Silage fermentation characteristics and microbial diversity of alfalfa (Medicago sativa L.) in response to exogenous microbiota from temperate grasses.

The objective of this study was to explore the microbiological factors that cause the difference in silage fermentation characteristics between grass and legume. Specifically, the effects of epiphytic microbiota from alfalfa, oat and Italian ryegrass on ensiling characteristics and microbial community of alfalfa were assessed. By γ-ray irradiation sterilization and microbiota transplantation technology, the sterile alfalfa was inoculated as follows: (i) aseptic water (STAL); (ii) epiphytic bacteria from alfalfa (ALAL); (iii) epiphytic bacteria from oat (ALOT); (iv) epiphytic bacteria from Ital

Chloroform extract of alfalfa (Medicago sativa) inhibits lipopolysaccharide-induced inflammation by downregulating ERK/NF-κB signaling and cytokine production.

Alfalfa (Medicago sativa L.) is commonly used as a traditional medicine and functional food. This study investigated the anti-inflammatory potential of alfalfa and the mechanisms involved. The chloroform extract of alfalfa aerial parts inhibited lipopolysaccharide (LPS)-stimulated immune responses more than ether, butanol, or water soluble extracts. Treatment with 1 μg/mL LPS increased nitrite concentrations to 44.3 μM in RAW267.4 macrophages, but it was reduced to 10.6 μM by adding 100 μg/mL chloroform extract. LPS treatment also increased the concentrations of tumor necrosis factor-α, interl…

Passage and digestibility of lucerne (Medicago sativa) hay in Japanese sika deer (Cervus nippon) and sheep under restricted feeding.

The mean retention time (MRT) of stained hay through the whole digestive tract and its digestibility were measured in Japanese sika deer (Cervus nippon) and were compared with those in sheep when lucerne (Medicago sativa) hay was fed at 10, 20 and 30 g/kg body-weight. The recoveries in faeces of plastic particles with five specific gravities were also measured in deer. MRT for deer was significantly shorter than that for sheep at 10 and 20 g/kg feeding levels. The digestibilities of dry matter, organic matter, and neutral-detergent fibre were significantly lower for deer than for sheep at 30, …

Contrasting responses of alfalfa (Medicago sativa) accessions to combined water and nutrient stress: integrated morphological, biochemical, and molecular analyses.

Combined water deficit and nutritional deficiencies pose significant challenges to the productivity of alfalfa (Medicago sativa). We evaluated natural variation in 14 Tunisian accessions under combined stress, and identifed two contrasting genotypes, Radhouane (tolerant) and Kettana 1 (sensitive), for detailed analysis. Multivariate analyses, including Principal Component Analysis and clustering, showed clear separation of the two genotypes based on growth traits, with Radhouane showing enhanced root biomass and relative water content, while Kettana 1 exhibited reduced stem length. Biochemical…

Physiological and transcriptomic analyses reveal the critical role of MsERF15 in negatively regulating cadmium stress tolerance in Alfalfa (Medicago sativa).

Cadmium (Cd) is a highly toxic heavy metal that poses a serious threat to plant growth and the quality of agricultural and animal products. Therefore, elucidating the response and regulatory mechanisms of plants to Cd stress is essential for controlling the safety of agricultural and animal products at the source. In this study, we used a Cd-tolerant alfalfa cultivar, Medicago sativa 'Zhaodong' (ZD), and a Cd-sensitive cultivar, M. sativa 'Zhongmu No.1' (ZM), to investigate their differential responses to Cd stress in terms of morphology, photosynthetic function, and oxidative damage through c…

In vitro Incubation with Rumen Fluid and Histological Staining of Select Cell Wall Components in Alfalfa (Medicago sativa L.) Stems.

The accumulation of cell wall components as plants grow limits forage nutrition for ruminants. In particular, lignin deposition in plant tissues impedes the breakdown of the cell wall carbohydrates cellulose and hemicellulose, thereby reducing the amount of energy available from forage. The highly digestible pectin component of the cell wall, however, is readily available for degradation. Information about lignin and pectin deposition in different stem cell types may provide mechanistic insights into the factors underlying variation in stem digestibility among alfalfa lines. In vitro incubatio

Metabolomics-Based Analysis Reveals Flavonoid-Mediated Defence in Alfalfa (Medicago sativa 'Gannong No. 5') Against Pea Aphid (Acyrthosiphon pisum) Infestation.

The pea aphid (Acyrthosiphon pisum) is a destructive phloem-feeding pest of alfalfa. Its overreliance on chemical insecticides necessitates the development of sustainable, plant-based resistance strategies. Plant flavonoids are important in induced defense, but their dynamic responses to aphid feeding and subsequent insecticidal effects remain unclear. Using the highly resistant alfalfa variety 'Gannong No. 5' (GN5), this study integrated behavioral assays, performance bioassays, untargeted and targeted metabolomics, and exogenous flavonoid feeding assays to investigate how pea aphid infestati

Time-Course Transcriptomic Analysis Identifies MsTIFY11B as a Promising Candidate Regulator of Salt-Alkali Tolerance in Alfalfa (Medicago sativa L.).

Soil salinization severely limits forage crop productivity, yet the regulatory networks that govern salt stress adaptation in alfalfa, a moderately salt-tolerant leguminous forage, remain largely unexplored. Here, we examined the physiological and transcriptomic dynamics of alfalfa leaves under 200 mM NaCl stress across three time points. Salt stress induced a progressive elevation of the Na+/K+ ratio, biphasic activation of antioxidant enzymes and concurrent accumulation of malondialdehyde. Time-course RNA-seq analysis identified 3631 differentially expressed genes (DEGs) and 132 core salt-re

Medicago Sativa Defensin1 as a tumor sensitizer for improving chemotherapy: translation from anti-fungal agent to a potential anti-cancer agent.

Plant defensins including Medicago Sativa defensin 1 (MsDef1) are cysteine-rich antifungal peptides which are known for potent broad-spectrum antifungal activity against bacterial or fungal pathogens of plants. The antimicrobial activities of these cationic defensins are attributed to their capacity to bind to cell membranes to create potentially structural defects tin the cell membranes to interact with intracellular target (s) and mediates cytotoxic effects. Our earlier work identified Glucosylceramide (GlcCer) of fungus F. graminearum as a potential target for biological activity. Multi-dru…

Alfalfa Plants (Medicago sativa L.) Expressing the 85B (MAP1609c) Antigen of Mycobacterium avium subsp. paratuberculosis Elicit Long-Lasting Immunity in Mice.

Mycobacterium avium subsp. paratuberculosis (MAP) is the etiological agent of Paratuberculosis, a contagious, untreatable, and chronic granulomatous enteritis that results in diarrhea, emaciation, and death in farmed ruminants (i.e., cattle, sheep, and goats). In this study, the Ag85B antigen from MAP was expressed in transgenic alfalfa as an attractive vaccine candidate. Agrobacterium-mediated transformation allowed the rescue of 56 putative transformed plants and transgenesis was confirmed in 19 lines by detection of the Ag85B gene (MAP1609c) by PCR. Line number 20 showed the highest Ag85B e

Whole genome sequencing and annotation of a non-pathogenic Fusarium brachygibbosum TZ1MST22 strain isolated from Medicago sativa in Southern Tunisian oases.

Fusarium sambucinum species complex (FSAMSC) includes not only major mycotoxin-producing pathogens but also non-pathogenic endophytic species that can promote plant growth and stress tolerance. However, the molecular mechanisms underlying these contrasting lifestyles remain poorly understood, underscoring the need for deeper molecular studies. A non-pathogenic endophytic Fusarium strain, TZ1MST22, isolated from Medicago sativa plants in the Tozeur region of Southern Tunisia, was sequenced using the Illumina NovaSeq 6000 sequencing technology. Here, we report a 39.8 Mb draft genome of TZ1MST22,…

Genome-Wide Identification of the PIN-LIKES (PILS) Gene Family in Alfalfa (Medicago sativa L.) and Its Expression Analysis Under Abiotic Stresses.

The PIN-LIKES (PILS) gene family is crucial for regulating auxin homeostasis and stress adaptation in plants; nevertheless, a comprehensive study on this family in alfalfa (Medicago sativa) remains insufficient. This research found 46 MsPILS genes within the tetraploid alfalfa genome and categorized them into four subfamilies. The genes are irregularly allocated throughout 16 chromosomes, with tandem duplications acting as a primary catalyst for family expansion. Analysis indicated that all MsPILS proteins contain the conserved Mem_trans domain. The promoter study revealed that MsPILS genes ha…
