 and 0.46 mN/m, respec
