Gastric mucus viscosity and peptic ulcer.

The following observations have been made: 1. The viscosity of a substance as complex as mucus requires study at different rates of shear and shear stresses. 2. Visible gastric mucus behaves as a pseudoplastic substance. 3. Duodenal ulcer patients have a higher gastric mucus viscosity than controls. 4. Pentagastrin stimulation in duodenal ulcer patients leads to the secretion of gastric mucus of low viscosity. 5. This decrease in the viscosity of gastric mucus on pentagastrin stimulation does not occur in patients with gastric ulcer. 6. Truncal vagotomy leads to an increase in the viscosity of

Characterization of Ovine Cervical Mucus and Implementation of a Sperm-Mucus Penetration Test Using Fluorescently Labeled Sperm.

In vaginal depositors, semen is deposited at the opening of the cervix, and sperm quickly enter the cervical canal, which is filled with cervical mucus in the lead up to ovulation. The outward flow of cervical mucus in the cervical lumen is a major challenge to sperm progression, and only a small percentage of the deposited sperm make their way into the uterus. Sperm-cervical mucus penetration tests are useful to assess the ability of both sperm and mucus to interact when in varying physiological states of the oestrous cycle. As sheep are vaginal depositors of semen, they are a useful model to

Airway mucus in pulmonary diseases: Muco-adhesive and muco-penetrating particles to overcome the airway mucus barriers.

Airway mucus is a complex viscoelastic gel that provides a defensive physical barrier and shields the airway epithelium by trapping inhaled foreign pathogens and facilitating their removal via mucociliary clearance (MCC). In patients with respiratory diseases, such as chronic obstructive pulmonary disease (COPD), cystic fibrosis (CF), non-CF bronchiectasis, and asthma, an increase in crosslinking and physical entanglement of mucin polymers as well as mucus dehydration often alters and typically reduces mucus mesh network pore size, which reduces neutrophil migration, decreases pathogen capture

Mucus Hypersecretion and Ciliary Impairment in Conducting Airway Contribute to Alveolar Mucus Plugging in Idiopathic Pulmonary Fibrosis.

Idiopathic pulmonary fibrosis (IPF) is a chronic lung disease attributed to the complex interplay of genetic and environmental risks. The muco-ciliary clearance (MCC) system plays a critical role in maintaining the conduit for air to and from the alveoli, but it remains poorly understood whether the MCC abnormalities in conducting airway are involved in IPF pathogenesis. In this study, we obtained the surgically resected bronchi and peripheral lung tissues from 31 IPF patients and 39 control subjects, and we sought to explore the morphologic characteristics of MCC in conducting airway by using

Pathological mucus and impaired mucus clearance in cystic fibrosis patients result from increased concentration, not altered pH.

Cystic fibrosis (CF) is a recessive genetic disease that is characterised by airway mucus plugging and reduced mucus clearance. There are currently alternative hypotheses that attempt to describe the abnormally viscous and elastic mucus that is a hallmark of CF airways disease, including: 1) loss of CF transmembrane regulator (CFTR)-dependent airway surface volume (water) secretion, producing mucus hyperconcentration-dependent increased viscosity, and 2) impaired bicarbonate secretion by CFTR, producing acidification of airway surfaces and increased mucus viscosity.A series of experiments was

Strategies for measuring airway mucus and mucins.

Mucus secretion and mucociliary transport are essential defense mechanisms of the airways. Deviations in mucus composition and secretion can impede mucociliary transport and elicit airway obstruction. As such, mucus abnormalities are hallmark features of many respiratory diseases, including asthma, cystic fibrosis and chronic obstructive pulmonary disease (COPD). Studying mucus composition and its physical properties has therefore been of significant interest both clinically and scientifically. Yet, measuring mucus production, output, composition and transport presents several challenges. Here

Roles and regulation of the mucus barrier in the gut.

The gastrointestinal tract is coated by a thick layer of mucus that forms the front line of innate host defense. Mucus consists of high molecular weight glycoproteins called mucins that are synthesized and secreted by goblet cells and functions primarily to lubricate the epithelium and protect it from damage by noxious substances. Recent studies have also suggested the involvement of goblet cells and mucins in complex immune functions such as antigen presentation and tolerance. Under normal physiological conditions, goblet cells continually produce mucins to replenish and maintain the mucus ba

Clinical issues of mucus accumulation in COPD.

Airway mucus is part of the lung's native immune function that traps particulates and microorganisms, enabling their clearance from the lung by ciliary transport and cough. Mucus hypersecretion and chronic productive cough are the features of the chronic bronchitis and chronic obstructive pulmonary disease (COPD). Overproduction and hypersecretion by goblet cells and the decreased elimination of mucus are the primary mechanisms responsible for excessive mucus in chronic bronchitis. Mucus accumulation in COPD patients affects several important outcomes such as lung function, health-related qual

Renal mucus gland cystadenomas in a horse.

A 35-year-old horse was submitted to the necropsy service at the University of Kentucky Livestock Disease Diagnostic Center. At necropsy, multiple 1-4-cm-diameter cystic structures were incidentally identified unilaterally in the right renal medulla and the cortex. On histologic examination, the cystic structures compressed the normal renal architecture, were lined by tall columnar epithelium that formed occasional papillary projections, and contained large amounts of mucicarmine and periodic acid-Schiff-positive mucinous material. The masses were diagnosed as renal mucus-gland cystadenomas. T

The repeatability of tear mucus ferning grading.

PURPOSE: Rolando's classification system for tear mucus ferning patterns subjectively assigns grades based on the size and spacing between ferns. Grade 1 and 2 patterns are considered "normal," whereas grade 3 and 4 patterns are often associated with keratoconjunctivitis sicca. This study was designed to examine the intraobserver and interobserver repeatability of Rolando's system. METHODS: Photographic slides (N = 418) of portions of tear ferning patterns were randomly assembled, numbered, and graded by two investigators (I1 and I2). I1 graded the slides twice; the slides were remixed under

The role of airway mucus in pulmonary toxicology.

Airway mucus is a complex airway secretion whose primary function as part of the mucociliary transport mechanism is to to serve as renewable and transportable barrier against inhaled particulates and toxic agents. The rheologic properties necessary for this function are imparted by glycoproteins, or mucins. Some respiratory disease states, e.g., asthma, cystic fibrosis, and bronchitis, are characterized by quantitative and qualitative changes in mucus biosynthesis that contribute to pulmonary pathology. Similar alterations in various aspects of mucin biochemistry and biophysics, leading to muc

Lysozyme in cervical mucus of patients with chorioamnionitis.

Lysozyme and lactoferrin, antimicrobial substances present in cervical mucus, play an important role in defense mechanisms against ascending infections in development of chorioamnionitis (CAM). In this study, we measured the concentration of lysozyme in cervical mucus, and studied its relationship with other factors, and obtained following results. The concentration of lysozyme in cervical mucus decreased more significantly in positive CAM in preterm labor than in negative CAM and control subjects (P < 0.001). A direct correlation between the concentrations of lysozyme and lactoferrin was obse

The dissolution of urinary mucus after cystoplasty.

Three agents have been tested for mucolytic activity to prevent or treat difficulties in bladder emptying following augmentation and substitution cystoplasty, particularly in patients emptying by intermittent self-catheterisation. Carbocysteine produced precipitation of mucus, which was found not to be helpful. N-acetylcysteine and urea both dissolved mucus, but urea proved to be more effective.

Role of cervical mucus in human infertility.

The physiological role of cervical mucus in enhancing spermatozoal penetration at mid-cycle has been established. However, the involvement of cervical mucus in primary infertility is still controversial. Although immunological reactions against spermatozoa in the female have been offered to account for the immobilization of spermatozoa--an observation termed 'hostility' of the mucus--this is not always demonstrable. In addition, there are a number of microbial antigens that are similar to spermatozoal antigens. Antibodies to these antigens are found in the serum of the majority of females and

Molecular fractionation of gastric mucus component by high-performance liquid chromatography: application to pig gastric mucus in vitro and to human gastric mucus collected by aspiration during gastric endoscopy.

The glycoprotein molecular composition of antral and fundic adherent mucus has been studied by high-performance liquid chromatography on a silica gel column. Preliminary assays with pig gastric mucus allowed us to demonstrate the reproducibility of the method. The mucolytic activity of pepsin on this mucus demonstrates its ability to detect degradation of its glycoprotein components. This method was applied to control the state of pig antral mucosa that has previously been used in an in vitro antacid evaluation procedure, and also study human fundic and antral mucus collected by aspiration fro

Mucus-Penetrating Nanostructured Lipid Carriers: An Effective Approach for Cannabidiol to Pass Across the Nasal Mucus-Mucosal Barrier.

Effective nose-to-brain delivery is limited by the mucus-mucosal barrier, which severely hinders drug transport after nasal administration. Although nanostructured lipid carriers (NLCs) offer promising solutions by improving their retention duration in the mucosal layer or promoting mucosal permeation and intracellular uptake, the nanoparticle-mucus interactions, epithelial uptake, and mechanisms supporting transcellular transport in the nasal cavity remain poorly learned. This study was developed to assess the interactions between mucin and NLCs, the absorption and transport capabilities of C
