Mitochondrial malate dehydrogenase of bovine cerebrum. Characterization and mechanisms of inhibition by silver ions.

Attempts were made to characterize mitochondrial malate dehydrogenase [L-malate: NAD+ oxidoreductase, EC 1.1.1.37] (M-MDH) purified from bovine cerebrum and to elucidate the mechanisms responsible for inhibition of the enzymic activity by Ag+. The molecular weights of the native enzyme and its subunits were 54,000-55,000 and 30,000-32,000, respectively. In general, the physiochemical and catalytic properties of bovine cerebral M-MDH was not very different from those of other corresponding mammalian enzymes. Incubation of the enzyme with Ag+ caused the loss of equivalent amounts of sulfhydryls

Exercise-Induced Rhabdomyolysis With Supplements (Creatine, β-Alanine, Citrulline Malate, and β-Ecdysterone).

Dietary supplements such as creatine, β-alanine, citrulline malate (CM), phosphatidic acid (PA), and β-ecdysterone are widely used to enhance exercise performance and muscle hypertrophy. Although generally considered safe, their combined use with high-intensity training may increase the risk of exertional rhabdomyolysis and secondary liver or kidney injury. We report the case of a 23-year-old physically active man admitted with severe myalgia, muscle weakness, fever, and dark brown urine following a prolonged (>4 hours) resistance training session. The patient regularly used multiple dietary s

OX-LDL causes vascular endothelial injury by interfering with the CREB1-MDH1B-malate axis.

BACKGROUND: Atherosclerosis (AS) is one of the most prevalent cardiovascular disorders, with endothelial dysfunction recognized as a central initiating event in its pathogenesis. Oxidized low-density lipoprotein (ox-LDL) is a key pathogenic factor that contributes to vascular injury. While the pro-atherogenic and oxidative effects of ox-LDL have been well described, the mechanisms by which it influences cellular energy metabolism remain largely unclear. OBJECTIVE: This study aims to elucidate how ox-LDL impairs endothelial cell function through repression of the transcription factor CREB1 and

Guard cell starch and malate metabolism facilitate stomatal opening in response to low CO2.

Guard cells adjust turgor to regulate stomatal aperture, integrating ion transport with metabolic adjustments. The effect of reduced CO2 on stomatal movement, expected under enhanced photosynthesis, remains largely unexplored. We exposed Arabidopsis thaliana plants to low CO2 before light onset and analyzed guard cell starch turnover, malate (Mal) accumulation, and stomatal conductance in wild-type and mutant lines. Low CO2 triggered rapid starch degradation and increased Mal levels in guard cells, accelerating stomatal opening. Mutants defective in starch degradation or hexose uptake showed i

Innovate and empower: the malate dehydrogenase course-based undergraduate research experiences and community of practice.

College science programs exhibit high rates of student attrition, especially among Students of Color, women, members of the LGBTQ+ community, and those with disabilities. Many of the reasons students choose to leave or feel pushed out of science can be mitigated through participation in faculty-mentored research. However, faculty resources are limited, and not every student has access to faculty mentoring due to systemic or structural barriers. By bringing authentic scientific research into the classroom context, course-based undergraduate research experiences (CUREs) expand the number of stud

Molecular Characteristics of the Malate Dehydrogenase (MDH) Gene Family in Spirometra mansoni (Cestoda: Diphyllobothriidea).

The plerocercoid larva of Spirometra mansoni can cause a parasitic zoonosis-sparganosis. Malate dehydrogenase (MDH) plays a very important role in the life activities of parasites. However, little is known about the MDH family in S. mansoni. We identified eight new MDH members in S. mansoni in this study. Clustering analysis divided SmMDHs into two groups and revealed patterns similar to the conserved motif organization. RT-qPCR suggested that five MDHs were highly expressed in the mature proglottid and that three MDHs were highly expressed in the gravid proglottid. Phylogenetic analysis revea

The malate-aspartate shuttle (Borst cycle): How it started and developed into a major metabolic pathway.

This article presents a personal and critical review of the history of the malate-aspartate shuttle (MAS), starting in 1962 and ending in 2020. The MAS was initially proposed as a route for the oxidation of cytosolic NADH by the mitochondria in Ehrlich ascites cell tumor lacking other routes, and to explain the need for a mitochondrial aspartate aminotransferase (glutamate oxaloacetate transaminase 2 [GOT2]). The MAS was soon adopted in the field as a major pathway for NADH oxidation in mammalian tissues, such as liver and heart, even though the energetics of the MAS remained a mystery. Only i

Bi-allelic GOT2 Mutations Cause a Treatable Malate-Aspartate Shuttle-Related Encephalopathy.

Early-infantile encephalopathies with epilepsy are devastating conditions mandating an accurate diagnosis to guide proper management. Whole-exome sequencing was used to investigate the disease etiology in four children from independent families with intellectual disability and epilepsy, revealing bi-allelic GOT2 mutations. In-depth metabolic studies in individual 1 showed low plasma serine, hypercitrullinemia, hyperlactatemia, and hyperammonemia. The epilepsy was serine and pyridoxine responsive. Functional consequences of observed mutations were tested by measuring enzyme activity and by cell

Glyoxylate detoxification is an essential function of malate synthase required for carbon assimilation in Mycobacterium tuberculosis.

The glyoxylate shunt is a metabolic pathway of bacteria, fungi, and plants used to assimilate even-chain fatty acids (FAs) and has been implicated in persistence of Mycobacterium tuberculosis (Mtb). Recent work, however, showed that the first enzyme of the glyoxylate shunt, isocitrate lyase (ICL), may mediate survival of Mtb during the acute and chronic phases of infection in mice through physiologic functions apart from fatty acid metabolism. Here, we report that malate synthase (MS), the second enzyme of the glyoxylate shunt, is essential for in vitro growth and survival of Mtb on even-chain

Molecular cloning, purification and immunogenicity of recombinant Brucella abortus 544 malate dehydrogenase protein.

The Brucella mdh gene was successfully cloned and expressed in E. coli. The purified recombinant malate dehydrogenase protein (rMDH) was reactive to Brucella-positive bovine serum in the early stage, but not reactive in the middle or late stage, and was reactive to Brucella-positive mouse serum in the late stage, but not in the early or middle stage of infection. In addition, rMDH did not react with Brucella-negative bovine or mouse sera. These results suggest that rMDH has the potential for use as a specific antigen in serological diagnosis for early detection of bovine brucellosis.

Hippocampus protein profiling reveals aberration of malate dehydrogenase in chlorpromazine/clozapine treated rats.

Chlorpromazine and clozapine are antipsychotic agents used to relieve symptoms of early-diagnosed schizophrenia. In this study, we used proteomics technology to screen the protein aberration in Sprague-Dawley rats treated with these two antipsychotic agents. Our goal was to identify the effects of antipsychotics on hippocampal proteins in rats. Protein expression profiles were compared in each experimental group using two-dimensional gel electrophoresis and identified using matrix-assisted laser desorption/ionisation time of flight mass spectrometry. Malate dehydrogenase, peroxiredoxin 3, vacu

Effect of partial obstruction of the rabbit urinary bladder on malate dehydrogenase and citrate synthase activity.

Previous studies have demonstrated that partial outlet obstruction in rabbits induced a significant decrease in oxidative metabolism in urinary bladder smooth muscle. The current experiments were designed to determine whether the decreased oxidative metabolism of obstructed bladder tissue is associated with alterations in the activities of specific mitochondrial enzymes. The activities of two important enzymes in the tricarboxylic acid cycle, malate dehydrogenase and citrate synthase, were measured in samples of bladder body and base from normal bladders and in bladders from rabbits in which p

Inhibition by (aminooxy)acetate of the malate-aspartate cycle in the isolated working guinea pig heart.

The quantitative importance of the malate-aspartate cycle and the sn-glycerol 3-phosphate cycle, with respect to the flux of cytosolic reducing equivalents across the mitochondrial membrane, was studied in isolated perfused guinea pig hearts. The heart preparations performed pressure-volume work and metabolized glucose, lactate, pyruvate or 3-hydroxybutyrate. With glucose or lactate as the substrate, (aminooxy)acetate, an inhibitor of the malate-aspartate cycle, caused left ventricular failure, manifested by reduced aortic pressure and cardiac output, in association with a decrease in myocardi

Identification and Bioinformatics Analysis of Malate Dehydrogenase and Pyruvate Kinase as Novel Allergens in Cicada Pupae.

Cicada pupae are a traditional and promising food source, and it is crucial to evaluate the associated allergenic risks. In this study, 13 potential allergens were identified based on proteomics combined with immunoinformatics tools in cicada pupae. These allergens were produced as recombinant proteins, and their allergenicity was investigated. Malate dehydrogenase (MDH) and pyruvate kinase (PK) were finally identified as new allergens via immunoblot experiments. The secondary structures of the two new allergens predominantly consist of α-helices and β-sheets. Five IgE-binding epitopes of MDH

WSSV-induced reversal of the malate-aspartate shuttle facilitates viral replication in shrimp hemocytes.

BACKGROUND: White spot syndrome virus (WSSV), one of the most devastating pathogens in global shrimp aquaculture, has been shown to hijack and reprogram host metabolic pathways to support its replication. Among the various host metabolic circuits, the malate-aspartate shuttle (MAS) is a key redox-balancing mechanism that facilitates the translocation of cytosolic NADH into mitochondria, thereby sustaining glycolysis and mitochondrial function. To date, however, the involvement of MAS in WSSV pathogenesis has not been documented. METHODS: In this study, we investigated the role of the MAS path
