Encephalitis-Associated Human Metapneumovirus Pneumonia in Adult, Australia.

Human metapneumovirus pneumonia, most commonly found in children, was diagnosed in an adult with encephalitis. This case suggests that testing for human metapneumovirus RNA in nasopharyngeal aspirate and cerebrospinal fluid samples should be considered in adults with encephalitis who have a preceding respiratory infection.

Diffuse Alveolar Hemorrhage Secondary to Human Metapneumovirus Pulmonary Infection.

A 67-year-old man presented with a week of flu-like symptoms, hypoxia, and fever. Respiratory viral panel was positive for human metapneumovirus. Initial chest imaging showed left lower lobe opacification, suggesting a bacterial superimposed on viral pneumonia. Despite antibiotics, the patient became tachycardic and increasingly hypoxic, requiring 40 L high-flow nasal cannula. Repeat imaging demonstrated worsening of a left lower lobe process. Elective bronchoscopy with bronchoalveolar lavage revealed hemorrhage. Subsequent autoimmune, bacterial, and fungal workup was negative. The patient was

Co-infection with SARS-CoV-2 and Human Metapneumovirus.

The novel coronavirus (now called SARS-CoV-2) initially discovered in Wuhan, China, has now become a global pandemic. We describe a patient presenting to an Emergency Department in Rhode Island on March 12, 2020 with cough and shortness of breath after a trip to Jamaica. The patient underwent nasopharyngeal swab for a respiratory pathogen panel as well as SARS-CoV-2 RT-PCR. When the respiratory pathogen panel was positive for human metapneumovirus, the patient was treated and discharged. SARS-CoV-2 RT-PCR came back positive 24 hours later. Although respiratory viral co-infection is thought to

Bleeding pneumonia: Diffuse alveolar hemorrhage due to human metapneumovirus.

Diffuse alveolar hemorrhage is a condition with high morbidity and mortality. The majority of cases are caused by pulmonary capillaritis associated with systemic vasculitis. Infection disease has also been associated with this condition. A 62-year-old woman with a history of chronic alcohol abuse presented with shortness of breath, hemoptysis, constipation, and icterus. Chest x-rays on admission showed diffuse patchy opacities concerning for diffuse alveolar hemorrhage. The patient quickly developed acute respiratory failure requiring intubation. PCR identified human metapneumovirus and bronch

Human metapneumovirus in lung transplant recipients: characteristics and outcomes.

Human metapneumovirus (hMPV) causes serious respiratory tract infections in lung transplant recipients (LTRs). We evaluated the characteristics and adverse drug reactions (ADR) of oral ribavirin therapy for hMPV infections in LTRs. LTRs with respiratory symptoms or suspected infection of unknown origin were routinely sampled with nasopharyngeal swabs (NPS) for virological and bacteriological analysis as part of a diagnostic workup. Medical records of hMPV polymerase chain reaction (PCR)-positive LTRs at the University Hospital of Zurich were reviewed retrospectively. Between January 2012 and J

Human metapneumovirus infection in hematopoietic stem cell transplant recipients.

UNLABELLED: Human metapneumovirus (hMPV) was described in 2001 and has been associated with both upper and lower respiratory tract infection (URTI and LRTI, respectively), especially in children, the elderly, and in immunocompromised patients. The objective of this study was to identify hMPV as the etiological agent of acute respiratory infection in hematopoietic stem cell transplant (HSCT) patients and to determine the clinical features of hMPV infection in these patients. METHODS: The study was performed retrospectively in 769 respiratory samples obtained from immunocompromised patients sub

Inhibition of human metapneumovirus replication by small interfering RNA.

BACKGROUND: Human metapneumovirus (hMPV) is a major respiratory viral pathogen in young children, elderly individuals and immunocompromised patients. Despite its major effects related to bronchiolitis, pneumonia and its potential role in recurrent wheezing episodes, there is still no commercial treatment or vaccine available against this paramyxovirus. METHODS: We tested a therapeutic strategy for hMPV that was based on RNA interference. RESULTS: An hMPV genome-wide search for small interfering RNAs (siRNAs) by computational analysis revealed 200 potentially effective 21-mer siRNAs. Initial

Human metapneumovirus infection in hospital referred South African children.

Human metapneumovirus (hMPV) was first described in Dutch children with acute respiratory symptoms. A prospective analysis of the epidemiology, clinical manifestation, and seroprevalence of hMPV and other respiratory viruses in South African children referred to hospital for upper or lower respiratory tract infection were carried out during a single winter season, by using RT-PCR, viral culture, and enzyme-linked immunosorbent assays. In nasopharyngeal aspirates from 137 children, hMPV was detected by RT-PCR in 8 (5.8%) children (2-43 months of age) as a sole viral pathogen, respiratory syncyt

Advancing human metapneumovirus vaccine development through fusion protein stabilization.

Human metapneumovirus (hMPV) is a major respiratory pathogen that causes a substantial global disease burden, particularly in infants, older adults, and immunocompromised individuals. Despite more than two decades of research, no licensed vaccines or antiviral therapies are available, underscoring a persistent unmet clinical need. The viral fusion (F) glycoprotein is the leading target for vaccine and immunotherapeutic development due to its essential role in viral entry and its ability to elicit neutralizing antibodies. However, hMPV-F is highly metastable and undergoes conformational transit

Human Metapneumovirus: An Underestimated Cause of Severe Respiratory Infection.

Since its identification in 2001, human metapneumovirus (hMPV) has been recognized as an important cause of acute respiratory tract infection in all age groups. Although infection is often mild in healthy adults, hMPV can cause severe lower respiratory tract disease, including pneumonia, in young children, older adults, individuals with chronic diseases and immunocompromising conditions, and residents of long-term care facilities. Accumulating evidence demonstrates that hMPV contributes substantially to hospitalization, intensive care unit admission, and mortality in these high-risk population

Determinants of respiratory syncytial virus and human metapneumovirus transmission.

SUMMARYRespiratory syncytial virus and human metapneumovirus are major causes of respiratory illness globally, particularly affecting children and older adults. Despite their clinical significance, substantial gaps persist in understanding the determinants of their transmission. This review examines the multifaceted factors shaping RSV and hMPV transmission, including viral structure and evolution, host behaviors and viral shedding dynamics, the influence of environmental conditions on viral survival, the roles of population- and community-level transmission networks, and the impact of SARS-Co

Lobar pneumonia due to human metapneumovirus: a case report.

Human metapneumovirus (hMPV) is a respiratory pathogen that can cause lower respiratory tract infections and pneumonia in immunocompetent adults. Pneumonia caused by hMPV is reportedly more likely to cause bronchial wall thickening and ground-glass opacity (GGO). A 44-year-old woman with no significant medical history developed fever, cough, and nausea. Computed tomography of the chest showed scattered GGOs in the right upper lobe and infiltrating shadows with air bronchograms in the left lingual and bilateral lower lobes. The patient was admitted to our hospital for further evaluation. Atypic

New Approaches for Immunization and Therapy against Human Metapneumovirus.

Human metapneumovirus (HMPV) is a paramyxovirus discovered in 2001 in the Netherlands. Studies have identified HMPV as an important causative agent of acute respiratory disease in infants, the elderly, and immunocompromised individuals. Clinical signs of infection range from mild upper respiratory illness to more serious lower respiratory illness, including bronchiolitis and pneumonia. There are currently no licensed therapeutics or vaccines against HMPV. However, several research groups have tested vaccine candidates and monoclonal antibodies in various animal models. Several of these approac

Human metapneumovirus and exacerbations of chronic obstructive pulmonary disease.

OBJECTIVE: Respiratory viruses are a common trigger for exacerbations of chronic obstructive pulmonary disease (COPD). Human metapneumovirus (hMPV) is a paramyxovirus associated with respiratory tract infections and wheezing. Our aim was to determine whether hMPV was associated with exacerbations of COPD. METHODS: The study was designed as an observational cohort study carried out in a 944-bed urban teaching hospital located in New Haven, Connecticut. Between December 2002 and May 2003, patients hospitalized due to an exacerbation of COPD were identified. Nasopharyngeal specimens obtained fro

[Human Metapneumovirus (hMPV) associated to severe bronchial asthmatic crisis].

Human Metapneumovirus (hMPV) is a recently reported agent of acute infection in the respiratory tract. It has been found in children as well as in young adults and elders. The clinical manifestations produced by hMPV are indistinguishable from those by common respiratory virus, and can evolve from asymptomatic infection into severe pneumonia. On the other hand, some authors have described cases of bronchial asthma exacerbation associated with hMPV infection. In this work we report a case of a child who presented a severe bronchial asthmatic crisis with a suspected viral associated infection. I

Human metapneumovirus: a ubiquitous and long-standing respiratory pathogen.

BACKGROUND: Human metapneumovirus (hMPV) is a recently described human pathogen first identified in respiratory specimens of young children suffering from respiratory syndromes ranging from mild to severe. METHODS AND RESULTS: Virological studies have reported the presence of hMPV infections in many countries from all continents. Seroprevalence studies have indicated that the virus has been circulating in humans for more than 50 years and that it infects virtually all children by the ages of 5-10 years. In young children, hMPV has been mainly associated with bronchiolitis but also with pneumo

Avian Metapneumovirus: A Review of Pathogenesis and Its Effect on Diagnosis.

Avian metapneumovirus (aMPV) is a highly contagious upper respiratory pathogen of poultry globally that emerged in the United States in January 2024 and rapidly spread to flocks throughout North America. In turkeys, the disease (turkey rhinotracheitis) initiates as an upper respiratory infection with high morbidity and frequent complication by bacterial coinfection that can lead to high mortality and significant economic loss. In chickens (swollen head syndrome), uncomplicated clinical disease is less severe than in turkeys. Chickens are less susceptible to the virus, although, once infected,

Molecular detection and genetic variability of human metapneumovirus in Uruguay.

Human metapneumovirus (hMPV) has been described as circulating among the Uruguayan population at least since 1998 based on serologic evidence. However, no isolation attempts, molecular detection, or genetic studies have been carried out so far in the country. In the present study, molecular detection of circulating hMPV in children hospitalized with acute respiratory tract infection in Montevideo-Uruguay was carried out by reverse transcription-polymerase chain reaction (RT-PCR) amplification of the hMPV nucleoprotein (N) gene from 217 nasopharyngeal aspirates. Genetic variability analysis of
