[Pulmonary infection due to Mycobacterium szulgai, a group 2 nontuberculous mycobacterium].

A 37-year-old male was admitted to our hospital because of abnormal findings seen in a chest radiograph from a physical examination. There was nothing unusual about his past history, and he had been healthy. Chest radiography showed an infiltrative shadow with a cavity in the right upper lobe. A smear test of the sputum was negative but a culture was positive for a mycobacterium, which was identified as M. szulgai. A diagnosis of atypical pulmonary mycobacteriosis caused by M. szulgai. The patient was treated with isoniazid, rifampicin and ethambutol, which gradually improved the infiltration

Phage infection, transfection and transformation of Mycobacterium avium complex and Mycobacterium paratuberculosis.

Mycobacterium avium complex strains and Mycobacterium paratuberculosis are closely related intracellular pathogens affecting humans and animals. M. avium complex infections are a leading cause of morbidity and mortality in AIDS patients, and M. paratuberculosis is the agent of Johne's disease in ruminants. Genetic manipulation of these micro-organisms would facilitate the understanding of their pathogenesis, the construction of attenuated vaccine strains and the development of new drugs and treatment methods. This paper describes the replication of mycobacterial shuttle phasmids and plasmids,

[Nontuberculous Mycobacterium pulmonary infection with pleural effusion caused by Mycobacterium kansasii].

A 28 year-old man was admitted to our hospital because of fever, cough and chest pain. A chest X-ray film taken on admission showed infiltrate in the left upper lung field with ipsilateral pleural effusion. Microscopical examinations of stained specimens of sputa and pleural effusions disclosed no acid-fast bacilli. The level of adenosine deaminase (ADA) in pleural effusion was 46.4 IU/l. A tuberculin skin test was moderately positive. The most probable diagnosis was pulmonary tuberculosis with pleural effusion. Isoniazid (INH) and rifampicin (RFP) were administered on the 5th hospital day and

Mycobacterium haemophilum and mycobacterium xenopi associated infection in a renal transplant patient.

The case is presented of a renal-transplant patient in Europe with a Mycobacterium haemophilum infection in association with M. xenopi infection. Clinical signs suggested the diagnosis of mycobacteriosis, which was confirmed by a skin biopsy. Despite antitubercular treatment which rapidly eliminated M. xenopi, the patient's condition did not improve until M. haemophilum was identified. Minimal inhibitory concentrations of various antimicrobial compounds showed a lack of efficacy of isoniazid, and rifampin had no clinical effect. The patient recovered only after careful surgical drainage of the

β-Aminopropioamidoximes derivatives as potential antitubercular agents against anthropozoonotic infections caused by Mycobacterium tuberculosis and Mycobacterium bovis.

BACKGROUND AND AIM: Tuberculosis (TB) remains a significant global health challenge, with increasing incidences of drug-sensitive (DS) and multidrug-resistant (MDR) TB. In addition, Mycobacterium bovis-induced zoonotic TB (zTB) presents treatment difficulties due to its resistance to pyrazinamide and the prolonged treatment duration required. This study aims to evaluate the antitubercular potential of β-aminopropioamidoxime derivatives against DS and MDR M. tuberculosis and M. bovis strains, and utilizing the SwissADME prognostic tool to predict the drug- and lead-likeness of the described com

Challenges in the Diagnosis and Treatment of Mycobacterium abscessus Infections.

The Mycobacterium abscessus complex (MABC) includes non-tuberculous mycobacteria that are widely distributed and clinically significant. Similar to tuberculosis, MABC can lead to skin and soft tissue infections and pulmonary diseases. These infections frequently occur in outbreaks, particularly among immunocompromised patients or those with preexisting pulmonary conditions. This review examines the recent progress in essential areas that define these infections as a significant challenge in medical practice, specifically the diagnostic modalities, antibiotic treatment options, and resistance o

Nitric Oxide-Releasing Liposomes for Treatment of Mycobacterium abscessus Infections.

Nontuberculosis mycobacteria (NTM) are ubiquitous, opportunistic pathogens that cause severe respiratory infection, primarily in elderly and immunocompromised populations. The second most prevalent NTM pathogen, Mycobacterium abscessus, is considered the most refractory due to its fast growth rate, intracellular survivability, and antibiotic resistance. Treatments are thus sparse and generally ineffective, promoting antibiotic resistance upon chronic use. Nitric oxide (NO) is an endogenously produced free radical that exerts antimicrobial effects against pathogens via several mechanisms of act

Methods and Models for Studying Mycobacterium tuberculosis in Respiratory Infections.

Respiratory infections, including tuberculosis, constitute a major global health challenge. Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), remains one of the leading causes of mortality worldwide. The disease's complexity is attributed to Mtb's capacity to persist in latent states, evade host immune defenses, and develop resistance to antimicrobial treatments, posing significant challenges for diagnosis and therapy. Traditional models, such as animal studies and two-dimensional (2D) in vitro systems, often fail to accurately recapitulate human-specific immune processes, particu

Prosthetic joint infections due to Mycobacterium tuberculosis: A retrospective study.

OBJECTIVE: Tuberculous prosthetic joint infection (PJI) is uncommon and often diagnosed late. The objective here is to describe the management of tuberculous PJI at an osteoarticular infection referral center. METHODS: A single-center retrospective study of patients managed between 1987 and 2016 was performed. RESULTS: We identified 9 patients with a median age of 80 years. The hip was involved in all 9 patients. A known history of tuberculosis was noted in 2 patients and tuberculosis was present at other sites in 4 patients (lung, n = 3; urinary tract and scrotum, n = 1; and spine, n = 1).

Identifying mixed Mycobacterium tuberculosis infections from whole genome sequence data.

BACKGROUND: Mixed, polyclonal Mycobacterium tuberculosis infection occurs in natural populations. Developing an effective method for detecting such cases is important in measuring the success of treatment and reconstruction of transmission between patients. Using whole genome sequence (WGS) data, we assess two methods for detecting mixed infection: (i) a combination of the number of heterozygous sites and the proportion of heterozygous sites to total SNPs, and (ii) Bayesian model-based clustering of allele frequencies from sequencing reads at heterozygous sites. RESULTS: In silico and in vitr

Mycobacterium marinum remains an unrecognized cause of indolent skin infections.

We identified 5 patients who had cutaneous lesions with cultures that yielded Mycobacterium marinum. It was discovered that all 5 patients had a home aquarium, and infection was preceded by trauma to the hand. However, the association between the development of the infection and exposure of the trauma site to the aquarium was not initially established until repeated questioning was performed. Skin biopsies or incision and drainage were performed for all patients, and the diagnosis was established by culture of the specimens. The mean time from initial presentation to diagnosis and initiation o

Source tracking Mycobacterium ulcerans infections in the Ashanti region, Ghana.

Although several studies have associated Mycobacterium ulcerans (MU) infection, Buruli ulcer (BU), with slow moving water bodies, there is still no definite mode of transmission. Ecological and transmission studies suggest Variable Number Tandem Repeat (VNTR) typing as a useful tool to differentiate MU strains from other Mycolactone Producing Mycobacteria (MPM). Deciphering the genetic relatedness of clinical and environmental isolates is seminal to determining reservoirs, vectors and transmission routes. In this study, we attempted to source-track MU infections to specific water bodies by mat

Mycobacterium haemophilum infections--New York City metropolitan area, 1990-1991.

During September 1990-March 1991, clinicians at a New York City (NYC) hospital diagnosed Mycobacterium haemophilum infections in four patients. Two cases occurred in persons with acquired immunodeficiency syndrome (AIDS), and two occurred in persons who had received bone marrow transplants and were on therapy to prevent a graft-versus-host reaction. In April 1991, three additional M. haemophilum infections in persons with AIDS were identified in the NYC area. Because M. haemophilum infection rarely has been recognized, intensive case finding was initiated in NYC as a result of these reports.

Therapy of Mycobacterium marinum infections. Use of tetracyclines vs rifampin.

We describe four patients with Mycobacterium marinum infections who did not respond to two- to six-week courses of therapy with tetracycline, minocycline, and doxycycline. All four patients had prompt responses to therapy with either rifampin alone (two patients) or rifampin in combination with ethambutol. Results of antimicrobial sensitivity tests may be helpful in guiding therapy. Rifampin may be the drug of choice for treatment of these infections.

Intraabdominal Mycobacterium tuberculosis vs Mycobacterium avium-intracellulare infections in patients with AIDS: distinction based on CT findings.

Abdominal CT scans of 71 patients with AIDS who had proved disseminated infection due to Mycobacterium tuberculosis (27 patients) or Mycobacterium avium-intracellulare (44 patients) were reviewed retrospectively to identify radiologic features that can be used to distinguish the two infections. CT findings in patients with disseminated M. tuberculosis included focal lesions in the liver (11%), spleen (30%), kidneys (19%), pancreas (7%), and gastrointestinal tract (15%) and lymph nodes with central or diffuse low attenuation (93%). CT findings in patients with disseminated M. avium-intracellula

Therapy of Mycobacterium abscessus Infections in Solid Organ Transplant Patients.

Mycobacterium abscessus complex (MABC), a rapidly growing Mycobacterium, is one of the most common causes of non-tuberculous mycobacteria (NTM) infections in the United States of America, and it has been associated with a wide spectrum of infections in immunocompetent and immunosuppressed individuals. Eradicating MABC is very challenging, even with prolonged combination therapies. The management of MABC infections in solid organ transplant (SOT) patients is usually complex given their net state of immunosuppression, associated comorbidities, and potential drug-drug interactions, among other th
