Metagenomic profiling of viral and microbial communities from the pox lesions of lumpy skin disease virus and sheeppox virus-infected hosts.

INTRODUCTION: It has been recognized that capripoxvirus infections have a strong cutaneous tropism with the manifestation of skin lesions in the form of nodules and scabs in the respective hosts, followed by necrosis and sloughing off. Considering that the skin microbiota is a complex community of commensal bacteria, fungi and viruses that are influenced by infections leading to pathological states, there is no evidence on how the skin microbiome is affected during capripoxvirus pathogenesis. METHODS: In this study, shotgun metagenomic sequencing was used to investigate the microbiome in pox

Influence of the lumpy skin disease virus (LSDV) superoxide dismutase homologue on host transcriptional activity, apoptosis and histopathology.

Lumpy skin disease virus (LSDV), a Capripoxvirus, is of economic importance in the cattle industry and is controlled by vaccination. A comparison was made of the host response to the two LSDV vaccines Neethling and Herbivac LS, with reference to the well-studied Orthopoxvirus, modified vaccinia Ankara (MVA), in a mouse model. Because the vaccines differ at the superoxide dismutase homologue (SOD) gene locus, recombinant SOD knock-out and knock-in nLSDV vaccines were constructed and all four vaccines were tested for the induction and inhibition of apoptosis. The SOD homologue was associated bot

Potential roles of Aedes aegypti (Diptera: Culicidae) and Aedes albopictus (Diptera: Culicidae) in lumpy skin disease virus transmission.

Lumpy skin disease (LSD) is an emerging viral disease affecting cattle and water buffalo, leading to significant socioeconomic impacts. In Southeast Asia, the first LSD outbreak was reported in Vietnam in 2020, and the disease has continued to expand to neighboring countries, including Thailand. Recently, LSD has been reported in wildlife such as yaks and giraffes. Despite being recognized for nearly a century, the mechanisms of lumpy skin disease virus (LSDV) transmission remain poorly understood. This study aimed to investigate the potential role of Aedes albopictus in LSDV transmission. Acc

Influence of Major Histocompatibility Complex (MHC) Diversity on Immune Modulation, Pathogenesis, and Control of Lumpy Skin Disease Virus.

INTRODUCTION: Lumpy Skin Disease Virus (LSDV), a member of the genus Capripoxvirus within the family Poxviridae, is an economically important transboundary viral pathogen affecting cattle and water buffalo. The disease causes severe production losses through decreased milk yield, infertility, hide damage, reduced growth performance, and occasional mortality. The rapid geographic spread of LSDV, together with its vectorborne transmission and emerging recombinant strains, has intensified the need for improved understanding of viral pathogenesis, host immune responses, and effective prevention st

Comparative evaluation of recombinant A4L, A27L, and A36R proteins for ELISA-based serodiagnosis of lumpy skin disease virus.

Lumpy skin disease (LSD) is a transboundary poxviral disease of cattle and buffalo that has been economically significant in various parts of Africa, the Middle East, Asia and some parts of Europe and therefore requires the development of reliable, high throughput diagnostic tools to support effective surveillance and control. Conventional serological assays are labor intensive, time consuming and involve handling of live virus, which makes them unsuitable for large-scale surveillance. Recombinant antigen based ELISAs offer a safer, scalable and high-throughput alternative; however, identifyin

Genetic Evolutionary Analysis of Lumpy Skin Disease Virus Strain Under Immune Pressure Exerted by Heterologous Goat Poxvirus Vaccines.

Lumpy skin disease (LSD) is an important infectious disease that threatens the global cattle industry. Recently, LSD has occurred frequently in Asia. The attenuated goat poxvirus (GTPV) vaccine is widely used to prevent LSD in cattle in China; however, sporadic cases of LSD still occur in immunized cattle. This study aimed to investigate the genetic evolution of isolated LSD virus (LSDV) strain under GTPV heterologous vaccine immune pressure. We isolated a new strain of LSDV, named LSDV/China/SX/2023, from a cattle herd immunized with a GTPV-attenuated vaccine in Shaanxi Province, Northwest Ch

Subclinical infection caused by a recombinant vaccine-like strain poses high risks of lumpy skin disease virus transmission.

Lumpy skin disease (LSD) is a transboundary viral infection, affecting cattle with characteristic manifestations involving multiple body systems. A distinctive characteristic of lumpy skin disease is the subclinical disease manifestation wherein animals have viremia and shed the virus through nasal and ocular discharges, while exhibiting no nodules but enlarged lymph nodes that are easily oversighted by inexperienced vets. Further research on the role of subclinically ill animals in the transmission of LSD virus (LSDV) can contribute to the development of more effective tools to control the di

Evaluation of haematological, serum biochemical and oxidative stress parameters in cattle naturally infected with lumpy skin disease virus.

Lumpy skin disease (LSD) is a vector-borne viral transboundary disease of cattle caused by the LSD virus (LSDV). Despite investigations on clinical and outbreak features of LSDV, information on disease pathogenesis and alternative changes in blood parameters are scarce. Keeping this in view, the current study was designed to determine haematological, serum biochemical, and oxidative stress parameters in naturally infected cattle with LSDV during the recent surge of outbreaks in Punjab, Pakistan. A total of 35 blood samples was collected from polymerase chain reaction-confirmed LSDV-infected ca

Inhibition of bovine and ovine capripoxviruses (Lumpy skin disease virus and Sheeppox virus) by ivermectin occurs at different stages of propagation in vitro.

Capripoxvirus diseases are listed as reportable diseases by World Organization for Animal Health (OIE). Lumpy skin disease virus (LSDV) and sheeppox virus (SPPV), which can only be distinguished by molecular analysis, cause moderately, severe, or sometimes fatal infections in cattle and sheep. Even though vaccines are the most effective way to control the infection, their effectiveness may decrease in some cases. Therefore, it is significant to explore antiviral drugs against these diseases along with the vaccine. This study aimed to investigate the antiviral efficiency of ivermectin (IVM) at

Development of a Duplex TaqMan-MGB qPCR Assay for Differential Detection of Chinese Epidemic Lumpy Skin Disease Virus Strains and Goatpox Virus.

Lumpy skin disease (LSD) is a transboundary viral disease of cattle, including Asian water buffalo and yaks, and certain wild ruminants (e.g., African buffalo, giraffe, wildebeest, eland, and Arabian oryx). It is caused by lumpy skin disease virus (LSDV), a member of the genus Capripoxvirus (family Poxviridae) together with goatpox virus (GTPV) and sheeppox virus (SPPV). High nucleotide identity and serological cross-reactivity among these viruses hinder differential diagnosis. The aim of this study was to develop a Duplex TaqMan-MGB qPCR Assay for Differential Detection of Chinese Epidemic Lu

Molecular evidence of prolonged shedding of lumpy skin disease virus (LSDV) in the semen of naturally infected cattle bulls.

Shedding of lumpy skin disease virus (LSDV) in the semen of experimentally infected cattle bulls has been proven, but research on LSDV shedding in semen following natural infection is scarce. This study aimed to assess the incidence and duration of LSDV shedding in the semen of bulls following natural infection. Testing of 78 cattle bulls from four LSD outbreak-affected breeding farms in India during 2022-2023 confirmed LSDV infection through LSDV real-time quantitative PCR and antibody ELISA. Subsequent real-time quantitative PCR analysis of semen samples identified LSDV genome in semen from

Evaluation of an interferon-gamma release assay for early detection of lumpy skin disease virus infection and vaccination in cattle.

Lumpy skin disease virus (LSDV) causes a nodular dermatitis in cattle and has high economic consequences in affected areas. Detection of LSDV exposure mostly relies on the humoral immune response, while the cell-mediated immune (CMI) response, an important hallmark of the immune reaction to LSDV, is neglected. We collected samples during 3 weeks post-vaccination of cattle with a Neethling-based live attenuated vaccine (LAV) and during 4 weeks post-LSDV infection under experimental conditions to i) investigate the development of the CMI response, ii) optimize an interferon-gamma release assay (

A Genome-Wide Screening of Novel Immunogenic TrLSDV103 Protein of Lumpy Skin Disease Virus and Its Application for DIVA.

Lumpy skin disease (LSD) is an infectious cattle disease caused by the lumpy skin disease virus (LSDV), posing a serious threat to the livestock industry. This study aimed to identify specific immunogenic targets for differential diagnosis and potential vaccine development. Using a phage display library covering the entire LSDV proteome, we screened sera from naturally LSDV-infected cattle and those vaccinated with the live attenuated goatpox virus vaccine (GTPV AV41) to identify differential antibody-binding viral peptides, leading to the identification of peptides within the LSDV103 protein.

Lumpy Skin Disease Virus (LSDV): evaluating global impact, Ethiopia's response strategies, and the promise of gamma-ray inactivated vaccines.

Lumpy Skin Disease (LSD), caused by the Lumpy Skin Disease Virus (LSDV), constitutes an escalating transboundary threat to cattle production, with disproportionately high economic impacts in Ethiopia. This review synthesizes recent global and national data to assess the socio-economic burden of LSDV, evaluate Ethiopia’s current control strategies, and examine the potential role of gamma-ray inactivated vaccines as supplementary candidates to existing immunization protocols. Globally, LSDV is associated with annual production losses of approximately USD 1.46 billion and trade disruptions exceed

Lumpy skin disease virus ORF142 suppresses the cGAS/STING-mediated IFN-I pathway through NBR1-mediated STING autophagic degradation.

Lumpy skin disease (LSD), caused by the Lumpy skin disease virus (LSDV), poses a significant threat to the global cattle farming industry. Viral proteins evolved the ability to escape host immune responses. Here, we demonstrate that LSDV infection inhibits interferon-β (IFN-β) production in Madin-Darby bovine kidney (MDBK) cells, even in the presence of Sendai virus (SEV) inducers. Through further investigation, we identify multiple genes at both ends of the LSDV genome that strongly inhibit IFN-β or NF-κB activation of the promoter. Notably, ORF142 selectively inhibits IFN-β promoter activity

Outbreak Investigation, Isolation, and Molecular Characterization of Lumpy Skin Disease Virus in Cattle from North West Oromia Region, Ethiopia.

Lumpy skin disease (LSD) is an economically significant viral disease because of its high morbidity and high production loss. Vaccination of cattle using LSD vaccines is a more effective disease preventive and control strategy in endemic countries such as Ethiopia. Despite high vaccination coverage, there is an increasing number of field reports of the disease outbreaks. Thus, an observational study was designed to investigate disease, characterize the disease-causing agent, and isolate the virus from a local isolate for future vaccine development. Wera Jarso and Amuru districts in North West
