Intravital two-photon microscopy of the native mouse thymus.

The thymus, a key organ in the adaptive immune system, is sensitive to a variety of insults including cytotoxic preconditioning, which leads to atrophy, compression of the blood vascular system, and alterations in hemodynamics. Although the thymus has innate regenerative capabilities, the production of T cells relies on the trafficking of lymphoid progenitors from the bone marrow through the altered thymic blood vascular system. Our understanding of thymic blood vascular hemodynamics is limited due to technical challenges associated with accessing the native thymus in live mice. To overcome th

Current Landscape of Advanced Imaging Tools for Pathology Diagnostics.

Histopathology relies on century-old workflows of formalin fixation, paraffin embedding, sectioning, and staining tissue specimens on glass slides. Despite being robust, this conventional process is slow, labor-intensive, and limited to providing two-dimensional views. Emerging technologies promise to enhance and accelerate histopathology. Slide-free microscopy allows rapid imaging of fresh, unsectioned specimens, overcoming slide preparation delays. Methods such as fluorescence confocal microscopy, multiphoton microscopy, along with more recent innovations including microscopy with UV surface

Advancements in organic fluorescent materials: unveiling the potential of peripheral group modification in dithienyl-diketopyrrolopyrrole derivatives for one- and two-photon bioimaging.

The quest for novel organic fluorescent materials capable of two-photon absorption (2PA) has intensified in recent years due to their promising applications in biological imaging. Two-photon fluorescence microscopy (2PFM) offers high spatial-temporal resolution, reduced photodamage, and deeper tissue penetration compared to conventional techniques. However, the development of bright two-photon molecular markers remains a challenge, necessitating compounds with high fluorescence quantum yield and 2PA cross-section (σ2PA). Strategies such as increasing π-conjugation have shown promise but are hi

High-resolution, label-free two-photon imaging of diseased human corneas.

The diagnosis of corneal diseases may be improved by monitoring the metabolism of cells and the structural organization of the stroma using two-photon imaging (TPI). We used TPI to assess the differences between nonpathological (NP) human corneas and corneas diagnosed with either keratoconus, Acanthamoeba keratitis, or stromal corneal scars. Images were acquired using a custom-built five-dimensional laser-scanning microscope with a broadband sub-15 femtosecond near-infrared pulsed excitation laser and a 16-channel photomultiplier tube detector in combination with a time-correlated single photo

Frontiers in Intravital Multiphoton Microscopy of Cancer.

BACKGROUND: Cancer is a highly complex disease which involves the co-operation of tumor cells with multiple types of host cells and the extracellular matrix. Cancer studies which rely solely on static measurements of individual cell types are insufficient to dissect this complexity. In the last two decades, intravital microscopy has established itself as a powerful technique that can significantly improve our understanding of cancer by revealing the dynamic interactions governing cancer initiation, progression and treatment effects, in living animals. This review focuses on intravital multipho

Phasor-Based Spatio-Spectral Segmentation for Hyperspectral Fluorescence Microscopy.

Fluorescence microscopy is essential for visualizing cellular and tissue structures. When combined with a hyperspectral modality, it enables the acquisition of emission spectra that provide insight into the composition of fluorophores. These spectra can be used for fluorophore-based image segmentation; however, accurate spectral separation becomes challenging when fluorophore emission spectra overlap and mix within individual pixels. We present a spatially-corrected density peak clustering (SC-DPC) method that combines phasor transformation with histogram-based density-peak detection and addit

Distinguishing between benign and malignant melanocytic nevi by in vivo multiphoton microscopy.

Monitoring of atypical nevi is an important step in early detection of melanoma, a clinical imperative in preventing the disease progression. Current standard diagnosis is based on biopsy and histopathologic examination, a method that is invasive and highly dependent upon physician experience. In this work, we used a clinical multiphoton microscope to image in vivo and noninvasively melanocytic nevi at three different stages: common nevi without dysplastic changes, dysplastic nevi with structural and architectural atypia, and melanoma. We analyzed multiphoton microscopy (MPM) images correspond

A hexaphenylbenzene based AIEE active two photon probe for the detection of hydrogen sulfide with tunable self-assembly in aqueous media and application in live cell imaging.

Supramolecular aggregates of hexaphenylbenzene derivative exhibit aggregation induced emission enhancement and modulation of self-assembled architecture from spherical to flower-like assembly in the presence of H2S. Furthermore, probe displays higher photostability, low toxicity and bright green fluorescence in two-photon microscopy (TPM) imaging for the detection of H2S in live HeLa cells.

Differentiating morphea from lichen Sclerosus by using multiphoton microscopy combined with U-Net model for elastic fiber segmentation.

This paper describes a methodology to differentiate morphea from lichen sclerosus based on examination with multiphoton microscopy (MPM) composed of two-photon excited fluorescence (TPEF) and second harmonic generation (SHG). Subcellular-resolution images were acquired by MPM from unstained lesion tissues then process spectral analysis to quantify the TPEF and SHG signals. Moreover, U-Net was employed to segment elastic fiber in TPEF images to combine with collagen fiber in SHG images for precise fiber quantification. Predictions of segmentation showed excellent performance on several evaluati

Intravital 2-Photon Microscopy of Diverse Cell Types in the Murine Tibia.

Intravital imaging allows the visualization of fluorescently labeled structures like cells, blood flow, and pathogens in a living organism. Nowadays, numerous methods for imaging in several organs are available. In this chapter, we present a method for intravital 2-photon microscopy of the murine tibial bone marrow. It enables the observation of hematopoietic cells including cells of the innate and adaptive immune system under physiological conditions. Motility analyses within this complex environment led to insights into their migratory potential as well as their interactions with other cells

Intravital Microscopy of the Beating Murine Heart to Understand Cardiac Leukocyte Dynamics.

Cardiovascular disease is the leading cause of worldwide mortality. Intravital microscopy has provided unprecedented insight into leukocyte biology by enabling the visualization of dynamic responses within living organ systems at the cell-scale. The heart presents a uniquely dynamic microenvironment driven by periodic, synchronous electrical conduction leading to rhythmic contractions of cardiomyocytes, and phasic coronary blood flow. In addition to functions shared throughout the body, immune cells have specific functions in the heart including tissue-resident macrophage-facilitated electrica

A lysosome targetable and acidic pH-activated fluorescent probe for two-photon imaging of hydrogen sulfide in live cells and animals.

The design strategy of targeted fluorescent probes usually involves modifying the probes with organelle-targeting groups, which then guide the probes to enter specific organelles by the traction of the targeting groups. However, the targeting ability of subcellular localization groups is not perfect, and the target analytes are not specifically distributed within the cells. Therefore, designing novel molecular fluorescent probes for highly specific and selective detection of target analytes within specific organelles is an important research direction that deserves attention. Although multiple

Dual deconvolution in multiphoton structured illumination microscopy for deep-tissue super-resolution imaging.

Imaging in thick biological tissues is often degraded by sample-induced aberrations, which reduce resolution and contrast, particularly in super-resolution techniques. While hardware-based adaptive optics (AO) using wavefront shaping can correct these aberrations, their complexity and cost hinder widespread adoption. Here, we present a computational AO framework for multiphoton structured illumination microscopy, enabling deep-tissue super-resolution imaging with minimal hardware modifications. By replacing the photodetector with a camera from the conventional laser-scanning multiphoton micros

One- and two-photon induced fluorescence of Pacific Blue-labeled human serum albumin deposited on different core size silver colloids.

We studied one- and two-photon induced fluorescence of Pacific Blue (PB)-labeled human serum albumin (HSA) in the presence of different size silver colloids. The PB fluorescence emission intensity was observed with small (30-40 nm) and large (about 120 nm) colloids and compared with PB emission in absence of colloids. For the system with a small core size colloids we did not detect any fluorescence enhancement with one-photon excitation and the enhancement observed with two-photon excitation was about 2.5-fold. In contrast, for large silver colloids we observed about a 2-fold increase in PB fl

Tunable Bessel beam two-photon fluorescence microscopy for high-speed volumetric imaging of brain dynamics.

High-speed volumetric imaging of the brain is essential for linking diverse cellular events to tissue-level functions. However, the brain's structural and dynamic heterogeneity-spanning microns to millimeters and milliseconds to hours-requires imaging techniques with tunable spatiotemporal resolution, flexible 3D sampling, and compatibility with targeted perturbations. Here, we present tunable Bessel beam two-photon fluorescence microscopy (tBessel-TPFM), a compact, low-cost, and versatile platform for intravital brain imaging across millimeter scale with subcellular resolution. tBessel-TPFM t

Two-Photon Fabrication of Donor-Specific Human Lamina Cribrosa Models.

PURPOSE: The lamina cribrosa (LC) is a complex network of collagenous beams that maintains mechanical homeostasis in response to fluctuations in intraocular pressure, thus protecting retinal ganglion cell axons exiting the eye. Understanding the structure and function of the LC may provide new insights into glaucomatous neurodegeneration. The purpose of this study is to utilize a two-photon fabrication technique to fabricate a model of the human LC (mLC) to scale. METHODS: Segmented multiphoton microscopy images of human LC tissues from a prior study were used to two-photon polymerize three m

Insights into cell-to-cell and cell-to-blood-vessel communications in the brain: in vivo multiphoton microscopy.

A complex and reciprocal communication of cells with each other and with relevant parts of the tissue stroma governs many biological processes in both health and disease. However, in the past, the study of these anatomical and molecular interactions has suffered from a lack of appropriate experimental models. An imaging methodology aimed at changing this should allow intravital display and quantification in an intact non-traumatized organ, imaging over a wide range of time spans including extended periods (i.e., months), many repetitive measurements of the same cell or area to permit the study

Integrated spectrometer design with application to multiphoton microscopy.

We present a prism-based spectrometer integrated into a multifocal, multiphoton microscope. The multifocal configuration facilitates interrogation of samples under different excitation conditions. Notably, the image plane of the microscope and the image plane of the spectrometer are coincident eliminating the need for an intermediate image plane containing an entrance slit. An EM-CCD detector provides sufficient gain for spectral interrogation of single-emitters. We employ this spectrometer to observe spectral shifts in the two-photon excitation fluorescence emission of single CdSe nanodots as
