AIM: To investigate the anti-tumor effects of antiangiogenic therapy (a combination of TNP-470, an antiangiogenic compound, with gemcitabine, an antimetabolite) on human pancreatic carcinoma xenografts and its mechanism.

In order to develop an in vivo model for growth of acoustic schwannomas, we studied tumor specimens from 10 patients, transplanted into a subcutaneous pocket of 67 nude mice. The number of tumors which survived or grew was 63 (94%). Obvious macroscopic growth was observed in 22 (33%), status quo in 28 (42%), and regression of tumor size in 13 (19%). The tumor disappeared in 4 cases (6%). Serial implantation was not possible due to the small amount of neuroma tissue in the surviving tumors. Burkitt's lymphoma (BL) cell lines estimated in a previous study as having a high, low and no tumourigenicity (7) were intravenously (i.v.) injected into preirradiated (480 rad) nude mice. Subcutaneous estrogen pellet implantation is commonly practiced to induce tumor promotion in nude (Nu-FOXn1(nu)) mice for xenograft tumor modeling. Footpad lesions of 3 nude mice infected by Mycobacterium leprae were studied at 9, 12, and 14 months after inoculation with light and electron microscope. The lesions were somewhat similar to those found in nodules in polar lepromatous leprosy. Striated muscles rather than nerves were the preferred site of the growth of M. leprae. Yet, M. leprae were identified in Schwann cells and endothelial cells, singly and in clumps. M. leprae filled macrophages, and free M. leprae were found in large numbers in the endoneurium without producing any significant demyelination.

The administration of melatonin to thymocyte-infused nude mice during the evening (dark cycle) of the circadian period restores transplantation immunity permanently, whereas inoculation during the morning cycle is quite ineffective. Transplantation of an intact pineal gland from normal donors into the mediastinal (athymic) tissue of nude mice leads to restoration of the capacity to reject skin grafts. Findings such as these attest to the genesis of transplantation immunity as linked to a thymus-directed establishment of a pineal circadian cyclicity.

Nude mice (nu/nu on BALB/cAn background), maintained under conventional conditions, developed an acute moist dermatitis of the nose and occasionally the ears, with heavy contamination with Staphylococcus pyogenes. Only homozygotes over 10 weeks old were affected. Treatment with parenteral ampicillin or oral oxytetracycline was ineffective. Other conditions associated with the same organism were conjunctivitis and periorbital abscesses.

Eight human upper urinary tract tumors were transplanted subcutaneously into congenitally athymic nude mice. Subcutaneous growth was obtained in three (NKPP 1, NKPP 2, and NKPU 1) at initial transplantation and they were all serially passaged up to 29, 28, and 5 passages, respectively.
