A constraint on understanding the pathogenesis of malignant catarrhal fever (MCF) is the limited number of tools to localize infected cells. The amount of detectable virus, visualized in the past either by immunohistochemistry or in situ hybridization (ISH), has been modest in fixed or frozen tissues. This complicates our understanding of the widespread lymphoid proliferation, epithelial necrosis/apoptosis, and arteritis-phlebitis that characterize MCF. In this work, we developed a probe-based in situ hybridization assay targeting 2 ovine herpesvirus 2 (OvHV-2) genes, as well as their respecti

An outbreak of malignant catarrhal fever (MCF) among bison sold at an auction market was studied for an 18-month period. Forty-five of 163 bison submitted for sale from 8 different bison farms died on 7 other destination farms. The outbreak began on day 50 after the sale, peaked between days 60 and 70, and ended on day 220. Twenty-one dead bison were confirmed to be MCF cases by clinical histories, pathology, and detection of ovine herpesvirus-2 DNA in their tissues with polymerase chain reaction assays. Twenty-four dead bison were classified as suspect MCF cases from clinical histories. No ca

Malignant catarrhal fever-like clinical disease was diagnosed in a free-ranging bighorn sheep ( Ovis canadensis ) from Alberta, Canada, in June 2015. Antemortem and gross pathology findings included muscle atrophy, marked weight loss, and bilaterally symmetric alopecia with hyperpigmentation and crusting over the face, medial surfaces of the pinnae, dorsal trunk, distal limbs, perineal area, and tail. Histologically, the skin lesions were characterized by granulomatous mural folliculitis with numerous multinucleated giant cells and fewer lymphocytes and eosinophils consistent with previous rep

We wished to determine the effect of of CpG ODN adjuvant on the magnitude and duration of protective immunity against alcelaphine herpesvirus-1 (AlHV-1) malignant catarrhal fever (MCF), a fatal lymphoproliferative disease of cattle. Immunity was associated with a mucosal barrier of virus-neutralising antibody. The results showed that CpG ODN included either with emulsigen adjuvant and attenuated AlHV-1 (atAlHV-1) or alone with atAlHV-1 did not affect the overall protection from clinical disease or duration of immunity achieved using emulsigen and atAlHV-1. This is in contrast to other similar

A 10-y-old Watusi ( Bos taurus africanus) steer housed at a drive-through game park in Winston, Oregon developed severe clinical illness including fever, marked nasal discharge, injected scleral and conjunctival membranes, plus oral hemorrhages and erosions. The animal responded poorly to supportive treatment and was euthanized. Additional gross findings at postmortem examination included papules and erosive lesions on the tongue, hemorrhagic large intestine, and multifocal cardiac hemorrhages. Histopathologic findings included multifocal lymphoplasmacytic vasculitis plus fibrin exudation in h

Malignant catarrhal fever (MCF) is a severe, usually fatal, acute systemic disease syndrome of certain domestic and wild ruminants caused by members of the family Gammaherpesvirinae. Two distinct but closely related viruses cause clinically indistinguishable syndromes: one that is indigenous to the widebeest and the other that apparently is indigenous to domestic sheep. Neither the pathogenesis nor the epidemiology of sheep-associated MCF (SA-MCF) is understood, primarily because of a lack of adequate detection methods for the etiologic agent or antibody against it. No acceptably documented is

Malignant catarrhal fever (MCF) is a viral infectious disease caused by specific members of the Macavirus genus that are referred to as the MCF virus (MCFV) complex group. This study determined the prevalence of MCFV-associated infections in cattle within the mesoregions of the state of Paraná, Southern Brazil, by analyzing the histopathologic patterns of renal lesions in association with positive immunoreactivity to intralesional antigens of MCFV. Intracytoplasmic MCFV antigens were identified in 41.7% (48/115) of the kidneys of cattle evaluated. Lymphocytic interstitial nephritis, vascular d

Malignant catarrhal fever (MCF) is a severe lymphoproliferative disease of domestic and wild ruminants caused by macaviruses, mainly ovine gammaherpesvirus 2 (OvHV-2) and alcelaphine gammaherpesvirus 1 (AlHV-1). Although relevant in sub-Saharan Africa, molecular epidemiological data remain limited, particularly in Namibia. This study investigated the occurrence and diversity of MCF viruses in Namibian livestock and contextualised local strains globally.Fifteen samples from cattle and sheep suspected of MCF were collected across five Namibian regions in 2023. Viral DNA was screened by duplex ne

BACKGROUND: Malignant catarrhal fever (MCF) is a sporadic infectious disease of domesticated and non-domesticated ruminants including: cattle, sheep, bison, cervids, antelope and buffaloes, and is caused by gammaherpesviruses. Between March and May 2023, several cases of sudden death were observed in endangered species of dorcas gazelle (Gazella dorcas osiris) in zoological garden in southeastern Poland. Given the rapid course of disease, MCF was suspected. During the epizootic investigation, other ruminants that could be a source of infection and routes of spread were examined, also taking in

Gammaherpesviruses (γHVs) include viruses that can induce lymphoproliferative diseases and tumors. These viruses can persist in the long term in the absence of any pathological manifestation in their natural host. Alcelaphine gammaherpesvirus 1 (AlHV-1) belongs to the genus Macavirus and asymptomatically infects its natural host, the wildebeest (Connochaetes spp.). However, when transmitted to several susceptible species belonging to the order Artiodactyla, AlHV-1 is responsible for the induction of a lethal lymphoproliferative disease, named wildebeest-derived malignant catarrhal fever (WD-MC

American bison (Bison bison) are particularly susceptible to developing fatal sheep-associated malignant catarrhal fever (SA-MCF) caused by ovine herpesvirus-2 (OvHV-2), a γ-herpesvirus in the Macavirus genus. This generally fatal disease is characterized by lymphoproliferation, vasculitis, and mucosal ulceration in American bison, domestic cattle (Bos taurus), and other clinically susceptible species which are considered non-adapted, dead-end hosts. The pathogenesis and cellular tropism of OvHV-2 infection have not been fully defined. An earlier study detected OvHV-2 open reading frame 25 (OR

Six members of the malignant catarrhal fever (MCF) virus group of ruminant rhadinoviruses have been identified to date. Four of these viruses are clearly associated with clinical disease: alcelaphine herpesvirus 1 (AlHV-1) carried by wildebeest (Connochaetes spp.); ovine herpesvirus 2 (OvHV-2), ubiquitous in domestic sheep; caprine herpesvirus 2 (CpHV-2), endemic in domestic goats; and the virus of unknown origin found causing classic MCF in white-tailed deer (Odocoileus virginianus; MCFV-WTD). Malignant catarrhal fever (MCF) is a highly fatal disease of cattle and other cloven-hoofed animals, most commonly caused by ovine herpesvirus 2 (OvHV-2), a member of the subfamily Gammaherpesvirinae. Despite nearly four decades since its identification, coding-complete genomic data for OvHV-2 remain limited. Recently, a Formosan sambar deer, an endemic subspecies in Taiwan, presented with characteristic MCF lesions and tested positive for OvHV-2 by PCR. We reconstructed the first coding-complete OvHV-2 genome from Taiwan (OvHV-2 TW01) using a hybrid whole-genome sequencing approach. The TW01

BACKGROUND AND AIM: Malignant catarrhal fever (MCF) is a highly fatal disease caused by ovine herpesvirus-2 (OvHV-2), with sheep acting as asymptomatic reservoir hosts. In Indonesia, MCF remains underdiagnosed due to its predominantly subclinical presentation, particularly in regions where mixed ruminant farming is practiced. Riau Province has a large population of Bali cattle, and sheep production is increasing, raising concerns regarding OvHV-2 spillover. This study aimed to determine the molecular prevalence of OvHV-2 infection in cattle and identify individual- and herd-level risk factors

Malignant catarrhal fever (MCF) is a fatal disease of cattle that, in East Africa, follows contact with wildebeest excreting alcelaphine herpesvirus 1 (AlHV-1). Recently an attenuated vaccine (atAlHV-1) was tested under experimental challenge on Friesian-Holstein (FH) cattle and gave a vaccine efficacy (VE) of approximately 90%. However testing under field conditions on an East African breed, the shorthorn zebu cross (SZC), gave a VE of 56% suggesting that FH and SZC cattle may respond differently to the vaccine. To investigate, a challenge trial was carried out using SZC. Additionally three a

In this study, the physical examination of 22 cattle revealed clinical signs of malignant catarrhal fever (MCF). Peripheral blood leukocyte (PBL) samples of the 22 cattle, and nasal (n = 7) and conjunctival (n = 9) swab samples from 16 sheep from two different farms, were taken for laboratory examination. The clinical diagnosis of MCF in cows was confirmed by the detection of ovine herpesvirus type 2 (OvHV-2) DNA by polymerase chain reaction (PCR). OvHV-2 DNA was detected by nested-PCR in PBL of one cow with clinical signs and nasal (1/7)-conjunctival(1/9) swab samples of two sheep housed in t

Alcelaphine gammaherpesvirus 1 (AlHV-1) asymptomatically persists in its natural host, the wildebeest. However, cross-species transmission to cattle results in the induction of an acute and lethal peripheral T cell lymphoma-like disease (PTCL), named malignant catarrhal fever (MCF). Our previous findings demonstrated an essential role for viral genome maintenance in infected CD8+ T lymphocytes but the exact mechanism(s) leading to lymphoproliferation and MCF remained unknown. To decipher how AlHV-1 dysregulates T lymphocytes, we first examined the global phenotypic changes in circulating CD8+

Ovine gammaherpesvirus 2 (OvGHV2) is the cause of sheep-associated malignant catarrhal fever (SA-MCF) in which sheep are the asymptomatic carrier-hosts with subclinical infections and/or clinical SA-MCF occurring in susceptible mammalian populations worldwide. Although SA-MCF is endemic in cattle throughout Brazil, there is only one report of OvGHV2-associated disease in water buffaloes (Bubalus bubalis), and comparatively few descriptions of infections by OvGHV2 in buffaloes worldwide. This study describes the molecular detection of OvGHV2 infections in asymptomatic water buffaloes and discus

Cross-species infection with ovine herpesvirus 2 (OvHV-2) in cattle causes malignant catarrhal fever (MCF). MCF may involve the central nervous system (CNS) with necrotizing arteritis and/or vasculitis described to be unique to MCF and discriminatory compared to other viral CNS infections. However, a systematic histopathological characterization of the neural form of MCF in cattle is lacking. We examined medulla oblongata (n = 9) or the entire brain (n = 9) of 18 cattle in which OvHV-2 was identified by quantitative polymerase chain reaction (qPCR), in order to pinpoint potential variations in
