Microinjection of Micrococcus luteus UV-endonuclease restores UV-induced unscheduled DNA synthesis in cells of 9 xeroderma pigmentosum complementation groups.

The UV-induced unscheduled DNA synthesis (UDS) in cultured cells of excision-deficient xeroderma pigmentosum (XP) complementation groups A through I was assayed after injection of Micrococcus luteus UV-endonuclease using glass microneedles. In all complementation groups a restoration of the UV-induced UDS, in some cells to the repair-proficient human level, was observed. Another prokaryotic DNA-repair enzyme, T4 endonuclease V, restored the UV-induced UDS in a similar way after microinjection into XP cells. Since both enzymes specifically catalyse only the incision of UV-irradiated DNA, we con

Synovial Fluid-derived Micrococcus Luteus G18 Exacerbates Osteoarthritis Progression by Promoting Chondrocyte Degradation via TLR2/JNK/AP-1 Signaling Pathway.

Traditionally considered a sterile environment, the articular cavity has this perception overturned with advances in multi-omics technologies-microbial communities are increasingly identified in once-thought sterile tissues, making the articular cavity a potential microbial niche. However, the presence and role of intra-articular microbes in osteoarthritis (OA) remain rarely studied. Synovial fluid microbiomes of OA patients at different stages are analyzed via 16S rRNA high-throughput sequencing, and the results show that microbial diversity is positively correlated with OA progression, with

The resuscitation-promoting factor (Rpf) from Micrococcus luteus and its putative reaction product 1,6-anhydro-MurNAc increase culturability of environmental bacteria.

Dormant bacterial cells do not divide and are not immediately culturable, but they persist in a state of low metabolic activity, a physiological state having clinical relevance, for instance in latent tuberculosis. Resuscitation-promoting factors (Rpfs) are proteins that act as signalling molecules mediating growth and replication. In this study we aimed to test the effect of Rpfs from Micrococcus luteus on the number and diversity of cultured bacteria using insect and soil samples, and to examine if the increase in culturability could be reproduced with the putative reaction product of Rpf, 1

Deciphering antibiotic resistance, quorum sensing, and biofilm forming genes of Micrococcus luteus from hemodialysis tunneled cuffed catheter tips of renal failure patients.

Catheter-related bloodstream infections create a significant challenge in healthcare system, often complicated by antibiotic resistance and biofilm formation of multi-drug resistance and virulent bacterial pathogens. This study focused on biofilm-forming efficiency, and underlying genetic mechanisms in Micrococcus luteus HL_Chru_C3, isolated from a hemodialysis catheter tip. The isolate exhibited resistance to multiple antibiotic classes, including beta-lactams and glycopeptides. Biofilm assays revealed that M. luteus HL_Chru_C3 formed optimum biofilms at high concentration of carbohydrates (5

Micrococcus luteus cells and cell walls induce anaphylactoid reactions accompanied by early death and serum cytokines in mice primed with muramyl dipeptide.

Micrococcus luteus strains at a dose of 500 microg of whole cells caused anaphylactoid reactions leading to death in some instances within 1 h in C3H/HeN mice primed with muramyl dipeptide (MDP, 100 microg). Tumor necrosis factor (TNF) and interleukin-6 (IL-6) were induced in the serum of half and of all the surviving mice, respectively. Cell wall specimens of M. luteus so far examined also caused anaphylactoid reactions accompanied by early death and one strain induced high levels of TNF and IL-6. Cytoplasmic membranes also induced IL-6. Essentially similar results were obtained with represen

Natural solution for the remediation of multi-metal contamination: application of natural amino acids, Pseudomonas fluorescens and Micrococcus yunnanensis to increase the phytoremediation efficiency.

Natural amino acids (NAA) have been rarely investigated as chelators, despite their ability to chelate heavy metals (HMs). In the present research, the effects of extracted natural amino acids, as a natural and environmentally friendly chelate agent and the inoculation of Pseudomonas fluorescens (PF) and Micrococcus yunnanensis (MY) bacteria were investigated on some responses of quinoa in a soil polluted with Pb, Ni, Cd, and Zn. Inoculation of PGPR bacteria enhanced plant growth and phytoremediation efficiency. Pb and Cd were higher in quinoa roots, while Ni and Zn were higher in the shoots.

Induction of necrosis factor-alpha and interleukin-6 in mice in vivo and in murine peritoneal macrophages and human whole blood cells in vitro by Micrococcus luteus teichuronic acids.

Earlier studies showed that Micrococcus luteus cells and cell walls induced anaphylactoid reactions leading to death, in some instances within 1 h, in C3H/HeN mice primed with muramyl dipeptide (MDP). They also induced serum cytokines in the surviving mice. The present study investigated the structural components responsible for these activities. Teichuronic acids, a component of M. luteus cell walls, induced tumour necrosis factor-alpha (TNF-alpha) and interleukin-6 (IL-6) in MDP-primed C3H/HeN mice. Peptidoglycans had little effect on the cytokine-inducing activities. Reducing teichuronic ac

Increased sensitivity of UV-repair-deficient human cells to DNA bound platinum products which unlike thymine dimers are not recognized by an endonuclease extracted from Micrococcus luteus.

We have studied the response of human cells in culture to cis platinum[II] diammine dichloride (cis Pt[II]) induced DNA damage. The survival data, measured as a function of cis Pt[II] dose were similar in a normal cell line (Human foetal lung) compared to a UV-sensitive, thymine dimer excision repair-deficient cell line (Xeroderma pigmentosum). However, there was a marked difference between the two cell lines when binding to DNA was plotted against dose of cis Pt[II] given for 1 h. When these findings were expressed as cell survival versus binding to DNA, a 4.1--fold difference between the slo

Coating silicon catheters with the optimized and stable carotenoid bioproduct from Micrococcus luteus inhibited the biofilm formation by multidrug-resistant Enterococcus faecalis via downregulation of GelE gene expression.

BACKGROUND: Microbial carotenoids have gained industry interest due to their safety and diverse biological activities; however, the low yield of carotenoids hinders their applications. Hence, this study focused on optimizing carotenoid pigment production from Micrococcus luteus strains by studying 54 physical and chemical independent conditions. The chronic infections by Enterococcus faecalis are related to its ability to form biofilms on the surface of several implanted medical devices, such as urinary catheters. Therefore, the potential antibacterial and antibiofilm activities of the purifie

Batch effects account for the main findings of an in utero human intestinal bacterial colonization study.

A recent study by Rackaityte et al. reported evidence for a low level of bacterial colonization, specifically of Micrococcus luteus, in the intestine of second trimester human fetuses. We have re-analyzed their sequence data and identified a batch effect which violates the underlying assumptions of the bioinformatic method used for contamination removal. This batch effect resulted in Micrococcus not being identified as a contaminant in the original work and being falsely assigned to the fetal samples. We further provide evidence that the micrographs presented by Rackaityte et al. are unlikely
