Use of an optimality model to solve the immunological puzzle of concomitant infection.

Immunological data indicate that different subsets of T-helper cells work best against different types of infection. Concomitant infection of a host may thus impose either conflicting or synergistic immune response requirements, depending upon the extent to which the component optimal immune responses differ. Drawing upon empirically-determined optimal responses to single-species infections, an optimality model is here used to generate testable hypotheses for optimal responses to concomitant infection. The model is based upon the principle that the joint immune response will minimize divergenc

Colorectal Organoids: Models, Imaging, Omics, Therapy, Immunology, and Ethics.

Colorectal epithelium was the first long-term 3D organoid culture established in vitro. Identification of the key components essential for the long-term survival of the stem cell niche allowed an indefinite propagation of these cultures and the modulation of their differentiation into various lineages of mature intestinal epithelial cells. While these methods were eventually adapted to establish organoids from different organs, colorectal organoids remain a pioneering model for the development of new applications in health and disease. Several basic and applicative aspects of organoid culture,

Immunological Effects of Diesel Particles in a Murine Model of Healthy Mice.

Introduction: Exposure to environmental pollutants such as diesel exhaust particles (DEP) increases the risk of respiratory disease exacerbation. However, the possible effects of these particles on the general population remain poorly understood. The present study aimed to assess the immunomodulatory and inflammatory effects of the inhalation of DEP in a model of healthy mice undergoing short-, mid- and long-term exposure. Materials and Methods: BALB/c ByJ mice were randomly divided into five experimental groups. The control group received three intranasal instillations of saline over 8 days w

Use of several immunological markers to model the probability of active tuberculosis.

Blood-based biomarkers tests are attractive alternative for diagnosing tuberculosis to assays depending on mycobacteria detection. Given several immunological markers we used logistic regression to model the probability of active tuberculosis in a cohort of patients with active or latent tuberculosis, showing an increased accuracy in distinguishing active from latent tuberculosis.

Proposed immunologic models of the inflammatory myopathies and potential therapeutic implications.

Several immune system cell types and processes have been recently identified in muscle in inclusion body myositis, dermatomyositis, and polymyositis. Plasmacytoid dendritic cells, the immune system's professional producer of the type 1 interferons alpha and beta, are present in dermatomyositis muscle and skin. These tissues also show high expression of transcripts and proteins from genes that are induced by interferon alpha and beta. Myeloid dendritic cells, which contribute to an immunologic synapse responsible for activation of the adaptive immune system, are abundant within muscle in inclus

Integrative models of bat rabies immunology, epizootiology and disease demography.

Bats are natural reservoirs of rabies. We address the maintenance of the disease in bat colonies by developing individual and population models that generate indicators of risk of rabies to bats, that provide dynamic estimates of effects of rabies on population densities, and that suggest consequences of viral exposures and infections in bats relative to physiological and ecological characteristics of bats in different habitats. We present individual models (within host) for the immune responses to a rabies virus challenge, an immunotypic disease model that describes the evolution of the disea

Immunological changes in the MPTP-induced Parkinson's disease mouse model.

The role of the central dopaminergic system in modulating immune response is not completely established. We examined the influence of central dopamine depletion on selected parameters of immune functions in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) treated and untreated mice. IgM antibody production of splenocytes to sheep red blood cells was reduced in MPTP-treated mice (P < 0.001). Proliferation of splenocytes in response to a wide range of mitogen concentrations (Concanavalin A, phytohaemagglutinin, lipopolysaccharide) was also significantly diminished in MPTP-treated mice. The guinea pig model of diisocyanate sensitization. I. Immunologic studies.

Two strains of guinea pigs were parenterally immunized with well-characterized diisocyanate-protein conjugates. Hapten-specific IgE antibodies were detected in the sera of English short-hair strain guinea pigs immunized with either toluene diisocyanate-human serum albumin (TDI-HSA) or hexamethylene diisocyanate-HSA (HDI-HSA) when these sera were analyzed by the 168 hr passive cutaneous anaphylaxis (PCA) technique followed by intravenous challenges with conjugates of respective ligands coupled to an unrelated carrier protein, transferrin. IgG1 antibodies and precipitating antibodies were demons

Immunologically induced tubulo-interstitial nephritis: experimental models in rats.

Experimental models of immunologically induced tubulo-interstitial nephritis (TIN) in rats are described. Antitubular basement membrane (anti-TBM) nephritis was demonstrated to be a TIN resulting from an antigen antibody reaction. Renal transplant rejection and the local graft-vs-host (GVH) reaction were cell-mediated immune reactions in which the antigen was possibly shared by kidney structures. A cellular immune reaction to locally applied antigens was shown to be the basis for another experimental model of TIN, possibly originating from activated mediator systems.

Burkitt's lymphoma - a human tumor model system for immunological studies.

Burkitt's lymphoma occurs mainly in parts of tropical Africa and has attracted the attention of experimental workers due to its epidemiological and clinical features, which indicate a viral etiology and a host immune response to the tumor. As a result of virological studies, Epstein-Barr virus (EBV) DNA has been demonstrated in almost all tested biopsies of African BL. This contrasts to the absence of EBV in all, or almost all, of the non-African Burkitt's lymphoma-like tumors, even though the number of tested tumors in this group is small, and to the lack of EBV in all other types of lymphoma

Depression of immunological responses due to surgery. A model in the guinea-pig.

The responses of guinea-pigs to in vivo sensitization immediately following cholecystectomy (as a standard surgical procedure) are significantly depressed. Generation of memory function on stimulation with sheep erythrocytes is radically impaired; the effect is also more marked in primary delayed hypersensitivity and thymus-dependent antibody responses than in the response to pneumococcal polysaccharide. Normal lymphocyte transfer tests using operated animals as recipients or donors are not depressed. The evidence is suggestive of dysfunction in the amplification or co-operative stages of immu

A Double-Humanized Murine Model in Bladder Cancer: A Novel Preclinical Model for Cancer Immunology Research.

BACKGROUND: Bladder cancer (BC) ranks as the 10th leading cause of cancer-related deaths in the United States. Despite significant advancements in managing BC, including antibody-drug conjugates and immune checkpoint blockers, a significant percentage of patients fail these treatments. This underscores the pressing need to develop methods for identifying novel therapeutic targets, predicting therapy responses, and optimizing personalized treatment strategies for patients. Patient-derived xenograft (PDX) models have the potential to address these challenges. However, these models suffer from li

Controlled human infectious models, a path forward in uncovering immunological correlates of protection: Lessons from enteric fevers studies.

Enteric infectious diseases account for more than a billion disease episodes yearly worldwide resulting in approximately 2 million deaths, with children under 5 years old and the elderly being disproportionally affected. Enteric pathogens comprise viruses, parasites, and bacteria; the latter including pathogens such as Salmonella [typhoidal (TS) and non-typhoidal (nTS)], cholera, Shigella and multiple pathotypes of Escherichia coli (E. coli). In addition, multi-drug resistant and extensively drug-resistant (XDR) strains (e.g., S. Typhi H58 strain) of enteric bacteria are emerging; thus, renewe

Spontaneous onset and transplant models of the Vk*MYC mouse show immunological sequelae comparable to human multiple myeloma.

BACKGROUND: The Vk*MYC transgenic and transplant mouse models of multiple myeloma (MM) are well established as a research tool for anti-myeloma drug discovery. However, little is known of the immune response in these models. Understanding the immunological relevance of these models is of increasing importance as immunotherapeutic drugs are developed against MM. METHODS: We set out to examine how cellular immunity is affected in Vk*MYC mouse models and compare that to the immunology of patients with newly diagnosed and relapsed/refractory MM. RESULTS: We found that there were significant immu

Preclinical demonstration of lentiviral vector-mediated correction of immunological and metabolic abnormalities in models of adenosine deaminase deficiency.

Gene transfer into autologous hematopoietic stem cells by γ-retroviral vectors (gRV) is an effective treatment for adenosine deaminase (ADA)-deficient severe combined immunodeficiency (SCID). However, current gRV have significant potential for insertional mutagenesis as reported in clinical trials for other primary immunodeficiencies. To improve the efficacy and safety of ADA-SCID gene therapy (GT), we generated a self-inactivating lentiviral vector (LV) with a codon-optimized human cADA gene under the control of the short form elongation factor-1α promoter (LV EFS ADA). In ADA(-/-) mice, LV E

Evaluating southern pigtail macaques as an immunological model for human antibody responses.

Antibody specificity is primarily derived from immunoglobulin (Ig) genes, which exhibit extensive genetic polymorphism both within and across species. Non-human primates (NHP) such as pigtail macaques are often used as preclinical models for immunological studies because of their genetic proximity to humans, despite a comparatively poor characterisation of their immunoglobulin loci and its impact on antibody specificity. To determine how pigtail macaque antibody responses differ from humans, we annotated the immunoglobulin loci of two pigtail macaques and compared BCR sequences and antigen bin

A Critical Oversight in Immunology Research: The Prevalence of Complement Deficiencies in Mouse Models.

The complement system is a crucial component of the innate immune response, playing a vital role in defending the body against pathogens and maintaining homeostasis. This complex network of proteins acts as a first line of defense, enhancing the ability of antibodies and phagocytic cells to clear microbes and damaged cells. It also participates in inflammation processes and leukocyte recruitment. However, findings regarding a common mutation in the complement component C5 in mouse strains have raised concerns about the validity of numerous studies in immunology, including infectious diseases,

Improving immunological insights into the ferret model of human viral infectious disease.

Ferrets are a well-established model for studying both the pathogenesis and transmission of human respiratory viruses and evaluation of antiviral vaccines. Advanced immunological studies would add substantial value to the ferret models of disease but are hindered by the low number of ferret-reactive reagents available for flow cytometry and immunohistochemistry. Nevertheless, progress has been made to understand immune responses in the ferret model with a limited set of ferret-specific reagents and assays. This review examines current immunological insights gained from the ferret model across

Modulation of Immunologic Response by Preventive Everolimus Application in a Rat CPB Model.

Everolimus (EVL) is widely used in solid organ transplantation. It is known to have antiproliferative and immunosuppressive abilities via inhibition of the mTOR pathway. Preventive EVL administration may lower inflammation induced by cardiopulmonary bypass (CPB) and reduce systemic inflammatory response syndrome (SIRS). After oral loading with EVL 2.5 mg/kg/day (n = 11) or placebo (n = 11) for seven consecutive days, male Wistar rats (400-500 g) were connected to a miniaturised heart-lung-machine performing a deep hypothermic circulatory arrest protocol.
