Sequencing of transcriptomes from two Miscanthus species reveals functional specificity in rhizomes, and clarifies evolutionary relationships.

BACKGROUND: Miscanthus is a promising biomass crop for temperate regions. Despite the increasing interest in this plant, limited sequence information has constrained research into its biology, physiology, and breeding. The whole genome transcriptomes of M. sinensis and M. sacchariflorus presented in this study may provide good resources to understand functional compositions of two important Miscanthus genomes and their evolutionary relationships. RESULTS: For M. sinensis, a total of 457,891 and 512,950 expressed sequence tags (ESTs) were produced from leaf and rhizome tissues, respectively, w

Recent diversification of Chrysoritis butterflies in the South African Cape (Lepidoptera: Lycaenidae).

Although best known for its extraordinary radiations of endemic plant species, the South African fynbos is home to a great diversity of phytophagous insects, including butterflies in the genus Chrysoritis (Lepidoptera: Lycaenidae). These butterflies are remarkably uniform morphologically; nevertheless, they comprise 43 currently accepted species and 68 currently valid taxonomic names. While many species have highly restricted, dot-like distributions, others are widespread. Here, we investigate the phylogenetic and biogeographic history underlying their diversification by analyzing molecular ma

A molecular timescale for evolution of cobamide biosynthesis.

Cobamides are essential nutrients for most organisms but are only biosynthesized by a limited number of taxa through aerobic and anaerobic pathways. Although the origin of these widespread shared cofactors changed ecosystems and the metabolisms of living organisms, little evolutionary information is available about the biosynthetic genes and the producers. Here, we established a timeframe for the emergence of cobamide biosynthesis genes and producers, using a series of Bayesian molecular clock analyses combined with phylogenetic reconciliation. We revealed the partial producers of tetrapyrrole

Adenovirus-vectored foot-and-mouth disease vaccine confers early and full protection against FMDV O1 Manisa in swine.

A human adenovirus (Ad5) vectored foot-and-mouth disease virus (FMDV) O1-Manisa subunit vaccine (Ad5-O1Man) was engineered to deliver FMDV O1-Manisa capsid and capsid-processing proteins. Swine inoculated with Ad5-O1Man developed an FMDV-specific humoral response as compared to animals inoculated with an empty Ad5-vector. Vaccinated animals were completely protected against homologous challenge at 7 or 21 days post-vaccination. Potency studies exhibited a PD50 of about 107 pfu/animal while a dose of 4×107pfu/animal fully protected swine against FMDV intradermal challenge. In-vitro cross-neutra

Phagocytic activity of marine and freshwater bivalves: in vitro exposure of hemocytes to metals (Ag, Cd, Hg and Zn).

We measured non-specific immune function of various bivalves from marine (Cyrtodaria siliqua, Mactromeris polynyma, Mesosdesma arctatum, Mya arenaria, Mya truncata, Mytilus edulis, Serripes groenlandicus, Siliqua costata) and freshwater environments (Dreissena polymorpha and Elliptio complanata). We used flow cytometry to quantify the phagocytosis of fluorescent microspheres by hemocytes exposed in vitro to increasing levels of various metal compounds (AgNO(3), CdCl(2), CH(3)HgCl, HgCl(2) and ZnCl(2)). In some species, low doses of mercury (organic and inorganic) and Zn suggest a hormesis-like

Genetic Diversification in a New Guinean Frog Genus (Mantophryne, Microhylidae) was Driven by Ancient Tectonic Activity and Climate Reorganisation.

Western Melanesia sits at the junction of three major tectonic plates, characterised by dynamic plate interactions and geological diversity. Tectonic activity in this region has given rise to modern New Guinea, a relatively young island with extreme topographic heterogeneity and evolutionarily complex biota. New Guinea-particularly the East Papuan Composite Terrane (EPCT)-supports exceptional frog diversity; however, evolutionary histories of many frogs of the EPCT are poorly understood. The microhylid frog genus Mantophryne currently includes five described species (M. axanthogaster, M. insig

Distribution of RPTLN Genes Across Reptilia: Hypothesized Role for RPTLN in the Evolution of SVMPs.

We report the cloning, full-length sequencing, and broad distribution of reptile-specific RPTLN genes across a number of Anapsida (Testudines), Diapsida (Serpentes, Sauria), and Archosauria (Crocodylia) taxa. The remarkable structural conservation of RPTLN genes in species that had a common ancestor more than 250 million years ago, their low transcriptional level, and the lack of evidence for RPTLN translation in any reptile organ investigated, suggest for this ancient gene family a yet elusive function as long noncoding RNAs. The high conservation in extant snake venom metalloproteinases (SVM

Exploratory Temporal and Evolutionary Insights into the Filoviridae Family Through Multiprotein Phylogeny.

Filoviruses are among the most lethal viral human pathogens known, with significant relevance to public health, yet their evolutionary history remains poorly resolved. This study applied a multiprotein molecular phylogenetic approach to investigate the evolutionary and temporal dynamics of the family Filoviridae. Amino acid sequences from the proteome and seven individual proteins (NP, VP35, VP40, GP, VP30, VP24, L) were analyzed using MEGA 12, with RelTime inference anchored on uniform calibrations, and integration of epidemiological data (cases, fatalities, case fatality). Rhinocerebral mucormycosis.

Rhinocerebral mucormycosis is a fungal diseases that has a 50% mortality. Its occurrence has increased, possibly because of greater use of chemotherapeutic agents that mya compromise the immunologic defenses of the host or alter the normal flora. The earliest signs, ulceration and pain, may appear in the mouth. In the patient described in this report, the autopsy showed that mucormycosis had entered the brain cells.

Conservation hints for Pinna nobilis from a century-old genetic time capsule.

The noble pen shell, Pinna nobilis, is an iconic marine bivalve endemic to the Mediterranean Sea, playing a key role as an ecosystem engineer. Over the past century, it has faced severe threats from overharvesting, pollution, and catastrophic mass mortality events. This study analysed 119 mitochondrial COI gene sequences from historical (1700s, 1920s, 1970s, 1990s) and modern (2000s) samples, including survivors of recent mass mortality crises. We standardised a protocol to extract DNA from ancient byssus samples over a century old and dated the emergence of the mitochondrial lineages of Pinna

A combination approach to treating fungal infections.

Azoles are antifungal drugs used to treat fungal infections such as candidiasis in humans. Their extensive use has led to the emergence of drug resistance, complicating antifungal therapy for yeast infections in critically ill patients. Combination therapy has become popular in clinical practice as a potential strategy to fight resistant fungal isolates. Recently, amphiphilic tobramycin analogues, C12 and C14, were shown to display antifungal activities. Herein, the antifungal synergy of C12 and C14 with four azoles, fluconazole (FLC), itraconazole (ITC), posaconazole (POS), and voriconazole (

Diaphorocleidus machacae n. sp. (Monogenea) Infecting the Gill Lamellae of Brycon guatemalensis (Characiformes: Bryconidae) from the Rio Lacantún Basin in Chiapas, Mexico.

BACKGROUND: There are currently nine known gill monogenean species of Diaphorocleidus Jogunoori et al. (Systematic Parasitology 58:115-124, 2004) (Dactylogyridae) which are dispersed within and/or among five fish families in the Neotropical Characiformes: Bryconidae, Acestrorhynchidae, Characidae, Iguanodectidae, and Hemiodontidae. During a fish parasitological study carried out in the Rio Lacantún from Chiapas, Mexico, a new species of Diaphorocleidus on the gill lamellae of Brycon guatemalensis Regan, 1908, was discovered. OBJECTIVE: Our objective was to morphologically describe the new par

A new genus and species of Hydrobiidae Stimpson, 1865 (Caenogastropoda, Truncatelloidea) from Peloponnese, Greece.

Minute caenogastropod brackish-water gastropods, formerly classified as Hydrobia, are important elements of the brackish-water fauna and were objects of intensive study for many years. Until now, five genera have been distinguished, most of them represented by a number of species, but rather indistinguishable without molecular data (cytochrome oxidase subunit I - COI). In the eastern Mediterranean region, they are still poorly studied. In this paper, we present a new species of "Hydrobia" from the brackish Moustos spring, Arkadia, eastern Peloponnese, Greece. The shell, protoconch, radula, fem

The new chimeric chiron genes evolved essential roles in zebrafish embryonic development by regulating NAD+ levels.

The origination of new genes is important for generating genetic novelties for adaptive evolution and biological diversity. However, their potential roles in embryonic development, evolutionary processes into ancient networks, and contributions to adaptive evolution remain poorly investigated. Here, we identified a novel chimeric gene family, the chiron family, and explored its genetic basis and functional evolution underlying the adaptive evolution of Danioninae fishes. The ancestral chiron gene originated through retroposition of nampt in Danioninae 48-54 million years ago (Mya) and expanded

Cryptic diversity in the Japanese mantis shrimp Oratosquilla oratoria (Crustacea: Squillidae): Allopatric diversification, secondary contact and hybridization.

Mounting evidence of cryptic species in the marine realm emphasizes the necessity to thoroughly revise our current perceptions of marine biodiversity and species distributions. Here, we used mitochondrial cytochrome oxidase subunit I (mtDNA COI) and nuclear ribosomal internal transcribed spacer (nrDNA ITS) to investigate cryptic diversity and potential hybridization in the Japanese mantis shrimp Oratosquilla oratoria in the Northwestern (NW) Pacific. Both mitochondrial and nuclear gene genealogies revealed two cryptic species in this morphotaxon, which was further confirmed by extensive popula

Foot-and-mouth disease virus vaccine with VP1 G-H loop substitution of the Cathay strain broadens antigen spectrum.

Recently, serotype O foot-and-mouth disease viruses (FMDVs) belonging to four lineages (O/ME-SA/PanAsia, O/ME-SA/Ind2001, O/SEA/Mya-98, and O/Cathay) co-circulate in China, and the emergence of new variant viruses belonging to the Cathay lineage renders the existing vaccines less effective, which poses a serious threat to the livestock industries. The surface-exposed G-H loop of the VP1 structural protein of FMDV plays an important role in inducing neutralizing antibodies, generation of antigenic variants, and virus attachment. Here, we generated three recombinant FMDVs in which the G-H loops

Chromosome-scale genome assembly of Artabotrys hexapetalus provides insights into its evolution and terpenoid biosynthesis.

Artabotrys hexapetalus is used as traditional Chinese medicine for the treatment of malaria. Its antimalarial activity has been shown to be the result of the sesquiterpene designated as yingzhaosu A (YZSA), which mainly occurs in the roots of A. hexapetalus. Despite its medicinal significance, the genetic basis and molecular mechanisms underlying the biosynthesis of its active terpenoids remain unclear. In this study, a high-quality chromosome-scale genome of A. hexapetalus was first reported. In total, 915.15 Mb of genome sequences were assembled, of which 911.58 Mb was sorted into 8 chromoso

Seascape Genomics of Elacatinus punctiulatus (Ginsburg, 1938): Understanding the Historical and Contemporary Drivers of Population Structure in the Gulf of California.

The genomic diversity and population structure of marine species represents a complex mosaic shaped by historical and contemporary environmental seascape features that maintain or alter it over time. The Gulf of California (GC) is an interior sea with a dynamic history during its formation and a contemporary environmental and oceanographic complexity; hence, it is a suitable system to test the effect of historical and contemporary factors on genomic diversity in marine species. We investigated the genomic seascape of the redhead goby (Elacatinus puncticulatus; Ginsburg, 1938), a cryptobenthic
