The application of 1.5-4 microM dantrolene decreased the threshold and the current sensitivity of the rhythmic hyperpolarizations that occur during depolarization of the sensory nerve terminal in the frog muscle spindle. The higher concentration provoked spontaneous rhythmic changes even without depolarization. Methylxanthines (5 mM caffeine, theophylline or pentylene-tetrazole) increased the threshold and the sensitivity. Electron microscopic observations of the dantrolene-treated spindles revealed numerous electron-dense deposits associated with the cytoplasmic membrane of the sensory termin

CONTEXT: Ankle sprains are common in athletes, with functional ankle instability (FAI) developing in approximately half of cases. The relationship between laxity and FAI has been inconclusive, suggesting that instability may be caused by insufficient sensorimotor function and dynamic restraint. Research has suggested that deafferentation of peripheral mechanoreceptors potentially causes FAI; however, direct evidence confirming peripheral sensory deficits has been elusive because previous investigators relied upon subjective proprioceptive tests.
OBJECTIVE: To develop a method for simultaneous

Muscle spindles are stretch receptors lying deep within the muscle belly involved in detecting changes in muscle length and playing a fundamental role in motor control, posture and synchronized gait. They are made up of an external capsule surrounding three to five intrafusal muscle fibres and a nuclear bag complex. Dysfunction of muscle spindles leads to abnormal proprioceptor function, which has been linked to aberrant bone and cartilage development, scoliosis, kyphosis and joint contractures. RYR1, the gene encoding the calcium release channel of the sarcoplasmic reticulum, is the most comm

Muscle stretch proprioceptors (muscle spindles) are required for stretch reflexes and locomotor control. Proprioception abnormalities are observed in many human neuropathies, but the mechanisms involved in establishing and maintaining muscle spindle innervation and function are still poorly understood. During skeletal muscle development, sensory (Ia-afferent) innervation induces contacted myotubes to transform into intrafusal muscle fibers that form the stretch receptor core. The transcriptional regulator Egr3 is induced in Ia-afferent contacted myotubes by Neuregulin1 (Nrg1)/ErbB receptor sig

1. The distribution of fusimotor axons to bag1, bag2 and chain muscle fibres in cat tenuissimus spindles has been studied using a modification of the glycogen-depletion technique of Edstrrom & Kugelberg (1968). Single fusimotor axons were stimulated intermittently at 40-100/sec for long periods (30-90 sec) during blood occlusion. Portions of muscle containing the activated spindles were quick-frozen, fixed in absolute ethanol during freeze-substitution, and then embedded in paraffin wax. Serial transverse sections were stained for glycogen using the periodic acid-Schiff method, and examined fo

Proprioceptive dysfunction is a common rehabilitation issue in neurological injury, and the muscle spindle (MS) is considered a critical structure in this dysfunction. The present study aimed to investigate the morphological alterations of MS in two classical neurological diseases: denervation and stroke, and to understand the role of MS in these two diseases preliminarily. The denervation and stroke model animals were constructed by using a surgical operation and middle cerebral artery occlusion (MCAO) protocol, respectively. The muscles from the affected-side limb and the healthy-side limb w

In some neurological conditions, like Parkinson's disease (PD) and Cerebral Palsy (CP), as well as with ageing, muscle spindles have been mentioned as participating in the pathological response of observed muscles. The aim of this review has therefore been to examine what is known about muscle spindle receptors, their function and how they are involved in regulating precise muscle movement in relation to these two conditions. Data from acoustic myography (AMG) studies with healthy controls (HC), CP and PD subjects have been re-examined with a view to identifying possible effects of changes in

Previous work had shown that some of the effects of catechol could be via the fusimotor system. In order to determine the extent of fusimotor involvement, recordings have been made from muscle spindle afferents in split dorsal root filaments of anaesthetised rats. Catechol failed to excite de-efferented muscle spindles therefore eliminating many possible non-fusimotor effects. Over 80% of spindle afferents with intact efferents showed increased discharge frequency 1 min after injection, this increase often following a biphasic pattern with a pronounced pre-myoclonic burst and decline followed

Arterial pulsations are known to modulate muscle spindle firing; however, the physiological significance of such synchronised modulation has not been investigated. Unitary recordings were made from 75 human muscle spindle afferents innervating the pretibial muscles. The modulation of muscle spindle discharge by arterial pulsations was evaluated by R-wave triggered averaging and power spectral analysis. We describe various effects arterial pulsations may have on muscle spindle afferent discharge. Afferents could be "driven" by arterial pulsations, e.g., showing no other spontaneous activity tha

Canine degenerative myelopathy (DM) is a progressive, adult-onset neurodegenerative disease, with similarities to superoxide dismutase 1 (SOD1)-associated ALS. Clinical signs of DM begin with asymmetric pelvic limb general proprioceptive ataxia and spastic paresis that progresses to flaccid tetraplegia and respiratory failure. Although pelvic limb muscle atrophy and peripheral neuropathy have been reported in DM, intramuscular SOD1 accumulation and muscle spindle pathology remain incompletely characterized. This study evaluated pathological changes in the tibialis cranialis muscle of Pembroke

Motor control requires sensory feedback, and the nature of this feedback has implications for the tasks of the central nervous system (CNS): for an approximately linear mechanical system (e.g., a freely standing person, a rider on a bicycle), if the sensory feedback does not contain the state variables (i.e., joint position and velocity), then optimal control actions are based on an internal dynamical system that estimates these states from the available incomplete sensory feedback. Such a computational system can be implemented as a recurrent neural network (RNN), and it uses a sensory model

INTRODUCTION: The masticatory muscles are considered to be important determinants of facial form, but little is known of the muscle spindle reflex characteristics and their relationship, if any, to face height. The aim of this study was to determine whether spindle reflexes, evoked by mechanical stimulation of an incisor and recorded on the masseter muscle, correlated with different facial patterns.
METHODS: Twenty-eight adult volunteers (16 women; ages, 19-38 years) underwent 2-N tap stimuli to their maxillary left central incisor during simulated mastication. Reflexes were recorded during l

The Riley-Day syndrome is the most common of the hereditary sensory and autonomic neuropathies (Type III). Among the well-recognized clinical features are reduced pain and temperature sensation, absent deep tendon reflexes and a progressively ataxic gait. To explain the latter we tested the hypothesis that muscle spindles, or their afferents, are absent in hereditary sensory and autonomic neuropathy III by attempting to record from muscle spindle afferents from a nerve supplying the leg in 10 patients. For comparison we also recorded muscle spindles from 15 healthy subjects and from two patien

Muscle spindles from the hind limb muscles of adult Ryr1(I4895T/wt) (IT/+) mice exhibit severe structural abnormalities. Up to 85% of the spindles are separated from skeletal muscle fascicles by a thick layer of connective tissue. Many intrafusal fibers exhibit degeneration, with Z-line streaming, compaction and collapse of myofibrillar bundles, mitochondrial clumping, nuclear shrinkage and pyknosis. The lesions resemble cores observed in the extrafusal myofibers of this animal model and of core myopathy patients. Spindle abnormalities precede those in extrafusal fibers, indicating that they a

Among the mechanisms that may result in modulation of the stretch reflex by the recent history of muscle contraction is the history dependence observed under some conditions in the response properties of muscle spindles. The present study was designed to test one report that in successive trials of muscle stretch-release, spindle afferent firing during stretch, i.e., the dynamic response shows no history dependence beyond the initial burst of firing at stretch onset. Firing responses of spindle afferents were recorded during sets of three consecutive trials of triangular stretch-release applie

The proprioceptive feedback associated with the performance of even quite simple movements is always generated by the whole set of muscles subjected to mechanical deformation (lengthening, shortening, contraction, etc.) during that particular movement. The question was addressed here as to how muscle spindle feedbacks arising from agonist and antagonist muscles may contribute to the coding of movement parameters such as the direction and velocity. For this purpose, the activity of single muscle spindle afferents located in the lateral peroneal nerve was analysed using the microneurographic tec

The goals of this review are to improve understanding of the aetiology of chronic muscle pain and identify new targets for treatments. Muscle pain is usually associated with trigger points in syndromes such as fibromyalgia and myofascial syndrome, and with small spots associated with spontaneous electrical activity that seems to emanate from fibers inside muscle spindles in EMG studies. These observations, added to the reports that large-diameter primary afferents, such as those innervating muscle spindles, become hyperexcitable and develop spontaneous ectopic firing in conditions leading to n

OBJECTIVES: To test the hypothesis that the firing rates and discharge variability of human muscle spindles are not affected by spinal cord injury.
METHODS: Tungsten microelectrodes were inserted into muscle fascicles of the peroneal nerve in six individuals with complete paralysis of the lower limbs following spinal cord injury: 12 afferents were spontaneously active at rest and 7 were recruited during passive muscle stretch. For comparison, recordings were made from 17 spontaneously active and 9 stretch-recruited afferents in 12 intact subjects.
RESULTS: Firing rates for the spontaneously
