The obligate-intracellular pathogen Chlamydia trachomatis (Ct) has undergone considerable genome reduction with consequent dependence on host biosynthetic pathways, metabolites and enzymes. Long-chain acyl-CoA synthetases (ACSLs) are key host-cell enzymes that convert fatty acids (FA) into acyl-CoA for use in metabolic pathways. Here, we show that the complete host ACSL family [ACSL1 and ACSL3-6] translocates into the Ct membrane-bound vacuole, termed inclusion, and remains associated with membranes of metabolically active forms of Ct throughout development. We discovered that three different

Immunoglobulin A nephropathy (IgAN), the most prevalent primary glomerular disease, is a major cause of end-stage renal disease. Ferroptosis, a novel cell death mode induced by iron-dependent phospholipid peroxidation, has been implicated in the progression of chronic kidney disease. Whereas, Acyl-CoA synthetase long-chain family member 4 (ACSL4) is a key enzyme in pro-ferroptotic lipid metabolism that plays a critical role in ferroptosis. In this study, the role of ACSL4-mediated ferroptosis in IgAN progression was investigated by identifying the cell subset ACSL4+ Loop of Henle cells (LOHs)

Inter-tumor and intra-tumor heterogeneity present significant challenges to achieving precise treatment in breast cancer (BC). To address this, we established a living biobank comprising 68 patient-derived organoids (PDOs) from 50 patients across various regions, accurately maintaining the histological, genomic, and transcriptomic characteristics of the original tumors. Paired profiling demonstrated high concordance in mutation burden, copy number alterations, and molecular subtypes, capturing both inter- and intra-tumor heterogeneity. Drug screening with four clinically used agents revealed p

Rabies, induced by rabies virus (RABV), still threaten global health all over the world, and no effective therapy is available for rabies currently. Recently, a series of natural plant components have been found to inhibit virus production. In this study, Z-Ligustilide, a natural component of Ligusticum chuanxiong Hort, was found to inhibit RABV replication. Initially, the concentration of cytotoxicity 50 % (CC50) of Z-Ligustilide in N2a and BSR cells were 429.9 μM and 335.5 μM, respectively, which both significantly restrict RABV production in a concentration-dependent manner. Moreover, Z-Lig

We present a family in which an initial clinical diagnosis of autosomal dominant pure hereditary spastic paraparesis (HSP) was made on the basis of a three generation pedigree in which both males and females presented with a spastic paraparesis. Subsequent biochemical and genetic analysis revealed that the family was in fact affected by the adrenomyeloneuropathy subtype of X-linked adrenoleukodystrophy. In the family described, both males and females were affected by a spastic paraparesis, and there was no male to male transmission, consistent with both autosomal dominant and X-linked inherita

Phytanic acid accumulates in excessive amounts in Refsum disease, a rare neurological disorder, due to a defect in its alpha-oxidation enzyme system in peroxisomes. The activation of phytanic acid to phytanoyl-CoA by phytanoyl-CoA ligase is a prerequisite for its alpha-oxidation. The studies described in this manuscript report that phytanoyl-CoA ligase in peroxisomes is an enzyme distinct from the previously reported acyl-CoA ligases.

Myocardial ischemia/reperfusion (I/R) injury remains a major clinical challenge, with ferroptosis emerging as a critical contributor to cardiomyocyte death. Notably, ring-finger protein 5 (RNF5), which functions as an E3 ubiquitin ligase that is recognized for its crucial involvement in the modulation of endoplasmic reticulum stress, unfolded protein responses, and inflammatory processes, has not been systematically investigated in the process of the onset and evolutionary trajectory of myocardial ischemia/reperfusion (MI/R) injury.Male 7-8 week-oldC57BL/6 mice were used to establish a myocard

Deregulated lipid metabolism contributes to leukemogenesis and the progression of acute myeloid leukemia (AML). By analyzing large-scale CRISPR-Cas9 screening data, we identified acyl-CoA synthetase long-chain family member 4 (ACSL4) as a selective vulnerability in lysine methyltransferase 2A-rearranged (KMT2Ar) AML. Functional validation using CRISPR interference and short hairpin RNA knockdown confirmed that ACSL4 loss impairs the growth of KMT2Ar but not non-KMT2Ar AML cells. ACSL4 knockdown reduced colony formation in cells derived from patients with KMT2Ar AML and murine MLL-AF9 cells and

Osteoporosis is driven by a pathological imbalance in bone remodeling, with osteoclast hyperactivation being a primary driver of bone loss. However, the intrinsic signaling nodes that govern osteoclast survival and function remain poorly understood. While loss-of-function mutations in Anoctamin 5 (Ano5) are associated with genetic skeletal disorders, its precise involvement in metabolic osteoporosis remains elusive. In this study, we utilized an Ano5-deficient ovariectomized (OVX) mouse model to investigate the role of Ano5 in bone homeostasis. We found that genetic depletion of Ano5 significa

Microglia-mediated neuroinflammation plays a crucial role in the progression of Parkinson’s disease (PD). Dysregulation of lipid droplet homeostasis is a significant factor affecting microglial inflammatory responses, but the mechanisms underlying lipid droplet imbalance in PD are currently unclear. Here, we report a subtype of microglia characterized by high expression of long-chain acyl-CoA synthetase 1 (ACSL1) through single-nucleus RNA sequencing analysis and machine learning algorithms, linking lipid metabolism to PD neuroinflammation. The results of multiple loss- and gain-of-function ex

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) is an increasingly prevalent condition and a major risk factor for chronic liver damage, potentially leading to steatohepatitis and HCC. It is already known that patients with MASLD show increased systemic and hepatic iron concentrations as well as perturbed lipid metabolism, suggesting the involvement of ferroptosis in the development and progression of MASLD. Consequently, inhibition of ferroptosis represents a potential therapeutic option for patients with MASLD.
METHODS: We investigated whether liver parenchymal

Ferroptosis, an iron-dependent form of programmed cell death driven by toxic lipid peroxide accumulation, plays a critical role in various diseases, making its modulation a promising therapeutic strategy. In this study, we identified defactinib, a specific inhibitor of FAK as a novel ferroptosis suppressors. We demonstrate that FAK/SRC-JNK signaling positively regulates ferroptosis by upregulating ACSL4, a critical mediator of ferroptosis. We reveal that a subset of JNK downstream transcription factors, including ATF2, NFATC1, NFATC3, and SMAD4, promote ferroptosis through direct binding to th

Chaperone-mediated autophagy (CMA) represents a critical lysosomal degradation pathway in the context of intervertebral disc degeneration (IVDD) associated with senescence. This study revealed a novel mechanism of CMA regulation involving targeted degradation of acyl-CoA synthetase long-chain family member 4 (ACSL4), which can delay nucleus pulposus cell (NPC) senescence and inhibit IVDD progression. Mechanistic investigations demonstrated that the acetyltransferase KAT2B can facilitate the acetylation of ACSL4 at lysine residues K500, K571, and K692. This post-translational modification serve

The prognosis of childhood medulloblastoma (MB) is often poor, and it usually requires aggressive therapy that adversely affects quality of life. microRNA-211 (miR-211) was previously identified as an important regulator of cells that descend from neural cells. Since medulloblastomas primarily affect cells with similar ontogeny, we investigated the role and mechanism of miR-211 in MB. Here we showed that miR-211 expression was highly downregulated in cell lines, PDXs, and clinical samples of different MB subgroups (SHH, Group 3, and Group 4) compared to normal cerebellum. miR-211 gene was ecto

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) and its progressive form, metabolic dysfunction-associated steatohepatitis, are highly prevalent and lack effective pharmacotherapies. Hepatocellular lipotoxicity-driven by the accumulation of saturated fatty acids (eg, palmitate)-promotes disease progression; however, the determinants of hepatocyte susceptibility remain incompletely defined.
METHODS: We performed a genome-wide CRISPR-Cas9 loss-of-function screening to identify the regulators of palmitate-induced lipotoxicity. The top candidates were validated using

Ferroptosis is a novel form of cell death characterized by iron-dependent oxidative damage, lipid peroxidation, and the accumulation of reactive oxygen species. There exists a close correlation between ferroptosis and gastric cancer (GC). Recent research has shown that many ferroptosis-related long non-coding RNAs (lncRNAs) affect the occurrence and progression of GC. However, the exact mechanism has not been elucidated. This study showed that a novel lncRNA LINC02266 was upregulated in GC cells and was a key lncRNA associated with GC prognosis and progression. Functional analysis indicated th

Ochratoxin A (OTA), a secondary fungal metabolite known for its nephrotoxic effects, is prevalent in various feeds and food items. Our recent study suggests that OTA-induced nephrotoxicity is linked to the Sigma-1 receptor (Sig-1R)-mediated mitochondrial pathway apoptosis in human proximal tubule epithelial-originated kidney-2 (HK-2) cells. However, the contribution of Sig-1R to OTA-induced nephrotoxicity involving other forms of regulated cell death, such as ferroptosis, remains unexplored. In this investigation, cell viability, malondialdehyde (MDA) levels, glutathione (GSH) levels, and prot
