GLK1 plays a positive role in regulating plant tolerance to alkali stress and salt-alkali mixed stress.

The Golden2-like 1 (GLK1) gene regulates photosynthesis and is associated with drought tolerance and cold tolerance. However, no report has determined the role of GLK1 in alkali tolerance. In this study, we found that MsGLK1 responds to alkali and salt-alkali mixed stresses in alfalfa (Medicago sativa) and carried out detailed functional analysis of GLK1 in the model plants Arabidopsis and Medicago truncatula. In Arabidopsis, the atglk1/2 double mutant exhibited reduced tolerance to these two stress treatments compared to the wild type. Conversely, overexpression of MsGLK1 in Arabidopsis displ

Functional divergence of plant SCAR/WAVE proteins is determined by intrinsically disordered regions.

Dynamic actin cytoskeleton reorganization enables plant developmental processes requiring polarized transport such as root hair and leaf trichome formation. The SCAR/WAVE complex plays a crucial role in regulating these dynamics through ARP2/3-mediated actin branching. SCAR/WAVE genes occur as small families across a wide range of plant species, but whether and how they fulfill different functions remains unclear. We use a systematic chimera approach to define the differential functionality of two closely related Medicago truncatula SCAR proteins in plant development. We show that SCAR/WAVE co

Calmodulin-Like Protein MfCML50 Interacts With Carveol Dehydrogenase in Medicago falcata to Regulate Cold Tolerance Through Mediating ROS Homeostasis.

Low temperature triggers Ca2+ signalling and reprogramming of gene expression and metabolism in plants. However, how the Ca2+ signal is transduced to the downstream metabolic pathways remains unknown. The involvement of a cold-induced calmodulin-like protein, MfCML50, from Medicago falcata in regulation of cold tolerance was examined in the present study. The interaction of MfCML50 with isopiperitenol/carveol dehydrogenase (MfCDH) was identified, which was dependent upon Ca2+ and led to activated activity of MfCDH. MfCDH catalysed the production of carvone from carveol, with Km value of 10.25

Estimating Rhizobial Fitness During Legume Symbiosis: Enriching Viable Undifferentiated Bacteria from Root Nodules.

Advances in understanding the evolutionary ecology of the rhizobia-legume mutualism have been constrained by methodological limitations in efficiently measuring relative strain frequencies alongside measurements of absolute population sizes of rhizobia living in nodules. To examine strain competition in natural and agricultural ecosystems that harbor multiple strains of rhizobia, an increasing number of manipulative and observational studies have recently begun to examine dozens or hundreds of strains simultaneously. Assessing the competitive fitness of multiple strains in legume nodules requi

ATP-Binding Cassette G Transporters SGE1 and MtABCG13 Control Stigma Exsertion.

Stigma exsertion is an important agricultural trait that facilitates the application of heterosis in crop breeding. Although several quantitative trait loci associated with stigma exsertion have been fine-mapped or cloned, the underlying genetic basis, particularly in legumes, remains unclear. In this study, we identified and characterized the exserted stigma mutant stigma exsertion1 (sge1) in the model legume Medicago truncatula The exserted stigma phenotype of sge1 is mainly caused by physical interaction between floral organs, in which normal petal and stamen elongation are inhibited due to

The APT1-NACsa3-GGP1 Module Enhances Salt Tolerance and Regulates Ascorbic Acid Biosynthesis in Medicago.

The unclear salt tolerance mechanism of alfalfa (Medicago sativa) is a major constraint to its molecular design breeding. Here, we identified a novel ATAF subfamily transcription factor, MtNACsa3, in the closely related diploid Medicago truncatula, and elucidated its function in salt tolerance. Phenotypic analysis of the MtNACsa3 mutants and overexpression lines showed its positive role in salt tolerance. Under salt or oxidative stress, MtNACsa3 relocates from the plasma membrane to the nucleus mediated by the de-S-acylation of Acyl-Protein Thioesterase 1 (MtAPT1). MtNACsa3 up-regulates the ex

Integrated bioinformatic and physiological analyses reveal the pivotal role of hydrogen sulfide in enhancing low-temperature tolerance in alfalfa.

Hydrogen sulfide (H2 S) is an important gaseous signal molecule that regulates plant growth and stress resistance. However, research on the H2 S synthase (HSase) genes is still limited in the model legume plant Medicago truncatula Gaertn. In the present study, a total of 40 HSase family members were first identified and analyzed in the M. truncatula genome, and these genes distributed across eight chromosomes and were clustered into five groups (I-V) based on their conserved gene structures and protein motifs. Expression analysis revealed that the MtHSase genes were expressed in all the tested

Spin-orbit coupling mediated transverse spin mode rotation in a uniaxial crystal.

Mutualism breakdown can be prevented if partner species preferentially select and reward partners that provide greater benefit. We examined these two components using the legume Medicago truncatula and its nitrogen-fixing symbiont Sinorhizobium meliloti. First, we re-analysed data from 202 accessions to show significant genetic variation in the capacity of Medicago to restrict strain diversity, finding that hosts with shorter nodules were more selective. A genome-wide association study on host selectivity identified genes including the hormone leginsulin, pectin degradation, multidrug and toxi

A new method to visualize CEP hormone-CEP receptor interactions in vascular tissue in vivo.

C-TERMINALLY ENCODED PEPTIDEs (CEPs) control diverse responses in plants including root development, root system architecture, nitrogen demand signalling, and nutrient allocation that influences yield, and there is evidence that different ligands impart different phenotypic responses. Thus, there is a need for a simple method that identifies bona fide CEP hormone-receptor pairings in vivo and examines whether different CEP family peptides bind the same receptor. We used formaldehyde or photoactivation to cross-link fluorescently tagged group 1 or group 2 CEPs to receptors in semi-purified Medi

Co-option of a conserved lateral-root development program by symbiotic signals.

Nod factors (NFs) are microbial signals originally identified for their key role in the nitrogen-fixing root nodule symbiosis in legumes. Beyond symbiosis, NFs also possess a conserved capacity to induce lateral-root formation across diverse plant species, including non-legumes. It is now well established that the nodule organogenesis program has co-opted several molecular mechanisms involved in root development, which raises the question of the developmental pathway controlled by NFs to trigger lateral-root formation and how it overlaps with nodule organogenesis in legumes. In Medicago trunca

Regulation of enteric endophytic bacterial colonization by plant defenses.

Bacterial endophytes reside within the interior of plants without causing disease or forming symbiotic structures. Some endophytes, such as Klebsiella pneumoniae 342 (Kp342), enhance plant growth and nutrition. Others, such as Salmonella enterica serovar Typhimurium (S. typhimurium), are human pathogens that contaminate raw produce. Several lines of evidence are presented here to support the hypothesis that plant defense response pathways regulate colonization by endophytic bacteria. An ethylene-insensitive mutant of Medicago truncatula is hypercolonized by Kp342 compared to the parent genotyp

MtING2 encodes an ING domain PHD finger protein which affects Medicago growth, flowering, global patterns of H3K4me3, and gene expression.

Flowering of the reference legume Medicago truncatula is promoted by winter cold (vernalization) followed by long-day photoperiods (VLD) similar to winter annual Arabidopsis. However, Medicago lacks FLC and CO, key regulators of Arabidopsis VLD flowering. Most plants have two INHIBITOR OF GROWTH (ING) genes (ING1 and ING2), encoding proteins with an ING domain with two anti-parallel alpha-helices and a plant homeodomain (PHD) finger, but their genetic role has not been previously described. In Medicago, Mting1 gene-edited mutants developed and flowered normally, but an Mting2-1 Tnt1 insertion

Molecular Insights into the Heme-Binding Potential of Plant NCR247-Derived Peptides.

Heme is involved in many critical processes in pathogenic bacteria as iron acquisition by these microorganisms is achieved by either direct uptake of heme or use of heme-binding proteins called hemophores. Exploring the underlying mechanisms on a molecular level can open new avenues in understanding the host-pathogen interactions. Any imbalance of the heme concentration has a direct impact on the bacterial growth and survival. Thus, heme-regulated proteins that are involved in heme homeostasis poise to be promising targets for research. Similarly, naturally occurring compounds, including cyste

Nodulation Signaling Pathway 2: A Proposed Multifunctional Signaling Hub for Coordinating Plant Architecture, Rhizosphere Chemistry, and Symbiotic Associations Under Nutrient Stress.

Under nutrient stress, plants adjust shoot branching, root remodeling, and symbiotic associations to optimize survival and growth. The GRAS-type transcription factor NODULATION SIGNALING PATHWAY 2 (NSP2) participates in these responses through context-dependent interactions with partners: NSP1, MYB40, and SLR1, and is indirectly repressed by SMAX1 via NSP1 and SLR1. NSP2 functions differ among species: it directly binds DNA in rice (Oryza sativa) strigolactone (SL) biosynthesis, whereas in M. truncatula it acts as a co-regulator in nodulation and flavonoid regulation, with NSP1 or MYB40 provid

Alfalfa varieties can weakly choose beneficial nitrogen-fixing bacteria from a population isolated from a single field.

In natural and agricultural systems, legumes recruit rhizobia from diverse soil populations to fix nitrogen in root nodules. A few legumes, including the model legume Medicago truncatula, can select and enrich beneficial rhizobia. Here, we investigated whether its perennial relative, Medicago sativa (alfalfa), a globally important forage crop, also possesses this ability. We developed a genetically variable collection of 117 Sinorhizobium meliloti strains sampled from three field-grown alfalfa varieties, performed multi-strain and single-strain inoculations in a nitrogen-free greenhouse experi

Priming Seeds with Indole and (Z)-3-Hexenyl Acetate Enhances Resistance Against Herbivores and Stimulates Growth.

A striking feature of plant ecology is the ability of plants to detect and respond to environmental cues such as herbivore-induced plant volatiles (HIPVs) by priming or directly activating defenses against future herbivores. However, whether seeds also respond to compounds that are common constituents of HIPV blends and initiate future plant resistance is unknown. Considering that seeds depend on other environmental cues to determine basic survival traits such as germination timing, we predicted that seeds exposed to synthetic constituents of HIPV blends would generate well-defended plants. We

Structural basis of methotrexate and pemetrexed action on serine hydroxymethyltransferases revealed using plant models.

Serine hydroxymethyltransferases (SHMTs) reversibly transform serine into glycine in a reaction accompanied with conversion of tetrahydrofolate (THF) into 5,10-methylene-THF (5,10-meTHF). In vivo, 5,10-meTHF is the main carrier of one-carbon (1C) units, which are utilized for nucleotide biosynthesis and other processes crucial for every living cell, but hyperactivated in overproliferating cells (e.g. cancer tissues). SHMTs are emerging as a promising target for development of new drugs because it appears possible to inhibit growth of cancer cells by cutting off the supply of 5,10-meTHF. Methot

The pgip family in soybean and three other legume species: evidence for a birth-and-death model of evolution.

BACKGROUND: Polygalacturonase-inhibiting proteins (PGIPs) are leucine-rich repeat (LRR) plant cell wall glycoproteins involved in plant immunity. They are typically encoded by gene families with a small number of gene copies whose evolutionary origin has been poorly investigated. Here we report the complete characterization of the full complement of the pgip family in soybean (Glycine max [L.] Merr.) and the characterization of the genomic region surrounding the pgip family in four legume species. RESULTS: BAC clone and genome sequence analyses showed that the soybean genome contains two pgip

Production of the Plant Hormone Auxin by Salmonella and Its Role in the Interactions with Plants and Animals.

The ability of human enteric pathogens to colonize plants and use them as alternate hosts is now well established. Salmonella, similarly to phytobacteria, appears to be capable of producing the plant hormone auxin via an indole-3-pyruvate decarboxylase (IpdC), a key enzyme of the IPyA pathway. A deletion of the Salmonella ipdC significantly reduced auxin synthesis in laboratory culture. The Salmonella ipdC gene was expressed on root surfaces of Medicago truncatula. M. truncatula auxin-responsive GH3::GUS reporter was activated by the wild type Salmonella, and not but the ipdC mutant, implying

Structural basis for size-selective perception of chitin in plants.

Plants detect microbes through pattern recognition receptors that perceive conserved microbial surface motifs known as microbe-associated molecular patterns (MAMPs). Lysin motif (LysM) receptors recognize and mediate downstream responses to chitinous MAMPs. In this study, we elucidated the mechanism for the specific recognition of long-chain chitin oligomers and identified a hallmark bridge domain characteristic of the CHITIN PERCEPTION (CHIP) receptor class. Structural analysis of receptor-ligand complexes, biochemistry, and in planta functional studies of inhibitory nanobodies revealed the m
