Neuroprotective effects of melanins and melanin derivatives from Calvatia Craniiformis and Xylaria plebeja against Parkinsonian toxins.

Fungal melanins have remarkable bioactivities, but their practical applications are limited, partly due to their hydrophobic nature. Melanin modification with amino acids was reported to improve their water solubility. This research aimed to extract, and modify the melanins from Calvatia craniiformis and Xylaria plebeja with arginine, and to evaluate their neuroprotective effects against Parkinsonian toxins. MTT assay showed that all melanins and melanin derivatives could protect SH-SY5Y neuroblastoma cells from 1 mM H2O2. Xylaria plebeja melanin and melanin derivative at 100 µg/mL increased t

Reply to "From melanin-positive to melanin-actionable: Spatial pigment phenotyping for precision melanoma phototherapy".

In a recent Letter to the Editor, Dutt et al. have presented interesting arguments to improve the application of different therapies to melanoma. Although sharing most of their arguments, we feel it is necessary to clarify better some points in relation to the photothermal therapy of cancer applied to melanomas. In particular, when it has to do with continuous photothermal excitation. In this case, in our opinion, the need to assess in depth the features of the melanoma target relaxes, because heat will diffuse from the heating particles and affect the whole treated volume in a more or less ho

Plumage colour mutations and melanins in the feathers of the Japanese quail: a first comparison.

The absorbance of melanin content from dorsal feathers was compared between wild-type Japanese quail and nine other quail plumage colours determined by single mutations in one of seven genes: extended brown (MC1R), yellow (ASIP), silver (MITF), lavender (MLPH), roux (TYRP1), imperfect albinism (SLC45A2) and rusty. As compared with wild-type quail, all mutations but extended brown decreased total melanins. The largest decrease was observed in quail with one of the dilution mutations at TYRP1, MLPH or SLCA45A2. No difference in eumelanins was found between the 10 plumage colours. Despite visible

Experimental autoimmune anterior uveitis (EAAU). III. Induction by immunization with purified uveal and skin melanins.

The pathogenicity of uveal tissue and melanin has been a controversial subject for a long time. The present new approach has elucidated some of the problems. Melanin granules have been extracted from bovine choroid, iris, hair and skin, and from human, monkey and rabbit choroid. The melanin granules have further been purified by detergent extractions, and are free from pathogenic retinal antigens. Lewis rats immunized with microgram doses bovine choroidal or iris melanin-protein (in Freund's complete adjuvant or Hunter's adjuvant, combined with pertussis toxin) develop severe experimental auto

Shedding light on melanins within in situ human eye melanocytes using 2-photon microscopy profiling techniques.

Choroidal melanocytes (HCMs) are melanin-producing cells in the vascular uvea of the human eye (iris, ciliary body and choroid). These cranial neural crest-derived cells migrate to populate a mesodermal microenvironment, and display cellular functions and extracellular interactions that are biologically distinct to skin melanocytes. HCMs (and melanins) are important in normal human eye physiology with roles including photoprotection, regulation of oxidative damage and immune responses. To extend knowledge of cytoplasmic melanins and melanosomes in label-free HCMs, a non-invasive 'fit-free' app

Melanin- and linewidth-corrected pulse-oximetry.

Current pulse oximeters tend to overestimate arterial saturation in darkly pigmented patients, increasing occult hypoxemia rates. One proposed mechanism is spectral coloring: melanin contributes to reshaping the detected spectrum of a broad-linewidth source, making the wavelength-dependent parameters behind calibration skin-tone-dependent. Diffuse optics theory allows for a melanin- and linewidth-corrected calibration equation, which to our knowledge has never been tested in vivo. In this work, we fit the empirical linear equation, the diffuse optics theoretical equation, and its corrected for

Non-melanin pigmentation: A narrative review.

Skin colour is determined by four biochromes: oxyhaemoglobin (red), reduced haemoglobin (blue), carotenoids (yellow), and, most importantly, the amount and type of melanin produced and its distribution in the skin. Pigmentary disorders comprise hypopigmentary, hyperpigmentary, and non-melanin pigmentary diseases. While the former two are discussed frequently, non-melanin pigmentary conditions are not unusual. Based on the aetiology and colour of pigmentation, these disorders can be categorised into blue-coloured lesions, hypopigmented lesions due to vascular aetiology, yellow-coloured lesions

[Distribution of melanin in the larvae of Schizothorax o'connori].

We examined the distribution of melanin during the development of the larvae of Schizothorax o'connori except the eyes with histological method. The results showed that after hatching, the appearance sequence of melanin in different organs were following an order of the outer membrane of neurocranium, the pericardial cavity and the dorsal skin, and the peritoneum and the spinal cord. Specifically, melanin appeared in the outer membrane of neurocranium around 5 DAH (days after hatching), in the pericardial cavity and the back skin at 7 DAH, and in the peritoneum and the spinal cord at 10 DAH. The physiology of melanin deposition in health and disease.

Eumelanin is the major pigment responsible for human skin color. This black/brown pigment is localized in membrane-bound organelles (melanosomes) found in specialized cells (melanocytes) in the basal layer of the epidermis. This review highlights the steps involved in melanogenesis in the epidermis and the disorders in skin pigmentation that occur when specific steps critical for this process are defective. Melanosomes, which contain tyrosinase, a major enzyme involved in melanin synthesis, develop through a series of steps in the melanocyte. They are donated from the melanocyte dendrites to t

Suppression of melanin production by expression of HSP70.

Skin hyperpigmentation disorders due to abnormal melanin production induced by ultraviolet (UV) irradiation are both a clinical and cosmetic problem. UV irradiation stimulates melanin production in melanocytes by increasing intracellular cAMP. Expression of heat shock proteins (HSPs), especially HSP70, is induced by various stressors, including UV irradiation, to provide cellular resistance to such stressors. In this study we examined the effect of expression of HSP70 on melanin production both in vitro and in vivo. 3-Isobutyl-1-methylxanthine (IBMX), a cAMP-elevating agent, stimulated melanin

Melanin-concentrating hormone and immune function.

To date, melanin-concentrating hormone (MCH) has been generally considered as peptide acting almost exclusively in the central nervous system. In the present paper, we revise the experimental evidence, demonstrating that MCH and its receptors are expressed by cells of the immune system and directly influence the response of these cells in some circumstances. This therefore supports the idea that, as with other peptides, MCH could be considered as a modulator of the immune system. Moreover, we suggest that this could have important implications in several immune-mediated disorders and affirm th

Role of melanin in the pathogenesis of cutaneous sporotrichosis.

In this study, we evaluate the clinical, histopathological and ultra structural aspects of the infection by field strain (MEL(+)) and mutated strain (MEL(-)) of Sporothrix schenckii. The development of sporotrichosis at the inoculation sites was similar for both groups of animals after the second week of the experimental period. Statistical differences appeared only at weeks 3 and 5. The pigmented isolate had greater tissue invasive capacity, promoting the formation of multifocal granulomas, whereas the albino isolate induced an enhanced inflammatory response, restraining the agent to the core

Melanin pigmentary disorders: treatment update.

Most of the melanin pigmentary disorders are cosmetically important and have a strong impact on the quality of life of affected individuals. This article examines recent advances in the treatment of melanin pigmentary disorder including hypermelanosis and hypomelanosis. The development of laser technologies has completed the use of the increasing number of bleaching agents in treating hyperpigmented lesions. The treatment of hypomelanotic disorders is still often disappointing, but new therapeutic options provide encouraging results.

Melanin pigmented oncocytic metaplasia of the nasopharynx.

A case of melanin pigmented oncocytic metaplasia is reported. A 62-year-old man presented with a history of discomfort of the ear of a 2 weeks' duration. Nasoscopic examination revealed a black nodule of about 5mm at the left Eustachian opening, and several black spots were discovered around the bilateral torus tubarius. The nodule was biopsied to determine the histology. Microscopically, there were oncocytic cells with abundant pigmented granules showing glandular pattern. Such a pigmented variant of benign oncocytic lesion is very rare.

Laccase and melanin in the pathogenesis of Cryptococcus neoformans.

Cryptococcosis, caused by an encapsulated fungus, Cryptococcus neoformans, has emerged as a life threatening infection in HIV positive individuals and other immunocompromised hosts. The present review describes laccase and its product melanin as an important virulence factor of Cryptococcus neoformans and illustrates the approaches used in elucidating the pathogenesis of cryptococcosis. Characterization of the biochemical pathways leading to melanin synthesis is summarized using biochemical and biomolecular approaches. Melanin synthesis is dependent on a single copper-dependent enzyme, laccase

Melanin and the regulation of mammalian photoreceptor topography.

Melanin, or products directly associated with it, regulates the maturation of the neural retina because in hypopigmented mammals the central retina fails to develop fully. To determine whether this deficit is reflected in the distribution of photoreceptors, their topography has been studied in the retinae of normally reared pigmented and albino ferrets and animals reared under reduced light conditions. In both strains, the general distribution of rods and cones was similar to that in the cat, cone density peaking in the central retina and rod density peaking in an annulus around the area centr

Presence of melanin in the cat pineal gland.

Light- and electron-microscopic features of pigmented cells in the cat pineal gland are described. These cells were observed throughout postnatal life from the second postnatal day to the oldest cats studied (up to 13 years old). No apparent relationship was observed among the amount of pigment and the animal age or sex. Pigmented cells showed a preferential localization at the ventral surface of the pineal gland near its distal end. The pineal pigment was histochemically identified as melanin. Pineal pigment granules showed ultrastructural features similar to melanocyte melanin granules.
