Effects of aflatoxin ingestion on the development of Moraxella bovis infectious bovine keratoconjunctivitis.

A study was conducted to determine whether measured doses of aflatoxin given under different schedules would influence the pathogenesis of Moraxella bovis induced infectious bovine keratoconjunctivitis (IBK). Calves were allotted to 4 groups (groups I-IV) of 9, 9, 9, and 8 calves, respectively. Group I calves were given aflatoxin for 11 consecutive days starting 5 days before their eyes were exposed to M. bovis. Group II calves were given aflatoxin for 5 consecutive days starting 7 days after their eyes were exposed to M. bovis. Group III calves were given aflatoxin for 5 consecutive days star

The pathophysiological effects of Moraxella bovis toxins on cattle, mice and guinea pigs.

In three experiments, cattle, mice and guinea pigs were inoculated with viable cultures of Moraxella bovis or fractions of this organism. Fractions were obtained by disruption of cells with a fractionator at 20,000 pounds per square inch, and separating the cell wall and cell sap fractions by differential centrifugation. Cell sap fractions were further separated by ultra-centrifugation, heating and precipitation with (NH(4))(2) SO(4). Different fractions induced different pathophysiological manifestations. The cell wall fractions caused localized lesions (necrosis) at the site of injection, an

Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry identification of Moraxella bovoculi and Moraxella bovis isolates from cattle.

Infectious bovine keratoconjunctivitis (IBK) is an economically significant disease caused by Moraxella bovis. Moraxella bovoculi, although not reported to cause IBK, has been isolated from the eyes of cattle diagnosed with IBK. Identification of M. bovis and M. bovoculi can be performed using biochemical or DNA-based approaches, both of which may be time consuming and inconsistent between laboratories. We conducted a comparative evaluation of M. bovoculi and M. bovis identification using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) with a databas

Characterization of hemolysin of Moraxella bovis using a hemolysis-neutralizing monoclonal antibody.

A concentrated bacterial culture supernatant from the hemolytic Moraxella bovis strain UQV 148NF was used to immunize mice and generate monoclonal antibodies (MAbs). One, MAb G3/D7, neutralized the hemolytic activity of M. bovis and recognized a 94-kDa protein by Western blot analysis in hemolytic M. bovis strains representing each of the different fimbrial serogroups. Exposure of corneal epithelial cells to M. bovis concentrated culture supernatants demonstrated a role for an exotoxin in the pathogenesis of infectious bovine keratoconjunctivitis, while neutralization of hemolytic and cytotoxi

Q pili enhance the attachment of Moraxella bovis to bovine corneas in vitro.

Moraxella bovis, the causative agent of infectious bovine keratoconjunctivitis, exhibits several virulence factors, including pili, haemolysin, leukotoxin, and proteases. The pili are filamentous appendages which mediate bacterial adherence. Prior studies have shown that Q-piliated M. bovis Epp63 are more infectious and more pathogenic than I-piliated and non-piliated isogenic variants, suggesting that Q pili per se, or traits associated with Q-pilin expression, promote the early association of Q-piliated bacteria with bovine corneal tissue. In order to better evaluate the role of Q pili in M.

Semiselective medium for isolation of Moraxella bovis from cattle with infectious keratoconjunctivitis.

The incorporation of 2.5 micrograms/ml of cloxacillin into 5% bovine blood agar provided an inexpensive, easily prepared culture medium for the primary isolation of Moraxella bovis from bovine lacrimal and nasal secretions. With this medium, the time required to identify and isolate M. bovis from large numbers of field specimens was substantially reduced, whereas the sensitivity of isolation was increased by 60%.

[Isolation of Chlamydia psittaci and Moraxella bovis from infectious keratoconjunctivitis in lambs].

Out of 189 lambs in the flock, 25 animals suffered from bilateral or unilateral conjunctivitis, or keratoconjunctivitis. By serological examination (RVK), positive levels of antibodies to the group-specific antigen of Chl. psittaci were found in three out of six lambs examined by laboratory methods. Bacteriological examination of eye smears of six lambs showed in four cases the infection by microorganisms of Moraxella bovis. Smears from the conjunctivas of these lambs were after preparation instilled in the yolk sacs of six to seven days old chicken embryos. One strain of Chlamydia psittaci wa

Effects of Moraxella bovis vaccination schedules on experimentally induced infectious bovine keratoconjunctivitis.

An oil-adjuvant Moraxella bovis bacterin was administered to weanling calves, using different vaccination schedules. Calves were given a booster vaccination after 3 weeks and were challenge exposed 2 weeks later with virulent M bovis recovered from calves with clinical infectious bovine keratoconjunctivitis (IBK). The effects of different routes of vaccination and homologous and heterologous challenge exposure on the incidence, severity, and duration of induced IBK was evaluated. All calves given a placebo developed clinical IBK. Calves vaccinated subcutaneously in the neck had the shortest du

Whole-bacterial cell enzyme-linked immunosorbent assay for cell-bound Moraxella bovis pili.

Infectious bovine keratoconjunctivitis (IBK), caused by Moraxella bovis, is a disease of major importance in cattle industry. M. bovis has several virulence factors among which pili are crucial antigen for the protective capacity of vaccines against this disease. The production of vaccines against IBK therefore requires a reliable technique for cellular piliation level assessment on cells to be included as vaccine components. In this study we describe a specific whole-bacterial cell enzyme-linked immunosorbent assay (bact-ELISA) capable of detecting pili antigen on M. bovis cell surface. A seq

Antimicrobial susceptibility of Moraxella bovis determined by agar disk diffusion and broth microdilution.

The antimicrobial susceptibility of 84 isolates of Moraxella bovis was evaluated by the standard agar disk diffusion and broth microdilution procedures. All isolates were resistant to cloxacillin by disk diffusion, with 97% of isolates having a minimal inhibitory concentration of greater than or equal to 2 micrograms/ml. Of the hemolytic isolates, 68% were resistant to streptomycin. A high frequency of susceptibility was recorded for all other antimicrobial agents tested. Quantitative data supported the use of sulfonamides, but not tylosin, for parenteral infectious bovine keratoconjunctivitis

Infectious bovine keratoconjunctivitis: effects of vaccination on Moraxella bovis carrier state in cattle.

A study was conducted to determine whether vaccination of cattle while they were undergoing an acute episode of infectious bovine keratoconjunctivitis (IBK), would cause vaccinated cattle to abort Moraxella bovis infection sooner than nonvaccinated cattle. Fourteen calves were allotted into two groups of seven calves each, and the eyes of each calf were exposed to a virulent culture of M bovis. Twenty days after calves were infected and showing signs of IBK, seven calves were vaccinated with M bovis pilus vaccine made from the exposure strain. Ocular and nasal discharges were collected and exa

A Field Trial of a Pilated Moraxella bovis Bacterin for the Prevention of Infectious Bovine Keratoconjunctivitis.

Two field trials were conducted to evaluate the efficacy of a Moraxella bovis bacterin in the control of infectious bovine keratoconjunctivitis. The bacterin did not affect the incidence of infectious bovine keratoconjunctivitis in either trial but did appear to reduce the severity of lesions, treatments required and withdrawals from pasture required in one trial. Possible reasons for the apparently contradictory results are offered. It is suggested that the bacterin may be of some value when used in conjunction with other preventive measures.

Molecular characterization of a secreted enzyme with phospholipase B activity from Moraxella bovis.

A candidate for a vaccine against infectious bovine keratoconjunctivitis (IBK) has been cloned and characterized from Moraxella bovis. The plb gene encodes a protein of 616 amino acids (molecular mass of ~65.8 kDa) that expresses phospholipase B activity. Amino acid sequence analysis revealed that PLB is a new member of the GDSL (Gly-Asp-Ser-Leu) family of lipolytic enzymes.

Immunogenicity of a Moraxella bovis bacterin containing attachment and cornea-degrading enzyme antigens.

An adjuvanted Moraxella bovis bacterin containing attachment antigens and cornea-degrading enzyme antigens protected cattle from infectious bovine keratoconjunctivitis (IBK) when experimentally challenged with homologous and heterologous challenge cultures of M. bovis. This bacterin also protected cattle against field exposure to M. bovis. Transmission electron microscopy and fluorescein labeled anti-M. bovis pili antiserum showed pili on the M. bovis bacterin strain. Scanning electron microscopy demonstrated a fibrillar glycocalyx.
