Integrative Analysis of Transcriptomic and Metabolomic Data for Identification of Pathways Related to Matrine-Induced Hepatotoxicity.

Matrine (MT) is a major bioactive compound extracted from Sophorae tonkinensis. However, the clinical application of MT is relatively restricted due to its potentially toxic effects, especially hepatotoxicity. Although MT-induced liver injury has been reported, little is known about the underlying molecular mechanisms. In this study, transcriptomics and metabolomics were applied to investigate the hepatotoxicity of MT in mice. The results indicated that liver injury occurred when the administration of MT (30 or 60 mg/kg, i.g) lasted for 2 weeks, including dramatically increased alanine aminotr

Matrine promotes oligodendrocyte development in CNS autoimmunity through the PI3K/Akt signaling pathway.

AIMS: Matrine (MAT), a quinolizidine alkaloid derived from the herb Radix Sophorae flavescens, has been recently found to be beneficial in experimental autoimmune encephalomyelitis (EAE), an animal model of multiple sclerosis, mainly through its anti-inflammatory effect. In the present study, we tested the effect of MAT on ongoing EAE and defined possible mechanisms underlying its effects on myelination and oligodendrocytes. MAIN METHODS: EAE was induced in C57BL/6 mice and MAT treatment was started at disease onset. Clinical scores were monitored daily; spinal cords and the corpus callosum b

Integrated metabolomics and gut microbiota analyses reveal the protective effects of matrine in ulcerative colitis.

BACKGROUND: Ulcerative colitis (UC) is a chronic inflammatory bowel disease driven by gut microbial dysbiosis and metabolic dysfunction. Matrine, a natural alkaloid with anti-inflammatory properties, shows therapeutic potential; however, its mechanisms involving the coordinated modulation of bacteria, fungi, and host intestinal luminal metabolism remain unclear. METHODS: We evaluated the therapeutic efficacy of matrine using a dextran sulfate sodium (DSS)-induced murine model of ulcerative colitis. Disease severity was assessed via the disease activity index, colon length, and histopathology.

Matrine induces ferroptosis and suppresses colorectal cancer through targeting the STAT3/SLC7A11 pathway.

Matrine, a natural alkaloid with recognized anti-cancer potential, has been reported to induce cancer cell death. However, the mechanism remains incompletely defined. In this study, we investigated the pharmacological effect of matrine on colorectal cancer (CRC) using both in vivo tumor‑bearing mouse models and in vitro cell cultures. Our results demonstrated that matrine effectively suppressed colorectal cancer progression in mice, accompanied by an increased area of tumor necrosis and enhanced infiltration of anti-tumor immune cells, including CD4+ and CD8+ T-cells. Importantly, matrine trea

Spheres, worms, and packing problems in the nanoscale assembly of ALS-linked protein Matrin-3.

Matrin-3 (MATR3) is an RNA-binding protein that forms higher-order assemblies and pathological inclusions associated with neurodegenerative diseases. In this issue of Molecular Cell, Sprunger et al.1 explore the assembly of MATR3 and find that it forms spherical and worm-like nanoscale assemblies distinct from condensates formed by macrophase separation.

Therapeutic potential of matrine and osthole against copper-promoted lung cancer cell malignancy.

Non-small cell lung cancer (NSCLC) is the major type of malignant tumor in the lungs. Emerging epidemiological evidence implicates environmental copper exposure as a potential risk modulator for NSCLC progression. This study investigated the effects of low-dose Copper (Cu) exposure on A549 cells and evaluated the therapeutic potential of two natural compounds, osthole and matrine. The results demonstrated that Cu exposure (5 μg/mL, 12 h) significantly promoted the migration, invasion and colony formation of A549 cells (P < 0.05), accompanied by a reduction in cell apoptosis, a decrease in ROS

Design, Synthesis, and Bioevaluation of Matrine Derivatives as Potential Anti-Hepatitis B Virus Agents.

Hepatitis B virus (HBV) is a causative reagent that frequently causes progressive liver diseases, leading to the development of acute hepatitis, chronic hepatitis, cirrhosis, and eventually hepatocellular carcinoma. Despite several antiviral drugs, including interferon-α and nucleotide derivatives, being approved for clinical treatment of HBV, critical issues remain unresolved, e.g., their low-to-moderate efficacy and adverse side effects, as well as resistant strains. In this study, twenty-three matrine derivatives were synthesized, and their antiviral effects against HBV were evaluated. Of t

Endothelial Matrin-3 Limits Cellular Senescence and Lipid Dysregulation to Protect Against Murine Atherosclerosis.

Matrin-3 is a DNA- and RNA-binding protein implicated in cardiovascular development. However, the role of endothelial Matrin-3 in atherosclerosis has not been previously studied. Here, we found that Matrin-3 silencing induces endothelial senescence, characterized by reduced cell proliferation, increased senescence-associated β-galactosidase activity, and impaired mitochondrial function in vitro. Quantitative proteomics and bulk RNA-sequencing identified hundreds of differentially expressed genes and proteins in human endothelial cells. Bioinformatic analyses revealed enrichment of pathways rel

Matrine-incorporated chitosan active biofilms with enhanced barrier and antimicrobial properties for postharvest fruit preservation.

Biodegradable active packaging biofilms with multifunctional preservation performance have attracted increasing attention for postharvest fruit preservation. In this study, matrine-incorporated chitosan active biofilms were developed using a citric acid-assisted casting strategy to improve the barrier, antioxidant, and antimicrobial properties of chitosan-based biofilms. Structural characterization demonstrated that matrine formed intermolecular hydrogen-bonding and electrostatic interactions with chitosan, resulting in a denser and more homogeneous biofilm network. Under the optimized formula

Sublethal effects of matrine and neemarin on the demography of Habrobracon hebetor developing on treated Helicoverpa armigera larvae.

Integrated pest management (IPM) relies on the compatibility of multiple control strategies. In controlling Helicoverpa armigera (Hübner) with the braconid parasitoid Habrobracon hebetor (Say), evaluating insecticide efficacy and compatibility is essential. Botanical insecticides may provide a safer alternative to synthetic chemicals due to reduced non-target effects. This study investigated the effects of sublethal concentrations of matrine and neemarin ingested by H. armigera larvae on H. hebetor developing on them, to assess their compatibility for potential integration into IPM programs. Design, synthesis and antifungal activity of novel matrine-hydroxamic acid derivatives containing benzene sulfonamide.

To address the urgent need for novel antibacterial drugs, herein, a series of 27 novel matrine derivatives incorporating hydroxamic acid and benzene sulfonamide moieties were designed and synthesized. Antimicrobial testing demonstrated exceptional inhibitory activity against Candida albicans, with the most potent compound (10g) showing a MIC value of 0.062 mg mL-1, which was significantly lower than that of the clinical antibiotic fluconazole (8.590 mg mL-1). 3D-QSAR analysis identified the phenylsulfonyl group as crucial for activity, particularly when substituted with a 4-(CH3)3 group. Combining with matrine restores ciprofloxacin efficacy against qnrS producing E. coli in vitro and in vivo.

The exposure risk of plasmid-mediated quinolone resistance (PMQR) genes increases the incidence of resistant bacterial infections, has resulted in clinical treatment failures with ciprofloxacin, necessitating urgent implementation of novel strategies for controlling this situation. Matrine serves as the principal constituent of the traditional Chinese herb Sophora flavescens Ait. and exhibits pharmacological activities including anti-inflammatory, antibacterial, anti-tumor, and hepatoprotective effects. However, the precise mechanism by which matrine exhibits antibacterial activity remains inc

Matrine Suppresses Arsenic-Induced Malignant Transformation of SV-HUC-1 Cells via NOX2.

Arsenic (As) has been classified as a carcinogen for humans. There is abundant evidence indicating that arsenic increases the risk of bladder cancer among human populations. However, the underlying mechanisms have yet to be fully understood and elucidated. NADPH oxidases (NOXs) are the main enzymes for ROS production in the body. NADPH Oxidase 2 (NOX2), which is the most distinctive and ubiquitously expressed subunit of NOXs, can promote the formation and development of tumors. The utilization of NOX2 as a therapeutic target has been proposed to modulate diseases resulting from the activation

Matrine induces V-ATPase-dependent cytoplasmic vacuolation and inhibits the function of the lysosome in leukemia cells.

Matrine is the main component extracted from legumes and has extensive anti-cancer effects; however, its molecular mechanism is unclear. In our study, we found that matrine induced vacuolation in leukemia cells is closely related to cell proliferation inhibition. Vacuolization was reversed after matrine removal. The neutral red staining assay indicated that the matrine-induced vacuoles were acidic, and the vacuoles originated mostly from the lysosome or endosome, as observed by transmission electron microscope (TEM) and fluorescence microscopy localization of LAMP-GFP. Furthermore, single-cell
