Subtype-specific plasticity of inhibitory circuits in motor cortex during motor learning.

Motor skill learning induces long-lasting reorganization of dendritic spines, principal sites of excitatory synapses, in the motor cortex. However, mechanisms that regulate these excitatory synaptic changes remain poorly understood. Here, using in vivo two-photon imaging in awake mice, we found that learning-induced spine reorganization of layer (L) 2/3 excitatory neurons occurs in the distal branches of their apical dendrites in L1 but not in the perisomatic dendrites. This compartment-specific spine reorganization coincided with subtype-specific plasticity of local inhibitory circuits. Somat

Dopaminergic projections from midbrain to primary motor cortex mediate motor skill learning.

The primary motor cortex (M1) of the rat contains dopaminergic terminals. The origin of this dopaminergic projection and its functional role for movement are obscure. Other areas of cortex receive dopaminergic projections from the ventral tegmental area (VTA) of the midbrain, and these projections are involved in learning phenomena. We therefore hypothesized that M1 receives a dopaminergic projection from VTA and that this projection mediates the learning of a motor skill by inducing cellular plasticity events in M1. Retrograde tracing from M1 of Long-Evans rats in conjunction with tyrosine hy

Motor cortex inhibition: a marker of ADHD behavior and motor development in children.

OBJECTIVE: Attention-deficit/hyperactivity disorder (ADHD) is a childhood-onset behavioral diagnosis in which children often fail to meet age norms in development of motor control, particularly timed repetitive and sequential movements, motor overflow, and balance. The neural substrate of this motor delay may include mechanisms of synaptic inhibition in or adjacent to the motor cortex. The primary objective of this study was to determine whether transcranial magnetic stimulation (TMS)-evoked measures, particularly short interval cortical inhibition (SICI), in motor cortex correlate with the pr

Repetitive stimulation of premotor cortex affects primary motor cortex excitability and movement preparation.

BACKGROUND: Repetitive transcranial magnetic stimulation can be used to explore functional connectivity between cortical areas. OBJECTIVE: To determine the effects of two theta burst stimulation (TBS) patterns (intermittent, iTBS; and continuous, cTBS) of left dorsal premotor cortex (PMd). METHODS: Left PMd was identified in 11 participants using functional magnetic resonance imaging (fMRI), during performance of complex sequential finger movements. Each participant received iTBS, cTBS, or sham TBS of left PMd in three separate sessions within a randomized, single-blind design. The speed and

Spike prediction on primary motor cortex from medial prefrontal cortex during task learning.

Objectives. Brain-machine interfaces (BMIs) aim to help people with motor disabilities by interpreting brain signals into motor intentions using advanced signal processing methods. Currently, BMI users require intensive training to perform a pre-defined task, not to mention learning a new task. Thus, it is essential to understand neural information pathways among the cortical areas in task learning to provide principles for designing BMIs with learning abilities. We propose to investigate the relationship between the medial prefrontal cortex (mPFC) and primary motor cortex (M1), which are acti

Transcranial stimulation of motor cortex in upper motor neurone syndrome: its relation to the motor deficit.

The purpose of this investigation was to clarify the functional significance of the fastest cortico-motoneuronal connections in chronic upper motor neurone syndromes. Using magneto-electrical stimulation of motor cortex the intactness of cortico-motoneuronal connections was assessed in 51 patients presenting with variable degrees of impairment. There was a gross correlation between clinical impairment of the patient and the degree of pathology of cortico-motoneuronal efferents. Covariation of clinical data with transcranial stimulation was better than covariation with the size of lesion on CT

Effects of TMS on different stages of motor and non-motor verb processing in the primary motor cortex.

The embodied cognition hypothesis suggests that motor and premotor areas are automatically and necessarily involved in understanding action language, as word conceptual representations are embodied. This transcranial magnetic stimulation (TMS) study explores the role of the left primary motor cortex in action-verb processing. TMS-induced motor-evoked potentials from right-hand muscles were recorded as a measure of M1 activity, while participants were asked either to judge explicitly whether a verb was action-related (semantic task) or to decide on the number of syllables in a verb (syllabic ta

Motor cortex rTMS modulates adult ocular dominance plasticity.

Well-established evidence supports cross-modal interactions of sensory brain plasticity. Here, we extend cross-modality beyond the boundaries of sensory regions and provide first-time evidence in adult healthy humans that motor plasticity influences homeostatic visual plasticity. To this end, we employed repetitive transcranial magnetic stimulation (rTMS), known to modulate the excitation/inhibition balance in the primary motor cortex (M1), while examining the impact of this intervention on ocular dominance plasticity induced by short-term monocular deprivation (MD). Specifically, participants

Sensory Entrained TMS (seTMS) Enhances Motor Cortex Excitability.

Transcranial magnetic stimulation (TMS) applied to the motor cortex has revolutionized the study of motor physiology in humans. Despite this, TMS-evoked electrophysiological responses show significant fluctuation, due in part to inconsistencies between TMS pulse timing and ongoing brain oscillations. Small or inconsistent responses to TMS limit mechanistic insights and clinical efficacy, necessitating the development of methods to precisely coordinate the timing of TMS pulses to the phase of relevant oscillatory activity. We introduce Sensory Entrained TMS (seTMS), a novel approach that uses m

Motor cortex stimulation for phantom limb pain treatment.

BACKGROUND: Phantom limb pain (PLP) is a chronic neuropathic pain syndrome experienced by individuals following limb amputation. Despite the use of various pharmacological treatments, including opioids, antidepressants, and anticonvulsants, effective pain relief remains challenging for many patients. Motor cortex stimulation (MCS) has emerged as a promising alternative for managing PLP. METHODS: We present the management of three patients with chronic, refractory PLP who underwent epidural MCS at Lille University Hospital Center. The quadripolar electrode lead was implanted into the epidural

Arterial vascularization of primary motor cortex (precentral gyrus).

BACKGROUND: The precentral gyrus (PG) is the primary motor area and is one of the most eloquent brain regions of neurosurgical interest. Although the arterial supply to the PG is generally known, contributions from different arterial branches such as the anterior cerebral artery (ACA), posterior cerebral artery (PCA), and middle cerebral artery (MCA) have not been comprehensively studied. The aim of the present study was to provide detailed information about the arteries of the PG. METHODS: Twenty adult human brains (40 hemispheres) were obtained, and ACA, MCA, and PCA were separately cannula

Visual deprivation effects on human motor cortex excitability.

Single and paired-pulse transcranial magnetic stimulation (TMS) were applied to the motor cortex of 12 healthy volunteers, who were instructed to relax under eyes-open and eyes-closed conditions with room lights on and after 30 min of blindfolding. Compared to the eyes-open condition, significantly larger motor-evoked potentials and less intracortical inhibition were observed during blindfolding. Visual deafferentation changes resting human motor cortex excitability and might be a novel way to promote brain plasticity. These results raise the issue of how widespread the effects of temporary de

Motor cortex excitability in patients with cerebellar degeneration.

OBJECTIVES: To study motor cortex (M1) excitability and the effect of subthreshold transcranial magnetic stimulation (TMS) in patients with cerebellar degeneration and normals performing a reaction time (RT) task. METHODS: Time to wrist flexion after a visual go-signal was measured. TMS was always delivered at 90% of resting motor evoked potential (MEP) threshold. In one experiment, test TMS was delivered at various intervals after the go-signal. In half the trials priming TMS was also given with the go-signal. A second experiment examined the effect on RT of M1 and occipital priming stimulat

Characterization of limb representation in the pig's motor cortex.

Due to its large gyrencephalic brain, the pig is increasingly used for neuroscience research, especially for the preclinical testing of novel neuroprostheses. However, our understanding of the pig's motor system remains limited compared with the common species used for neuroscience research. Here, we aimed to characterize the forelimb and hindlimb representation of the pig motor cortex using intracortical microstimulation (ICMS). Three domestic pigs (Sus scrofa) were placed in a modified stereotactic frame and maintained under intravenous propofol sedation. We mapped the motor cortex using ICM

Constraints on somatotopic organization in the primary motor cortex.

Since the 1870s, the primary motor cortex (M1) has been known to have a somatotopic organization, with different regions of cortex participating in control of face, arm, and leg movements. Through the middle of the 20th century, it seemed possible that the principle of somatotopic organization extended to the detailed representation of different body parts within each of the three major representations. The arm region of M1, for example, was thought to contain a well-ordered, point-to-point representation of the movements or muscles of the thumb, index, middle, ring, and little fingers, the wr

Magnetic stimulation of the motor cortex in cervical spondylosis.

We report a new technique of transcranial magnetic stimulation of the motor cortex to measure conduction within central motor pathways of 67 patients with cervical spondylosis or disk herniation. There were upper motor neuron signs in 34 patients (51%) and x-ray evidence of cervical cord compression in 44 (66%). Muscle action potentials (MAPs) to cortical stimulation were abnormal in 84% of patients with, and 22% of those without, radiologic signs of cervical cord compression. Median nerve somatosensory evoked potentials were altered in only 25% of patients. The frequency of MAP alterations co

Distinct Motor Cortex Somatotopy in Experimental Alzheimer's Disease.

Alzheimer's Disease (AD) and related dementias (AD/RDs) impact cortical motor and sensory biology whereas the precise changes to these systems, and their clinical relevance, remain under debate. We hypothesized that cortical representations of complex and simple movements are differently altered during disease progression in 5XFAD mice, a well-established model of AD. Motor cortex somatotopy was determined in 5XFAD and Wild-Type Control (WT Control) mice at 6 and 12 months (mos.) of age using long-duration intracortical microstimulation (LD-ICMS) to systematically identify cortical sites evoki

RTP801 regulates motor cortex synaptic transmission and learning.

BACKGROUND: RTP801/REDD1 is a stress-regulated protein whose upregulation is necessary and sufficient to trigger neuronal death in in vitro and in vivo models of Parkinson's and Huntington's diseases and is up regulated in compromised neurons in human postmortem brains of both neurodegenerative disorders. Indeed, in both Parkinson's and Huntington's disease mouse models, RTP801 knockdown alleviates motor-learning deficits. RESULTS: We investigated the physiological role of RTP801 in neuronal plasticity and we found RTP801 in rat, mouse and human synapses. The absence of RTP801 enhanced excita

Regional motor cortex dysfunction in amyotrophic lateral sclerosis.

OBJECTIVE: The pathophysiological processes underlying amyotrophic lateral sclerosis (ALS) need to be better understood, although cortical dysfunction has been implicated. Previous transcranial magnetic stimulation (TMS) studies have assessed cortical dysfunction from the hand. The aim of the present study was to determine whether cortical dysfunction was evident across representations of three body regions, and to relate these changes to clinical features of ALS. METHODS: In this cross-sectional study, threshold tracking TMS was undertaken in 60 sporadic ALS patients, with motor evoked poten
