Advances in metabolic engineering for the production of sustainable aviation fuels.

Sustainable aviation fuels (SAF) are critical for decarbonising the hard-to-abate aviation sector, which significantly contributes to global CO2 emissions. Conventional SAF production routes, such as Hydroprocessed Esters and Fatty Acids, Fischer-Tropsch and Alcohol-to-Jet, offer drop-in compatibility but are constrained by feedstock availability, high costs and environmental impacts. This review highlights, as promising alternatives, microbial bioproduction via precision fermentation of SAF-relevant compounds from low-cost feedstocks, with reduced land use and enhanced circularity. Here, we f

Two routes for tyrosol production by metabolic engineering of Corynebacterium glutamicum.

BACKGROUND: The phenolic compound tyrosol is widely used in the pharmaceutical industry, owing to its beneficial effects on human health and its use as a precursor for key pharmaceuticals, including β1-receptor blockers. Tyrosol can be found in olive oil, but despite its natural biosynthesis in plants, low extraction efficiencies render microbial production a more viable alternative. RESULTS: Here, we engineered the L-tyrosine overproducing Corynebacterium glutamicum strain AROM3 for the de novo production of tyrosol. Two routes were established and compared: one via 4-OH-phenylpyruvate as in

Metabolic Engineering of Escherichia coli for Efficient Production of Benzyl Alcohol.

Benzyl alcohol (BALC) is a naturally occurring compound widely used in the fragrance, pharmaceutical, and chemical industries. Traditional chemical synthesis involves toxic materials and costly purification, and the resulting products are often labeled as "artificial fragrances", which are generally less favored by consumers due to concerns about their safety, sustainability, and environmental impact. Biosynthesis offers a sustainable alternative for "naturally derived" BALC, but existing pathways face insufficient precursors and bottleneck enzyme, limiting titers to 114 mg/L. To address these

Engineering of xylose metabolic pathways in Rhodotorula toruloides for sustainable biomanufacturing.

The oleaginous yeast Rhodotorula toruloides is a promising microbial cell factory for the sustainable production of biofuels and value-added chemicals from renewable carbon sources. Unlike the conventional yeast Saccharomyces cerevisiae, R. toruloides can naturally metabolize xylose, the second most abundant sugar in lignocellulosic hydrolysates. However, its native xylose metabolism is inefficient, characterized by slow xylose uptake and accumulation of D-arabitol. Moreover, despite its phenotype, research on the enzymes involved in xylose metabolism has yet to reach a consensus. Therefore, t

Engineering an Artificial Myxopyronin Derivative with Enhanced Metabolic Stability via Mutasynthesis.

The rise of multidrug-resistant pathogens, such as Staphylococcus aureus and Mycobacterium tuberculosis, underscores an urgent need for therapeutic innovation. The antibiotic development pipeline targeting these bacteria is critically limited, with most discovered candidates exhibiting structurally similar features of prominent chemical entities and with well-established molecular targets or binding modes. The myxobacterial α-pyrone antibiotics, myxopyronins, represent a highly promising compound class due to their ability to inhibit RNA polymerase by binding to the "switch region", a distinct

Systemic metabolic engineering of Enterobacter aerogenes for efficient 2,3-butanediol production.

2,3-Butanediol (2,3-BDO) is an important gateway molecule for many chemical derivatives. Currently, microbial production is gradually being recognized as a green and sustainable alternative to petrochemical synthesis, but the titer, yield, and productivity of microbial 2,3-BDO remain suboptimal. Here, we used systemic metabolic engineering strategies to debottleneck the 2,3-BDO production in Enterobacter aerogenes. Firstly, the pyruvate metabolic network was reconstructed by deleting genes for by-product synthesis to improve the flux toward 2,3-BDO synthesis, which resulted in a 90% increase o

Metabolic engineering of Bacillus subtilis for production of D-lactic acid.

Poly lactic acid (PLA) based plastics is renewable, bio-based, and biodegradable. Although present day PLA is composed of mainly L-LA, an L- and D- LA copolymer is expected to improve the quality of PLA and expand its use. To increase the number of thermotolerant microbial biocatalysts that produce D-LA, a derivative of Bacillus subtilis strain 168 that grows at 50°C was metabolically engineered. Since B. subtilis lacks a gene encoding D-lactate dehydrogenase (ldhA), five heterologous ldhA genes (B. coagulans ldhA and gldA101, and ldhA from three Lactobacillus delbrueckii) were evaluated. [Metabolic engineering of Escherichia coli for efficient production of nicotinamide riboside].

Nicotinamide riboside (NR), an essential precursor for nicotinamide adenine dinucleotide (NAD+), has garnered increasing attention for its anti-aging properties and diverse pharmacological activities. Developing an efficient, sustainable, and environmentally friendly synthesis approach is of great significance for the industrial production of NR. In this study, we developed a modular metabolic engineering strategy to construct an engineered strain capable of synthesizing NR from nicotinamide efficiently. First, six genes, including rihA, were deleted to block the degradation pathways of NR and

Systematic metabolic engineering of Komagataella phaffii for methanol-to-lactate bioconversion.

Methanol is an attractive C1 feedstock for sustainable biomanufacturing. However, efficient conversion of methanol into value-added products remains challenging due to metabolic compartmentalization, accumulation of toxic intermediates, and carbon loss in methylotrophic yeasts. In this study, we established a methanol-to-lactate bioconversion platform in Komagataella phaffii through a systematic metabolic engineering strategy that integrates methanol assimilation, detoxification, and downstream carbon flux redirection. Initial expression of a heterologous lactate dehydrogenase enabled efficien

Metabolic engineering of Ruminiclostridium papyrosolvens for 2,3-butanediol production from lignocellulose.

Ruminiclostridium papyrosolvens is an anaerobic, mesophilic, and cellulolytic bacterium capable of secreting cellulosomes for efficient lignocellulose degradation, making it a promising candidate for consolidated bioprocessing (CBP). However, its native cellulolytic activity is repressed by cellobiose, and its natural fermentation products are limited to low-value compounds. In this study, we firstly discovered that the cellulosomal cip-cel gene cluster is subject to cellobiose-mediated carbon catabolite repression (CCR). By replacing its native promoter with the endogenous cellobiose/cellodex

Metabolic Engineering of E. coli for Enhanced Diols Production from Acetate.

Effective employment of renewable carbon sources is highly demanded to develop sustainable biobased manufacturing. Here, we developed Escherichia coli strains to produce 2,3-butanediol and acetoin (collectively referred to as diols) using acetate as the sole carbon source by stepwise metabolic engineering. When tested in fed-batch experiments, the strain overexpressing the entire acetate utilization pathway was found to consume acetate at a 15% faster rate (0.78 ± 0.05 g/g/h) and to produce a 35% higher diol titer (1.16 ± 0.01 g/L) than the baseline diols-producing strain. Moreover, singularly

Refining and illuminating acetogenic Eubacterium strains for reclassification and metabolic engineering.

BACKGROUND: The genus Eubacterium is quite diverse and includes several acetogenic strains capable of fermenting C1-substrates into valuable products. Especially, Eubacterium limosum and closely related strains attract attention not only for their capability to ferment C1 gases and liquids, but also due to their ability to produce butyrate. Apart from its well-elucidated metabolism, E. limosum is also genetically accessible, which makes it an interesting candidate to be an industrial biocatalyst. RESULTS: In this study, we examined genomic, phylogenetic, and physiologic features of E. limosum

Metabolic engineering of Komagataella phaffii for the efficient utilization of methanol.

BACKGROUND: Komagataella phaffii, a type of methanotrophic yeast, can use methanol, a favorable non-sugar substrate in eco-friendly bio-manufacturing. The dissimilation pathway in K. phaffii leads to the loss of carbon atoms in the form of CO2. However, the ΔFLD strain, engineered to lack formaldehyde dehydrogenase-an essential enzyme in the dissimilation pathway-displayed growth defects when exposed to a methanol-containing medium. RESULTS: Inhibiting the dissimilation pathway triggers an excessive accumulation of formaldehyde and a decline in the intracellular NAD+/NADH ratio. Here, we desi

Metabolic engineering of Escherichia coli to efficiently produce monophosphoryl lipid A.

Monophosphoryl lipid A (MPL), mainly isolated from Salmonella minnesota R595, has been used as adjuvant in several vaccines. In this study, an Escherichia coli strain that can efficiently produce the MPL has been constructed. The gene clusters related to the biosynthesis of O-antigen, core oligosaccharide, enterobacterial common antigen, and colanic acid were sequentially removed to save the carbon source and to increase the activity of PagP in E. coli MG1655. Then, the genes pldA, mlaA, and mlaC related to the phospholipid transport system were further deleted, resulting in the strain MW012.
