The effects of selective antagonists of serotonin 5-HT7 and 5-HT1A receptors on MK-801-induced impairment of learning and memory in the passive avoidance and Morris water maze tests in rats: mechanistic implications for the beneficial effects of the novel atypical antipsychotic lurasidone.

We have previously reported that lurasidone, a novel atypical antipsychotic with potent serotonin 5-HT(7) antagonist and 5-HT(1A) partial agonist activities, is superior to other antipsychotics in improving the N-methyl-D-aspartate (NMDA) receptor antagonist MK-801-induced learning and memory impairment in the passive avoidance (PA) and Morris water maze (MWM) tests in rats. In this study, we investigated the effects of selective antagonists of 5-HT(7) and 5-HT(1A) receptors (SB-656104-A and WAY-100635, respectively) on MK-801-induced learning and memory impairment in the same tests. In the PA

Refinement of the Barnes and Morris water maze protocols improves characterization of spatial cognitive deficits in the lithium-pilocarpine rat model of epilepsy.

Temporal lobe epilepsy (TLE) often causes cognitive impairment, especially a decline in spatial memory. Reductions in spatial memory and learning are also common in rodent models of TLE. The Morris water maze and the Barnes maze are the standard methods for evaluating spatial learning and memory in rodents. However, animals with TLE may exhibit agitation, distress, and fail to follow the paradigmatic context of these tests, making the interpretation of experimental data difficult. This study optimized the procedure of the Morris water maze and the Barnes maze to evaluate spatial learning and m

Latent learning improves foraging efficiency in ants.

Learning allows animals to react to changing environmental conditions and to respond to stimuli in an adaptive way, for instance by giving novel stimuli positive or negative value. However, there is a form of learning that does not require reward or punishment: latent learning. This knowledge is hidden at the time of learning but it manifests later when a suitable incentive appears. Latent learning helps animals to navigate their environments, thus improving foraging efficiency, especially when resources are unpredictably distributed. Although latent learning has been well documented in verteb

Rat maze performance after infusion of cross-linked hemoglobin solution.

Neurotoxicity and behavioral performance degradation have previously been observed in rats after exchange transfusions with unmodified stroma-free hemoglobin solutions. We evaluated a diaspirin cross-linked stroma-free hemoglobin solution (HbXL) for evidence of neurotoxicity. Rats that were trained to complete a water alley maze received a clinically-relevant dose (20 ml/kg) of the 14% HbXL solution on top of their normal blood volume. After post-treatment memory testing in the water maze, the same rats were challenged to learn an elevated radial-arm maze. The HbXL and control groups showed no

Reward-Based Spatial Learning in Teens With Bulimia Nervosa.

OBJECTIVE: To assess the functioning of mesolimbic and fronto-striatal areas involved in reward-based spatial learning in teenaged girls with bulimia nervosa (BN) that might be involved in the development and maintenance of maladaptive behaviors characteristic of the disorder. METHOD: We compared functional magnetic resonance imaging blood oxygen level-dependent response in 27 adolescent girls with BN to that of 27 healthy, age-matched control participants during a reward-based learning task that required learning to use extra-maze cues to navigate a virtual 8-arm radial maze to find hidden r

Water T-maze: a useful assay for determination of repetitive behaviors in mice.

BACKGROUND: Repetitive behavior is a term used to describe a wide variety of invariant and inappropriate behaviors that occur in many diverse conditions, including autism. It is necessary to utilize and/or design rodent behavioral assays that exploit individual types of repetitive behavior so that underlying pathology and therapeutic measures can be determined. A variety of high-throughput assays to investigate lower order repetitive behaviors are available for rodents, whereas, fewer assays are available to investigate higher order repetitive behaviors, such as perseverative behavior. Magnesium deficiency reduces fear-induced conditional lick suppression in mice.

The consequences of broad-scale alterations in magnesium (Mg2+) levels on learning and memory are poorly understood. We have recently demonstrated that adult male mice maintained on an Mg2+-deficient diet exhibit reduced conditional freezing behavior. The purpose of the present study was to determine if the detrimental effect of Mg2+ deficiency in mice extended to another measure of conditional fear, conditioned lick suppression (CLS), as well as to another form of learning, spatial learning in the swim maze task. Adult male C57Bl/6J mice were provided with a normal or Mg2+-deficient diet and

The effects of chronic nicotine on spatial learning and bromodeoxyuridine incorporation into the dentate gyrus of the rat.

RATIONALE: Nicotine is reported to improve learning and memory in experimental animals. Improved learning and memory has also been related to increased neurogenesis in the dentate gyrus (DG) of the hippocampal formation. Surprisingly, recent studies suggest that self-administered nicotine depresses cell proliferation in the DG. OBJECTIVE: To test the hypothesis that the effects of nicotine on cell proliferation in the DG and learning and memory depend upon the nicotine dose administered. METHODS: Rats were chronically infused from subcutaneous osmotic mini pumps with nicotine (0.25 or 4 mg k

Sex differences in the effects of 10 Hz and 40 Hz transcranial alternating current stimulation on spatial cognition in mice.

BACKGROUND: Sex differences are crucial to understanding neuropsychiatric disorders, yet they are often overlooked in the development of therapies. Transcranial alternating current stimulation (tACS) shows promise for cognitive enhancement, but its sex-specific effects are largely unknown. METHODS: In this study, the effects of 10 Hz and 40 Hz tACS on spatial cognition were examined in male and female mice using three tests: the Y-maze to evaluate spatial recognition memory, the Barnes maze to evaluate spatial learning and memory related to punishment, and the reversal Barnes maze to evaluate

Differences in learning between hyperprolinemic mice and their congenic controls.

These experiments expanded earlier work on hyperprolinemic mice which showed learning deficits. The following behavioral tasks were used: step-through, passive avoidance; T-maze acquisition; shuttlebox acquisition, and radial-arm maze. Mouse species included PRO/Re-bb (genetically hyperprolinemic mice) and PRO/Re-aa (congenic nonhyperprolinemic controls) obtained from the Jackson Breeding Laboratories. Hyperprolinemic mice were impaired in acquiring T-maze and shuttlebox footshock avoidance behavior. One-trial passive avoidance behavior did not clearly differentiate between the groups. Radial

Protective role of hemokinin-1 against age-dependent spatial memory and attention decline in mice.

Age-related cognitive decline is a major social and medical challenge, but its mechanisms remain poorly understood; prevention and therapy are unsatisfactory. The tachykinin hemokinin-1 (HK-1), encoded by the Tac4 gene, is expressed in brain regions involved in learning and memory and has been implicated in neuroimmune regulation, but its contribution to age-dependent cognitive impairment remains unclear. Here, we investigated whether endogenous HK-1 modulates cognitive performance during aging. Young (3-5 months), middle-aged (12-15 months), and old (18-20 months) male and female C57BL/6 J wi

Characterization of learning and memory deficits in C57BL/6 mice infected with LP-BM5, a murine model of AIDS.

Mice infected with an immunosuppressive murine leukemia virus mixture, LP-BM5 show a profound immunosuppression described as murine acquired immune deficiency syndrome (AIDS). In the present study, we characterized learning and memory deficits in C57BL/6 mice infected with LP-BM5. Spontaneous alternation behavior in a Y-maze and latent learning (spatial attention) in a water-finding test, as well as spatial reference and reversal learning in a water maze test, were significantly impaired in the mice infected with LP-BM5. These deficits appeared in the absence of any motoric and visual impairme

Neurotoxic effects of repetitive inhibition of oxidative phosphorylation in young adults surfacing with deficits of spatial learning in old age.

Little is known about whether events in youth impact performance in old age. We examined spatial navigation in young (4.5 months) and middle-aged (9 months) CD-1 mice in a complex maze after treatment with 3-nitropropionate (3-np; 20 mg/kg body weight; 9 injections intraperitoneally [i.p.] every other day). Young mice treated with 3-np were examined in a mirror version of this maze in old age (22 months) and with a nonreference memory task of an eight-arm radial maze. The performance of young mice was affected to a small degree by treatment with 3-np. However, the performance of middle-aged mi

Behavioural retardation in the neuropathology of mental retardation.

A series of experiments, involving diverse perinatal treatments of either rats or mice, have been performed in order to investigate the effects of these treatments upon certain selected spontaneous and learned behaviours in the laboratory in order to derive animal models of behavioural retardation. Rat dams were administered either methylazoxymethanol, metallic mercury, organic tin or neuroleptic compounds, and the offspring of these dams were studied on the behavioural tests at adult ages, prenatal studies. Newborn rat pups were administered either 6-OHDA (at various doses), or metallic mercu

Novel measures of Morris water maze performance that use vector field maps to assess accuracy, uncertainty, and intention of navigational searches.

Most commonly used behavioral measures for testing learning and memory in the Morris water maze (MWM) involve comparisons of an animal's residence time in different quadrants of the pool. Such measures are limited in their ability to test different aspects of the animal's performance. Here, we describe novel measures of performance in the MWM that use vector fields to capture the motion of mice as well as their search pattern in the maze. Using these vector fields, we develop quantitative measures of performance that are intuitive and more sensitive than classical measures. First, we describe

Fetal hydantoin syndrome in rats: dose-effect relationships of prenatal phenytoin on postnatal development and behavior.

Sprague-Dawley rats were gavaged once daily on days 7-18 of gestation with, 100, 150, or 200 mg/kg of phenytoin. Only the highest dose of phenytoin decreased maternal weight during gestation or increased offspring mortality up to weaning. Offspring were evaluated for activity prior to weaning (pivoting and photocell) and afterwards (figure 8, open-field, and hole-board), dynamic righting development, maze learning (Biel maze), and visual discrimination (Y maze), and for startle reaction to both auditory and tactile stimuli. The highest dose of phenytoin produced increased activity on all tests
