Tumor heterogeneity drives cancer progression, dissemination, therapeutic adaptation, and relapse, yet many clinically relevant cell states are rare, transient, context-dependent, and obscured by population-averaged analysis. This review examines how microfluidic platforms preserve biologically meaningful linkages among cell identity, molecular state, secreted output, functional phenotype, perturbation history, and microenvironmental context, which are frequently disrupted by conventional workflows. We first define analytical requirements imposed by tumor heterogeneity, then examine microwell-

We developed a low-cost polymer-film spiral inertial microfluidic device for the effective size-dependent separation of malignant tumor cells. The device was fabricated in polymer films by rapid laser cutting and chemical bonding. After fabricating the prototype device, the separation performance of our device was evaluated using particles and cells. The effects of operational flow rate, cell diameter, and cell concentration on the separation performance were explored. Our device successfully separated tumor cells from polydisperse white blood cells according to their different migration modes

Magnetic relaxation biosensing (MRB) is inherently limited by relaxation noise arising from magnetic bead heterogeneity and diffusion-mediated averaging in bulk aqueous environments. Here, we present a microdroplet-confined magnetic relaxation biosensing (mc-MRB) strategy that enhances signal stability and measurement sensitivity by physically regulating both magnetic perturbers and their relaxation microenvironment. A refined magnetophoretic-inertial microfluidic module removes low-moment and aggregated beads, yielding a homogeneous immunomagnetic nanobead population. Subsequent confinement w

In this paper, we propose a microfluidic platform toseparate magnetic particles with a constant volumetric flow condition. In order to realize this architecture, three main functions for a droplet manipulation based system are integrated into a single device: synchronization of droplets by matching a location of droplets; lateral electro-coalescence; and magnetic particle manipulation. For an optimized condition of this device, a droplet generation was controlled by varying a droplet size at a fixed flow rate ratio 0.86. The electro-coalescence efficiency and maximum throughput are investigate

Microfluidics can achieve the spatiotemporal manipulation of particles and cells in the microscale fluids, but highly relies on the accuracy of the pumping systems. To overcome this issue, a pump-free, hydraulic-amplification oscillatory microfluidic (PHOMF) device is presented, which can be actuated by fingers to handle particles and cells within the microchannel. The PHOMF device has a hydraulic-amplification module for pressure transfer and a soft microchannel module for the generation of oscillatory flows. This is made possible by the periodic transfer of finger-driven liquid pressure to t

Systemic lupus erythematosus (SLE) is a chronic autoimmune disease, with anti-double-stranded DNA (anti-dsDNA) antibodies as its serological biomarkers. However, conventional anti-dsDNA antibody detection methods, which mainly rely on antibody-binding assays, often suffer from limited sensitivity and specificity, cumbersome procedures, and poor suitability for accurate clinical analysis. Herein, we developed an integrated detection system combining a circularly permuted fluorescent protein (cpFP)-based biosensor with a microfluidic chip for rapid and reliable anti-dsDNA antibody detection. Understanding the impact of forces generated by blood flow on biological processes in the circulatory system, such as the invasion of human red blood cells by malaria parasites, is currently limited by the lack of experimental systems that integrate them. Recent systematic quantification of the growth of Plasmodium falciparum, the species that causes the majority of malaria mortality, under a range of shaking conditions has shown that parasite invasion of erythrocytes is affected by the shear stress to which the interacting P. falciparum merozoites and their target red blood cells are exposed.

Single-cell technologies are emerging as a complementary approach to conventional laboratory-scale cultivation setups. Particularly in microfluidic cultivation and screening, precisely controlled environmental conditions and defined minimal media become important for quantitative studies. Nevertheless, conventional bulk media optimized for other scales, are often employed despite differences in nutrient requirements across varying scales and cultivation modes. This study demonstrates the limitations of using bulk media for single-cell analysis of Corynebacterium glutamicum and highlights the n

Red blood cell (RBC) aggregation and viscosity-related flow resistance are important hemorheological parameters for assessing blood flow abnormalities, but their simultaneous measurement often requires multiple pumps or intermittent flow stoppage. In this study, we propose a single syringe pump microfluidic sensing method for simultaneous evaluation of RBC aggregation and transient flow response under continuous pulsatile blood delivery. The device consists of a single inlet, a main straight channel, a bifurcated test channel, and a big outlet. An optimized pulsatile-flow profile was applied b

Capillary flow-driven microfluidic devices enable autonomous fluid transport for bioanalytical applications by using surface tension and wettability in microscale channels and porous structures, eliminating the need for external pumps. This review develops a coherent physical framework for capillary devices, based on the Young-Laplace equation, Jurin's law, the Lucas-Washburn equation, and Darcy's law, that relates interfacial curvature, capillary pressure, viscous dissipation, and permeability to fluid behavior in both hollow channels and porous media. From this framework, we derive practical

PCR is indispensable in basic science and biotechnology for in-orbit life science research. However, manpower and resources are limited in space. To address the constraints of in-orbit PCR, we proposed an oscillatory-flow PCR technique based on biaxial centrifugation. Oscillatory-flow PCR remarkably reduces the power requirements of the PCR process and has a relatively high ramp rate. A microfluidic chip that could perform dispensing, volume correction, and oscillatory-flow PCR of four samples simultaneously using biaxial centrifugation was designed. An automatic biaxial centrifugation device

Integrating nucleic acid extraction in amplification-based point-of-care diagnostics will be a significant feature for next-generation point-of-care virus detection devices. However, extracting DNA efficiently on a microfluidic chip poses many technological and commercialization challenges, including manual steps, multiple instruments, pretreatment processes, and the use of organic solvents (ethanol, IPA) that inhibit detection, which is not viable with routine testing such as viral load monitoring of transplant patients for post-operative care. This paper presents a microfluidic system capabl

Microbial habitats in nature are often characterized by low concentrations of mixed nutrients, spatial heterogeneity, and temporal fluctuations. However, traditional laboratory culturing methods fail to replicate these conditions. Batch cultures cannot sustain growth in low-nutrient environments, while chemostats maintain steady-state growth with a single limiting nutrient but are challenging to implement when the goal is to maintain defined low concentrations of nutrient mixtures or to introduce rapid fluctuations. Microfluidic systems generate dynamic environments but yield insufficient biom

The content release of single giant liposomes (40-100 μm diameter) confined in a microfluidic device was investigated by combining amperometric, fluorescence, and electrochemiluminescence (ECL) monitoring techniques. Accordingly, a microfluidic device was designed to guide, isolate, and immobilize without altering a single liposome in contact with a semitransparent platinum electrode, in a micrometric channel compatible with luminescence microscopy observations. Sealed giant asymmetrical liposomes were made from DOPG/DOPC phospholipids (i.e., 1,2-dioleoyl-sn-glycerol-3-phospho-(1'-rac-glycerol

The pathogenic bacterium Pseudomonas aeruginosa is a major cause of numerous nosocomial infections, and its growing antimicrobial resistance has led to it posing a significant public health threat. This article presents a comprehensive protocol detailing the use of a microfluidic system for the real-time visualization and quantification of biofilm development in two key P. aeruginosa strains, PAO1 and PA14. The method employs optically transparent, multichannel microchannel plates to subject bacterial cultures to a continuous, steady flow of media, including tryptic soy broth (TSB) or modified

Digital microfluidics (DMF) enables programmable manipulation of discrete droplets for multi-step biochemical workflows; however, integrating polymerase chain reaction (PCR) into reconfigurable DMF systems remains challenging due to evaporation, surface fouling, and loss of droplet mobility during PCR. Here, we present a dual-phase pinning system (DPPS) for stable and reversible static-site PCR in DMF. In this approach, an aqueous PCR droplet is locally encapsulated within an immiscible lubricating phase and stabilized through combined mechanical confinement and continuous electrowetting actua

We report the results of a label-free analysis of ribonuclease activity using droplet-based microfluidics. The ribonucleolytic activity of ribonucleases (RNases) plays a critical role in cellular functions such as development, survival, growth and differentiation. Altered ribonucleolytic activity and/or the expression level of the RNase A family are known to be associated with pancreatic, bladder, ovarian and thyroid cancers among others. For this reason, the RNase A family is a meaningful protein biomarker that can be used in the diagnosis of cancer and as a target for new drug screening. The

We present the use of a simple microfluidic technique to separate living parasites from human blood. Parasitic trypanosomatids cause a range of human and animal diseases. African trypanosomes, responsible for human African trypanosomiasis (sleeping sickness), live free in the blood and other tissue fluids. Diagnosis relies on detection and due to their often low numbers against an overwhelming background of predominantly red blood cells it is crucial to separate the parasites from the blood. By modifying the method of deterministic lateral displacement, confining parasites and red blood cells

Microfluidic tools enable manipulation of microliter-scale volumes within a stable microenvironment. Microfluidics has been widely adopted in various biological fields for, among other applications, controlled delivery of sensory, chemical, and mechanical stimuli to the observed organisms. In this chapter, we describe the design and usage of a custom-made double-layered PDMS microfluidic device, named BUGchip, developed for the purpose of studying plant root growth and metabolic responses in rapidly changing conditions with a high time-scale resolution. We demonstrate the utility of this devic
