Mitophagy as an active regulator of cardiac metabolic reprogramming.

Mitophagy is increasingly recognized as a context-dependent regulator of cardiac metabolic adaptation rather than solely as a disposal pathway for damaged mitochondria. By coupling mitochondrial turnover to substrate selection, redox control, and inflammatory signaling, mitophagy can influence fatty acid oxidation (FAO), glycolysis, and oxidative phosphorylation (OXPHOS) in cardiomyocytes, vascular endothelial cells, and immune cells. In this review, the term Mitophagy-Metabolic Rewiring Axis (MMRA) is used as an integrative conceptual framework-not as a newly discovered pathway or theory-to o

Metabolic Reprogramming by Engineered Probiotics Potentiates Tumor Chemodynamic Immunotherapy.

Engineered bacteria hold promise for remodeling immunosuppressive tumor microenvironments through innate immunogenicity and localized therapeutic delivery. While bacterial-derived metabolites are increasingly recognized as key mediators, their specific roles in interkingdom crosstalk remain underexplored. Here, we develop a programmable probiotic platform leveraging Lactobacillus gasseri (LG)-tumor metabolic interplay to potentiate bladder cancer therapy. The selected LG strain demonstrates superior tumor-colonizing ability and intrinsic H2O2/lactate biosynthesis, creating an optimal tumor mic

Metabolic reprogramming and immune regulation in acute myeloid leukemia.

The most prevalent kind of acute leukemia in adults is acute myeloid leukemia (AML). While some individuals have had better effectiveness due to advancements in targeted medications, recurrence after remission and inadequate treatment specificity continue to be significant therapeutic problems. By controlling essential metabolic pathways and metabolites, metabolic reprogramming, a crucial strategy for cellular adaptability to energy needs, modifies cellular metabolic rhythms. In addition to being involved in immune cell proliferation, differentiation, and effector function, this pathway is als

HDAC3 mediates retinal endothelial cell metabolic reprogramming and angiogenesis.

Pathological retinal neovascularization (NV) contributes to vision loss in diabetic retinopathy (DR) and retinopathy of prematurity, which are the leading causes of blindness in working-age adults and children, respectively. Retinal hypoxia is a key driver of pathological neovascularization that results in uncontrolled vessel sprouting and the formation of immature and leaky blood vessels. Anti-vascular endothelial growth factor and laser therapies are the standard of care to mitigate vision loss, but their limited effectiveness underlies the need to identify new therapeutic targets. The goal

Metabolic reprogramming of NK cells drives anti-tumor immunity.

Cellular metabolism critically controls immune cell functionality, necessitating dynamic metabolic adaptation to sustain effector responses under microenvironmental fluctuations. Natural killer (NK) cells, innate lymphocytes essential for tumor immunosurveillance, exhibit functional plasticity intrinsically linked to metabolic reprogramming. While NK cell-based immunotherapy show promise due to MHC-independent cytotoxic, their clinical efficacy diverges markedly between hematological and solid tumor-a disparity rooted in tumor microenvironment (TME) driven immunosuppression. Conserved lipid metabolic reprogramming confers hypoxic and aging resilience.

The Arctic ground squirrel (AGS, Urocitellus parryii), an extreme hibernator, exhibits remarkable resilience to stressors like hypoxia and hypothermia, making it an ideal model for studying cellular metabolic adaptation. The underlying mechanisms of AGS resilience are largely unknown. Here, we use lipidomic and metabolomic profiling to discover specific downregulation of triglyceride lipids and upregulation of the lipid biosynthetic precursor malonic acid in AGS neural stem cells (NSC) versus murine NSCs. Inhibiting lipid biosynthesis recapitulates hypoxic resilience of squirrel NSCs. Extendin

The role of metabolic reprogramming in inflammatory bone diseases.

Inflammatory bone diseases are characterized by bone destruction, resulting from an imbalance between bone resorption and formation due to chronic inflammation. The dynamic equilibrium among osteoblasts, osteoclasts, and various immune cells is crucial for disease progression. Metabolic reprogramming is a key factor involved in regulating the differentiation and function of these cells, and recent research has increasingly revealed its role in inflammatory bone diseases. This review focuses on the complex roles of glucose, lipid, and amino acid metabolic reprogramming in cellular processes, co

Glucose Metabolic Reprogramming of Macrophages Against Mycobacterium tuberculosis Infection.

Tuberculosis (TB) is a global infectious disease caused by Mycobacterium tuberculosis (Mtb). Serving as the primary effector cells, macrophages play a crucial role in host immune responses against Mtb. During Mtb infection, macrophages undergo extensive metabolic reprogramming, notably glycolysis, the pentose phosphate pathway (PPP) and the tricarboxylic acid (TCA) cycle, to adapt to the challenges posed by the pathogen, with glucose metabolic rewiring being particularly critical. This review focuses on the dynamic reprogramming of glucose metabolism in macrophages during Mtb infection, highli

Metabolic reprogramming and its clinical implication for liver cancer.

Cancer cells often encounter hypoxic and hypo-nutrient conditions, which force them to make adaptive changes to meet their high demands for energy and various biomaterials for biomass synthesis. As a result, enhanced catabolism (breakdown of macromolecules for energy production) and anabolism (macromolecule synthesis from bio-precursors) are induced in cancer. This phenomenon is called "metabolic reprogramming," a cancer hallmark contributing to cancer development, metastasis, and drug resistance. HCC and cholangiocarcinoma (CCA) are 2 different liver cancers with high intertumoral heterogenei

Metabolic reprogramming of fibro/adipogenic progenitors facilitates muscle regeneration.

In Duchenne muscular dystrophy (DMD), the absence of the dystrophin protein causes a variety of poorly understood secondary effects. Notably, muscle fibers of dystrophic individuals are characterized by mitochondrial dysfunctions, as revealed by a reduced ATP production rate and by defective oxidative phosphorylation. Here, we show that in a mouse model of DMD (mdx), fibro/adipogenic progenitors (FAPs) are characterized by a dysfunctional mitochondrial metabolism which correlates with increased adipogenic potential. Using high-sensitivity mass spectrometry-based proteomics, we report that a sh

Oxidative Stress and Metabolic Reprogramming in Cr(VI) Carcinogenesis.

Cr(VI)-containing compounds are well-established lung carcinogens. Chronic exposure of the normal human epithelial cells is able to induce malignant cell transformation, the first stage of metal carcinogenesis. These Cr(VI)-transformed cells exhibit increased level of antioxidants, reduced capacity of generating reactive oxygen species (ROS), and development of apoptosis resistance, promoting tumorigenesis of Cr(VI)-transformed cells, the second stage of metal carcinogenesis. The mechanism of Cr(VI) induced carcinogenesis is still under investigation. Recent studies indicate that ROS play a po

Epigenetic reprogramming in metabolic disorders: nutritional factors and beyond.

Environmental factors (e.g., malnutrition and physical inactivity) contribute largely to metabolic disorders including obesity, type 2 diabetes, cardiometabolic disease and nonalcoholic fatty liver diseases. The abnormalities in metabolic activity and pathways have been increasingly associated with altered DNA methylation, histone modification and noncoding RNAs, whereas lifestyle interventions targeting diet and physical activity can reverse the epigenetic and metabolic changes. Here we review recent evidence primarily from human studies that links DNA methylation reprogramming to metabolic d

Targeting metabolic reprogramming in cancer: Past, present, and future.

Cancer cells undergo profound metabolic reprogramming to sustain uncontrolled proliferation within a nutrient-limited and often hypoxic tumor microenvironment (TME). Metabolic rewiring is an active driver of oncogenesis, immune evasion, epigenetic remodeling, and therapy resistance. Over the past century, our understanding of tumor metabolism has grown from Warburg's seminal description of aerobic glycolysis to a comprehensive adaptive network. Cancer cells coordinate glucose catabolism, mitochondrial oxidative metabolism, fatty acid synthesis and oxidation, amino acid catabolism, nucleotide b

Metabolic reprogramming underlying EGFR-TKI resistance in lung cancer.

Epidermal growth factor receptor tyrosine kinase inhibitors (EGFR-TKIs) have significantly improved the clinical efficacy in non-small cell lung cancer (NSCLC) patients with EGFR mutations. However, acquired resistance to EGFR-TKIs remains an unavoidable therapeutic bottleneck. The metabolic reprogramming underlying this resistance is crucial for understanding the resistance mechanisms and identifying new therapeutic targets. Recently, an increasing number of studies have focused on the impact of metabolic reprogramming on acquired EGFR-TKI resistance. Collectively, this review reveals the cor

Mucin-induced metabolic reprogramming in Pseudomonas aeruginosa clinical isolates.

Mucins modulate microbial metabolism, which is crucial in many human infections. It is unknown whether the metabolic alterations induced by mucin are universal or unique across varied clinical isolates of bacterial pathogens. We experimentally profiled a representative set of seven clinical Pseudomonas aeruginosa isolates in a synthetic cystic fibrosis growth medium in the presence or absence of physiological mucin, using high-throughput transcriptomic analyses. We observed mucin-induced transcriptional changes across many central and peripheral metabolic pathways in all the clinical isolates.

The dual role of AMPK in cancer metabolic reprogramming.

Metabolic dysregulation is a core hallmark of tumor cells, which reshapes energy metabolism patterns to meet the demands of rapid proliferation, invasion, metastasis, and drug resistance. AMP‑activated protein kinase (AMPK), as a central regulator of cellular energy homeostasis, senses changes in the intracellular AMP/ATP ratio and extensively participates in the regulation of glucose metabolism, lipid metabolism, amino acid metabolism, and autophagy. In the field of oncology, the role of AMPK is controversial. Initially, because it is associated with liver kinase B1 (LKB1) and exhibits growth

Metabolic reprogramming in hepatocellular carcinoma: mechanisms and therapeutic implications.

Hepatocellular carcinoma features extensive metabolic reprogramming. This includes alterations in major biochemical pathways such as glycolysis, the pentose phosphate pathway, amino acid metabolism and fatty acid metabolism. Moreover, there is a complex interplay among these altered pathways, particularly involving acetyl-CoA (coenzyme-A) metabolism and redox homeostasis, which in turn influences reprogramming of other metabolic pathways. Understanding these metabolic changes and their interactions with cellular signaling pathways offers potential strategies for the targeted treatment of hepat
