Development of microbial-human enterocyte interaction: cholera toxin.

Diarrhea in infants and children is a major health hazard worldwide. Certain toxigenic diarrheas occur more commonly and are manifested more severely during the neonatal period. We have previously studied the regulation of cholera toxin-induced secretion in animal models during development. In those studies we have shown that cholera toxin stimulates a much greater secretion by immature compared with mature small intestine, and the mechanism appears to be an up-regulation of postreceptor signal transduction molecules (adenyl cyclase and Gsalpha) leading to an elevated cAMP level. In this study

The Gut Microbiota in Parkinson's Disease: Mechanistic Insights into Microbial-Host Interactions.

Parkinson's disease (PD) is a multifactorial neurodegenerative disorder characterized by progressive motor and non-motor manifestations, including early gastrointestinal dysfunction. Growing evidence implicates the gut microbiota as an active modulator of host immune tone and neurodegenerative vulnerability, extending beyond descriptive taxonomic associations toward functional and metabolic mechanisms. PD-associated dysbiosis is consistently characterized by altered microbial functional capacity, including reduced short-chain fatty acid (SCFA) production, enrichment of pro-inflammatory metabol

Microbial interactions: Crosstalk in the fungal mycobiome through a novel small protein.

Although Candida glabrata reproduces asexually, it has maintained the canonical mating signaling pathway. A new paper shows that C. glabrata can use this pathway to secrete a novel small protein, Yhi1, that induces hyphal formation in Candida albicans, with implications for colonization and virulence.

Linking complex microbial interactions and dysbiosis through a disordered Lotka-Volterra model.

The rapid advancement of environmental sequencing technologies, such as metagenomics, has significantly enhanced our ability to study microbial communities. The eubiotic composition of these communities is crucial for maintaining ecological functions and host health. Species diversity is only one facet of a healthy community's organization; together with abundance distributions and interaction structures, it shapes reproducible macroecological states, that is, joint statistical fingerprints that summarize whole-community behavior. Despite recent developments, a theoretical framework connecting

Microbial adaptation and host signaling: Bacteroides-bile acid interactions in gut health.

Bile acids are amphipathic molecules synthesized from cholesterol that act not only as digestive agents but also as key signaling molecules regulating host metabolic and immune pathways via FXR (farnesoid X receptor) and TGR5 (G protein-coupled bile acid receptor). In the colon, microbial metabolism profoundly transforms bile acids, reshaping their composition, physicochemical properties, and signaling capacity. Among gut microbes, Bacteroides spp. are abundant, metabolically versatile, and immunologically active, displaying high resilience to bile acids through efflux pumps, membrane adaptati

Interspecific interactions modulate atrazine and S-metolachlor toxicity on aquatic microbial autotrophs.

Atrazine and S-metolachlor are herbicides frequently detected in agricultural streams, sometimes at concentrations of concern, as they may affect photosynthetic organisms. In this study, we exposed a coculture of the green alga Enallax costatus, the diatom Gomphonema parvulum and the cyanobacteria Phormidium sp. and Microcystis aeruginosa, to a gradient of atrazine and S-metolachlor (0, 10, 100 and 1000 µg·L-1) for 7 days. The results showed that species interactions within a coculture can modulate herbicide toxicity compared to responses observed in monocultures. Green algae and cyanobacteria

Synthetic microbial community mimicking kefir for investigating community dynamics and interspecies interactions.

Kefir grains serve as natural dairy starter cultures, surviving in high-altitude environments with low temperatures and limited oxygen, while maintaining robust fermentation capabilities. In this study, we reconstructed a synthetic microbial community (SMC) within the kefir microbial ecosystem and explored the strategies that keep this SMC stable and functioning within the complex environment. We investigated the interactions among kefir species by comparing their symbiotic capabilities, milk acidification properties, and fermentation profiles during growth in both individual cultures and co-c

Comprehensive gut microbiota composition and microbial interactions among the three age groups.

There is a growing interest in studying the microbiota associated with aging by integrating multiple longevity researches while minimizing the influence of confounding factors. Here, we reprocessed metagenomic sequencing data from four different aging research studies and evaluated potential confounding factors in order to minimize the batch effect. Subsequently, we detected the diversity and abundance of the gut microbiome in three different age cohorts. Out of 1053 different bacteria species, only four showed substantial depletion across different age groups: Ligilactobacillus ruminis, Turic

Roles of AGD2a in Plant Development and Microbial Interactions of Lotus japonicus.

Arabidopsis AGD2 (Aberrant Growth and Death2) and its close homolog ALD1 (AGD2-like defense response protein 1) have divergent roles in plant defense. We previously reported that modulation of salicylic acid (SA) contents by ALD1 affects numbers of nodules produced by Lotus japonicus, but AGD2's role in leguminous plants remains unclear. A combination of enzymatic analysis and biological characterization of genetic materials was used to study the function of AGD2 (LjAGD2a and LjAGD2b) in L. japonicus. Both LjAGD2a and LjAGD2b could complement dapD and dapE mutants of Escherichia coli and had a

Relay Architectures and Ecological Strategies of Quorum Sensing Effects on Microbial Interactions.

Microbial communities assemble in heterogeneous habitats, where quorum sensing (QS) influences interactions beyond signalling populations. Yet QS studies are commonly organized by signal type, circuit, or phenotype, obscuring the routes by which a QS event reaches neighbouring populations and why they prevail in particular settings. This Review synthesizes evidence from host-associated, terrestrial, marine, and aquatic systems to develop a two-dimensional framework for QS-mediated ecological effects. The framework distinguishes three relay architectures by which QS activity in one population a

Deciphering fungal-microbial interactions: From microscale experimental models to integrative systems biology.

Fungal-microbial interactions drive key ecosystem functions and offer vast, untapped potential for the bio-economy. However, translating these natural consortia into controllable applications is hindered by the structural and metabolic complexity of the interplay. This review navigates the complexity landscape of these interactions, examining the technological trajectory from the mechanistic foundation of 1:1 dialogue to the rational design of synthetic consortia. We discuss the transition from static cultivation toward advanced microfluidic ("fungi-on-a-chip") and biomimetic platforms that re

Gut microbial-host isozymes: A novel perspective on gut microbiota-host interactions.

The interaction between gut microbiota and host health has garnered significant attention since its initial discovery. Dysbiosis of the gut microbiota is implicated in various diseases, particularly metabolic disorders such as diabetes, as well as neuro-, cardiovascular-, and hepatic-metabolic diseases. In recent years, the concept of microbial-host isozymes (MHIs) has emerged as a novel research area within the microbiome field. These enzymes, encoded by intestinal microbiota, can replicate the functions of host enzymes and contribute to disease development, presenting potential new therapeut

Altered gut mycobiome and cross-kingdom microbial interactions in systemic lupus erythematosus.

BACKGROUND: Systemic lupus erythematosus (SLE) is a complex autoimmune disorder shaped by host genetics and environmental exposures, including the gut microbiota. While bacterial dysbiosis in SLE is well characterized, the role of the gut mycobiome and its cross-kingdom interactions remains largely unexplored. METHODS: Using fecal metagenomic sequencing from 117 SLE patients and 115 healthy controls (HCs), we established a non-redundant fungal genome catalog and revealed significant alterations in fungal composition, function, and cross-kingdom ecology. RESULTS: Fungal diversity was increased

Disease-specific loss of microbial cross-feeding interactions in the human gut.

Many gut microorganisms critical to human health rely on nutrients produced by each other for survival; however, these cross-feeding interactions are still challenging to quantify and remain poorly characterized. Here, we introduce a Metabolite Exchange Score (MES) to quantify those interactions. Using metabolic models of prokaryotic metagenome-assembled genomes from over 1600 individuals, MES allows us to identify and rank metabolic interactions that are significantly affected by a loss of cross-feeding partners in 10 out of 11 diseases. When applied to a Crohn's disease case-control study, o

Metabolic modeling of the International Space Station microbiome reveals key microbial interactions.

BACKGROUND: Recent studies have provided insights into the persistence and succession of microbes aboard the International Space Station (ISS), notably the dominance of Klebsiella pneumoniae. However, the interactions between the various microbes aboard the ISS and how they shape the microbiome remain to be clearly understood. In this study, we apply a computational approach to predict possible metabolic interactions in the ISS microbiome and shed further light on its organization. RESULTS: Through a combination of a systems-based graph-theoretical approach, and a constraint-based community m

Microbial-gut interactions in health and disease. Progress in enteric vaccine development.

Enteric infections resulting in diarrhoea are among the most important causes of morbidity and mortality, particularly in children in developing countries. They are also a common cause of disease among travellers to Africa, Asia and Latin America. Recently, effective, live and inactivated oral and parenteral vaccines against some of the most severe enteric infections-cholera and typhoid fever-have been licensed in several countries. Different candidate vaccines against rotavirus, Shigella and ETEC diarrhoea have also been developed and tested for safety and immunogenicity in developed as well

Microbial Metabolite-Stimulated Bitter Taste Receptor T2R14 Signaling Is Modulated by CFTR Interactions.

Bitter taste receptors (TAS2Rs or T2Rs) are a subset of G protein-coupled receptors (GPCRs) that play a key role in responding to microbial presence at epithelial surfaces. We previously reported that mutations in the cystic fibrosis transmembrane conductance regulator (CFTR) affect innate immune signaling from the bitter taste receptor T2R14, but the mechanisms remain understudied. Here we employ multiple biophysical tools to investigate T2R14 and CFTR interactions and its effect on signaling. The data indicate that either the N-terminus or the NBD2/C-terminus of CFTR can independently intera
