Monocyte accessory cell function in patients infected with the human immunodeficiency virus.

Previous studies suggested that peripheral blood monocytes (Mo) from HIV-infected patients were poor accessory cells (AC), although most of these studies were limited by using autologous T cells as responders. Using allogeneic T cells from uninfected volunteers as responders, the current studies demonstrate that Mo from infected individuals were comparable to Mo from uninfected volunteers as AC in Con A and pokeweed mitogen-stimulated lymphocyte proliferation assays, but were inferior to normal Mo in stimulating a mixed leukocyte reaction. This deficiency was not explained by HIV Mo-induced su

Membrane and transformation characteristics of lymphocytes isolated from the synovial membrane and paired peripheral blood of patients with rheumatoid arthritis.

Membrane and transformation characteristics of lymphocytes isolated from the synovial membrane and from paired peripheral blood samples, obtained from patients with classical rheumatoid arthritis, were studied. Synovial tissue lymphocytes were isolated by a new technique. Two suspensions of peripheral blood lymphocytes were studied: one isolated by Ficoll-Isopaque density gradient centrifugation, the other enriched in T cells by an additional step of 1 hour nylon wool column filtration. All suspensions were characterised by the percentages of mononuclear phagocytic cells, and T and B lymphocyt

HLA-D region products are expressed in endothelial cells.

The mixed lymphocyte endothelial cell culture was studied by the primed lymphocyte typing (PLT) technique. By comparing the HLA-D/DR specificity of the secondary response when using either peripheral blood mononuclear cells (PBM) or endothelial cells from umbilical cords for priming or restimulation of lymphocytes, it was found that PBM from newborn would induce a clear-cut specificity for HLA-D/DR when used for priming a well a for restimulation. No HLA-D/DR specificity was seen, however, when endothelial cells were used for restimulation of lymphocytes primed to HLA-D/DR on PBM. On the other

Selective defects in T cell function in ataxia-telangiectasia.

We have studied three patients with ataxia-telangiectasia (AT) and found two of them to have a normal mixed leucocyte culture stimulating and responding ability. However, all three patients and one parent had defective cell-mediated lympholysis (CML), even in the face of a potent proliferative response to allogeneic leucocytes. None of these patients showed significant proliferative responses to common microbial antigens (tetanus toxoid, Candida albicans, purified protein derivative (PPD), diphtheria toxoid, influenza). Our studies indicate tha the T cell defect in AT preferentially affects ce

Specific elimination of alloreactive T cells by an anti-interleukin-2 receptor B chain-specific immunotoxin.

Graft-versus-host disease and graft rejection remain the two principal causes of morbidity and mortality after major-histocompatibility-complex-mismatched bone marrow transplantation. Human and animal models suggest that both CD4+ and CD8+ T cell subsets present in the donor inoculum are responsible for their initiation. Since the human mixed lymphocyte culture (MLC) and the HLA-restricted cytotoxicity may reflect cellular interactions occurring during GVHD and graft rejection, inhibitions of these responses may represent useful approaches for screening functional T cell depletion in experimen

Activated T cells in HTLV-I-associated myelopathy: autologous mixed lymphocyte reaction.

We examined the autologous mixed lymphocyte reaction in human T-cell lymphotropic virus type I-associated myelopathy to elucidate the known increase in spontaneous proliferation of peripheral blood lymphocytes. Proliferative responses in the autologous mixed lymphocyte reaction test were significantly increased (34,205 cpm) in cells from 9 patients with the myelopathy, as compared with findings (18,695 cpm) in the control subjects. When non-T-cell fractions were depleted from autologous mixed lymphocyte reaction cultures, proliferative responses were completely suppressed in the control subjec

Enhancement of the autologous mixed lymphocyte reaction in patients with Chagas' heart disease.

Autologous mixed lymphocyte reactions (AMLR) were studied in 18 individuals chronically infected with Trypanosoma cruzi. These individuals were further classified into three clinical groups: the asymptomatic indeterminate form (n = 5); the mega disease form (n = 5); and the cardiomyopathy form (n = 8). While patients with mega disease showed a normal proliferative response when compared with normal controls, the indeterminate group showed a lowered response in sharp contrast with the heart disease group, which presented a very high proliferative response to autologous non-T cells. These abnorm

The MLC response of patients with infectious mononucleosis to Epstein-Barr virus-transformed cells.

Lymphocytes of 6 patients with infectious mononucleosis were found to be capable of responding in mixed lymphocyte culture (MLC) to Epstein-Barr virus (EBV)--transformed lymphoblastoid cell lines and Raji cells, whereas their response to PWM-induced blasts and non-T cells was significantly depressed. This means that despite the suppression of cellular reactivity, observed in infectious mononucleosis, the specific response to EBV-infected cells remains unaffected.

Factors contributing to impairment of the mixed lymphocyte reaction in leprosy.

Whereas the mixed lymphocyte reaction was essentially normal in inactive lepromatous leprosy and tuberculoid leprosy, it was severely impaired in active lepromatous leprosy. The impairment was found to be contributed by certain unknown factors in their plasma and subnormal reactivity of their T lymphocytes. The plasma derived from active lepromatous leprosy patients depressed the reaction of normal cells and normal plasma enhanced the reaction of active lepromatous lymphocytes. The cellular factor was studied by using a one-way reaction in which one of the two lymphocyte preparations was inact

Cellular immunity in patients with the Wiskott-Aldrich syndrome before and after administration of transfer factor: a follow-up study.

Four patients with Wiskott-Aldrich syndrome were treated with transfer factor (TF), in an attempt to improve their clinical condition. Before and during treatment, cellular reactivity was followed in vivo (delayed type skin reactivity) and in vitro (lymphocyte transformation). In all patients positive skin reactions were occasionally observed during TF treatment. The lymphocyte reactivity in vitro to phytohaemagglutinin (PHA) was slightly diminished, the responding capacity of the mixed lymphocyte culture (MLC) severely impaired and the response to bacterial, viral and fungal antigens absent b

The cells involved in cell-mediated and transplantation immunity in the normal outbred rabbit. XIII. The identity of the responder cells and the role of phagocytic cells in the mixed leucocyte culture reaction.

The ability of rabbit spleen cells depleted of thymus, bone marrow or appendix-derived cells to respond in the one-way mixed leucocyte reaction (MLR) has been investigated. The specific subpopulations of lymphoid cells were eliminated by lysis of these cells in the presence of specific antisera and complement. Phagocytic cells (monocytes and heterophils) were removed with a strong magnet after incubation with carbonyl iron particles. The results indicate that the MLR-responding cells are thymus-derived and that neither bone marrow-derived cells nor appendix-derived cells are essential for the

Induction of an immature T-cell phenotype in malignant helper T cells by cocultivation with epidermal cell cultures.

The possible inductive effect of epidermal cells on T-cell maturation has been examined employing an in vitro cocultivation technique. Mononuclear cells from 6 patients with cutaneous T-cell lymphoma (CTCL) and from 12 healthy volunteers were studied. In the 6 CTCL patients, all showed an expansion of the helper T-cell subpopulation and in one patient with leukemic CTCL, there was almost complete replacement of peripheral blood mononuclear cells by malignant cells with a helper T-cell phenotype. Epidermal cells derived from normal human skin were cultured to confluent monolayers, and were cocu

Variation in the CD4/non-T cell autologous mixed lymphocyte reaction with disease activity and rheumatoid factor titer in patients with juvenile rheumatoid arthritis.

Variation in the CD4/non-T autologous mixed lymphocyte reaction (AMLR) with disease activity in juvenile rheumatoid arthritis (JRA) was evaluated by repeated study of 5 patients with rheumatoid factor (RF) negative and 8 patients with seropositive polyarticular onset. Results of repeated studies of adult controls fell within the limits of 1 SD from the previously established normal mean. In contrast, greater variation occurred in results of patients with JRA. Patients with RF negative JRA showed no correlation with disease activity but in seropositive patients stimulation indices in the AMLR i

Human mesenchymal stem cells suppress induction of cytotoxic response to alloantigens.

Mesenchymal stem cells (MSC) fail to induce allogeneic responses in mixed lymphocyte reaction assays. Because MSC express HLA class I molecules, here we investigated whether they could be recognized as allogeneic targets by cytolytic T lymphocytes (CTL). With this aim, CTL precursor (CTLp) frequencies were measured following stimulation of T cells with either allogeneic mononuclear cells (MNC) or MSC originated from the same human bone marrow donor. Lysis of MSC was measured at day 10 of culture in standard chromium release assays. In addition, allogeneic PHA blast T cells or B-EBV lymphoblast

Lymphocyte aberrations in ankylosing spondylitis.

Because the etiology of IgA and IgG hyperimmunoglobulinemia in ankylosing spondylitis (AS) remains unknown, the cellular immune system has been further studied by measuring the lymphocyte and its subpopulations in peripheral bloods of 50 definite AS patients and 40 normal controls. The system's functions have also been assessed in terms of proliferative responses to phytomitogens (Con A, PHA and PWM) and autologous mixed lymphocyte reaction (AMLR). The following results were obtained: The number of lymphocytes, total T, active T, OKIa1, OKT3, OKT4 and OKT8 cells in peripheral bloods of AS pati

Defect in the generation of cytotoxic T cells in lepromatous leprosy.

Cytotoxic T cells are consistently produced in normal individuals after in vitro stimulation by a pool of mitomycin-treated normal lymphocytes. Patients suffering from lepromatous leprosy (LL), presenting with large amounts of Mycobacterium leprae and without a history of erythema nodosum leprosum (ENL) are unable to generate such cytotoxic T cells, while lepromatous patients with ENL which, in the present study were all deprived of M. leprae, react normally.
