Photochemically Mediated Toluene Oxidation through a Copper Complex.

A method is described to photochemically oxidize toluene selectively to benzaldehyde, an essential compound in the chemical industry. Copper(I) complexes with different ligands were applied in combination with [Ru(bipy)3 ](PF6 )2 and dioxygen as the oxidant. As a result, a "dioxygen adduct" copper complex, for example, a peroxido complex, is formed as the active species. The copper(II) complex obtained after oxidation can be photochemically reduced to the starting copper(I) species, and the process can be repeated continuously. The ligand tris(2-methylpyridyl)amine (tmpa) led to the highest co

Inflammasome Complexes: Crucial mediators in osteoimmunology and bone diseases.

Inflammasome complexes play a vital role in the inflammatory response by producing interleukin-1β and inducing cell pyroptosis. During the long period of co-evolution of the skeletal and immune systems, inflammasome complexes bridged the bone-immune interaction, which is crucial for osteoimmunology and bone diseases. Additionally, the M1/M2 ratio, the Th17/Treg ratio, and osteoclastogenesis are regulated by inflammasomes in multiple bone diseases. Particularly, based on different disease features and inflammasome functions, osteoclastogenesis can be categorized as site-specific inflammation-re

The Role of Ate Complexes in the Copper-Mediated Trifluoromethylation of Alkynes.

Trifluoromethylation reactions have recently received increased attention because of the beneficial effect of the trifluoromethyl group on the pharmacological properties of numerous substances. A common method to introduce the trifluoromethyl group employs the Ruppert-Prakash reagent, that is, Si(CH3 )3 CF3 , together with a copper(I) halide. We have applied this method to the trifluoromethylation of aromatic alkynes and used electrospray-ionization mass spectrometry to investigate the mechanism of these reactions in tetrahydrofuran, dichloromethane, and acetonitrile as well as with and withou

Hereditary nephritis mimicking immune complex-mediated glomerulonephritis.

The defining ultrastructural features of hereditary nephritis are "basket weave" lamellation or thinning of glomerular basement membranes. Electron-dense deposits are not seen and immunofluorescence (IF) is generally negative. In this study, we report 5 cases of hereditary nephritis in which substantial amounts of glomerular electron-dense deposits were identified on electron microscopy, with corresponding positive IF staining in 4 cases, suggesting immune complex-mediated glomerulonephritis. However, no case had histological evidence of glomerular endocapillary or extracapillary proliferation

The MRN complex: coordinating and mediating the response to broken chromosomes.

The MRE11-RAD50-NBS1 (MRN) protein complex has been linked to many DNA metabolic events that involve DNA double-stranded breaks (DSBs). In vertebrate cells, all three components are encoded by essential genes, and hypomorphic mutations in any of the human genes can result in genome-instability syndromes. MRN is one of the first factors to be localized to the DNA lesion, where it might initially have a structural role by tethering together, and therefore stabilizing, broken chromosomes. This suggests that MRN could function as a lesion-specific sensor. As well as binding to DNA, MRN has other r

Koebnerization as a cutaneous manifestation of immune complex-mediated vasculitis.

Two unusual examples of the cutaneous manifestations of vasculitis are presented. In both cases lesions occurred on previously traumatized skin and on normal skin of the dependent areas. Lesional skin biopsy specimens obtained from the koebnerized sites and from the other dependent sites revealed evidence of vascular injury in both patients. A diagnosis of leukocytoclastic vasculitis was made in one patient and pityriasis lichenoides et varioliformis acuta in the other. Direct immunofluorescence microscopy of lesional skin specimens from both patients demonstrated dermal vascular immune deposi

Immune complex-mediated rheumatic diseases: the evidence and the enigmas.

Clearly, many of the connective tissue disorders are intimately associated with either focal tissue evidence of immune complex deposition or markedly elevated levels of complexes during acute disease activity. In many diseases, such as systemic lupus erythematosus or rheumatoid arthritis, the immune complexes have been firmly established as basic elements in pathogenesis. In other disorders, however, the presence or measurable elevation of circulating immune complex materials may represent an epiphenomenon that reflects tissue injury secondary to the underlying inflammatory process. Much more

Immune-complex mediated colitis in rabbits. An experimental model.

An experimental colitis in rabbits is described, following the intravenous injection of preformed immune complexes of human serum albumin (HSA) and anti-HSA into non-sensitised rabbits. Tissue damage was localised to the colon by the Auer technique of inducing local non-specific inflammation, by the rectal instillation of dilute formalin. Formalin alone gave transient changes that reverted to normal within 24 hours. In rabbits given intravenous immune complexes formed in antigen-excess, a severe colitis was initiated, with histological features including mucosal ulceration, mixed inflammatory

Tracing the paths: a systematic review of mediators of complex trauma and complex post-traumatic stress disorder.

UNLABELLED: Complex trauma is associated with complex-posttraumatic stress disorder (CPTSD). While dissociative processes, developmental factors and systemic factors are implicated in the development of CPTSD, there are no existing systematic reviews examining the underlying pathways linking complex trauma and CPTSD. This study aims to systematically review evidence of mediating factors linking complex trauma exposure in childhood (birth to eighteen years of age) and subsequent development of CPTSD (via self-reports and diagnostic assessments). All clinical, at-risk and community-sampled artic

Distinct but milder phenotypes with choreiform movements in siblings with compound heterozygous mutations in the transcription preinitiation mediator complex subunit 17 (MED17).

Two siblings born to non-consanguineous parents showed nystagmus and sudden opistotonic posturing from the early infancy, and subsequent developmental delay and marked choreiform movements with hypotonia in the childhood. The brother had a mild postnatal microcephaly. Brain MRI of the sister showed mild delay of myelination, dilated anterior horn and mild cerebellar atrophy. Whole exome sequencing (WES) revealed compound heterozygous mutations in MED17 gene in both siblings: c.1013-5A>G and c.1484T>G mutations transmitted from their father and mother, respectively. The c.1013-5A>G mutation cau

Pivotal role for a tail subunit of the RNA polymerase II mediator complex CgMed2 in azole tolerance and adherence in Candida glabrata.

Antifungal therapy failure can be associated with increased resistance to the employed antifungal agents. Candida glabrata, the second most common cause of invasive candidiasis, is intrinsically less susceptible to the azole class of antifungals and accounts for 15% of all Candida bloodstream infections. Here, we show that C. glabrata MED2 (CgMED2), which codes for a tail subunit of the RNA polymerase II Mediator complex, is required for resistance to azole antifungal drugs in C. glabrata. An inability to transcriptionally activate genes encoding a zinc finger transcriptional factor, CgPdr1, a

[The role of the AP2 Adaptor Protein Complex in clathrin-mediated endocytosis].

This work presents an overall characterization of the endocytosis process, including its various forms, stages, and biological significance. Particular emphasis is placed on clathrin-mediated endocytosis (CME) and the adaptor protein complexes AP1–AP5 that participate in this pathway. Their structure, subcellular localization, function, as well as the disorders and diseases associated with them are addressed. The main attention is devoted to the AP2 complex, which is indispensable in CME. The conformational changes of this complex and their importance in the initiation and progression of clath

SARS-CoV-2 Immune Complex-Mediated Neutrophil Activation.

Understanding disease pathogenesis is essential to developing therapies in patients with infections that cause critical illness. Herein, we show that SARS-CoV-2-specific antibody levels and markers of neutrophil activation are associated with disease severity in patients hospitalized with COVID-19. We also provide a link between the adaptive and innate immune response by demonstrating an association between antibody levels and multiple markers of neutrophil degranulation and NETosis. We further demonstrate through a series of in vitro assays that SARS-CoV-2 antigen-antibody immune complexes ca

The complex nature of lncRNA-mediated chromatin dynamics in multiple myeloma.

Extensive genome-wide sequencing efforts have unveiled the intricate regulatory potential of long non-protein coding RNAs (lncRNAs) within the domain of haematological malignancies. Notably, lncRNAs have been found to directly modulate chromatin architecture, thereby impacting gene expression and disease progression by interacting with DNA, RNA, and proteins in a tissue- or condition-specific manner. Furthermore, recent studies have highlighted the intricate epigenetic control of lncRNAs in cancer. Consequently, this provides a rationale to explore the possibility of therapeutically targeting

Immune complex-mediated neutrophil activation in patients with polymyalgia rheumatica.

OBJECTIVE: Neutrophils are important in host defence. However, neutrophils are also linked to inflammation and organ damage. The purpose of this study was to assess whether markers of neutrophil activation are increased in PMR. METHODS: Levels of immune complexes (IC), calprotectin and neutrophil extracellular traps (NETs) were measured in plasma of healthy individuals (n = 30) and patients with PMR (n = 60), at flare and upon treatment with glucocorticoids using ELISA. Plasma-mediated neutrophil activation was assessed in presence of an FcγRIIA inhibitory antibody (IV.3). RESULTS: Plasma le

PHF10 subunit of PBAF complex mediates transcriptional activation by MYC.

The PBAF complex, a member of SWI/SNF family of chromatin remodelers, plays an essential role in transcriptional regulation. We revealed a disease progression associated elevation of PHF10 subunit of PBAF in clinical melanoma samples. In melanoma cell lines, PHF10 interacts with MYC and facilitates the recruitment of PBAF complex to target gene promoters, therefore, augmenting MYC transcriptional activation of genes involved in the cell cycle progression. Depletion of either PHF10 or MYC induced G1 accumulation and a senescence-like phenotype. Our data identify PHF10 as a pro-oncogenic mechani
