This study identifies metabolic and protein phenotypic alterations in gastrocnemius, tibialis anterior and diaphragm muscles of Col6a1(-/-) mice, a model of human collagen VI myopathies. All three muscles of Col6a1(-/-) mice show some common changes in proteins involved in metabolism, resulting in decreased glycolysis and in changes of the TCA cycle fluxes. These changes lead to a different fate of α-ketoglutarate, with production of anabolic substrates in gastrocnemius and tibialis anterior, and with lipotoxicity in diaphragm. The metabolic changes are associated with changes of proteins invo

Tendons are transitional tissues linking muscle to bone, enabling locomotion and fine motor control. The cellular biology across the Achilles tendon unit is poorly understood, yet is critical for interpreting normal function and pathological changes across its microanatomically defined functional zones. We generated a spatially resolved transcriptomic atlas of adult (age 45-76) nontendinopathic human Achilles tendon, sampling the tendon-bone junction (enthesis), midbody, myotendinous junction, and adjoining muscle. Six fibroblast subtypes were identified, with distinct transcriptional profiles

Tendon connects muscle to bone and functions to transmit muscular forces across joints to stabilize or move those joints. Tendons in the foot and ankle are subject to enormous loads and consequently make up a substantial portion of the body's tendon injuries. Understanding the mechanisms of these injuries requires an understanding of the relative rates of muscle, tendon, osteotendinous junction, and myotendinous junction adaptation. This article provides the practitioner with an overview of tendon anatomy, physiology, healing, and repair and correlates didactic and clinical aspects so that pra

Fluoroquinolones are widely utilized antibiotics, but there is an increasing association with musculoskeletal complications, most notably Achilles tendon injury. We present the case of a 53-year-old woman with rheumatoid arthritis and a history of thyroid cancer who developed recurrent right calf pain and myotendinous strains approximately five months after sequential therapy with ciprofloxacin and levofloxacin. These episodes were localized to the gastrocnemius-soleus junction and occurred with minimal activity, without evidence of tendon rupture on MRI. Her condition was managed conservative

Rectus femoris musculotendinous junction injuries occur at the interface between the muscle fibers and the tendon of the rectus femoris muscle. These types of injuries can result from acute trauma or repetitive microtrauma, leading to muscle retraction, fibrosis, muscle fiber degeneration, and chronic inflammation. Management of these injuries typically involves conservative treatment, including rest, ice, compression, and elevation, followed by a structured rehabilitation program. Surgical intervention is required when conservative measures fail and in cases of complete tendon avulsion or chr

Previous studies of acute muscle injury with active stretch used cyclic stretching or stretching the muscle to complete muscle-tendon dissociation. This study tried to determine minimal force required for skeletal muscle injury with one active stretch to establish an injury "threshold." Tibialis anterior and extensor digitorum longus rabbit muscles were actively stretched at 10 cm/sec to 60%, 70%, 80%, or 90% of the force required to passively fail tibialis anterior and extensor digitorum longus muscles of the control (contralateral) limb. Maximal isometric contractile force, tensile propertie

The hamstring muscle complex is made by a group of posterior biarticular thigh muscles, originating at the ischial tuberosity, which extend the hip and flex the knee joint. Proximal hamstring injuries are frequent among athletes, commonly involving their long myotendinous junction during an eccentric contraction. In this pictorial essay, we describe the ultrasound technique to visualize the normal anatomy of the proximal hamstring muscle-tendon-bone complex and present ultrasound findings in patients with traumatic injuries and tendinopathies.

An adult rhabdomyoma was light and electron microscopically studied. The lesional cells presented well-known structural details, such as abundance of mitochondria and glycogen, myofilaments and myofibrils, hypertrophied Z-bands and masses of Z-band material. Triads were randomly scattered in the cytoplasm and also related to the individual sarcomeres. In sarcomeres the triads were regularly placed near to the A-I-junctions. This peculiarity of mammalian skeletal muscle fibers may yield a criterion to distinguish between cardiac and extracardiac rhabdomyomas. Circumscribed surface areas of the

The long head of the biceps femoris (BFlh) is among the most frequently injured components of the hamstring complex, particularly at the myotendinous junctions (MTJs). Despite its clinical relevance, the gross morphology of the BFlh-including its tendon structure and anatomic variability-remains insufficiently characterized. This study aimed to provide a detailed anatomic and morphometric analysis of the BFlh, focusing on the proximal and distal tendons and their integration with surrounding muscle fibers. Thirty-five formalin-fixed male cadavers were dissected via a posterior longitudinal app

OBJECTIVE: Hamstring injuries are common in sports. Although management and outcomes are sport specific, clinical evaluation alone is a poor guide for treatment planning and prognostication. Cross-sectional imaging has added value in these cases.
CONCLUSION: Specifically, the location (tendon attachment, classic or intramuscular myotendinous junction, or extramuscular portion of the tendon), specific muscles involved, and anatomic extent are factors that can influence the immediate treatment, expected convalescent period, and risk of recurrence in these athletes.

Injuries of the gastrocnemius muscle and the common calcaneal tendon in dogs require effective stabilization of the tarsocrural joint to allow proper healing and prevent mechanical overload. This report described a novel minimally invasive technique for temporary internal tarsal stabilization using synthetic tape in two dogs. No comparison with other stabilization techniques was carried out. Both dogs presented with hindlimb lameness and partial plantigrade stance secondary to a subtotal injury of the lateral myotendinous gastrocnemius, and were treated without primary tenorrhaphy. Internal st

Magnetic resonance imaging of skeletal muscles was performed in 11 patients with polymyositis. Two types of muscle lesions were revealed. The first, inflammation, showed increased signal intensity on T2-weighted images and iso-intensity on T1-weighted images. The second, fatty replacement, showed increased signal intensity on both images. The coronal sections could elucidate the extension of the lesion in each affected muscle. Inflammation was relatively diffuse, while homogeneous fatty replacement tended to begin at the lower myotendinous junctions.

A 23-year-old amateur football player presented 9 months after acute onset of severe pain and a lump in the posterior right knee whilst lifting a heavy box. He had been unable to return to playing football or climbing the stairs. Clinically, a Baker's cyst was suspected. MRI scan, the imaging modality of choice, was essentially normal. A subsequent ultrasound (US) scan demonstrated abnormal dynamic bunching of the muscle fibres at the distal semimembranosus myotendinous junction on resisted isometric contraction, most likely due to a previous tear isolated to the distal myotendinous junction.

BACKGROUND: Magnetic resonance imaging (MRI) is the preferred study of choice for pectoralis major ruptures. Because this is a rare injury, no large case series have evaluated the efficacy of MRI for diagnosing and characterizing pectoralis major ruptures. We hypothesized that MRI would be accurate for diagnosis of the location and grade of pectoralis major tears.
METHODS: The study included 36 operative cases of pectoralis major ruptures with detailed descriptions of tear location and grade and satisfactory preoperative MRIs. Two musculoskeletal fellowship-trained radiologists interpreted th

Hydroxyapatite crystal deposition disease and its pathophysiology are poorly understood; however, it is known that calcific tendinitis is the most common condition related to this deposition in the upper limb. Movement of the crystals toward the myotendinous junction is rare; to the best of our knowledge, only 19 cases have been described in the literature, one of which was a case of intramuscular deposition without migration. The purpose of this technical note is to describe the technical details of the arthroscopic treatment of intramuscular calcium hydroxyapatite deposits without intratendi

OBJECTIVE: To describe the MR appearance of a series of throwing-related injuries to the subscapularis muscle-tendon complex among baseball players.
MATERIALS AND METHODS: A retrospective review of MR scans of the shoulder in players from 1 professional baseball organization over the course of 5 years was performed to identify cases with findings suggestive of subscapularis injury. These findings were graded and the medical record was reviewed to assess clinical findings, treatment, and follow-up. Preinjury baseline measurements of arm external rotation at 90° of abduction were compared to me

Mammalian extraocular muscles contain singly innervated twitch muscle fibers (SIF) and multiply innervated nontwitch muscle fibers (MIF). In monkey, MIF motoneurons lie around the periphery of oculomotor nuclei and have premotor inputs different from those of the motoneurons inside the nuclei. The most prominent MIF motoneuron group is the C group, which innervates the medial rectus (MR) and inferior rectus (IR) muscle. To explore the organization of both cell groups within the C group, we performed small injections of choleratoxin subunit B into the myotendinous junction of MR or IR in monkey

CASE: A 36-year-old male competitive powerlifter sustained asynchronous bilateral Achilles tendon ruptures after using 2 types of selective androgen receptor modulators (SARMs). Both tendon ruptures occurred near the myotendinous junction and were treated with open surgical repair and an initial period of immobilization followed by progressive weightbearing and rehabilitation; no postoperative complications were observed.
CONCLUSION: Previous studies have reported that anabolic androgenic steroids have a deleterious impact on tendon structure and function. This case suggests that SARM compoun

Localized differences in tissue degeneration throughout intact and torn rotator cuff tendons have not been well quantified. The objective of this study was to investigate histological differences in localized degeneration in tendons with and without rotator cuff tears isolated to the supraspinatus tendon. Four intact shoulders and four shoulders with rotator cuff tears isolated to the supraspinatus tendon were dissected down to the infraspinatus and supraspinatus tendons. Biopsies were taken throughout the tendon insertion, mid-substance, myotendinous junction, and around the tear if present.
