Photosensitive response mouse model: A potential tool for studying mechanisms of photosensitive epilepsy and initial screening of anti-epileptic drugs.

Numerous studies have investigated the mechanisms underlying photosensitive epilepsy, yet they remain poorly understood. Progress in this field has been substantially hampered by the scarcity of valid animal models and the difficulty in establishing them. Here we presented flashing lights at varying frequencies to head-fixed mice while recording local field potentials (LFPs) in the primary visual cortex (V1). We identified approximately 5 Hz as the resonant frequency for eliciting photosensitive responses. By modulating the luminance within the behavioural chamber we found that the response am

Transcranial ultrasound stimulation of motor networks in Parkinson's disease informed by local field potential dynamics.

Transcranial ultrasound stimulation (TUS) is a promising noninvasive technique for modulating deep brain targets and circuits with high spatial precision. For its successful clinical translation, confirmation of target engagement, together with a deeper understanding of the effects of TUS, is essential. To advance these goals, we obtained direct measures of neural activity using electrodes implanted in the subthalamic nucleus (STN) in patients with Parkinson's disease (PD) during TUS of deep and superficial targets, guided by magnetic resonance imaging-based acoustic modeling and real-time neu

Intracranial mapping identifies high-frequency cortical dynamics underlying visual discomfort.

High-contrast striped patterns near 3 cycles per degree are well established as a cause of visual discomfort and perceptual distortions, which in clinical populations can manifest as symptoms such as migraines or seizures. This sensitivity has been linked to cortical hyperexcitability, characterized by abnormally increased neural responses to visual input. Although gamma-band oscillations in visual cortex are known to reflect excitatory-inhibitory dynamics associated with pattern sensitivity, the contribution of higher-frequency neural activity and large-scale network interactions has received

Shared Spectral Parameters Between EEG and Basal Ganglia LFP Distinguish Sleep-Specific Oscillations in Parkinson's Disease.

BACKGROUND: In Parkinson's disease (PD), sleep-related oscillatory dynamics between basal ganglia and cortical nodes may inform sleep dysfunction. The subthalamic nucleus (STN) is highly interconnected with human sleep circuitry and is a current target of interest for sleep neuromodulation. METHODS: To investigate correlated cortical-subcortical activity, we recorded simultaneous PSG and STN local field potentials (LFP) in 19 patients undergoing deep brain stimulation (DBS) for PD. We evaluated the correlation in spectral power between STN-LFP and EEG during polysomnogram (PSG)-defined stages

40 Hz noninvasive transcranial ultrasound stimulation modulates multiscale nonlinear dynamics of hippocampal CA1 neural oscillations of Alzheimer's mouse model.

Objective.Aberrant nonlinear dynamics of hippocampal neural oscillations are a hallmark of early network dysfunction in Alzheimer's disease (AD), with critical roles in memory processing. Although transcranial ultrasound stimulation (TUS) has shown cognitive benefits in AD models and patients, its effects on the nonlinear dynamics of hippocampal CA1 oscillations and their multiscale organization remain unclear.Approach.Here, we applied 40 Hz noninvasive TUS to the hippocampal CA1 region of AD mouse model and recorded local field potentials before, during, and after stimulation. Analyses integr

Vestibular modulation of the tail of the rat striatum.

Fragmented and piecemeal evidence from animal and human studies suggests that vestibular information is transmitted to the striatum, a part of the basal ganglia that degenerates in Parkinson's Disease. Nonetheless, surprisingly little is known about the precise effects of activation of the vestibular system on the striatum. Electrophysiological studies have yielded inconsistent results, with many studies reporting only sparse responses to vestibular stimulation in the dorsomedial striatum. In this study, we sought to elucidate the effects of electrical stimulation of the peripheral vestibular

Variability-dominated auditory cortical dysfunction and targeted VNS modulation in a Mecp2+/- model of Rett syndrome.

OBJECTIVE: Rett syndrome is a neurodevelopmental disorder caused by mutations in the X-linked transcriptional regulator MECP2, which causes widespread abnormalities in cortical network activity and sensory processing. In rodent models, Mecp2+/- rats recapitulate key clinical features of Rett syndrome, including disrupted auditory cortical responses and impaired spike timing fidelity. Given its potential to normalize abnormal cortical activity, vagus nerve stimulation (VNS) may offer a promising therapeutic approach. This study examined auditory local field potential (LFP) responses in Mecp2+/-

Synchronous Integrated Detection System for Animal Gait and Cortical Functional Network Connectivity.

Deciphering the spatiotemporal patterns of gait behavior and synchronized cortical functional networks is highly significant in neuroscience research. However, current small-animal gait analyzers cannot synchronize gait and cortical functional network connectivity (CFNC), thus limiting our understanding of CFNC dynamics under different gaits. To address this issue, we developed a small-animal gait analyzer that can synchronize gait with whole-brain local field potentials in real time. Using this device, we precisely analyzed the real-time dynamic changes in the CFNC during fine gait movements

Parkinson's disease severity is encoded by non-linear oscillatory interactions across cortical-subcortical networks.

Parkinson's Disease (PD) is characterized by motor symptoms linked to dysregulated neural activity within basal ganglia-thalamocortical (BGTC) circuits. While clinical assessments such as the Unified Parkinson's Disease Rating Scale (UPDRS) effectively measure symptom severity, they are not designed to provide insights into underlying neuropathophysiology. We hypothesize that PD severity is encoded by nonlinear interactions across BGTC neurophysiological biomarkers. We recorded simultaneous preiriolandic electrocorticography and globus pallidus internus (GPi) local field potentials in 47 patie

Cognitive and Emotional Determinants of Subthalamic Oscillations During Freezing While Turning in Parkinson's Disease.

Freezing of gait (FoG) in Parkinson's disease (PD) is a disabling phenomenon that, beyond motor impairment, involves cognitive and emotional processes. However, their unique mechanisms and contributions to FoG remain unclear. In this study, we examined whether the severity of cognitive and emotional symptoms was associated with subthalamic nucleus (STN) oscillatory activity during freezing while turning. Local field potentials (LFPs) were recorded from the STN of 13 individuals with PD during both effective turning and freezing while turning. Given that the modulation of STN-LFPs during cognit

Prospective multi-phase observational study evaluating local field potentials to guide deep brain stimulation programming in dystonia at a UK Tertiary Neurosciences Centre (LFP-DYT): a protocol.

INTRODUCTION: Deep brain stimulation (DBS) for dystonia is effective but programming optimisation can take months. Local field potentials (LFPs) recorded by the Medtronic Percept device may provide biomarkers to guide stimulation. This study will prospectively evaluate whether chronic LFP profiles correlate with clinical outcomes and can inform DBS programming strategies. METHODS AND ANALYSIS: LFP-DYT is a single-centre, multi-phase observational study at Newcastle upon Tyne National Health Service (NHS) Foundation Trust. An internal pilot (Cohort 1) will refine recording workflows, followed

Targeted optical uncaging of an adenosine A1 receptor agonist regulates excitability of hippocampal slices derived from epileptic mice.

Photocaged adenosine A1 receptor (A1R) agonists combined with spatiotemporally selective illumination represents a promising therapeutic strategy for mesial temporal lobe epilepsy (mTLE). This ex vivo study investigated whether the A1R agonist N6-cyclopentyladenosine (CPA) suppresses hippocampal excitability in the intrahippocampal kainic acid (IHKA) mouse model and whether photocaged CPA (cCPA) enables subregion-selective and closed-loop reduction of hippocampal excitability. Hippocampal slices from male IHKA and saline-injected control mice were exposed to CPA (10-900 nM) while electrically

Polarization-engineered displacement sensing of nanoparticles via tracking scattering C-points.

We propose a polarization-engineered displacement sensing scheme that connects the three-dimensional polarization landscape of a tightly focused field with the scattering C-points of an embedded dipolar scatterer. The particle's displacement is encoded as angular shifts of the C-points in the far field. To engineer local polarization states, we first use circularly polarized light to localize the spin-rotating polarization landscape at the focal center, yielding a sensitive, nearly linear evolution of C-points. Furthermore, by employing structured light, we confine the polarization ellipticity

Noninvasive deep brain stimulation using theta-burst transcranial magnetic-acoustic coupling to modulate cortical rhythms in Parkinsonian mice.

Modulating the subthalamic nucleus (STN) through neural circuits can suppress abnormal discharge rhythms in the primary motor cortex (M1) in Parkinson's disease (PD), thereby enhancing motor function. However, a clinically viable noninvasive deep brain stimulation method for PD has yet to be realized. We developed a noninvasive transcranial magnetic-acoustic coupling stimulation (TMAS) system, employing a theta burst stimulation (TBS) mode, to examine its effects of simulated electrical parameters on cortical rhythms in PD. The theta burst-TMAS (TBTMAS) system was established, and its physical

Salicylate induced tinnitus: behavioral measures and neural activity in auditory cortex of awake rats.

Neurophysiological studies of salicylate-induced tinnitus have generally been carried out under anesthesia, a condition that abolishes the perception of tinnitus and depresses neural activity. To overcome these limitations, measurement of salicylate induced tinnitus were obtained from rats using schedule induced polydipsia avoidance conditioning (SIPAC) and gap pre-pulse inhibition of acoustic startle (GPIAS). Both behavioral measures indicated that tinnitus was present after treatment with 150 and 250 mg/kg of salicylate; measurements with GPIAS indicated that the pitch of the tinnitus was ne

Implications of Disruptions in HIV-Related Metrics.

Rigorous and timely HIV-related surveillance is essential to monitoring progress toward national, state, and local programmatic goals, making midcourse corrections, and informing policymaking. Decades of work have gone into making HIV surveillance a leading example in public health measurement systems. However, some disruption of, and uncertainty about this key surveillance system has begun. Here, we described the uncertainty regarding the National HIV Behavioral Surveillance (NHBS) system, estimation of HIV incidence, and tracking of HIV-related health disparities. Further, we articulate pote

Efficacy of Liposomal Bupivacaine in Ultrasound-Guided Peripheral Nerve Blocks for Postoperative Pain Management in Foot and Ankle Surgery.

BACKGROUND: In response to the opioid epidemic, many surgical specialties have adopted nonopioid pain management strategies. Ultrasound (US)-guided peripheral nerve blocks (PNBs) are effective in reducing pain and opioid consumption postsurgery. Liposomal bupivacaine (LB), shown effective in shoulder surgery, was approved in November 2023 for use in US-guided lower extremity blocks. This study compares the efficacy of LB in US-guided PNBs, surgeon-administered LB field blocks, and a control group using PNB with local anesthetic only. METHODS: This prospective, randomized, single-anonymized co
