Fatal disseminated cryptococcosis in a HIV negative elderly Indian woman--an early diagnosis by polarizing microscopy.

An elderly HIV negative woman with disseminated cryptococcosis is reported. The clinical presentation was unusual. Rapid diagnosis is possible if Maltese crosses are demonstrated on polarizing microscopy of tissue sections or smears. Cutaneous involvement in cryptococcosis is uncommon, however when present, can be a valuable and relatively easily available source of material from which Cryptococci can be demonstrated.

Polarized light microscopy and digital image processing identify a multilaminar structure of the hamster zona pellucida.

The zona pellucida (zona) is a glycoprotein coat that envelopes the oocyte and embryo, binds sperm during fertilization and facilitates transfer of the embryo through the Fallopian tube. Before implantation can occur, the blastocyst must hatch from the zona. Several lines of evidence suggest that the zona is multilaminar. We hypothesized that the multilaminar structure of the zona filaments could be imaged non-destructively with the polarized light microscope. A recent modification of the polarized light microscope (pol-scope), which combines innovations in polarization optics with novel image

The effect of speed of deoxygenation on the percentage of aligned hemoglobin in sickle cells. Application of differential polarization microscopy.

We have used differential polarization microscopy, which provides images of linear dichroism, to measure the percentage of aligned hemoglobin (Hb) in 1086 deoxygenated red blood cells from subjects with sickle cell anemia. The percentage was found to be only slightly dependent on the speed of deoxygenation, thus showing that the percentage of aligned Hb was thermodynamically controlled (as has been found previously for the percentage of polymerized Hb in vitro). A slight decrease in the percentage of aligned Hb due to increasing speed of deoxygenation is primarily due to the increase in the nu

Test characteristics of Raman spectroscopy integrated with polarized light microscopy for the diagnosis of acute gouty arthritis.

OBJECTIVES: We studied the performance of Raman spectroscopy integrated with polarized light microscopy (iRPolM) as a next-generation technique for synovial fluid analysis in gout. METHODS: This is a prospective study, including consecutive synovial fluid samples drawn from any peripheral swollen joint. Diagnostic accuracy was compared to the 2015 ACR/EULAR Gout classification criteria as a reference test and to polarized light microscopy (PLM) analysis by a rheumatologist. Synovial fluid was analysed with iRPolM after unblinding the PLM results. RESULTS: Two hundred unselected consecutive p

Correlative light and scanning electron microscopy (CLSEM) for analysis of bacterial infection of polarized epithelial cells.

Infection of mammalian host cells by bacterial pathogens is a highly dynamic process and microscopy is instrumental to reveal the cellular and molecular details of host-pathogen interactions. Correlative light and electron microscopy (CLEM) combines the advantages of three-dimensional live cell imaging with ultrastructural analysis. The analyses of adhesion to, and invasion of polarized epithelial cells by pathogens often deploys scanning electron microscopy (SEM), since surface structures of the apical brush border can be analyzed in detail. Most available CLEM approaches focus on relocalizat

Cancer cell identification in tissue imaging via phasor map analysis in polarization-resolved two-photon excitation fluorescence microscopy.

While cancer characteristics can vary significantly across types, methods that distinguish malignant cells from normal ones hold promise by targeting shared cellular anomalies. Among these, morphological differences play a key role in driving the aggressive behavior and altered function typical of cancer cells. Detecting and analyzing such cells within complex, densely packed tissue environments requires advanced imaging techniques. Polarization-resolved fluorescence microscopy offers rich insights into cellular composition, molecular binding affinities, and structural organization, particular

Polarization-Resolved Second Harmonic Generation Microscopy of Silk Fibers Is Sensitive to β-Sheet Orientation and Molecular Structure.

Spider silk biomaterials have generated significant interest due to their high strength, biocompatibility, and biodegradability. However, the complex multiscale structure of silk fibers creates difficulties in investigating the relationship between the structure and mechanical properties in silks. Previous work has shown that silks at the focus of an ultrafast laser produce a significant second harmonic generation (SHG) signal. This presents an exciting opportunity since polarization-resolved SHG microscopy (PSHG) is a technique that has shown high sensitivity local molecular structure and org

Picrosirius red, polarized light microscopy, and curvelet transform analysis: An improved method for liver fibrosis assessment.

Liver fibrosis is a hallmark of chronic liver diseases, characterized by excessive extracellular matrix deposition and structural alterations. Conventional histological assessments rely on qualitative or semi-quantitative scoring systems that may lack sensitivity for subtle fibrotic changes. Here, we present an improved fibrosis evaluation method integrating Picrosirius Red (PSR) staining, polarized light (PL) microscopy, and curvelet transform-based fiber analysis (CT-FIRE). This approach provides a quantitative and reproducible assessment of collagen fiber morphology and composition. Applied

Structural differences between immature and mature articular cartilage of rabbits by microscopic MRI and polarized light microscopy.

This study aimed to determine the structural features between immature and mature articular cartilage from the humeral and femoral joints of rabbits. Specimens of articular cartilage (n = 6 for immature tissue, n = 6 for mature tissue) that were still attached to the underlying bone from a humerus (shoulder joint) or femur (knee joint) were imaged using microscopic MRI (µMRI) and polarized light microscopy (PLM). Quantitative µMRI data with a pixel resolution of 11.7-13.2 µm revealed a number of differences between the immature and mature cartilage, including total thickness, and T2 and T1ρ re

Correlative Light and Scanning Electron Microscopy to Study Interactions of Salmonella enterica with Polarized Epithelial Cell Monolayers.

Live cell fluorescence imaging is the method of choice to visualize dynamic cellular processes in time and space, such as adhesion to and invasion of polarized epithelial cells by Salmonella enterica sv. Typhimurium. Scanning electron microscopy provides highest resolution of surface structures of infected cells, providing ultrastructure of the apical side of host cells and infecting Salmonella. Combining both methods toward correlative light and scanning electron microscopy (CLSEM) enables new insights in adhesion and invasion mechanisms regarding dynamics over time, and high spatial resoluti

Incremental lines in root cementum of human teeth: an approach to their role in age estimation using polarizing microscopy.

Age estimation is an important factor in the identification of an individual in forensic science. The hard tissues of the human dentition are able to resist decay and degradation long after other tissues are lost. This resistance has made teeth useful indicators for age calculation. Recent research indicates that tooth cementum annulations (TCA) may be used more reliably than any other morphological or histological traits of the adult skeleton for age estimation. The purpose of this study was to examine the correlation between age and the number of incremental lines in human dental cementum an

Instant polarized light microscopy pi (IPOLπ) for quantitative imaging of collagen architecture and dynamics in ocular tissues.

Collagen architecture determines the biomechanical environment in the eye, and thus characterizing collagen fiber organization and biomechanics is essential to fully understand eye physiology and pathology. We recently introduced instant polarized light microscopy (IPOL) that encodes optically information about fiber orientation and retardance through a color snapshot. Although IPOL allows imaging collagen at the full acquisition speed of the camera, with excellent spatial and angular resolutions, a limitation is that the orientation-encoding color is cyclic every 90 degrees (π/2 radians). Real-Time Scanning Ion Conductance Microscopy Reveals MARCKS-Driven Morphological Remodeling and Nanomechanical Softening during LPS-Induced Macrophage Polarization.

Macrophages initiate acute immune responses to danger signals through highly plastic polarization and dynamic remodeling of cell mechanics and morphology. While physical cues are increasingly recognized as coregulators of macrophage activation alongside biochemical signals, the role of pathogen-induced biophysical remodeling in shaping immune function remains poorly understood. In this study, we employed noncontact scanning ion conductance microscopy (SICM) to monitor real-time nanomechanical and morphological changes in immortalized bone marrow-derived macrophages (iBMDMs) at single-cell reso

Live Fluorescence Lifetime Imaging Microscopy (FLIM) of the Epithelial Polarity and Cell-Specific Segmentation in the Small Intestinal Organoids.

Apical-basal polarity is essential for gastrointestinal epithelium function, where the apical membrane mediates nutrient absorption, host-microbiota interactions, and pathogen defense. However, conventional intestinal organoid culture displays an "apical-in" topology, limiting functional research with respect to the apical membrane. F-actin labeling with fluorescent phalloidin, a well-established apical marker, is also incompatible with live imaging. To address these limitations, in this chapter we describe an extracellular matrix (ECM) removal protocol to achieve "apical-out" topology in porc

Real-time imaging of optic nerve head collagen microstructure and biomechanics using instant polarized light microscopy.

Current tools lack the temporal or spatial resolution necessary to image many important aspects of the architecture and dynamics of the optic nerve head (ONH). We evaluated the potential of instant polarized light microscopy (IPOL) to overcome these limitations by leveraging the ability to capture collagen fiber orientation and density in a single image. Coronal sections through the ONH of fresh normal sheep eyes were imaged using IPOL while they were stretched using custom uniaxial or biaxial micro-stretch devices. IPOL allows identifying ONH collagen architectural details, such as fiber inte

Wide-field imaging of birefringent synovial fluid crystals using lens-free polarized microscopy for gout diagnosis.

Gout is a form of crystal arthropathy where monosodium urate (MSU) crystals deposit and elicit inflammation in a joint. Diagnosis of gout relies on identification of MSU crystals under a compensated polarized light microscope (CPLM) in synovial fluid aspirated from the patient's joint. The detection of MSU crystals by optical microscopy is enhanced by their birefringent properties. However, CPLM partially suffers from the high-cost and bulkiness of conventional lens-based microscopy, and its relatively small field-of-view (FOV) limits the efficiency and accuracy of gout diagnosis. Here we pres
