Theory and Simulations of Atomic Force Microscopy-Based Two-dimensional Electronic Spectroscopy.

Probing excited states and ultrafast dynamics in heterogeneous and interfacial systems requires enhanced spatial and temporal resolution. Coherent two-dimensional electronic spectroscopy (2DES) offers a subpicosecond temporal resolution but its spatial resolution is restricted by the diffraction limit. Atomic force microscopy (AFM) overcomes the diffraction limit and routinely attains a sub-10 nm spatial resolution. In this Letter we establish a theoretical framework for the AFM-detected 2DES spectroscopy─a technique that is yet to be experimentally realized but promises to deliver high tempor

Temperature-Dependent Chain Structures during Solution-Grown Crystallization via Atomic Force Microscopy.

Semi-crystalline polymers are widely utilized due to their favorable cost, and their crystallization behavior is significantly influenced by crystallization temperature. In this study, we employed AFM-based SMFS (single-molecule force spectroscopy) to investigate single-chain folding structures in Polyethylene Oxide (PEO) at various crystallization temperatures (Tc) to elucidate the temperature effect. We utilized force-volume (FV) mode for SMFS, which enables the measurement of single-chain information while simultaneously obtaining information about the surface morphology of the crystal. By

Molecular-level studies of extracellular matrix proteins conducted using atomic force microscopy.

Extracellular matrix (ECM) proteins provide anchorage and structural strength to cells and tissues in the body and, thus, are fundamental molecular components for processes of cell proliferation, growth, and function. Atomic force microscopy (AFM) has increasingly become a valuable approach for studying biological molecules such as ECM proteins at the level of individual molecules. Operational modes of AFM can be used to acquire the measurements of the physical, electronic, and mechanical properties of samples, as well as for viewing the intricate details of the surface chemistry of samples. Applying the Atomic Force Microscopy Technique in Medical Sciences-A Narrative Review.

Penetrating deep into the cells of the human body in real time has become increasingly possible with the implementation of modern technologies in medicine. Atomic force microscopy (AFM) enables the effective live imaging of cellular and molecular structures of biological samples (such as cells surfaces, components of biological membranes, cell nuclei, actin networks, proteins, and DNA) and provides three-dimensional surface visualization (in X-, Y-, and Z-planes). Furthermore, the AFM technique enables the study of the mechanical, electrical, and magnetic properties of cells and cell organelle

Atomic force microscopy reveals morphological and mechanical properties of schistosoma mansoni tegument.

Schistosoma mansoni, an intravascular parasitic worm and the causative agent of schistosomiasis, relies on its tegument (outer layer) for survival and host interaction. This study explored the morphology and mechanical properties of S. mansoni tegument using Atomic Force Microscopy (AFM). Notably, we employed the PeakForce Quantitative Nanomechanical Mapping (PF-QNM) mode in air, enabling simultaneous acquisition of 3D topography and mechanical property contrasts (adhesion, elastic modulus). Additionally, nanoindentation (AFM contact mode) was performed on female worm tegument for elastic modu

Quantification of bush-cricket acoustic trachea mechanics using Atomic Force Microscopy nanoindentation.

Derived from the respiratory tracheae, bush-crickets' acoustic tracheae (or ear canals) are hollow tubes evolved to transmit sounds from the external environment to the interior ear. Due to the location of the ears in the forelegs, the acoustic trachea serves as a structural element that can withstand large stresses during locomotion. In this study, we report a new Atomic Force Microscopy Force Spectroscopy (AFM-FS) approach to quantify the mechanics of taenidia in the bush-cricket Mecopoda elongata. Mechanical properties were examined over the longitudinal axis of hydrated taenidia, by indent

Method to Quantify Nanoscale Surface Charge in Liquid with Atomic Force Microscopy.

A theory is presented to obtain surface charge density on nanoscale objects from data in the snap-to-contact portion of an atomic force microscope force-separation curve. The mathematical model takes into account the tip's dielectric constant using the Self-Consistent Sum of Dipoles theory which includes the charge-charge interaction and the charge-dipole interaction with electrolyte-induced exponentially decaying screening, Debye and London dipolar force, and fluid viscosity including confined fluid layers to account for energy dissipation. Using previously published experimental data, the ma

Atomic Force Microscopy Investigations of DNA Lesion Recognition in Nucleotide Excision Repair.

AFM imaging is a powerful technique for the study of protein-DNA interactions. This single molecule method allows the simultaneous resolution of different molecules and molecular assemblies in a heterogeneous sample. In the particular context of DNA interacting protein systems, different protein complex forms and their corresponding binding positions on target sites containing DNA fragments can thus be distinguished. Here, an application of AFM to the study of DNA lesion recognition in the prokaryotic and eukaryotic nucleotide excision DNA repair (NER) systems is presented. The procedures of D

Adhesion of resin-based sealers to dentine: an atomic force microscopy study.

AIM: To evaluate the effect of several final irrigants on the adhesion force (Fad) between dentine and resin-based sealers by means of atomic force microscopy (AFM). METHODOLOGY: Twelve distal roots of mandibular molars were used. The roots were smoothened and cut to obtain 36 specimens. During chemomechanical preparation of the root canals, 5.25% NaOCl was used as the irrigant. The specimens were then divided into six groups according to the final irrigant used: control group - immersed in distilled water (DW) for 1 min; chlorhexidine (CHX) group - in 2% CHX for 1 min; NaOCl group - in 5.25%

Local micromechanical properties of decellularized lung scaffolds measured with atomic force microscopy.

Bioartificial lungs re-engineered from decellularized organ scaffolds are a promising alternative to lung transplantation. Critical features for improving scaffold repopulation depend on the mechanical properties of the cell microenvironment. However, the mechanics of the lung extracellular matrix (ECM) is poorly defined. The local mechanical properties of the ECM were measured in different regions of decellularized rat lung scaffolds with atomic force microscopy. Lungs excised from rats (n=11) were decellularized with sodium dodecyl sulfate (SDS) and cut into ~7μm thick slices. The complex el

Atomic force microscopy-based cell nanostructure for ligand-conjugated quantum dot endocytosis.

While it has been well demonstrated that quantum dots (QDs) play an important role in biological labeling both in vitro and in vivo, there is no report describing the cellular nanostructure basis of receptor-mediated endocytosis. Here, nanostructure evolution responses to the endocytosis of transferrin (Tf)-conjugated QDs were characterized by atomic force microscopy (AFM). AFM-based nanostructure analysis demonstrated that the Tf-conjugated QDs were specifically and tightly bound to the cell receptors and the nanostructure evolution is highly correlated with the cell membrane receptor-mediate

Enhancement of topographic images obtained in liquid media by atomic force microscopy.

The open liquid-cell atomic force microscope (AFM) has potential for studies of biomaterials and surface morphology in liquid media, and a variety of fluids can be used as buffer solutions. The dependence of image distortion on fluid properties (kinematic viscosity) has been studied with edge friction force obtained in lateral images and will shortly appear elsewhere [Appl. Phys. Lett. 2006, 88, 173121]. Previous studies indicate that the scan rate should be slower for obtaining a nondistorted image. However, the time required for the scan is greatly increased. Therefore, we introduced the vec

Atomic force microscopy and confocal laser scanning microscopy on the cytoskeleton of permeabilised and embedded cells.

We describe a technical method of cell permeabilisation and embedding to study the organisation and distribution of intracellular proteins with aid of atomic force microscopy and confocal laser scanning microscopy in identical areas. While confocal laser scanning microscopy is useful for the identification of certain proteins subsequent labelling with markers or antibodies, atomic force microscopy allows the observation of macromolecular structures in fixed and living cells. To demonstrate the field of application of this preparatory technique, cells were permeabilised, fixed, and the actin cy

Evaluation of the influence of albumin on the mineralization of a glass by Atomic Force Microscopy.

Bioactive glasses have been used as a graft material that can stimulate the formation of a new bone. In vitro tests usually give sensible indications about the potential bioactivity of these glasses. In the present work the influence of egg albumin on the formation of a Ca-P precipitate on a glass of the system SiO(2)-CaO-MgO-P(2)O(5) was evaluated. The samples were immersed in simulated body fluid (SBF) that simulates the composition of human plasma, with and without albumin. After immersion in this solution for 7 and 14 days, the glass was characterized by X-Ray Diffraction (XRD) and Atomic

Measuring Molecular Forces With Atomic Force Microscopy 1: Solvent Influence on Hydrophobic Interactions.

Molecular forces drive phenomena such as self-assembly, aggregation, and protein folding, where hydrophobic interactions are paramount. However, the origin of the hydrophobic mechanism remains unknown. Advances in techniques like atomic force microscopy (AFM) have improved our ability to study this topic. Hydrophobic interactions are stronger and longer ranged than van der Waals (vdW) forces, potentially arising from water structuring, polarization, and entropic effects. In this primer, fluorocarbon surfaces were prepared via chemical vapor deposition (CVD) on gold to explore the impact of wat

Single integrin molecule adhesion forces in intact cells measured by atomic force microscopy.

Cross-talk between cells and the extracellular matrix is critically influenced by the mechanical properties of cell surface receptor-ligand interactions; these interactions are especially well defined and regulated in cells capable of dynamically modifying their matrix environment. In this study, attention was focused on osteoclasts, which are absolutely dependent on integrin extracellular matrix receptors in order to degrade bone; other bone cells, osteoblasts, were used for comparison. Integrin binding forces were measured in intact cells by atomic force microscopy (AFM) for several RGD-cont

Differential susceptibility of Streptococcus dentisani to dental surfaces: An atomic force microscopy study.

BACKGROUND: Given frequent dental restorations, understanding the interactions of probiotic Streptococcus dentisani with enamel and dental materials is key, in contrast to the well-studied Streptococcus mutans. This knowledge is vital for the potential applications in promoting oral health of S. dentisani. OBJECTIVE: This study aims to evaluate and compare S. mutans and S. dentisani initial adhesion, proliferation, and colonization on dental enamel and commonly used dental materials: nickel-chromium alloy, porcelain, lithium disilicate, autocured, and thermocured acrylics, using atomic force

Interaction of βL- and γ-Crystallin with Phospholipid Membrane Using Atomic Force Microscopy.

Highly concentrated lens proteins, mostly β- and γ-crystallin, are responsible for maintaining the structure and refractivity of the eye lens. However, with aging and cataract formation, β- and γ-crystallin are associated with the lens membrane or other lens proteins forming high-molecular-weight proteins, which further associate with the lens membrane, leading to light scattering and cataract development. The mechanism by which β- and γ-crystallin are associated with the lens membrane is unknown. This work aims to study the interaction of β- and γ-crystallin with the phospholipid membrane wit

Atomic force microscopy analysis of native infectious and inactivated SARS-CoV-2 virions.

SARS-CoV-2 is an enveloped virus responsible for the Coronavirus Disease 2019 (COVID-19) pandemic. Here, single viruses were analyzed by atomic force microscopy (AFM) operating directly in a level 3 biosafety (BSL3) facility, which appeared as a fast and powerful method to assess at the nanoscale level and in 3D infectious virus morphology in its native conformation, or upon inactivation treatments. AFM imaging reveals structurally intact infectious and inactivated SARS-CoV-2 upon low concentration of formaldehyde treatment. This protocol combining AFM and plaque assays allows the preparation
