Splicing is dynamically regulated during limb development.

Modifications to highly conserved developmental gene regulatory networks are thought to underlie morphological diversification in evolution and contribute to human congenital malformations. Relationships between gene expression and morphology have been extensively investigated in the limb, where most of the evidence for alterations to gene regulation in development consists of pre-transcriptional mechanisms that affect expression levels, such as epigenetic alterations to regulatory sequences and changes to cis-regulatory elements. Here we report evidence that alternative splicing (AS), a post-

Prolonged Myocardial Regenerative Capacity in Neonatal Opossum.

BACKGROUND: Early neonates of both large and small mammals are able to regenerate the myocardium through cardiomyocyte proliferation for only a short period after birth. This myocardial regenerative capacity declines in parallel with withdrawal of cardiomyocytes from the cell cycle in the first few postnatal days. No mammalian species examined to date has been found capable of a meaningful regenerative response to myocardial injury later than 1 week after birth. METHODS: We examined cardiomyocyte proliferation in neonates of the marsupial opossum (Monodelphis domestica) by immunostaining at v

Paternal X inactivation does not correlate with X chromosome evolutionary strata in marsupials.

BACKGROUND: X chromosome inactivation is the transcriptional silencing of one X chromosome in the somatic cells of female mammals. In eutherian mammals (e.g. humans) one of the two X chromosomes is randomly chosen for silencing, with about 15% (usually in younger evolutionary strata of the X chromosome) of genes escaping this silencing. In contrast, in the distantly related marsupial mammals the paternally derived X is silenced, although not as completely as the eutherian X. A chromosome wide examination of X inactivation, using RNA-seq, was recently undertaken in grey short-tailed opossum (Mo

Heterochronic limb patterning in marsupials reveals flexibility in the processes underlying lateral plate mesoderm morphogenesis.

Marsupial embryos develop under intense functional constraints: neonates are born after a short gestation but must immediately crawl to the teat using precociously developed forelimbs. To meet this demand, marsupials have evolved extreme acceleration of limb morphogenesis, yet the cellular and molecular mechanisms underlying this shift remain unresolved. Using two distantly related marsupials, the fat-tailed dunnart (Sminthopsis crassicaudata) and the gray short-tailed opossum (Monodelphis domestica), we show that this acceleration extends upstream to the earliest stages of lateral plate mesod

Monkeypox zoonotic associations: insights from laboratory evaluation of animals associated with the multi-state US outbreak.

At the onset of the 2003 US monkeypox outbreak, virologic data were unavailable regarding which animal species were involved with virus importation and/or subsequent transmission to humans and whether there was a risk for establishment of zoonotic monkeypox in North America. Similarly, it was unclear which specimens would be best for virus testing. Monkeypox DNA was detected in at least 33 animals, and virus was cultured from 22. Virus-positive animals included three African species associated with the importation event (giant pouched rats, Cricetomys spp.; rope squirrels, Funisciuris sp.; and

Detection of anti-Toxoplasma gondii antibodies in small wild mammals from preserved and non-preserved areas in the Caatinga biome, a semi-arid region of Northeast Brazil.

This study was conducted to determine the seroprevalence of anti-Toxoplasma gondii antibodies in 152 free-living small wild mammals from distinct regions in the Caatinga biome, a semi-arid region in the Northeast of Brazil: the National Park of Serra das Confusões (NPSC), which is a preserved area in the state of Piauí, and the municipalities of Petrolina and Lagoa Grande, two non-preserved areas in the state of Pernambuco. Using the modified agglutination test (MAT), we found that 5.3% (4/75) and 3.3% (2/60) of small wild mammals were positive for IgG anti-T. gondii antibodies in the NPSC and

Rodents and other small mammal reservoirs in plague foci in northeastern Brazil.

INTRODUCTION: Plague is an acute, infectious zoonotic disease, primarily of wild rodents and their fleas, that affects humans and other mammals. In Brazil, several plague foci are located in the northeast region. Plague surveillance based on monitoring of rodents was discontinued in 2007, and the current information on rodent populations is unsatisfactory. Our purpose was to update the information on rodents and other small mammals in plague foci in northeastern Brazil. METHODOLOGY: Nine surveys in the historically most important northeastern plague areas were conducted in 2013-2015. RESULTS

Seroprevalence of Toxoplasma gondii in wild marsupials and rodents from the Atlantic forest of Pernambuco state, northeastern region, Brazil.

Felids are important in the epidemiology of Toxoplasma gondii because they are the only hosts that can excrete the environmentally resistant oocysts in their feces. Cats acquire T. gondii infection in nature by ingesting tissues of small mammals and birds. Serum samples of 223 feral marsupials and 174 feral rodents captured in 7 segments of the Atlantic Forest of the State of Pernambuco, northeastern region of Brazil, and in urban areas of the municipality of Recife were examined for antibodies to T. gondii by the modified agglutination test (MAT). Antibodies (MAT ≥ 25) were found in 6.7% (15

Chiasmatic specificity in the regenerating mammalian optic nerve.

The mammalian central nervous system is capable of regenerating; however, there is no evidence that the regenerating axons can navigate along their normal pathways and reestablish topographically organized projections: essential for functional return of vision. Here retinal ganglion cells in the opossum Monodelphis were birthdated with tritiated thymidine on the sixth postnatal day (P6), before being lesioned in the temporal retina at P8. Retrograde tracing with horseradish peroxidase injected into the ipsilateral optic tract at P24 showed that the temporal crescent had reformed behind the ret

Paracoccidioides brasiliensis Infection in Small Wild Mammals.

Paracoccidioidomycosis (PCM) is a systemic mycosis prevalent in Brazil and other Latin American countries. The etiological agents of PCM are the thermo-dimorphic fungi Paracoccidioides brasiliensis and P. lutzii. Taking into account that the natural habitat of Paracoccidioides spp. is still undefined, domestic and wild animals could be useful as indicators of Paracoccidioides spp. presence in endemic areas. The objective of this study was to evaluate the infection of small wild mammals by P. brasiliensis in an endemic area for human PCM. Samples from 38 wild mammals from different species such

Recent advances in sunlight-induced carcinogenesis using the Xiphophorus melanoma model.

Unlike breast and prostate cancers, the nature and sequence of critical genetic and epigenetic events involved in the initiation and progression of melanoma are not well understood. A contributing factor to this dilemma, especially given our current understanding of the importance of UV light in melanoma etiology, is the lack of quality UV-inducible melanoma animal models. In this study we elaborate on the capability of UV light to induce cutaneous malignant melanomas (CMM) in Xiphophorus fishes, which were previously found to develop melanomas after acute neonatal UVB irradiation. In two sepa

Use of an Argentinean Wildlife Tissue Collection for Epidemiological Studies of Histoplasmosis.

Histoplasmosis is a worldwide systemic endemic mycosis caused by several cryptic species included within the Histoplasma capsulatum complex. Domestic and wild mammals are susceptible to infection by this fungus and could be used as indicators of its presence in the environment. The aim of the study was to identify the natural reservoirs of H. capsulatum in the Argentinean Humid Pampas eco-region analyzing a wildlife frozen-tissue collection and trace its distribution patterns over time and space. Tissue samples from 34 small wild mammals caught in the Humid Pampas were analyzed using two molec

Metabolic adaptation to hypoxia: cost and benefit of being small.

Following metabolic size allometry, the specific metabolic rate of mammals increases with decreasing body mass, resulting in a steeper metabolic fall-off and a faster exhaustion of energy reserves under hypoxic conditions. However, both mammalian hibernators and fetuses are able to temporarily "switch-off" Kleiber's rule as an adaptation to limited food or oxygen supply. Further exceptions to the usual metabolic size relationship are observed in newborn mammals. For instance, neonatal mouse hearts exhibit slower calorimetric "dying curves" under conditions of ischemia, although their aerobic t

Amblyomma triste Koch, 1844 (Acari: Ixodidae): hosts and seasonality of the vector of Rickettsia parkeri in Uruguay.

Amblyomma triste Koch, 1844, the tick species most frequently involved in human bites in Uruguay, has been implicated as a vector of human rickettsiosis. Seasonal abundance of adult A. triste was examined by standard flagging at three sites where human tick bites and cases of the disease have been reported. Adult tick activity occurred from August to February (end of winter to mid summer in the southern hemisphere) with a peak in spring. Activity declined in step with decreasing temperatures and photoperiod during winter. This period of activity coincides with seasonal outbreaks of human ricke

Toxoplasma gondii in domestic and wild animals from forest fragments of the municipality of Natal, northeastern Brazil.

Toxoplasmosis stands out as a global disease that has felines as definitive hosts. In the municipality of Natal, Rio Grande do Norte State, Brazil, two parks are notable for their ecological and social importance. This study aimed to investigate the presence of Toxoplasma gondii in short hair cats, bats and small non-volant mammals in these two ecological reserves. Altogether, biological samples were obtained from 154 mammals, 92 wild animals from both areas and 62 domestic cats of the Parque da Cidade. In total, 22 (53.7%) non-volant wild mammals, 11 (21.5%) bats and 28 (52.8%) cats were posi

Feeding preferences of Lutzomyia longipalpis (Diptera: Psychodidae), the sand fly vector, for Leishmania infantum (Kinetoplastida: Trypanosomatidae).

Leishmania infantum, the causative agent of visceral leishmaniasis (VL) in Brazil, is spread mostly by the bite of the sand fly Lutzomyia longipalpis (Lutz & Neiva). We trapped sand flies in endemic neighborhoods near Natal, Brazil, where cases of human and dog VL were documented. Amplification of species-specific cytochrome b (Cyt b) genes by polymerase chain reaction revealed that sand flies from rural and periurban areas harbored blood from different sources. The most common source ofbloodmeal was human, but blood from dog, chicken, and armadillo was also present. We tested the preference f

The contribution of cross-modal sensory input to reaching and grasping in blind and sighted short-tailed opossums.

Reaching and grasping are essential goal-directed behaviors that require the integration of visual, olfactory, and somatosensory inputs. Although the loss of vision can profoundly disrupt sensory-guided behaviors, mammals often exhibit compensatory cross-modal plasticity allowing them to reach and grasp with ease. To examine how early sensory loss shapes reaching behavior, we performed bilateral enucleations in short-tailed opossums (Monodelphis domestica) at postnatal day 4, before retino-thalamic and thalamocortical connections have formed. We assessed performance in early blind (EB) and sig
