Functional mapping of the Trypanosoma cruzi serinome by fluorophosphonate activity-based protein profiling.

Serine hydrolases (SHs) constitute one of the largest enzyme superfamilies in eukaryotes, yet their roles in Trypanosoma cruzi, the causative agent of Chagas disease, remain largely uncharacterized. Here, we report an activity-based chemoproteomic map of the T. cruzi epimastigote serinome by combining genome-informed in silico curation with whole-cell activity-based protein profiling (ABPP) using a panel of cell-permeable fluorophosphonate (FP)-alkyne probes. Whole-cell labelling followed by label-free quantitative proteomics (LFQ-MS) identified 37 enriched SH-like proteins, including 35 with

Analysis of regulatory protease sequences identified through bioinformatic data mining of the Schistosoma mansoni genome.

BACKGROUND: New chemotherapeutic agents against Schistosoma mansoni, an etiological agent of human schistosomiasis, are a priority due to the emerging drug resistance and the inability of current drug treatments to prevent reinfection. Proteases have been under scrutiny as targets of immunological or chemotherapeutic anti-Schistosoma agents because of their vital role in many stages of the parasitic life cycle. Function has been established for only a handful of identified S. mansoni proteases, and the vast majority of these are the digestive proteases; very few of the conserved classes of reg

Artificial targeting of autophagy components to mitochondria reveals both conventional and unconventional mitophagy pathways.

Macroautophagy/autophagy enables lysosomal degradation of a diverse array of intracellular material. This process is essential for normal cellular function and its dysregulation is implicated in many diseases. Given this, there is much interest in understanding autophagic mechanisms of action in order to determine how it can be best targeted therapeutically. In mitophagy, the selective degradation of mitochondria via autophagy, mitochondria first need to be primed with signals that allow the recruitment of the core autophagy machinery to drive the local formation of an autophagosome around the

Sex-specific cardiometabolic remodeling induced by DPP-4 inhibition under high-fat diet: insights from cardiac proteomics.

BACKGROUND AND AIM: Dipeptidyl peptidase-4 (DPP-4) inhibitors are widely used in the management of type 2 diabetes and are generally considered cardiovascularly safe. However, their mechanistic actions on the heart under metabolic stress remain incompletely understood. METHODS AND RESULTS: In this study, we investigated sex-specific cardiac proteomic responses to the DPP-4 inhibitor linagliptin (L) in a mouse model of diet-induced obesity. Male and female C57BL/6 N mice were fed a normal chow or high-fat diet for 15 weeks and received L (0.003% w/w in the diet; estimated exposure ∼120 μg/day)

Transcriptome profile analysis reveals putative molecular mechanisms of 5-aminolevulinic acid toxicity.

5-aminolevulinic acid (5-ALA) is the first precursor of the heme biosynthesis pathway, accumulated in acute intermittent porphyria (AIP), an inherited metabolic disease characterized by porphobilinogen deaminase deficiency. An increased incidence of hepatocellular carcinoma (HCC) has been reported as a long-term manifestation in symptomatic AIP patients. 5-ALA is an α-aminoketone prone to oxidation, yielding reactive oxygen species and 4,5-dioxovaleric acid. A high concentration of 5-ALA presents deleterious pro-oxidant potential. It can induce apoptosis, DNA damage, mitochondrial dysfunction,

Tumor-derived acetylcholine promotes immune evasion in hepatocellular carcinoma by protecting regulatory T cells from ferroptosis.

Hepatocellular carcinoma (HCC) is characterized by immune suppression and limited durability of systemic therapy, yet how tumor metabolic remodeling supports regulatory T-cell (Treg) persistence remains unclear. Here, we identify a cholinergic immune-evasion pathway that links acetylcholine (ACh) accumulation to Treg ferroptosis resistance in HCC. Sorafenib-resistant patient-derived xenografts showed enhanced Treg-associated signatures together with enrichment of ferroptosis- and cholinergic signaling-related pathways. Clinical HCC tissues exhibited cholinergic remodeling, characterized by red

Enzyme replacement therapy with recombinant pro-CTSD (cathepsin D) corrects defective proteolysis and autophagy in neuronal ceroid lipofuscinosis.

CTSD (cathepsin D) is one of the major lysosomal proteases indispensable for the maintenance of cellular proteostasis by turning over substrates of endocytosis, phagocytosis and autophagy. Consequently, CTSD deficiency leads to a strong impairment of the lysosomal-autophagy machinery. In mice and humans CTSD dysfunction underlies the congenital variant (CLN10) of neuronal ceroid lipofuscinosis (NCL). NCLs are distinct lysosomal storage disorders (LSDs) sharing various hallmarks, namely accumulation of protein aggregates and ceroid lipofuscin leading to neurodegeneration and blindness. The most

Copper metabolism in cell death and autophagy.

Copper is an essential trace element in biological systems, maintaining the activity of enzymes and the function of transcription factors. However, at high concentrations, copper ions show increased toxicity by inducing regulated cell death, such as apoptosis, paraptosis, pyroptosis, ferroptosis, and cuproptosis. Furthermore, copper ions can trigger macroautophagy/autophagy, a lysosome-dependent degradation pathway that plays a dual role in regulating the survival or death fate of cells under various stress conditions. Pathologically, impaired copper metabolism due to environmental or genetic

CD200 is a potent new growth factor in Multiple Myeloma dependent on IGF1 receptor signaling.

CD200 is an immune checkpoint aberrantly expressed in 70% of newly diagnosed patients with multiple myeloma (MM) and associated with a poor outcome. CD200 expression by tumor cells inhibits immune response through interaction with CD200R and participates in resistance to immune based therapies. However, its role in tumorigenesis remains poorly understood. In this study, we show that MM cells overexpressing CD200 present a significant proliferative advantage in serum free culture medium and clonogenic assays. The cell growth advantage mediated by CD200 in MM cells is dependent on the presence o

Epigenetic modification in diabetic kidney disease.

Diabetic kidney disease (DKD) is a common microangiopathy in diabetic patients and the main cause of death in diabetic patients. The main manifestations of DKD are proteinuria and decreased renal filtration capacity. The glomerular filtration rate and urinary albumin level are two of the most important hallmarks of the progression of DKD. The classical treatment of DKD is controlling blood glucose and blood pressure. However, the commonly used clinical therapeutic strategies and the existing biomarkers only partially slow the progression of DKD and roughly predict disease progression. Therefor

Sympathetic nervous system in tumor progression and metabolic regulation: mechanisms and clinical potential.

Tumor progression is characterized by profound metabolic alterations and dynamic interactions within the tumor microenvironment (TME), which enable rapid proliferation, immunoinvasion, and metastasis. The sympathetic nervous system (SNS), which has been best known for its role in stress regulation, has emerged as a critical regulator of tumor metabolism. The SNS influences glucose, lipid and glutamine metabolism in tumor cells and stromal components by releasing neurotransmitters such as norepinephrine (NE), creating a pro-tumor metabolic and immunosuppressive microenvironment. SNS signaling e

Inhibition of cytochrome P450 epoxygenase promotes endothelium-to-mesenchymal transition and exacerbates doxorubicin-induced cardiovascular toxicity.

BackgroundDoxorubicin (DOX) is a potent chemotherapy widely used in treating various neoplastic diseases. However, the clinical use of DOX is limited due to its potential toxic effect on the cardiovascular system. Thus, identifying the pathway involved in this toxicity may help minimize chemotherapy risk and improve cancer patients' quality of life. Recent studies suggest that Endothelial-to-Mesenchymal transition (EndMT) and endothelial toxicity contribute to the pathogenesis of DOX-induced cardiovascular toxicity. However, the molecular mechanism is yet unknown. Given that arachidonic acid a

Tumor Targeting with Peptide-Drug Conjugates: Showcasing Key Progress and Hurdles.

Peptide-drug conjugates (PDCs) are modular, targeted therapeutics composed of a homing peptide linked to a cytotoxic or modulating drug payload via a cleavable/non-cleavable linker. PDCs utilize peptide targeting to enhance the delivery of potent drugs to tumors, providing advantages such as superior tissue penetration, reduced immunogenicity, and simpler manufacture compared to antibody-drug conjugates (ADCs). A comparison of PDCs versus ADCs highlights that PDCs' small size (~1-3 kDa) enables deeper tumor penetration and faster clearance, whereas ADCs (~150 kDa) benefit from prolonged circul
