Purification and characterization of component A of the methane monooxygenase from Methylococcus capsulatus (Bath).

Methylococcus capsulatus (Bath) possesses a multi-component methane monooxygenase which catalyzes in vivo the conversion of methane to methanol. Component A of this enzyme system, believed to be the oxygenase component, has been purified to near homogeneity (95%). The native protein has a molecular weight of approximately 210,000 and is comprised of three subunits of Mr = 54,000, 42,000, and 17,000, which appear to be present in stoichiometric amounts suggesting an alpha 2, beta 2, gamma 2 arrangement in the native protein. Purified preparations of the protein are virtually colorless and exami

Phenotypic characterization of copper-resistant mutants of Methylosinus trichosporium OB3b.

Cultures of Methylosinus trichosporium OB3b grown in the presence of very low concentrations of copper synthesize a soluble methane monooxygenase (sMMO) that efficiently catalyzes the oxidation of trichloroethylene and other organic pollutants. Recently, we isolated five M. trichosporium OB3b mutants that express sMMO activity when grown in the presence of elevated copper concentrations (P.A. Phelps, S. K. Agarwal, G. E. Speitel, Jr., and G. Georgiou, Appl. Environ. Microbiol. 58:3701-3708, 1992). Here we show that, in contrast to the results for the wild-type cells, the addition of copper to

Oxygen activation catalyzed by methane monooxygenase hydroxylase component: proton delivery during the O-O bond cleavage steps.

The effects of solvent pH and deuteration on the transient kinetics of the key intermediates of the dioxygen activation process catalyzed by the soluble form of methane monooxygenase (MMO) isolated from Methylosinus trichosporium OB3b have been studied. MMO consists of hydroxylase (MMOH), reductase, and "B" (MMOB) components. MMOH contains a carboxylate- and oxygen-bridged binuclear iron cluster that catalyzes O2 activation and insertion chemistry. The diferrous MMOH-MMOB complex reacts with O2 to form a diferrous intermediate compound O (O) and subsequently a diferric intermediate compound P

Screening methane-oxidizing bacteria from municipal solid waste landfills and simulating their effects on methane and ammonia reduction.

Municipal solid waste landfills are not only a crucial source of global greenhouse gas emissions; they also produce large amounts of ammonia (NH3), hydrogen sulfide, and other odorous gases that negatively affect the regional environment. Several types of methane-oxidizing bacteria (MOB) were proved to be effective in mitigating methane emission from landfills. Nevertheless, more MOB species and their technical parameters for best mitigating methane still need to be explored. In landfills, methane is simultaneously generated with ammonia, which may impede the CH4 bio-oxidizing process of MOB.

Effect of nitrogen source on growth and trichloroethylene degradation by methane-oxidizing bacteria.

The effect of nitrogen source on methane-oxidizing bacteria with respect to cellular growth and trichloroethylene (TCE) degradation ability were examined. One mixed chemostat culture and two pure type II methane-oxidizing strains, Methylosinus trichosporium OB3b and strain CAC-2, which was isolated from the chemostat culture, were used in this study. All cultures were able to grow with each of three different nitrogen sources: ammonia, nitrate, and molecular nitrogen. Both M. trichosporium OB3b and strain CAC-2 showed slightly lower net cellular growth rates and cell yields but exhibited highe

Trichloroethylene degradation and mineralization by pseudomonads and Methylosinus trichosporium OB3b.

To examine the trichloroethylene (C2HCl3)-degrading capability of five microorganisms, the maximum rate, extent, and degree of C2HCl3 mineralization were evaluated for Pseudomonas cepacia G4, Pseudomonas cepacia G4 PR1, Pseudomonas mendocina KR1, Pseudomonas putida F1, and Methylosinus trichosporium OB3b using growth conditions commonly reported in the literature for expression of oxygenases responsible for C2HCl3 degradation. By varying the C2HCl3 concentration from 5 microM to 75 microM, Vmax and Km values for C2HCl3 degradation were calculated as 9 nmol/(min mg protein) and 4 microM for P.

Transcriptomic and Metabolomic Responses to Carbon and Nitrogen Sources in Methylomicrobium album BG8.

Methanotrophs use methane as their sole carbon and energy source and represent an attractive platform for converting single-carbon feedstocks into value-added compounds. Optimizing these species for biotechnological applications involves choosing an optimal growth substrate based on an understanding of cellular responses to different nutrients. Although many studies of methanotrophs have examined growth rate, yield, and central carbon flux in cultures grown with different carbon and nitrogen sources, few studies have examined more global cellular responses to different media. Here, we evaluate

Evaluation of toxic effects of aeration and trichloroethylene oxidation on methanotrophic bacteria grown with different nitrogen sources.

In this study we evaluated specific and nonspecific toxic effects of aeration and trichloroethylene (TCE) oxidation on methanotrophic bacteria grown with different nitrogen sources (nitrate, ammonia, and molecular nitrogen). The specific toxic effects, exerted directly on soluble methane monooxygenase (sMMO), were evaluated by comparing changes in methane uptake rates and naphthalene oxidation rates following aeration and/or TCE oxidation. Nonspecific toxic effects, defined as general cellular damage, were examined by using a combination of epifluorescent cellular stains to measure viable cell

Capacity for biodegradation of CFCs and HCFCs in a methane oxidative counter-gradient laboratory system simulating landfill soil covers.

The attenuation of methane and four chlorofluorocarbons was investigated in a dynamic methane and oxygen counter-gradient system simulating a landfill soil cover. Soil was sampled at Skellingsted Landfill, Denmark. The soil columns showed a high capacity of methane oxidation with oxidation rates of 210 g m(-2) d(-1) corresponding to a removal efficiency of 81%. CFC-11 and to a lesser extent also CFC-12 were degraded in the active soil columns. The average removal efficiency was 90% and 30% for CFC-11 and CFC-12, respectively. Soil gas concentration profiles indicated that the removal was due t

Single cell protein production potential of enriched microbial populations from rice paddy soils and roots: Insights into protein yield enhancement by Methylophilaceae.

This study aimed to determine key factors for enriching methylotrophic populations for single-cell protein (SCP) production and to determine the predominant active methylotrophs. Microbial populations from rice paddy soils and roots, hotspots for CH4 oxidation, underwent serial incubation by feeding CH4 and O2. The protein content was primarily influenced by the incubation stage, with a lesser effect from the rice plant roots as an inoculum source, reaching a maximum of 20 % in biomass dominated by Methylomonadaceae and Methylophilaceae. While Methylomonadaceae as CH4 oxidizers exhibited lower

Formate dehydrogenase from the methane oxidizer Methylosinus trichosporium OB3b.

Formate dehydrogenase (NAD+ dependent) was isolated from the obligate methanotroph Methylosinus trichosporium OB3b. When the enzyme was isolated anaerobically, two forms of the enzyme were seen on native polyacrylamide gels, DE-52 cellulose and Sephacryl S-300 columns; they were approximately 315,000 and 155,000 daltons. The enzyme showed two subunits on sodium dodecyl sulfate-polyacrylamide gels. The Mr of the alpha-subunit was 53,800 +/- 2,800, and that of the beta-subunit was 102,600 +/- 3,900. The enzyme (Mr 315,000) was composed of these subunits in an apparent alpha 2 beta 2 arrangement.

Biological conversion of propane to 2-propanol using group I and II methanotrophs as biocatalysts.

Propane is the main component of liquefied petroleum gas and is derived from crude oil processing. Methanotrophic bacteria can convert various alkanes using methane monooxygenase enzyme to primary alcohols. These are further oxidized to various aldehydes by alcohol dehydrogenases or methanol dehydrogenases. In this study, 2-propanol was produced from propane using the whole cells of Methylosinus trichosporium OB3b, Methylomicrobium alcaliphilum 20Z, and Methylomonas sp. DH-1 as the biocatalysts. The biocatalytic process of converting propane to 2-propanol was optimized by the use of several in

Detection of soluble methane monooxygenase producing Methylosinus trichosporium OB3b by polymerase chain reaction.

The soluble methane monooxygenase (sMMO) enzyme complex of methanotrophs cometabolizes haloaliphatic compounds such as trichloroethylene. Two 18-mer oligonucleotides as primary primers and a nested primer of the same length were selected to amplify specific DNA sequences of the sMMO gene cluster using polymerase chain reaction (PCR). Two DNA fragments of sizes 270 and 400 base pairs were obtained when purified DNA from the methanotroph Methylosinus trichosporium OB3b was used as template. The primers were specific for sMMO sequences of M. trichosporium, since none of the 13 bacterial isolates

Haloalkene oxidation by the soluble methane monooxygenase from Methylosinus trichosporium OB3b: mechanistic and environmental implications.

The soluble, three-protein component methane monooxygenase purified from Methylosinus trichosporium OB3b is capable of oxidizing chlorinated, fluorinated, and brominated alkenes, including the widely distributed ground-water contaminant trichloroethylene (TCE). The oxidation rates for the chloroalkenes were observed to be comparable to that for methane, the natural substrate, and up to 7000-fold higher than those reported for other well-defined biological systems. The competitive inhibitor tetrachloroethylene was found to be the only chlorinated ethylene not turned over. However, this appears

Inhibition of dimethyl ether and methane oxidation in Methylococcus capsulatus and Methylosinus trichosporium.

Metal-chelating or -binding agents inhibited the oxidation of dimethyl ether and methane, but not methanol, by cell suspensions of Methylococcus capsulatus and Methylosinus trichosporium. Evidence suggests that the involvement of metal-containing enzymatic systems in the initial step of oxidation of dimethyl ether and methane.

[Methanotrophic bacteria of acid sphagnum bogs].

Acid sphagnum bogs cover a considerable part of the territory of Russia and are an important natural source of biogenic methane, which is formed in their anaerobic layers. A considerable portion of this methane is consumed in the aerobic part of the bog profile by acidophilic methanotrophic bacteria, which comprise the methane filter of sphagnum bogs and decrease CH4 emission to the atmosphere. For a long time, these bacteria escaped isolation, which became possible only after the elucidation of the optimal conditions of their functioning in situ: pH 4.5 to 5.5; temperature, from 15 to 20 degr

Characterization of a methane-utilizing bacterium from a bacterial consortium that rapidly degrades trichloroethylene and chloroform.

A mixed culture of bacteria grown in a bioreactor with methane as a carbon and energy source rapidly oxidized trichloroethylene and chloroform. The most abundant organism was a crescent-shaped bacterium that bound the fluorescent oligonucleotide signature probes that specifically hybridize to serine pathway methylotrophs. The 5S rRNA from this bacterium was found to be 93.5% homologous to the Methylosinus trichosporium OB3b 5S RNA sequence. A type II methanotrophic bacterium, isolated in pure culture from the bioreactor, synthesized soluble methane monooxygenase during growth in a copper-limit

Methyloraptor flagellatus gen. nov., sp. nov., novel Ancalomicrobiaceae-affiliated facultatively methylotrophic bacteria that feed on methanotrophs of the genus Methylococcus.

A morphologically conspicuous microbial association was detected in a bioreactor running in a continuous mode with methanotrophic bacteria of the genus Methylococcus and natural gas as a growth substrate. The association consisted of spherical Methylococcus cells colonized by elongated rods, which produced rosette-like aggregates and inhibited the cultivation process. An isolate of these bacteria, strain S20T, was obtained and identified as belonging to the alphaproteobacterial family Ancalomicrobiaceae but displaying only a distant relationship (93.9-95.1 % 16S rRNA gene sequence similarity)
