Sensing pressure in the cardiovascular system: Gq-coupled mechanoreceptors and TRP channels.

Despite the central physiological importance of cardiovascular mechanotransduction, the molecular identities of the sensors and the signaling pathways have long remained elusive. Indeed, how pressure is transduced into cellular excitation has only recently started to emerge. In both arterial and cardiac myocytes, the diacylglycerol-sensitive canonical transient receptor potential (TRPC) subunits are proposed to underlie the stretch-activated depolarizing cation channels. An indirect mechanism of activation through a ligand-independent conformational switch of Gq-coupled receptors by mechanical

Vagal afferent discharge from mechanoreceptors in different regions of the ferret stomach.

1. The rate and volume of gastric filling was estimated in conscious ferrets by measuring the amount of milk they would drink after an overnight fast. The mean volume was 94.5 +/- 7.5 ml. at a rate of 13.0 +/- 0.74 ml./min. An intragastric infusion rate of 10 ml./min to a total of 50 ml. was selected as a standard distension stimulus. 2. Action potentials were recorded from single gastric afferent fibres in the cervical vagus. All but two of thirty-six afferent units were tonically active when the stomach was deflated. 3. Afferent fibres arising from receptors in the antrum showed modulation i

Esophageal mucosal sensory nerves and potential mechanoreceptors in patients with ineffective esophageal motility.

BACKGROUND: Ineffective esophageal motility (IEM) is the most common motility disorder. However, little is known about its pathophysiology. Vagal afferent nerves convey esophageal intraluminal bolus information to solitary nucleus, which is likely to be involved with esophageal primary and secondary peristalsis (SP). We hypothesized that altered mucosal sensory afferents underlie the pathogenesis of IEM. METHODS: We prospectively collected esophageal biopsies from 38 patients with proton pump inhibitor-refractory reflux symptoms from January to December 2019. All patients underwent high-resol

Periodontal Mechanoreceptor Contribution to Minimal Holding Force Between Incisor Teeth.

BACKGROUND: The functional importance of periodontal mechanoreceptors (PMRs) is highlighted by impaired force control when periodontal input is reduced or absent. Previous studies used handheld transducers to measure the minimum holding force between the incisors. In the present study, a high-resolution force transducer was held passively between the incisors against gravity. OBJECTIVE: To quantify the contribution of anterior PMRs to minimum holding force and examine the effects of body position and periodontal health. METHODS: Twenty healthy adults (12 females, 8 males; 23.2 ± 1.9 years) p

Persistence of mechanoreceptors in ACL remnants: a histological and immunohistochemical analysis of 128 patients.

BACKGROUND: Anterior cruciate ligament (ACL) remnants may retain sensory neural elements after rupture, potentially contributing to proprioceptive function and graft reinnervation following reconstruction. However, the persistence of mechanoreceptors in remnant tissues and the factors influencing their preservation remain incompletely understood. This study aimed to evaluate the presence of neural structures in ACL remnants using histological and immunohistochemical methods and to determine the association between mechanoreceptor preservation, injury duration, and remnant attachment pattern.

Possible Role of the Myelinated Neural Network in the Parietal Peritoneum in Rats as a Mechanoreceptor.

A network of myelinated nerve fibers in the peritoneum covers the abdominal wall. We studied the topographic distribution of this network, explored the fibers' destination in the central nervous system, and examined the markers in these fibers in order to identify the nature of the sensation conveyed by the network of nerve fibers in rats. We used Sihler's method, which stains myelinated fibers in whole mount materials, and observed a dense nerve network and endings toward the peritoneal cavity in the peritoneum that covers the abdomen's lateral bulge. We studied the axonal transport of choler

Calcium concentration changes during sensory transduction in spider mechanoreceptor neurons.

Most mechanoreceptor neurons encode mechanical signals into action potential trains within the same cell. Evidence suggests that intracellular calcium ion concentration, [Ca2+], increases during mechanotransduction, either by direct entry through mechanically activated channels or indirectly through voltage-activated calcium channels. However, little is known about the amounts of calcium involved or its roles in mechanotransduction. We estimated [Ca2+] in mechanoreceptor neurons of the spider, Cupiennius salei, during mechanical stimulation using Oregon Green BAPTA-1, and a single-compartment

The role of PGE2 in the sensitization of mechanoreceptors in normal and inflamed ankle joints of the rat.

The role of PGE2 in the sensitization of high-threshold tarsal joint mechanoreceptors (putative nociceptors) has been investigated in 11 arthritic and 16 normal rats. Injections of a low dose of Freund's complete adjuvant at multiple sites into the tissues surrounding the ankle joint induced a chronic unilateral monoarthritis in the injected limb. Measurements of both spontaneous activity and responses of tarsal joint mechanoreceptors to repeated graded mechanical stimuli were made. All of the mechanoreceptors examined had afferent fibres with conduction velocities in the C- or A-delta range.

C-fiber low-threshold mechanoreceptors versus C-fiber nociceptors: distinct properties and signatures.

Mammalian skin is innervated by distinct classes of low-threshold mechanoreceptors (LTMRs) and nociceptors, which are broadly categorized according to axonal conduction velocity into Aβ-, Aδ-and C-fiber populations. C-LTMRs encode innocuous mechanical stimuli and, in humans (where they are termed C-tactile [CT] afferents), contribute to affective/social touch, whereas nociceptors detect potentially tissue-damaging stimuli. For decades, C-LTMRs were thought to be confined to hairy skin. However, approximately a decade ago, we provided the first electrophysiological evidence for a rare populatio

Clustering of slowly adapting type II mechanoreceptors in human peripheral nerve and skin.

The intrafascicular organization of human peripheral nerves was studied with percutaneous microneurography in the median, radial and peroneal nerves with one-surface or two-surface thin diameter concentric needle electrodes. Data from 33 recording sites containing 47 slowly adapting type II (SAII) units were analysed. At many sites two, sometimes even three, neighbouring SAII units were recorded from the explored nerve fascicle and they had adjacent or even overlapping cutaneous receptive fields. Among pairs of SAII units found at the same site, one unit often had ongoing discharge, whereas th

Development of nonunions in the rat fibula after removal of periosteal neural mechanoreceptors.

Periosteal proprioceptive nerve receptors may act as mechanoreceptors of long bones during adaptive remodeling after fracture. They may also contribute to the mechanisms of coordinated functional activity of fractured limbs and thus inhibit harmful overloading of fracture callus. The area of proprioceptive nerve receptors around the distal part of the rat fibula was stripped surgically, and a standard fracture of the fibular shaft was produced. Animals failed to unite their fractures and developed mainly atrophic nonunions. Atrophy of ununited fragments was due to osteoclastic bone resorption.

Mechanoreceptor plexin D1 regulates lymphatic valve morphogenesis and lymphedema pathogenesis.

Lymphatic valves are essential for maintaining tissue fluid homeostasis, and their dysfunction leads to lymphedema, a morbid and disfiguring disease without a cure. Mechanical forces due to lymph flow are required for proper lymphatic valve development, yet it remains unclear how lymphatic endothelial cells (LECs) sense and decode mechanical signals. In this study, we identify the cell guidance semaphorin receptor plexin D1 (PLXND1) as a lymphatic mechanosensor required for lymphatic valve morphogenesis. Conditional genetic ablation of Plxnd1 in LECs caused major defects in lymphatic valve dev

Vascular mechanoreceptor magnetic activation, hemodynamic evidence and potential clinical outcomes.

There is sufficient proof that static magnetic fields (SMFs) of different parameters have a significant effect on the cardiovascular system. The sometimes contradictory, opposite-directional nature of SMF's hemodynamic effect generates uncertainty; therefore, an explanation of the underlying mechanisms is required. Following SMF selective carotid baroreceptors or microvascular net exposure, both high and low blood pressure (BP)/vascular tone starting conditions showed a return to normal. Beyond the previous descriptions of SMF's simple hemodynamic results, the current study aims to clarify the

Semicircular Canals Circumvent Brownian Motion Overload of Mechanoreceptor Hair Cells.

Vertebrate semicircular canals (SCC) first appeared in the vertebrates (i.e. ancestral fish) over 600 million years ago. In SCC the principal mechanoreceptors are hair cells, which as compared to cochlear hair cells are distinctly longer (70 vs. 7 μm), 10 times more compliant to bending (44 vs. 500 nN/m), and have a 100-fold higher tip displacement threshold (< 10 μm vs. <400 nm). We have developed biomechanical models of vertebrate hair cells where the bundle is approximated as a stiff, cylindrical elastic rod subject to friction and thermal agitation. Our models suggest that the above differ

Intracellular characterization of identified sensory cells in a new spider mechanoreceptor preparation.

1. We have developed an isolated mechanoreceptor-organ preparation in which the intact sensory structures are available for mechanical stimulation and electrical recording. The anterior lyriform slit sense organ on the patella of the spider, Cupiennius salei Keys., consists of seven or eight cuticular slits, each innervated by a pair of large bipolar sensory neurons. The neurons are fusiform, and the largest somata are < or = 120 microns long. The innervation of the organ was characterized by light microscopy of neurons backfilled with neuronal tracers. Intracellular recording was used to meas

[Effect of stimulation of splanchnic mechanoreceptors on the activity of purkinje cells (author's transl)].

Climbing and mossy fibre activity in Purkinje cells of cat cerebellum (vermis part of lobules V and VI) were recorded by means of extracellular microelectrodes. Responses were obtained by stimulation of different types of mechanoreceptors in the gastro-intestinal tract: muscular receptors connected with C fibres, peritoneal movement receptors connected with A gamma delta or B fibres, Pacinian corpuscules connected with A beta fivres. The characteristics of the splanchnic fibre projections on this cerebellar area were defined. Splanchno-somatic, splanchno-visceral, and splanchno-cortical conver

Properties of intraoral mechanoreceptors represented in the mesencephalic nucleus of the fifth nerve in the cat.

1. The activity of neurones in the mesencephalic nucleus of the fifth nerve that respond to forces applied to the teeth were recorded using extracellular microelectrodes; the properties of these neurones have been studied. 2. Electrophysiological evidence consistent with the view that primary afferent intraoral mechanoreceptor fibres have their cell bodies in the trigeminal mesencephalic nucleus is presented. 3. Two groups of intraoral mechanoreceptor neurones were found. The first group, the periodontal mechanoreceptor neurones, which have been described by previous workers, responded to elec
