Current cutting-edge omics techniques on musculoskeletal tissues and diseases.

Musculoskeletal disorders, including osteoarthritis, rheumatoid arthritis, osteoporosis, bone fracture, intervertebral disc degeneration, tendinopathy, and myopathy, are prevalent conditions that profoundly impact quality of life and place substantial economic burdens on healthcare systems. Traditional bulk transcriptomics, genomics, proteomics, and metabolomics have played a pivotal role in uncovering disease-associated alterations at the population level. However, these approaches are inherently limited in their ability to resolve cellular heterogeneity or to capture the spatial organization

Sarcopenia in rheumatic and musculoskeletal diseases: pathophysiology, diagnosis, and management.

Sarcopenia, defined by the progressive decline of skeletal muscle mass, strength, and performance, is an important, yet often overlooked complication of rheumatic and musculoskeletal diseases (RMDs). It arises from a multifactorial interplay of chronic systemic inflammation, oxidative stress, hormonal dysregulation, mitochondrial dysfunction, and prolonged glucocorticoid therapy. Elevated pro-inflammatory cytokines, such as IL-6, TNF-α, and IL-1β, promote catabolic pathways, impair regeneration, and accelerate muscle degradation. The prevalence of sarcopenia in RMDs may reach 40%, particularly

Osteostatin, a peptide for the future treatment of musculoskeletal diseases.

Nowadays, the treatment of musculoskeletal diseases represents a major challenge in the developed world. Diseases such as osteoporosis, osteoarthritis and arthritis have a high incidence and prevalence as a consequence of population aging, and they are also associated with a socioeconomic burden. Many efforts have been made to find a treatment for these diseases with various levels of success, but new approaches are still needed to deal with these pathologies. In this context, one peptide derived for the C-terminal extreme of the Parathormone related Peptide (PTHrP) called Osteostatin can be u

Hydrogen Sulfide in Musculoskeletal Diseases: Molecular Mechanisms and Therapeutic Opportunities.

Significance: Musculoskeletal diseases seriously affect global health, but their importance is greatly underestimated. These diseases often afflict the elderly, leading to disability, paralysis, and other complications. Hydrogen sulfide (H2S) plays an important role in the occurrence and development of musculoskeletal diseases, which may have potential therapeutic significance for these diseases. Recent Advances: Recently, it has been found that many musculoskeletal diseases, such as osteoporosis, periodontitis, muscle atrophy, muscle ischemia-reperfusion injury, muscle contraction under high

Mast Cell Involvement in the Pathogenesis of Selected Musculoskeletal Diseases.

In recent years, there has been a noteworthy revival of interest in the function of mast cells (MCs) in the human body. It is now acknowledged that MCs impact a wide array of processes beyond just allergies, leading to a shift in research direction. Unfortunately, some earlier conclusions were drawn from animal models with flawed designs, particularly centered around the receptor tyrosine kinase (Kit) pathway. Consequently, several subsequent findings may have been unreliable. Thus, what is now required is a re-examination of these earlier findings. Nevertheless, the remaining data are fascina

Eight pillars of oncorheumatology: Crossroads between malignancies and musculoskeletal diseases.

RELATIONSHIP BETWEEN MALIGNANCIES AND MUSCULOSKELETAL DISEASES: Oncorheumatology is the meeting point of tumor formation and rheumatic musculoskeletal diseases (RMD). Multiple interactions exist between these two medical specialties. One major field is the topic of malignancies associated with rheumatic diseases, while the other topic covers the development of musculoskeletal disease in cancer patients. Within the first group, secondary malignancies may be associated with rheumatic diseases. Mostly sustained inflammation is responsible for transition into cancer. Tumor-associated antigens (TAA

Surgical Management of Orthopedic and Musculoskeletal Diseases of Feedlot Calves.

Injuries, infections, and disorders of the musculoskeletal system are common in feedlot calves. These conditions often are amenable to surgical treatment with return of the calf to productivity. Weight gain and carcass quality are expected to be significantly adversely affected by pain and debilitation. The goal of surgical management of disorders of the joints, muscles, and feet should be resolution of the inciting cause, mitigation of pain, and restoration of form and function. If these are achieved, calves should return to acceptable, if not normal, feed intake, rate of gain, and carcass qu

[Roles of Musculoskeletal Ultrasonography in the Management of Rheumatic Diseases].

Inflammation of soft tissues and the subsequent structural damage are the characteristic features of many rheumatic conditions. However, the conventional measures to evaluate these features are not accurate, potentially causing under- and over-diagnosis or treatment. Musculoskeletal ultrasonography, on the other hand, directly visualizes features which are characteristic to rheumatic conditions, such as synovitis, tenosynovitis, bursitis, enthesitis, crystal depositions, bone erosions, and osteophytes/enthesophytes. By visualizing these key features, ultrasound facilitates a more accurate eval

Role of diagnostic ultrasound in the assessment of musculoskeletal diseases.

The wide availability and recent improvement in technology coupled with portability, low cost and safety makes ultrasound the first choice imaging investigation for the evaluation of musculoskeletal diseases. Diagnostic use of ultrasound findings is greatly enhanced by knowledge of the clinical presentation. Conversely, ultrasound skills with its prerequisite anatomical knowledge make the clinical diagnosis more precise and reduce uncertainty in the choice of therapy. Therefore, it is essential for rheumatologists to acquire ultrasonography skills in order to improve patient care. Ultrasound e

Histone Deacetylase 4: The Epigenetic Hub in Degenerative Musculoskeletal Diseases.

Aging has rendered Degenerative Musculoskeletal Diseases (DMDs) an increasingly significant public health burden. Growing evidence suggests that epigenetic dysregulation plays a crucial mechanistic role in the pathogenesis of DMDs. Histone Deacetylase 4 (HDAC4), known for its regulatory roles in the nervous, cardiovascular, and cancer systems, has also emerged as a central coordinator of musculoskeletal physiology and pathology. It dynamically modulates cellular metabolism in cartilage, bone, and muscle tissues. This review aims to provide a comprehensive overview of current research on HDAC4

Radiomics in non-oncologic musculoskeletal diseases: from pixels to practice.

Radiomics has emerged as a powerful tool in medical imaging, enabling the extraction of high-dimensional data from radiologic images to uncover quantitative relationships between image features and underlying pathology. Although it is most established in oncology, radiomics is now being widely adopted for non-oncologic musculoskeletal conditions, including osteoporosis, osteoarthritis, rheumatologic diseases, nerve pathologies, and trauma-related injuries. This review provides an overview of the principles and workflows of radiomics, emphasising its applications beyond oncology. It highlights

[Shoulder pain in inflammatory rheumatic diseases : Importance of musculoskeletal ultrasound].

Shoulder pain is a frequent reason for presentation in medical practices; however, it can sometimes be challenging to differentiate between an inflammatory versus mechanical or degenerative cause of shoulder pain. A swelling of the shoulder joint is only palpable in cases of pronounced clinical findings. In rheumatoid arthritis (RA) and psoriatic arthritis (PsA) the shoulder joints can be involved, which is early and frequent, particularly in the case of RA and is associated with a more severe course of the disease. Polymyalgia rheumatica (PMR) is typically manifested with shoulder and hip gir

Different training instructions in persons with rheumatic and musculoskeletal diseases.

INTRODUCTION: People with rheumatic and musculoskeletal diseases are advised to do aerobic exercise for symptom relief and to reduce the risk of cardiovascular disease. Continuous exercise at an intensity causing a rate of perceived exertion of 15, on a 6-20-point Borg scale, exemplifies such exercise. Also, the instruction "Now you need to increase your heart rate" is used before aerobic exercise. However, the exercise intensity caused by that instruction is unknown. We tested the hypothesis that intensity during cycling, initiated with the instruction "Now you need to increase your heart rat

Rare Causes of Musculoskeletal Pain: Thinking beyond Common Rheumatologic Diseases.

OBJECTIVES: Rare metabolic bone diseases can present with symptoms mimicking more common rheumatological conditions including spondyloarthritis, osteoarthritis, and fibromyalgia. Increasing awareness of these rare diseases within the rheumatology community is vital to ensure that affected patients are diagnosed and appropriately treated. The literature includes several reports of tumour-induced osteomalacia initially diagnosed as rheumatic disease, but other rare diseases such as X-linked hypophosphatemia (XLH) and hypophosphatasia (HPP) also deserve attention. Here, we describe two cases of a

Diabetes mellitus-related musculoskeletal disorders: Unveiling the cluster of diseases.

The current study ushers in a comprehensive review in clinical research to demonstrate the prevalence of musculoskeletal (MSK) complications in diabetes mellitus and the most relevant clinical aspects. In particular, revealing the early symptoms of the disorders, the pathology lurking behind the complications and their optimal management. In diabetes mellitus, MSK complications are common and are largely due to similar pathogenetic factors responsible for the internal organ complications associated with diabetes leading to chronic low-intensity inflammatory processes. MSK disorders develop by

Impact of Cadmium Mediated by Tobacco Use in Musculoskeletal Diseases.

Tobacco use has a negative impact on health due to its relationship with the development of high-mortality diseases, such as pulmonary cancer. However, the effect of cadmium (Cd), present in tobacco smoke, on the development of joint diseases has been scarcely studied. The objective of this review is to discuss the evidence regarding the mechanisms by which Cd exposure, through tobacco smoke, may lead to the development of osteoarthritis (OA), osteoporosis (OP), and rheumatoid arthritis (RA). There's evidence suggesting a string association between moderate to severe OA development and tobacco

Dehydroepiandrosterone and age-related musculoskeletal diseases: Connections and therapeutic implications.

Musculoskeletal disorders related to ageing are one of the most common causes of mortality and morbidity among elderly individuals worldwide. The typical constitutive components of the musculoskeletal system, including bone, muscle, and joints, gradually undergo a process of tissue loss and degeneration as a result of life-long mechanical and biological stress, ultimately leading to the onset of a series of age-related musculoskeletal diseases, including osteoporosis (OP), sarcopenia, and osteoarthritis (OA). Dehydroepiandrosterone (DHEA), a precursor of androgen secreted mainly by the adrenal
