Clonal spread of Mycobacterium monacense strains among patients with extrapulmonary mycobacteriosis.

BACKGROUND: This study presents the phylogenetic and antimicrobial susceptibility characterization of Mycobacterium monacense, a rare nontuberculous mycobacterium (NTM), cultured from clinical extrapulmonary samples. METHODS: Eight Mycobacterium monacense isolates were identified between 2019 and 2023 in the Western Cape province of South Africa. Whole-genome sequencing (WGS) was applied to assess phylogenetic relatedness, identify virulence factors, and characterize the resistome of the isolates. Antimicrobial susceptibility testing (AST) was performed using the GenoType NTM-DR line probe as

Mycobacterium vaccae ATCC 15483T administered to nulliparous C57BL/6N mice prior to mating protects their male offspring against the negative consequences of prenatal stress.

The prenatal period is a vulnerable phase for the offspring's development in utero and ex utero. Accordingly, negative environmental influences such as chronic maternal stress pose a major risk factor for the offspring to develop a variety of mental and physical disorders later in life. Many of these pathologies are accompanied by an over-reactive immune system and chronic low-grade inflammation, and prospective human and mechanistic animal studies strengthen the idea that an exaggerated immune (re)activity plays a causal role in their pathogenesis. In line with the latter, we have shown in mi

A study of mycobacterial species causing lymphadenitis.

This prospective study evaluated the non-tuberculous mycobacteria (NTM) cases of lymphadenitis. A total of 76 isolates of mycobacteria were obtained from 200 lymph node aspirates suspected of tuberculosis, 74 of which were Mycobacterium tuberculosis, one was Mycobacterium fortuitum and one Mycobacterium kansasii. These results highlight the importance of NTM in HIV-negative patients as a case of lymphadenitis, and indicates the re-emergence of NTM as potential lymph node pathogens in this part of the country. Further studies on a larger scale are needed to delineate the association between NTM

Zoonotic diseases of fish and their prevention and control.

Fish and aquatic-derived zoonotic diseases have caused considerable problems in the aquaculture industry and fishery worldwide. In particular, zoonotic diseases can pose widespread threats to humans. With the world's growing population and potential global trade of aquaculture and fish, the risk of environmental contamination and development of fish and aquatic-derived zoonoses in humans are increasing. The important causes of zoonoses include bacteria, parasites, viruses, and fungi. The zoonotic bacterial agents are divided into two main groups: Gram-positive (Mycobacteriaceae, Streptococcace

Atypical Mycobacterial infections in children: the case for early diagnosis.

BACKGROUND & AIMS: Atypical Mycobacteria (ATB) are a miscellaneous collection of Mycobacteriaceae which also includes M. tuberculosis, M. bovis and M. leprae. In the paediatric population, ATB infections present with non-tender unilateral lymphadenopathy in a systemically well child. Initially the disease may be mistaken for a staphylococcal or streptococcal abscess. Inappropriate surgical incision and drainage is often performed and specimens may be sent for routine histopathology and bacteriology analysis only without considering Mycobacterial infection. The simple incision and drainage proc

Differential patterns of antibiotic resistance, virulence, and dissemination risks in floating and sedimented plastispheres.

The plastisphere, a unique ecological niche on plastic surfaces, enriches microbial antibiotic resistance genes (ARGs) and virulence factors (VFs), posing environmental and health risks. Although aquatic sediment is a major sink for plastic contaminants, the resistance, virulence and dissemination potentials of sedimented plastispheres remain poorly characterized compared to floating plastics. Through investigation of metagenomes from two sites in the Pearl River in China, one of the world's plastic pollution hotspots, we report that water plastisphere showed 2.4 and 3.6 times more ARG and VF

Deciphering key traits and dissemination of antibiotic resistance genes and degradation genes in pharmaceutical wastewater receiving environments.

Discharge of pharmaceutical wastewater significantly affects the receiving environments. However, the development of antibiotic resistance and microbial enzymatic degradation in wastewater-receiving soils and rivers remains unclear. This study investigated a sulfonamide-producing factory to explore the distribution of antibiotic resistance genes (ARGs) in the receiving river and soil environments (0-100 cm depth), and the potential hosts of sadABC genes (sulfonamide-degrading genes) as well as their phylogenetic characterization. We identified plentiful ARGs (28 types and 1065 subtypes) and th

Microbial Communities Associated with Methylmercury Degradation in Paddy Soils.

Bioaccumulation of the neurotoxin methylmercury (MeHg) in rice has raised worldwide concerns because of its risks to human health. Certain microorganisms are able to degrade MeHg in pure cultures, but the roles and diversities of the microbial communities in MeHg degradation in rice paddy soils are unknown. Using a series of microcosms, we investigated MeHg degradation in paddy soils from Hunan, Guizhou, and Hubei provinces, representing three major rice production regions in China, and further characterized one of the soils from the Hunan Province for microbial communities associated with MeH

A Case of Pediatric Subcutaneous Abscess Caused by Mycobacterium mageritense Infection.

Soft tissue infections with Mycobacterium mageritense are uncommon. We report the case of a 5-year-old girl who developed a subcutaneous abscess in her right ankle caused by M. mageritense. She had a history of acute encephalopathy and adrenal insufficiency and was hospitalized for acute pancreatitis. During hospitalization, the patient developed fever and tachycardia. Blood culture was positive for gram-positive bacilli. Although initial testing with matrix-assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF MS) reported a different organism, a repeat test identif

Histopathology of laboratory-reared Nothobranchius fishes: Mycobacterial infections versus neoplastic lesions.

Short-lived killifishes of the genus Nothobranchius Peters, 1868 (Cyprinodontiformes) are considered promising model organisms for biomedical research on ageing and tumorigenesis. We conducted histopathological analysis of 411 adult individuals from three Nothobranchius species to study details on spontaneous age-related neoplastic lesions. Light microscopy based on H&E and toluidine blue-stained sections revealed (a) non-proliferative liver changes with pronounced vacuolation of hepatocytes; (b) proliferation of kidney haemopoietic tissue contributing to excretory system damage; (c) prolifera

Effects of Mycobacterium vaccae vaccine in a mouse model of tuberculosis: protective action and differentially expressed genes.

BACKGROUND: Tuberculosis is a leading cause of death worldwide. BCG is an effective vaccine, but not widely used in many parts of the world due to a variety of issues. Mycobacterium vaccae (M. vaccae) is another vaccine used in human subjects to prevent tuberculosis. In the current study, we investigated the potential mechanisms of M. vaccae vaccination by determining differentially expressed genes in mice infected with M. tuberculosis before and after M. vaccae vaccination. METHODS: Three days after exposure to M. tuberculosis H37Rv strain (5 × 105 CFU), adult BALB/c mice randomly received e

Mycobacterium mageritense infection following mastectomy and breast reconstruction requiring triple antibiotic therapy and surgical debridement.

A 40-year-old woman was referred to infectious disease specialists for a Mycobacterium mageritense skin infection following mastectomy and bilateral reconstruction with deep inferior epigastric perforator flap. Her case demonstrates the difficulty in treating non-tuberculosis mycobacterial infections, especially the rarely seen species. She failed to respond to dual antibiotic therapy containing imipenem-cilastin despite reported sensitivity. Additionally, her course was complicated by intolerance to various regimens, including gastrointestinal distress, a drug rash with eosinophilia and syste

Fungal, Mycobacterial, and Nocardia infections and the eye: an update.

Although relatively uncommon, fungi, atypical Mycobacteria, and Nocardia have been isolated from a variety of infections of eye including keratitis, scleritis, canaliculitis, dacryocystitis, endophthalmitis and orbital cellulites. The organisms typically cause a slowly progressive disease. The diseases caused by the organisms can pose both diagnostic and therapeutic challenges. In this manuscript we will describe updates on important aspects of the ocular infections caused by these organisms.

Mycobacterium vaccae NCTC 11659 Attenuates Early Life Adversity-Induced Hippocampal Neuroinflammation in Mice.

INTRODUCTION: Early life adversity (ELA) is a potent risk factor for negative mental health outcomes, in part through its enduring effects on neuroimmune function. The hippocampus is particularly sensitive to stress across the lifespan, and neuroinflammatory signaling has been implicated as a mediator linking a life stress history to vulnerability. Accordingly, we hypothesized that ELA, in particular when followed by chronic psychosocial stress during adulthood (CAS), would enhance expression of inflammatory genes in the hippocampus and that these effects would be ameliorated by intervention w

Predicted functional analysis of rumen microbiota suggested the underlying mechanisms of the postpartum subacute ruminal acidosis in Holstein cows.

BACKGROUND: The relationships between the postpartum subacute ruminal acidosis (SARA) occurrence and predicted bacterial functions during the periparturient period are still not clear in Holstein cows. OBJECTIVES: The present study was performed to investigate the alterations of rumen fermentation, bacterial community structure, and predicted bacterial functional pathways in Holstein cows. METHODS: Holstein cows were divided into the SARA (n = 6) or non-SARA (n = 4) groups, depending on whether they developed SARA during the first 2 weeks after parturition. Reticulo-ruminal pH was measured c
