Galantamine-Memantine Combination in the Treatment of Parkinson's Disease Dementia.

Parkinson's disease (PD) is a progressive neurodegenerative disorder that affects over 1% of population over age 60. It is defined by motor and nonmotor symptoms including a spectrum of cognitive impairments known as Parkinson's disease dementia (PDD). Currently, the only US Food and Drug Administration-approved treatment for PDD is rivastigmine, which inhibits acetylcholinesterase and butyrylcholinesterase increasing the level of acetylcholine in the brain. Due to its limited efficacy and side effect profile, rivastigmine is often not prescribed, leaving patients with no treatment options. PD

Galantamine-Memantine combination in the treatment of Alzheimer's disease and beyond.

Alzheimer's disease (AD) is the most prevalent form of dementia in the elderly population worldwide. Despite the major unmet clinical need, no new medications for the treatment of AD have been approved since 2003. Galantamine is an acetylcholinesterase inhibitor that is also a positive allosteric modulator at the α4β2 and α7nACh receptors. Memantine is an N-methyl-d-aspartate receptor modulator/agonist. Both galantamine and memantine are FDA-approved medications for the treatment of AD. The objective of this review is to highlight the potential of the galantamine-memantine combination to condu

[Akatinol memantin in Alzheimer's disease: clinico-immunological correlates].

16 patients with Alzheimer's disease were treated with akatinol memantine. The levels of production of interleukin-1 (IL-1) in lymphocyte culture were evaluated both before and after the treatment that lasted for 2-4.5 months. Before the therapy a significant (p < 0.05) increase in the production of IL-1 was observed in patients with mild and moderate dementia, with early onset of the disease and with the duration of the disease up to 9 years as compared with a control group (24 healthy individuals). After the treatment the production of IL-1 was decreased in patients with moderate dementia, w

[Effect of memantine on central neurotransmitter systems. Review of the results].

In the present review the effects of 1,3-dimethyl-5-amino-adamantane (DMAA, D-145, Memantine, Memantine) on various neurotransmitter systems were presented. The most marked effect of DMAA seems to be the stimulation of dopaminergic neurons as reflected by the respective behavioural changes and biochemical effects. The stimulatory action on noradrenaline and serotonin neurons (at least partially indirectly through the activation of dopamine system) as well as on GABA neurons is apparent. No direct effects on Dopamine, alpha 1, alpha 2, Serotonin, GABA, benzodiazepine and opiate receptors were f

Efficacy of memantine prodrug microemulsion in a Preclinical model of tendinopathy.

Tendinopathy is a complex musculoskeletal pathology characterized by pain and dysfunction in the tendon. Tendinopathy management includes pharmacological treatments, which, however, have numerous side effects and low efficacy. New pharmacological approaches are essential to overcome these drawbacks. This study aimed to test the activity of the newly synthesized memantine-prodrug Memit against chronic tendon pain, which releases H2S and memantine at the therapeutic site, formulated in a microemulsion for peri-tendon administration. Memit (1 % w/w) was successfully formulated in a microemulsion

Differential effects of hypothermia and memantine in neonatal hypoxia-ischemia.

Therapeutic hypothermia (TH) is the standard treatment for neonatal hypoxia-ischemia (HI), but its efficacy is limited. Understanding the molecular pathways involved in cell death and survival, while considering sex-related differences, is essential to optimize therapeutic strategies. This study evaluated the neuroprotective effects of memantine, a non-competitive NMDA receptor antagonist, alone or in combination with TH, in male and female neonatal rats subjected to HI. Seven-day-old pups (P7) underwent right common carotid artery occlusion followed by 90 min of hypoxia (8% O2). Memantine (20

Unfavorable effects of memantine on the skeletal system in female rats.

Memantine is an N-methyl-D-aspartate (NMDA) receptor antagonist used in the treatment of Alzheimer's disease (AD). NMDA receptors are expressed on bone cells. The aim of the present study was to investigate the effects of memantine on the rat musculoskeletal system. Taking into account that most of female AD patients are postmenopausal, the study was carried out on intact and ovariectomized (estrogen-deficient) rats. Mature Wistar rats were divided into following groups: non-ovariectomized (NOVX) control rats, NOVX rats treated with memantine, ovariectomized (OVX) control rats, and OVX rats tr

An evaluation of memantine ER + donepezil for the treatment of Alzheimer's disease.

Alzheimer's disease (AD) is the most common neurodegenerative disease worldwide and carries an immense societal burden. Unfortunately, no curative or disease-modifying treatment has yet been discovered. The currently approved medications are symptomatic. They include two classes: the cholinesterase inhibitors, such as donepezil, and the NMDA receptor antagonist memantine. Most evidence has shown that combining both classes is superior to monotherapy but may complicate the treatment regimen for patients and families. Namzaric®, a fixed dose combination of donepezil and memantine extended-releas

Memantine for the treatment of general neuropathic pain: a narrative review.

Neuropathic pain (NP) is difficult to treat and is associated with a decline in quality of life. NP aetiologies are numerous and a number of pathologies display neuropathic characteristics. Of the various N-methyl-d-aspartate antagonists that are alternatives to be recommended in first-line NP treatment, memantine has the safest side-effect profile and has long been approved in Alzheimer's disease. The review covers memantine studies in postherpetic neuralgia, diabetic pain, postoperative pain, complex regional pain syndrome, chronic phantom limb pain, opioid-refractory pain and fibromyalgia.

Effects of memantine on the excitation-inhibition balance in prefrontal cortex.

Memantine is one of the few drugs currently approved for treatment of Alzheimer's disease (AD). The clinical effects of memantine are thought to be associated with inhibition of NMDA receptors (NMDARs). Surprisingly, other open-channel NMDAR blockers have unacceptable side effects that prevent their consideration for AD treatment. One of the mechanisms proposed to explain the therapeutic benefits of memantine involves preferential decrease of excitatory drive to inhibitory neurons in the cortical circuitry and consequent changes in balance between excitation and inhibition (E/I). In this study

Outcomes of Alzheimer's disease therapy with acetylcholinesterase inhibitors and memantine.

INTRODUCTION: Alzheimer's disease (AD) is a world-wide health problem with implications for an increasing number of people and countries. Populations suffering from AD financially strain the healthcare budgets of rich and poor countries alike. Moreover, no effective treatment is available and current drugs merely slow the progression of cognitive function deterioration and overall health status toward an inevitable end point. An increasing number of novel approaches have been tested in numerous clinical trials, but none of them has proved safe and effective for treating AD. AREAS COVERED: Thi

Crossover trial of gabapentin and memantine as treatment for acquired nystagmus.

We conducted a masked, crossover, therapeutic trial of gabapentin (1,200mg/day) versus memantine (40 mg/day) for acquired nystagmus in 10 patients (aged 28-61 years; 7 female; 3 multiple sclerosis [MS]; 6 post-stroke; 1 post-traumatic). Nystagmus was pendular in 6 patients (4 oculopalatal tremor; 2 MS) and jerk upbeat, hemi-seesaw, torsional, or upbeat-diagonal in each of the others. For the group, both drugs reduced median eye speed (p < 0.001), gabapentin by 32.8% and memantine by 27.8%, and improved visual acuity (p < 0.05). Each patient improved with 1 or both drugs.
