Negligible senescence in naked mole rats may be a consequence of well-maintained splicing regulation.

Naked mole-rats (NMRs) have amongst the longest lifespans relative to body size of any known, non-volant mammalian species. They also display an enhanced stress resistance phenotype, negligible senescence and very rarely are they burdened with chronic age-related diseases. Alternative splicing (AS) dysregulation is emerging as a potential driver of senescence and ageing. We hypothesised that the expression of splicing factors, important regulators of patterns of AS, may differ in NMRs when compared to other species with relatively shorter lifespans. We designed assays specific to NMR splicing

The Distribution of Gastrointestinal Parasites in Two Populations of Common Mole-Rats ( Cryptomys hottentotus hottentotus).

The spread of parasites through a host population is based on the variation in behavior and immune function between individuals and is rarely uniform. We studied the gastrointestinal parasites of common mole-rats ( Cryptomys hottentotus hottentotus, Lesson 1826) from 2 sites and assessed the levels of infection based on host sex, breeding status, and season. Only nematode species were found: Neoheligmonella sp. and Mammalakis macrospiculum (Ortlepp, 1939) and a single specimen of Trichuris sp., all of which have direct life cycles. Parasite burden and species richness was greater in the mesic

Retinal ganglion cell survival and axon regeneration after optic nerve injury in naked mole-rats.

In the adult mammalian central nervous system (CNS), axonal damage often triggers neuronal cell death and glial activation, with very limited spontaneous axon regeneration. In this study, we performed optic nerve injury in adult naked mole-rats, the longest living rodent, with a maximum life span exceeding 30 years, and found that injury responses in this species are quite distinct from those in other mammalian species. In contrast to what is seen in other mammals, the majority of injured retinal ganglion cells (RGCs) survive with relatively high spontaneous axon regeneration. Furthermore, inj

THE STATIC ELECTROCARDIOGRAM OF ZOO-KEPT NAKED MOLE-RATS (HETEROCEPHALUS GLABER) UNDER ALFAXALONE-KETAMINE ANESTHESIA.

The naked mole-rat (Heterocephalus glaber) has become a subject of interest in cardiovascular research due to its unique biological properties. However, the ECG of this species has not been thoroughly described. The objective of this study was to characterize the ECG of ten clinically healthy naked mole-rats. Each animal was anesthetized with ketamine and alfaxalone, and ECGs were recorded in sternal recumbency using a hexaxial lead system. ECG resembled those of other rodents with a median heart rate of 174 beats/min (range: 131-202 beats/min), positive Q wave, R wave, and S wave (QRS) comple

Naked mole-rats have distinctive cardiometabolic and genetic adaptations to their underground low-oxygen lifestyles.

The naked mole-rat Heterocephalus glaber is a eusocial mammal exhibiting extreme longevity (37-year lifespan), extraordinary resistance to hypoxia and absence of cardiovascular disease. To identify the mechanisms behind these exceptional traits, metabolomics and RNAseq of cardiac tissue from naked mole-rats was compared to other African mole-rat genera (Cape, Cape dune, Common, Natal, Mahali, Highveld and Damaraland mole-rats) and evolutionarily divergent mammals (Hottentot golden mole and C57/BL6 mouse). We identify metabolic and genetic adaptations unique to naked mole-rats including elevate

Acute pH alterations do not impact cardiac mitochondrial respiration in naked mole-rats or mice.

Energetically demanding conditions such as hypoxia and exercise favour anaerobic metabolism (glycolysis), which leads to acidification of the cellular milieu from ATP hydrolysis and accumulation of the anaerobic end-product, lactate. Cellular acidification may damage mitochondrial proteins and/or alter the H+ gradient across the mitochondrial inner membrane, which may in turn impact mitochondrial respiration and thus aerobic ATP production. Naked mole-rats are among the most hypoxia-tolerant mammals, and putatively experience intermittent environmental and systemic hypoxia while resting and ex

Evoked auditory potentials from African mole-rats and coruros reveal disparity in subterranean rodent hearing.

Hearing in subterranean rodents exhibits numerous peculiarities, including low sensitivity and restriction to a narrow range of comparatively low frequencies. Past studies provided two conflicting hypotheses explaining how these derived traits evolved: structural degeneration and adaptive specialization. To further elucidate this issue, we recorded auditory brainstem responses from three species of social subterranean rodents that differ in the degree of specialization to the underground habitat: the naked mole-rat (Heterocephalus glaber) and the Mashona mole-rat (Fukomys darlingi), which repr

Cessation of reproduction-related spine elongation after multiple breeding cycles in female naked mole-rats.

The breeding female or "queen" naked mole-rat has a uniquely elongated body morphology attributed to the lengthening of the lumbar vertebral column that occurs during pregnancy. It is unknown whether this vertebral growth is a continuous process, or associated only with early reproductive experience. We compared pregnancy-related bone elongation in nascent primiparous queens and established queens to determine if this vertebral expansion was a lifelong process in these females. We also investigated the impact of lactation on vertebral elongation in these mole-rats because it is known to be a t

The isotopic ecology of African mole rats informs hypotheses on the evolution of human diet.

The diets of Australopithecus africanus and Paranthropus robustus are hypothesized to have included C4 plants, such as tropical grasses and sedges, or the tissues of animals which themselves consumed C4 plants. Yet inferences based on the craniodental morphology of A. africanus and P. robustus indicate a seasonal diet governed by hard, brittle foods. Such mechanical characteristics are incompatible with a diet of grasses or uncooked meat, which are too tough for efficient mastication by flat, low-cusped molars. This discrepancy, termed the C4 conundrum, has led to the speculation that C4 plant

Multimodal imaging reveals no evidence for magnetite-based magnetoreceptors in the mole-rat eye.

Magnetoreception is widespread across animals, but its sensory mechanism remains unresolved. One leading hypothesis proposes that single-domain magnetite is coupled to mechanosensitive ion channels. In subterranean Ansell's mole-rats, such receptors have been proposed to reside in the cornea or retina. We screened for magnetite in the mole-rat eye by combining iron imaging via enhanced Prussian blue staining and synchrotron X-ray fluorescence microscopy (XFM) with magnetic imaging via MRI-quantitative susceptibility mapping (MRI-QSM) and quantum-diamond microscopy. The retina and cornea contai

Absence of a glutamatergic channel arrest mechanisms in hypoxic naked mole-rat cortex.

In the brain of hypoxia-intolerant vertebrates, hypoxia induces neuronal depolarization, chronic activation of voltage-gated excitatory glutamatergic receptors, and rapid accumulation of Ca2+ in the cytosol, which induces downstream activation of cell death pathways. Conversely, deleterious Ca2+ accumulation and cell death is avoided in the brain of hypoxia-tolerant vertebrates. One neuroprotective adaptation that is present in some of the most hypoxia-tolerant vertebrates is channel arrest, whereby Ca2+ ion influx through glutamate receptors is reduced in hypoxia, and cytotoxic accumulation o

Progeroid Syndrome with Signs of Autophagy Dysfunction in the Naked Mole Rat (Heterocephalus glaber).

The naked mole rat is considered a unique non-aging mammalian species and is widely used in laboratories to study the biology of longevity. Previously, our group was the first to describe a new fatal disease in the naked mole rat, termed "idiopathic cachexia." A detailed study of pathological changes in the organs of affected animals, combined with the data on gene expression changes, allows us to interpret this disease as a highly specific variant of accelerated aging (progeroid syndrome or progeria) in these animals. Symptoms of the disease include cachexia, cataracts, lipofuscinosis, and ap

Intestinal barrier function in the naked mole-rat: an emergent model for gastrointestinal insights.

The intestinal barrier plays a crucial role in homeostasis by both facilitating the absorption of nutrients and fluids and providing a tight shield to prevent the invasion by either pathogen or commensal microorganisms. Intestinal barrier malfunction is associated with systemic inflammation, oxidative stress, and decreased insulin sensitivity, which may lead to the dysregulation of other tissues. Therefore, a deeper understanding of physiological aspects related to an enhanced barrier function is of significant scientific and clinical relevance. The naked mole-rat has many unusual biological f

Extreme Physiology Extreme Tolerance to Hypoxia, Hypercapnia, and Pain in the Naked Mole-Rat.

Challenging environmental conditions can drive the evolution of extreme physiological traits. The naked mole-rat has evolved to survive and thrive in a low oxygen, high carbon dioxide environment that would be deadly to humans and most other mammals. The naked mole-rat's lifestyle is unusual in that this species combines subterranean living and living in large, social groups of up to 300 + individuals. Many respiring animals in a closed environment can lead to depletion of oxygen (hypoxia) and accumulation of carbon dioxide (hypercapnia). Naked mole-rats display a variety of physiological trai

The brains of six African mole-rat species show divergent responses to hypoxia.

Mole-rats are champions of self-preservation, with increased longevity compared with other rodents their size, strong antioxidant capabilities and specialized defenses against endogenous oxidative stress. However, how the brains of these subterranean mammals handle acute in vivo hypoxia is poorly understood. This study is the first to examine the molecular response to low oxygen in six different species of hypoxia-tolerant mole-rats from sub-Saharan Africa. Protein carbonylation, a known marker of DNA damage (hydroxy-2'-deoxyguanosine), and antioxidant capacity did not change following hypoxia

Ectoparasite Burdens of the Damaraland Mole-Rat (Fukomys damarensis) from Southern Africa.

Damaraland mole-rats (Fukomys damarensis) of the family Bathyergidae are widely distributed subterranean rodents in sub-Saharan Africa. No parasites have ever been reported for this species and only 1 ectoparasite is described for the entire genus. In the current study ectoparasites were collected from individuals captured at 3 localities in South Africa and Namibia to document the ectoparasite community of F. damarensis, investigate their aggregation patterns, and evaluate the influence of season on ectoparasite burden. A total of 2,071 arthropods from 9 mite taxa and 1 louse species (Eulinog

Melatonin secretion in the Mashona mole-rat, Cryptomys darlingi--influence of light on rhythmicity.

The hormone melatonin is synthesised and secreted from the pineal gland in darkness and triggers the daily and seasonal timing of various physiological and behavioural processes. The Mashona mole-rat, Cryptomys darlingi, lives in subterranean burrows that are completely sealed and is therefore rarely, if ever, exposed to light under natural conditions. Hence, this species is of particular interest for studies on rhythms of melatonin secretion. We investigated how plasma melatonin concentrations of the Mashona mole-rat responded to exposure to a long-term standard photoperiod of 12 h light, 12

The glutamatergic drive to breathe is reduced in severe but not moderate hypoxia in Damaraland mole-rats.

Damaraland mole-rats (Fukomys damarensis) are a hypoxia-tolerant fossorial species that exhibit a robust hypoxic metabolic response (HMR) and blunted hypoxic ventilatory response (HVR). Whereas the HVR of most adult mammals is mediated by increased excitatory glutamatergic signalling, naked mole-rats, which are closely related to Damaraland mole-rats, do not utilize this pathway. Given their phylogenetic relationship and similar lifestyles, we hypothesized that the signalling mechanisms underlying physiological responses to acute hypoxia in Damaraland mole-rats are like those of naked mole-rat

Enhanced mitochondrial buffering prevents Ca2+ overload in naked mole-rat brain.

Deleterious Ca2+ accumulation is central to hypoxic cell death in the brain of most mammals. Conversely, hypoxia-mediated increases in cytosolic Ca2+ are retarded in hypoxia-tolerant naked mole-rat brain. We hypothesized that naked mole-rat brain mitochondria have an enhanced capacity to buffer exogenous Ca2+ and examined Ca2+ handling in naked mole-rat cortical tissue. We report that naked mole-rat brain mitochondria buffer >2-fold more exogenous Ca2+ than mouse brain mitochondria, and that the half-maximal inhibitory concentration (IC50) at which Ca2+ inhibits aerobic oxidative phosphorylati
