The Potential of Flos sophorae immaturus as a Pigment-Stabilizer to Improve the Monascus Pigments Preservation, Flavor Profiles, and Sensory Characteristic of Hong Qu Huangjiu.

Hong Qu Huangjiu (HQW) is distinguished by its inclusion of Monascus pigments, meaning that photosensitivity strongly affects the sensory quality of the wine. In this study, the effects of Flos sophorae immaturus (FSI) on the stability of Monascus pigments, the flavor profiles, and the sensory characteristics of HQW were investigated. After sterilization, the addition of FSI increased the preservation rate of Monascus pigments in HQW by up to 93.20%, which could be accounted for by the synergy of rutin and quercetin in FSI. The total content of the volatile flavor compounds in HQW increased si

Spatially resolved metabolomics integrated with multi-omics analysis suggests a coordinated metabolic framework for lipid transformation and methyl ketone formation in Monascus-fermented cheese.

Methyl ketones play a crucial role in shaping the characteristic aroma of Monascus-fermented cheese (MC). However, their formation pathways within complex solid-state fermentation systems are not yet fully understood. Spatial metabolomics was integrated with lipidomics, sensomics, metagenomics, and metaproteomics to investigate lipid transformation and methyl ketone formation during MC ripening. Glycerophospholipids showed distinct spatial distribution patterns during mid-ripening, supporting spatially heterogeneous lipid transformation. Temporal analysis revealed sequential dynamics, with ear

Integrated non-targeted metabolomics, enzyme activities and morphological analysis to elucidate the mechanisms underlying the enhancement of activity and flavor of Monascus purpureus fermented Undaria pinnatifida.

Monascus purpureus-fermentation enhanced antioxidant activity and flavor of Undaria pinnatifida. This study employed an integrated approach combining non-targeted metabolomics, enzyme activity assays, and scanning electron microscopy (SEM) to elucidate the mechanisms. Results show that fermentation enhanced the activities of cellulase and pectinase, which effectively initiated the degradation of the cell wall. Morphological study by SEM confirmed this result. This upstream enzymatic change triggered the liberation of intracellular bioactive compounds. The increase in total phenolic and flavono

The noncovalent conjugations of human serum albumin (HSA) with MS/AK and the effect on anti-oxidant capacity as well as anti-glycation activity of Monascus yellow pigments.

Monascin (MS) and ankaflavin (AK), as typical yellow lipid-soluble pigments identified from Monascus-fermented products, have been confirmed to possess diverse biological activities such as anti-oxidation, reversing diabetes, and anti-atherosclerosis, and have received increasing attention in recent years. Certainly Monascus-fermented product with a high content of MS/AK is also a concern. The current work explored interactions between MS/AK and human serum albumin (HSA) as well as their influence on the anti-oxidant properties of MS/AK. Moreover, the anti-glycation potential of Monascus-ferme

Monascin and Ankaflavin of Monascus purpureus Prevent Alcoholic Liver Disease through Regulating AMPK-Mediated Lipid Metabolism and Enhancing Both Anti-Inflammatory and Anti-Oxidative Systems.

Alcohol metabolism causes an excessive accumulation of liver lipids and inflammation, resulting in liver damage. The yellow pigments monascin (MS) and ankaflavin (AK) of Monascus purpureus-fermented rice were proven to regulate ethanol-induced damage in HepG2 cells, but the complete anti-inflammatory and anti-fatty liver mechanisms in the animal model are still unclear. This study explored the roles of MS and AK in improving alcoholic liver injury. MS and AK were simultaneously fed to evaluate their effects and mechanisms in C57BL/6J mice fed the Lieber-DeCarli liquid alcohol diet for 6 weeks.

Integrated physiology, transcriptomics and targeted metabolomics reveal a potential quorum sensing network involved in the initial spore density-dependent regulation of morphogenesis and polyketide biosynthesis in Monascus.

Density dependence is a key characteristic of quorum sensing (QS) in fungi; however, no relevant reports have been found in Monascus. Therefore, this study aimed to investigate the effects of initial spore density on the morphological development and polyketide secondary metabolism of Monascus purpureus to elucidate the regulatory role of QS. At the high initial spore density, more active conidial development and secondary metabolism were observed in the early fermentation stage, accompanied by rougher hyphal surfaces, increased secretion and larger vacuoles.
