The effect of carbamate local anaesthetics on vascular smooth muscle.

The effect of carbamate local anaesthetics (pentacaine and heptacaine) on contractile responses induced by transmural nerve stimulation, exogenous noradrenaline (NA) and KCl, as well as on noradrenaline-induced Ca2+ mobilization, were investigated in isolated rabbit blood vessels under isometric conditions. Pentacaine and heptacaine inhibited the neurogenic contractions and those elicited by exogenous NA and KCl. The neurogenic contractions were more sensitive to the local anaesthetics than those evoked by exogenous NA. The onset of inhibition was slow and developed gradually. This inhibition

CaMKIIδ splice variants differentially regulate vascular smooth muscle cell motility.

Vascular smooth muscle (VSM) cell migration promotes physiological and pathophysiological vascular remodeling. A previous study from our laboratory demonstrated that both CaMKIIδ and FYN, a SRC- family kinase, promote VSM cell migration in vitro. CaMKIIδ co-localized with FYN and physically interacted in an activation-dependent manner, with CaMKIIδ activation resulting in FYN activation and dissociation of the interaction. The CAMK2D gene undergoes extensive alternative splicing, including an exon encoding a 21-amino acid C-terminus, resulting in the expression of CaMKIIδ2 and related variants

Sphingosine kinase 1 inhibition aggravates vascular smooth muscle cell calcification.

Medial vascular calcification is common in chronic kidney disease patients and linked to hyperphosphatemia. Upon phosphate exposure, intricate signaling events orchestrate pro-calcific effects in the vasculature mediated by vascular smooth muscle cells (VSMCs). Sphingosine kinase 1 (SPHK1) produces sphingosine-1-phosphate (S1P) and is associated with complex effects in the vascular system. The present study investigated a possible involvement of SPHK1 in VSMC calcification. Experiments were performed in primary human aortic VSMCs under pro-calcific conditions, with pharmacological inhibition o

Mineralocorticoid Receptors in Vascular Smooth Muscle: Blood Pressure and Beyond.

After half a century of evidence suggesting the existence of mineralocorticoid receptors (MR) in the vasculature, the advent of technology to specifically knockout the MR from smooth muscle cells (SMCs) in mice has elucidated contributions of SMC-MR to cardiovascular function and disease, independent of the kidney. This review summarizes the latest understanding of the molecular mechanisms by which SMC-MR contributes to (1) regulation of vasomotor function and blood pressure to contribute to systemic and pulmonary hypertension; (2) vascular remodeling in response to hypertension, vascular inju

Intracellular glutamine level determines vascular smooth muscle cell-derived thrombogenicity.

BACKGROUND AND AIMS: The everolimus-eluting stent (EES), one of the effective stents for in-stent restenosis (ISR), has a lower incidence of stent thrombosis; however, the underlying mechanism remains unknown. This study aimed to identify the effects of everolimus on vascular metabolism and thrombogenicity and examine their mechanistic link. METHODS: EESs and bare-metal stents were implanted in rabbit iliac arteries with smooth muscle cell (SMC)-rich neointima induced by endothelial denudation. Four weeks after stent implantation, the stented arteries were examined for histological analysis a

TRPML1 channels initiate Ca2+ sparks in vascular smooth muscle cells.

TRPML1 (transient receptor potential mucolipin 1) is a Ca2+-permeable, nonselective cation channel localized to the membranes of endosomes and lysosomes and is not present or functional on the plasma membrane. Ca2+ released from endosomes and lysosomes into the cytosol through TRPML1 channels is vital for trafficking, acidification, and other basic functions of these organelles. Here, we investigated the function of TRPML1 channels in fully differentiated contractile vascular smooth muscle cells (SMCs). In live-cell confocal imaging studies, we found that most endosomes and lysosomes in freshl

Vascular Kv7 channels control intracellular Ca2+ dynamics in smooth muscle.

Voltage-gated Kv7 (or KCNQ) channels control activity of excitable cells, including vascular smooth muscle cells (VSMCs), by setting their resting membrane potential and controlling other excitability parameters. Excitation-contraction coupling in muscle cells is mediated by Ca2+ but until now, the exact role of Kv7 channels in cytosolic Ca2+ dynamics in VSMCs has not been fully elucidated. We utilised microfluorimetry to investigate the impact of Kv7 channel activity on intracellular Ca2+ levels and electrical activity of rat A7r5 VSMCs and primary human internal mammary artery (IMA) SMCs. MicroRNAs orchestrating senescence of endothelial and vascular smooth muscle cells.

During organism aging, the process of cellular senescence is triggered by critical stressors such as DNA damage, oncogenes, oxidative stress, and telomere erosion, and vascular cells are not exempted. Senescent cells stop proliferating but remain metabolically active producing pro-inflammatory signals in the environment collectively named senescence-associated secretory phenotype (SASP) that contribute to the amplification of the response to the neighbor and distant cells. Although the shift toward senescence is protective against tumors and needed during wound healing, the accumulation of sen

Cyclic guanosine monophosphate compartmentation in human vascular smooth muscle cells.

AIMS: The role of different vascular subtypes of phosphodiesterases (PDE) in cGMP compartmentalization was evaluated in human smooth muscle cells. METHODS AND RESULTS: To understand how the cGMP conveys different information we infected smooth muscle cells with adenovirus containing mutants of the rat olfactory cyclic nucleotide-gated (CNG) channel-subunit and we recorded the associated cGMP-gated current (ICNG). The whole cell configuration of patch clamp technique was used to measure the ICNG and also the potassium current (IK) in human umbilical artery smooth muscle cells (HUASMC). ANP (0.

Towards the therapeutic use of vascular smooth muscle progenitor cells.

Recent advances in the development of alternative proangiogenic and revascularization processes, including recombinant protein delivery, gene therapy, and cell therapy, hold the promise of greater efficacy in the management of cardiovascular disease in the coming years. In particular, vascular progenitor cell-based strategies have emerged as an efficient treatment approach to promote vessel formation and repair and to improve tissue perfusion. During the past decade, considerable progress has been achieved in understanding therapeutic properties of endothelial progenitor cells, while the thera

Tormentic acid reduces vascular smooth muscle cell proliferation and survival.

The triterpene tormentic acid (TA) has been reported to exhibit anticancer, anti-inflammatory and anti-atherogenic properties, and minimal toxicity has been detected in in vivo. Vascular smooth muscle cell (VSMC) proliferation and apoptosis resistance are hallmarks of vasculoproliferative diseases, such as post-angioplasty restenosis. The present study was designed to assess the effects of TA on the phenotype of cultured VSMC. The exposure of VSMC to TA (30 muM) significantly increased apoptosis of serum-deprived A7r5 cells, whereas cell survival in the presence of 10% fetal bovine serum was l

Annexin-mediated matrix vesicle calcification in vascular smooth muscle cells.

In bone, osteoblasts and chondrocytes synthesize matrix vesicles (MVs) that interact with collagen to initiate calcification. MVs have been identified in human calcified arteries but are poorly characterized. The objective of this study is to determine the role of annexins and fetuin-A in MV formation and activity during calcification in bovine vascular smooth muscle cells (BVSMCs). BVSMCs were treated with control or calcification (high phosphorus) media, and cellular MVs were isolated by collagenase digestion and secreted MVs were isolated from cultured media by ultracentrifugation. Cyclic AMP-adenosine pathway inhibits vascular smooth muscle cell growth.

In this study we determined whether cAMP is metabolized to adenosine in vascular smooth muscle cells and whether cAMP-derived adenosine modulates vascular smooth muscle cell growth. Confluent smooth muscle cells were exposed to cAMP (0.01 to 30 mumol/L) in the presence and absence of 3-isobutyl-1-methylxanthine (IBMX, 1 mmol/L; an inhibitor of both extracellular and intracellular phosphodiesterase), alpha, beta-methyleneadenosine 5'-diphosphate (AMP-CP, 100 mumol/L; an ecto-5'-nucleotidase inhibitor), and 1,3-dipropyl-8-p-sulfophenyl-xanthine (DPSPX, 100 mumol/L; a xanthine that can inhibit ex

Mitochondrial Function Changes in hiPSCs-Derived Vascular Smooth Muscle Cells.

Utilizing human-induced pluripotent stem-cell-derived vascular smooth muscle cells (hiPSC-VSMCs) to address seed cell source in tissue-engineered vascular grafts necessitates promoting cellular maturation. Morphological changes and functional remodeling of mitochondria are crucial to cellular differentiation and maturation. However, our current understanding of these changes during the transition from hiPSCs to VSMCs is limited. Our findings revealed that after hiPSC-VSMC differentiation, mitochondria transitioned from perinuclear to cytoplasmic distribution, increased in number, and changed s

Uncovering a Sex-Specific cAMP Landscape in Vascular Smooth Muscle.

The second messenger, cyclic adenosine monophosphate (cAMP), plays a crucial role in regulating cellular function, including in vascular smooth muscle (VSM). While extensive research has focused on the compartmentalization of cAMP signaling in various cell types, the influence of biological sex on distinct cAMP pools and their functional implications in the VSM remains largely unexplored. To address this knowledge gap, we employed a multiscale experimental approach spanning targeted cAMP biosensors, wire myography, live-cell calcium imaging, proximity ligation assays, and in vivo ultrasound im

Smooth Muscle Cell MAPK14 Promotes Vascular Calcification During Hyperlipidemic Aging.

Vascular calcification is a hallmark of vascular aging that is accelerated by hyperlipidemia and contributes to adverse cardiovascular outcomes. Although vascular smooth muscle cells (VSMCs) are key mediators of vascular calcification, the signaling pathways linking aging-associated stress to osteogenic remodeling remain incompletely understood. To define the role of MAPK14 (p38α) in vascular aging, we integrated bulk RNA sequencing of young and aged mouse aortas with published single-nucleus RNA sequencing data from VSMC-specific Mapk14 knockout (KO) mice. Bulk RNA-seq identified age-associat

Robotic handling preserves iPSC-derived vascular smooth muscle cell differentiation.

To tackle the global cardiovascular disease epidemic, vascular cell culture requires scalable, efficient, and reproducible methods for differentiating cells. However, no studies have validated whether automation, essential for scale-up, preserves differentiation quality of cells. Here, we developed a differentiation protocol for induced pluripotent stem cell (iPSC)-derived vascular smooth muscle cells (iVSMCs) that is applicable to both manual, and automated robotic culture. iPSC lines generated from male and female donors across various ages were used to optimize the generation of iVSMCs. iVS
