The duck plague virus UL7 protein mediates RIG-I degradation to block host antiviral responses and promote viral pathogenesis.

Duck plague (DP), which is caused by duck plague virus (DPV), is an acute, highly contagious disease with an extremely high mortality rate, and poses a serious threat to the waterfowl industry. DPV, which is an immunosuppressive virus, can significantly suppress host innate immune responses during the late stages of infection. However, the specific mechanisms by which the DPV UL7 protein functions in the viral replication cycle and immune evasion remain unclear. This study demonstrated that the DPV UL7 protein markedly inhibited IFN-β promoter activity, downregulated the transcription levels o

Marek's disease virus infection alters the composition of exosomes in chicken fibroblasts and promotes viral replication.

Marek's disease (MD) is a highly contagious lymphoproliferative disease caused by Marek's disease virus (MDV), typically characterized by severe immunosuppression. In this study, exosomes were isolated and purified from chicken embryo fibroblasts (CEFs) infected with the virulent MDV strain Md5 (Exo-Md5) and uninfected controls (Exo-NC), respectively. Combining small RNA sequencing, mass spectrometry, and Western blot analysis, we confirmed that MDV infection significantly remodels the exosomal cargo composition. Exo-Md5 exhibited a predominant downregulation of miRNAs. Subsequent GO and KEGG

A polymerase chain reaction assay for detection of virulent and attenuated strains of duck plague virus.

Sequence analysis of duck plague virus (DPV) revealed that there was a 528bp (B fragment) deletion within the UL2 gene of DPV attenuated vaccine strain in comparison with field virulent strains. The finding of gene deletion provides a potential differentiation test between DPV virulent strain and attenuated strain based on their UL2 gene sizes. Thus we developed a polymerase chain reaction (PCR) assay targeting to the DPV UL2 gene for simultaneous detection of DPV virulent strain and attenuated strain, 827bp for virulent strain and 299bp for attenuated strain. This newly developed PCR for DPV

Cross-Species Antiviral Activity of Goose Interferons against Duck Plague Virus Is Related to Its Positive Self-Feedback Regulation and Subsequent Interferon Stimulated Genes Induction.

Interferons are a group of antiviral cytokines acting as the first line of defense in the antiviral immunity. Here, we describe the antiviral activity of goose type I interferon (IFNα) and type II interferon (IFNγ) against duck plague virus (DPV). Recombinant goose IFNα and IFNγ proteins of approximately 20 kDa and 18 kDa, respectively, were expressed. Following DPV-enhanced green fluorescent protein (EGFP) infection of duck embryo fibroblast cells (DEFs) with IFNα and IFNγ pre-treatment, the number of viral gene copies decreased more than 100-fold, with viral titers dropping approximately 100

Skin involvement in lymphomas caused by Marek's disease virus infection in Silkie chickens.

The Silkie is a typical Chinese breed of chicken. In 2012, batches of Silkies were found to have diffuse tumor-like nodules on their skin after feather removal, when they were slaughtered at about 60 days old. Gross examination showed no visible neoplastic lesions on the visceral organs and peripheral nerves, except slight splenomegaly in individual chickens. The disease was prevalent, with high condemnation rates for skin lesions, which caused great economic losses to the company. Tissues, including skin, visceral organs, and peripheral nerves, were collected for histologic examination. Hepar

Replication kinetics of Marek's disease vaccine virus in feathers and lymphoid tissues using PCR and virus isolation.

CVI988 (Rispens), an avirulent strain of Marek's disease virus, is the most widely used vaccine against Marek's disease. The kinetics of replication of CVI988 was examined in tissues of chickens vaccinated at either 1 day or 14 days of age and sampled regularly up to 28 days post-vaccination. Age at vaccination had no significant effect on the kinetics of CVI988 virus replication. During the cytolytic phase of infection (1-7 days), virus levels peaked in the spleen, bursa and thymus with very close correlation among these organs. Virus load in peripheral blood lagged behind and did not reach h

Duck plague virus US2 promotes p62-mediated autophagic degradation of RIG-I to suppress antiviral signaling.

Duck plague virus (DPV) is a highly contagious pathogen that causes severe immunosuppression and high mortality in waterfowl, resulting in substantial economic losses to the poultry industry. However, the mechanisms by which DPV evades host innate immune responses remain incompletely understood. In this study, we investigated the role of the DPV tegument protein US2 in regulating host antiviral responses. pUS2 significantly suppressed IFN-β promoter activation induced by poly(I:C) and poly(dA:dT) and reduced the transcription of IFN-β and interferon-stimulated genes (ISGs), including OASL and

Marek's disease virus-1 unique gene LORF1 is involved in viral replication and MDV-1/Md5-induced atrophy of the bursa of Fabricius.

Marek's disease virus (MDV), an alphaherpesvirus, causes severe immunosuppression and T cell lymphomas in chickens, known as Marek's disease (MD), an economically important poultry disease primarily controlled by vaccination. Importantly, it also serves as a comparative model for studying herpesvirus-induced tumor formation in humans. MDV encodes more than 100 genes, most of which have unknown functions. MDV LORF1 is unique to serotype I MDV (MDV-1), lacking homologs in other herpesviruses, and has not been explored yet. To this end, an infectious bacterial artificial chromosome (BAC) harborin

A targeted mutation in ICP8 attenuates duck plague virus by disrupting replication and proposes a vaccine strategy.

Controlling duck plague (DP) is essential for waterfowl health and industry productivity. The herpesviral single-stranded DNA-binding protein ICP8 is essential for replication compartment assembly, yet its exploitation for vaccine development in avian herpesviruses is limited. In this study, we generated two ICP8 mutants (H825A and F829A/P830A) in duck plague virus (DPV) via BAC-based homologous recombination. Using structural modeling, in vitro replication kinetics, co-immunoprecipitation, and in vivo duckling models, we assessed impacts on replication complex assembly, virulence attenuation,

Duck plague virus UL24 protein initiates K48/K63-linked IRF7 polyubiquitination to antagonize the innate immune response.

Duck plague virus (DPV), which is the causative agent of duck viral enteritis, is highly infectious and can cause severe disease and death in ducks, geese and other waterfowl. Several tegument proteins of DPV have been shown to affect the cyclic GMP-AMP synthase (cGAS)-STING signaling pathway to modulate host innate immune responses. DPV UL24, an important DPV tegument protein, can inhibit the activity of the IFN-β promoter. However, the mechanism by which the DPV UL24 protein regulates the host innate immune response remains unclear. In this study, we found that the UL24 protein can significa

Construction of the recombinant duck enteritis virus delivering capsid protein VP0 of the duck hepatitis A virus.

Duck hepatitis A virus type 1 (DHAV-1) disease causes significant economic losses to the duck industry. Duck enteritis virus (DEV) is frequently used as a viral vector for aquatic poultry vaccination, but no recombinan DEV expressing DHAV-1 VP0 has been developed. In this study, we established a system for rescuing the DEV C-KCE vaccine strain by transfecting cells with six fosmid DNAs. We generated a recombinant virus (rDEV-ul41VP0) by inserting the VP0 gene of DHAV-1 into the ul41 region in the DEV C-KCE genome. DHAV-1 VP0 was stably expressed in the rDEV-ul41VP0 infected cells, but did not

DPV pUL15 possesses a potential NLS, which is important for the location of the terminase complex and for viral proliferation and genome cleavage.

In herpesvirus, the terminase subunit pUL15 is involved in cleavage of the viral genome concatemers in the nucleus. Previous studies have shown that herpes simplex virus 1 (HSV-1) pUL15 can enter the nucleus without other viral proteins and help other terminase subunits enter the nucleus. However, this study revealed that duck plague virus (DPV) pUL15 cannot localize independently to the nucleus and can only be localized in the nucleus in the presence of pUL28 and pUL33. However, the data suggested the presence of a potential nuclear localization signal (NLS) in DPV pUL15, which is important f

Differential Virus-Specific IFN-Gamma Producing T Cell Responses to Marek's Disease Virus in Chickens With B19 and B21 MHC Haplotypes.

Marek's disease virus (MDV), the etiologic agent for Marek's disease (MD), causes a deadly lymphoproliferative disease in chickens. Causes of the well-documented association between genetically defined lines of chicken and resistance to MD remain unknown. Here, the frequencies of IFN-gamma producing pp38 and MEQ-specific T cell responses were determined in line N (B21 haplotype; MD-resistant) and line P2a (B19 haplotype, MD-susceptible) chickens after infection with vaccine and/or virulent (RB1B) strains of MDV using both standard ex vivo and cultured chIFN-gamma ELISPOT assays. Notably, MDV i

Is MS an auto-immune disease or a chronic gammaherpesvirus infection?

The geographical-epidemiological findings of MS over the past 75 years are reviewed. Evidence concerning PR latitude-gradients, clusters, migration patterns, and enhancement-protective factors are critically reviewed and analyzed. The evidence in genetics, twin-studies and auto-immune theories are also thoroughly scrutinized as to their possible involvement as causative factors in MS- and found to be somewhat lacking. Environmental discoveries in the Faroes and Key West clusters are reviewed for possible similar environmental causes by a deadly agent. Exogenous-environmental candidate agents o

Downregulation of the host ribonucleotide reductase subunit M2 via the Wnt/β-catenin pathway is a novel mechanism for duck plague virus replication and a potential antiviral target.

Duck plague virus (DPV), a highly contagious alphaherpesvirus, poses a significant threat to the global waterfowl industry. A comprehensive understanding of virus-host interactions is critical for developing effective control strategies. While DPV replication is known to depend on host cellular machinery, the mechanisms by which it manipulates host metabolism to its advantage remain poorly defined. Using RNA-seq analysis of DPV-infected duck embryo fibroblasts (DEFs), we identified a significant downregulation of genes involved in nucleotide metabolism, particularly the ribonucleotide reductas

Multidecadal High Mortality Disease Events in Australian Domestic Geese Associated with a Novel Alphaherpesvirus, Designated Anatid Alphaherpevirus 2.

Herpesviruses are ubiquitous viruses which infect a wide range of hosts. Novel herpesviruses are being increasingly detected in free-ranging bird populations and there are growing concerns for cross-species infection and spillover events. Herein, multiple sporadic outbreaks of mortality caused by a herpesvirus are described in domestic geese in Queensland, Australia. Goose herpesvirus was initially detected in 1989 in south-east Queensland, and this article details four further recent outbreaks and reports novel genome sequencing and phylogeny of the preliminarily designated anatid alphaherpes

Virulence attenuation of intestinal pathogenicity via combined gene deletion in duck enteritis vaccine strain restores gut microbiota balance and enhances safety.

There is an increasing need for a new generation of effective and safe vaccines, in the context of large-scale poultry farming and the prevalence of infectious diseases. With this in mind, we developed, for the first time, a duck enteritis virus (DEV) mutant, ΔTK-ΔgI/gE-ΔgG/gJ, through the deletion of multiple virulence genes. The resulting gene-deletion strain exhibited replication kinetics similar to those of the parent strain and was found to be safe in various animal models, offering a strategy for rapidly generating attenuated DEV strains. Previously, our team reported that DEV infection

An evolutionary perspective on the nucleocytoplasmic shuttling mechanism of duck plague virus UL54 protein.

Duck plague virus (DPV), a member of the alpha-herpesvirus, causes duck plague (DP), thereby posing a serious threat to the waterfowl industry. The DPV UL54-encoded protein shuttles between the nucleus and cytoplasm to modulate viral replication. However, the role of the UL54 protein in the evolutionary process remains unknown. This study is conducted from an evolutionary perspective to explore the nuclear-cytoplasmic shuttling characteristic of UL54 protein. First, we analyzed the co-evolution on DPV whole genome and its UL54 gene using molecular evolutionary methods. Next, we constructed phy

The UL31 protein of duck plague virus promotes viral replication and interacts with ICP4, ICP22, and VP16 proteins.

Duck plague is a highly contagious viral disease caused by duck plague virus (DPV) that poses a significant threat to the waterfowl industry. In herpesviruses, pUL31 plays a key role in the nuclear egress of viral nucleocapsids. However, it remains unclear whether pUL31 participates in other stages of the viral life cycle. Therefore, this study aimed to elucidate the functions of pUL31 in the DPV life cycle beyond its established role in facilitating nucleocapsid nuclear egress. In this study, glutathione S-transferase (GST) pull-down coupled with high-performance liquid chromatography-tandem

Duck plague virus US3 kinase phosphorylates and induces STING degradation to inhibit innate immune responses.

Duck plague virus (DPV) causes the highest mortality rate among aquatic birds; however, its antago nistic mechanism against antiviral innate immune responses remains elusive. In this study, we systematically screened and found that most DPV genes have inhibitory potential for duck cyclic guanosine monophosphate-adenosine monophosphate synthetase (cGAS)/stimulator of interferon (IFN) gene (STING) pathway-mediated antiviral responses, with the DPV US3 kinase showing the strongest inhibitory activity. Co-immunoprecipitation and immunoblotting assays demonstrated that DPV US3 interacted with STING
