Mast cell stabilizer ketotifen [4-(1-methyl-4-piperidylidene)-4h-benzo[4,5]cyclohepta[1,2-b]thiophen-10(9H)-one fumarate] prevents mucosal mast cell hyperplasia and intestinal dysmotility in experimental Trichinella spiralis inflammation in the rat.

Trichinella spiralis infection in rats induces hypermotility and an abnormal response to cholecystokinin (CCK) similar to motor disturbances observed in irritable bowel syndrome. Mast cell hyperplasia is also characteristic of this experimental model. The aim of our study was to correlate mast cell activity with the development of dysmotility and to demonstrate whether the mast cell stabilizer ketotifen [4-(1-methyl-4-piperidylidene)-4H-benzo[4,5]cyclohepta[1,2-b]thiophen-10(9H)-one fumarate] could prevent the development of intestine hypermotility. Sprague-Dawley rats were infected with T. sp

Glehnia littoralis and imperatorin attenuate allergic airway inflammation via mast cell stabilization and transcriptome-defined suppression of FcεRI/TLR-MAPK/NF-κB signaling.

BACKGROUND: Allergic airway inflammation is driven by mast cell activation and IgE-FcεRI signaling. Current pharmacological treatments often present limited efficacy or long-term adverse effects. Glehnia littoralis Fr. Schmidt ex Miq. (GL) is a traditional medicinal herb used for respiratory ailments, but its anti-allergic mechanisms remain largely undefined. PURPOSE: We aimed to evaluate the anti-allergic potential of GL and its bioactive compound, imperatorin, specifically investigating their ability to stabilize mast cells and modulate inflammatory signaling. STUDY DESIGN: This study inte

[Treatment of eye allergies].

Seasonal atopic conjunctivitis is treated with antihistamines, cromoglycate and short courses of corticosteroids, in severe cases with subcutaneous or sublingual immunotherapy. Chronic conjunctivitis requires year-round treatment with mast cell stabilizers, antihistamines or topical corticosteroids. Long-term treatment of atopic blepharoconjunctivitis consists of tacrolimus or pimecrolimus cream. For atopic keratoconjunctivitis corticosteroid and, if necessary, cyclosporine eye drops are needed. First-line therapy of vernal conjunctivitis involves mast cell stabilizers and, if necessary, corti

Revisiting pharmacological actions of disodium cromoglycate in light of GPR35: New therapeutic contexts for classical drugs.

Mast cell stabilizers such as disodium cromoglycate have been used for decades as prophylactic anti-allergic agents, yet their molecular targets remained unknown, and their intracellular mechanisms were long debated. Identification of GPR35 as a receptor engaged by multiple mast cell stabilizers provided a new framework to reinterpret classical pharmacology. Here, historical and recent evidence linking mast cell stabilizers to GPR35 is integrated and pharmacological characteristics of GPR35 are described in the context of species difference and intracellular signaling. GPR35 is expressed in ep

[Vernal keratoconjunctivitis and possibilities its treatment].

The authors present a case of vernal keratoconjunctivitis (VKC) in 5 years old boy. Despite intensive topical pharmacological therapy (mast-cell stabilizers, antihistamines, steroids in acute exacerbations) unsatisfied reduction of the symptoms was observed and cornea became involved in the left eye (LE). Repeated surgical abrasion of the papillary hypertrophy was performed with temporaly relief. We decided to provide a cryocoagulation of the papillary hypertrophy in the LE. We did not see any pre and post operative complications, significant improvement was observed and the patient is now (13

Mast cell, a promising therapeutic target in tubulointerstitial fibrosis.

Tubulointerstitial fibrosis is a final common pathway to the eventual structural desolation of kidneys. However, the mechanism involved in this phenomenon is still poorly understood, and current therapies are ineffective or only marginally effective. Mast cell has a variety of physiological and pathological functions through the production of heparin, histamine, neutrophil chemoattractants, immunoregulatory cytokines, and mast cell-specific serine proteases tryptase and chymase. The survival and proliferation of mast cell are dependent upon stem cell factor. Presently, mast cells are known to

Induction of Mast Cell Accumulation by Tryptase via a Protease Activated Receptor-2 and ICAM-1 Dependent Mechanism.

Mast cells are primary effector cells of allergy, and recruitment of mast cells in involved tissue is one of the key events in allergic inflammation. Tryptase is the most abundant secretory product of mast cells, but little is known of its influence on mast cell accumulation. Using mouse peritoneal model, cell migration assay, and flow cytometry analysis, we investigated role of tryptase in recruiting mast cells. The results showed that tryptase induced up to 6.7-fold increase in mast cell numbers in mouse peritoneum following injection. Inhibitors of tryptase, an antagonist of PAR-2 FSLLRY-NH

Advances in the Management of Mediator-Related Symptoms in Non-Advanced Systemic Mastocytosis.

Systemic mastocytosis (SM) is a rarely occurring clonal mast-cell disorder defined by aberrant mast-cell accumulation and episodic or chronic mediator release, giving rise to a broad range of manifestations from pruritus and flushing to gastrointestinal symptoms and anaphylaxis. In non-advanced SM, mediator-related symptoms are the major source of morbidity and substantially impair quality of life. Traditional symptom-directed therapies-including antihistamines, leukotriene modifiers, and mast-cell stabilizers-remain the foundation of care, but a subset of patients experience persistent, refra

Mucosal permeability and mast cells as targets for functional gastrointestinal disorders.

The intestinal mucosa is constantly exposed to harmful luminal content, and uptake is closely controlled and regulated by neuro-immune factors. If control is broken, it might lead to ongoing enhanced mucosal permeability, potentially resulting in functional gastrointestinal disorders. The importance of mast cells in the regulation of the mucosal barrier has become obvious, and increased numbers and more activated mast cells have been observed in irritable bowel syndrome, functional dyspepsia and gastroesophageal reflux disease. To target the disturbed mucosal permeability, directly or via mast

Mast cell mediation of visceral sensation and permeability in irritable bowel syndrome.

Abnormalities of mast cell structure or function may play prominent roles in irritable bowel syndrome (IBS) symptom genesis. Mast cells show close apposition to sensory nerves and release bioactive substances in response to varied stimuli including infection, stress, and other neuroendocrine factors. Most studies focus on patients who develop IBS after enteric infection or who report diarrhea-predominant symptoms. Three topics underlying IBS pathogenesis have been emphasized in recent investigations. Visceral hypersensitivity to luminal stimulation is found in most IBS patients and may contrib

Low-dose diet supplement of a natural flavonoid, luteolin, ameliorates diet-induced obesity and insulin resistance in mice.

SCOPE: Mast cells play important roles in diet-induced obesity and diabetes, and some synthetic mast cell stabilizers can improve related metabolic disturbances in mice. Luteolin (LU) is a potent natural mast cell stabilizer. However, a direct correlation between LU and these common metabolic diseases is not established. METHODS AND RESULTS: Male C57BL/6 mice were fed low-fat diet, high-fat diet (HFD), HFD with 0.002 and 0.01% LU for 12 wk, respectively. Dietary LU suppressed HFD-induced body weight gain, fat deposition, and adipocyte hypertrophy. Meanwhile, glucose intolerance and insulin se

Vernal keratoconjunctivitis presenting unilaterally.

A 7-year-old boy had itching, foreign body sensation, and redness in his right eye. Unilateral cobblestone papillae and a shield ulcer were found. Topical antihistamines, mast cell stabilizers, and steroids led to marked improvement. Unilateral vernal keratoconjunctivitis should be included in the differential diagnosis of unilateral giant papillary conjunctivitis.

Juvenile primary extranasopharyngeal angiofibroma, presenting as cheek swelling.

BACKGROUND: Dental pulp inflammation is a very complex process due to its situation in low compliance (confined environment) surrounded by mineralized dentin. Mast cells are one of the mediators of inflammation. Immunohistochemical localization of mast cells by anti-tryptase antibodies in formalin-fixed paraffin-embedded sections has been shown to be highly specific. The aim of the present study is to quantify the mast cells in inflamed and noninflamed human pulp tissue using immunohistochemical analysis. MATERIALS AND METHODS: Immunohistochemical localization of mast cells by anti-tryptase a

Ocular itch associated with allergic conjunctivitis: latest evidence and clinical management.

Allergic conjunctivitis is one of the most common allergic conditions worldwide. Its incidence is increasing due to changing climate, pollution, increased pollen loads, and the subject's heightened immunological sensitivity in response to these environmental changes. The pathophysiology predominantly involves immunoglobulin E-related mast-cell activation, with release of histamine and other mediators contributing to the propagation of the response by calling in other immune cells and further inflammation. This article presents the evolution of ocular allergy treatments, from vasoconstrictors,

Innervation of enteric mast cells by primary spinal afferents in guinea pig and human small intestine.

Mast cells express the substance P (SP) neurokinin 1 receptor and the calcitonin gene-related peptide (CGRP) receptor in guinea pig and human small intestine. Enzyme-linked immunoassay showed that activation of intramural afferents by antidromic electrical stimulation or by capsaicin released SP and CGRP from human and guinea pig intestinal segments. Electrical stimulation of the afferents evoked slow excitatory postsynaptic potentials (EPSPs) in the enteric nervous system. The slow EPSPs were mediated by tachykinin neurokinin 1 and CGRP receptors. Capsaicin evoked slow EPSP-like responses tha
