Natural Product Toosendanin Suppresses the Malignant Development of Skin Melanoma by Targeting BNC2 for Degradation.

Toosendanin (TSN) is a highly potent natural anticancer compound derived from the fruit of Fructus Meliae Toosendan. In this study, TSN inhibited melanoma cell proliferation and cell-cycle progression, induced apoptosis, and suppressed metastasis in both in vitro and in vivo models. TSN is a potent PI3K inhibitor; here, we identify basonuclin 2 (BNC2) as a transcription factor critically involved in TSN's suppression of PIK3CA transcription in melanoma cells. In clinical samples and melanoma datasets, BNC2 expression is substantially elevated in skin cutaneous melanoma and correlates with poor

Toosendanin interferes with pore formation of botulinum toxin type A in PC12 cell membrane.

AIM: Botulinum neurotoxins (BoNT) abort the process of neurotransmitter release at presynaptic motor nerve terminals, causing muscle paralysis. The ability of botulinum toxin to produce its effect is dependent on the ability of the light chain to cleave the SNARE proteins associated with transmitter release. Translocation of the light chain protease through the heavy chain-formed channel is a pivotal step in the intoxication process. Toosendanin (TSN), a triterpenoid derivative extracted from a Chinese traditional medicine, has been demonstrated to be an effective cure for experimental botulis

Toosendanin induced hepatotoxicity via triggering PERK-eIF2α-ATF4 mediated ferroptosis.

Toosendanin (TSN) is the main active compound of Melia toosendan Sieb et Zucc with various bioactivities. In this study, we investigated the role of ferroptosis in TSN-induced hepatotoxicity. The characteristic indicators of ferroptosis were detected including reactive oxygen species (ROS), lipid-ROS, glutathione (GSH), ferrous ion and the expression of glutathione peroxidase 4 (GPX4), which showed that TSN caused ferroptosis in hepatocytes. The results of qPCR analysis and western blotting assay showed that TSN-induced activation of protein kinase R-like endoplasmic reticulum kinase (PERK)- e

Biological effects of toosendanin, a triterpenoid extracted from Chinese traditional medicine.

Toosendanin (TSN) is a triterpenoid extracted from Melia toosendan Sieb et Zucc, which was used as a digestive tract-parasiticide and agricultural insecticide in ancient China. TSN was demonstrated to be a selective presynaptic blocker and an effective antibotulismic agent. By interfering with neurotransmitter release through an initial facilitation followed by a subsequent depression, TSN eventually blocks synaptic transmission at both the neuro-muscular junction and central synapses. Despite sharing some similar actions with botulinum neurotoxin (BoNT), TSN has a marked antibotulismic effect

Inhibitors of intracellular signaling pathways that lead to stimulated epidermal pigmentation: perspective of anti-pigmenting agents.

Few anti-pigmenting agents have been designed and developed according to their known hyperpigmentation mechanisms and corresponding intracellular signaling cascades. Most anti-pigmenting agents developed so far are mechanistically involved in the interruption of constitutional melanogenic mechanisms by which skin color is maintained at a normal and unstimulated level. Thus, owing to the difficulty of confining topical application to a specific hyperpigmented skin area, potent anti-pigmenting agents capable of attenuating the natural unstimulated pigmentation process have the risk of leading to

Anti-Inflammatory Effect of 21α-Methylmelianol In Vitro and In Vivo via NF-κB/STAT Signaling Pathway Modulation.

Melia azedarach L., also known as the chinaberry tree, has long been used in traditional medicine to treat inflammatory conditions. In this study, we preliminarily investigated the anti-inflammatory effect of 21α-methylmelianol (MMN), a tirucallane-type triterpenoid isolated from M. azedarach, in lipopolysaccharide (LPS)-mediated RAW264.7 macrophages in vitro and in a dextran sodium sulfate (DSS)-stimulated ulcerative murine colitis model in vivo. MMN reduced LPS-stimulated nitric oxide production without inducing cytotoxicity. It significantly attenuated LPS-induced protein expression of reac

Integrated Proteomics, Biological Functional Assessments, and Metabolomics Reveal Toosendanin-Induced Hepatic Energy Metabolic Disorders.

Toosendanin (TSN), a compound from Melia toosendan, exhibits severe hepatotoxicity, which restricts its clinical application. However, the mechanism is not clear. Our previous research found that covalent modification of TSN for proteins might be a possible reason using human liver microsomes, and the glycolytic enzymes, triosephosphate isomerase 1 (TPIS) and α-enolase (ENOA), were responsible for the hepatotoxicity. In this study, we tried to prove these findings in cell and animal models by integration of proteomics, metabolomics, and biological methods. Proteomics analysis in rats showed th

Integrated analysis of microRNA and mRNA expression profiles highlights the complex and dynamic behavior of toosendanin-induced liver injury in mice.

Triterpenoid Toosendanin (TSN) exhibits a plenty of pharmacological effects in human and great values in agriculture. However, the hepatotoxicity caused by TSN or Melia-family plants containing TSN used in traditional Chinese medicine has been reported, and the mechanisms of TSN-induced liver injury (TILI) still remain largely unknown. In this study, the dose- and time-dependent effects of TSN on mice liver were investigated by an integrated microRNA-mRNA approach as well as the general toxicological assessments. As the results, the dose- and time-dependent liver injury and alterations in glob

Evaluating larvicidal, ovicidal and growth inhibiting activity of five medicinal plant extracts on Culex pipiens (Diptera: Culicidae), the West Nile virus vector.

Mosquitoes, one of the deadliest animals on the planet, cause millions of fatalities each year by transmitting several human illnesses. Synthetic pesticides were previously used to prevent the spread of diseases by mosquitoes, which was effective in protecting humans but caused serious human health problems, environmental damage, and developed mosquito pesticide resistance. This research focuses on exploring new, more effective, safer, and environmentally friendly compounds to improve mosquito vector management. Phytochemicals are possible biological agents for controlling pests and many are t

Toosendanin sensitizes hepatocyte ferroptosis via dual inhibition of Nrf2 O-GlcNAcylation and USP7-driven deubiquitination.

Toosendanin (TSN) is the key bioactive component of Melia toosendan Sieb. et Zucc known for alleviating pain and expelling roundworms, but severe hepatotoxicity limited its further application. O-GlcNAcylation is a dynamic and reversible post-translational modification which has emerged as an important regulatory mechanism in cellular response to liver injury. In this study, we investigated the effect of aberrant O-GlcNAcylation and augmented O-GlcNAc signaling via Thiamet G (TMG) on TSN-induced ferroptosis in HepaRG cells. The O-GlcNAc transferase (OGT) expression and global O-GlcNAcylation l

Comment on "Automatic rib fracture detection on postmortem CT data using deep learning".

Lopez-Melia et al. recently reported a feasibility study on automatic rib fracture detection in postmortem computed tomography (PMCT) using nnDetection, a self-configuring three-dimensional object detection framework based on deep learning (DL), and explored the impact of domain shift between clinical and postmortem datasets. This comment discusses three issues that may refine the interpretation of their findings and inform future work. First, we argue that the relationship to previous PMCT DL studies by Ibanez et al., including RiFNet and a subsequent fracture-type classifier, should be clari

The anti-botulism triterpenoid toosendanin elicits calcium increase and exocytosis in rat sensory neurons.

Toosendanin, a triterpenoid from Melia toosendan Sieb et Zucc, has been found before to be an effective anti-botulism agent, with a bi-phasic effect at both motor nerve endings and central synapse: an initial facilitation followed by prolonged depression. Initial facilitation may be due to activation of voltage-dependent calcium channels plus inhibition of potassium channels, but the depression is not fully understood. Toosendanin has no effect on intracellular calcium or secretion in the non-excitable pancreatic acinar cells, ruling out general toosendanin inhibition of exocytosis. In this st
